Adaptation Of The Doubly Labeled Water Method For Subjects Consuming Isotopically Enriched Water, Randall J. Gretebeck, Dale A. Schoeller, Rick A. Socki, Janis Davis-Street, Everett K. Gibson, Leslie O. Schulz, Helen W. Lane Feb 1997

Adaptation Of The Doubly Labeled Water Method For Subjects Consuming Isotopically Enriched Water, Randall J. Gretebeck, Dale A. Schoeller, Rick A. Socki, Janis Davis-Street, Everett K. Gibson, Leslie O. Schulz, Helen W. Lane

College of Nursing Faculty Research and Publications

The use of doubly labeled water (DLW) to measure energy expenditure is subject to error if the background abundance of the oxygen and hydrogen isotope tracers changes during the test period. This study evaluated the accuracy and precision of different methods by which such background isotope changes can be corrected, including a modified method that allows prediction of the baseline that would be achieved if subjects were to consume water from a given source indefinitely. Subjects in this study were eight women (4 test subjects and 4 control subjects) who consumed for 28 days water enriched to resemble drinking water …

Functional Analysis Of Surfactant Protein B (Sp-B) Promoter. Sp1, Sp3, Ttf-1, And Hnf-3Α Transcription Factors Are Necessary For Lung Cell-Specific Activation Of Sp-B Gene Transcription, Ramgopol K. Margana, Vijayakumar Boggaram

School of Medical and Biological Sciences Faculty Publications and Presentations

Surfactant protein B (SP-B) is essential for maintenance of biophysical properties and physiological function of pulmonary surfactant. SP-B mRNA expression is restricted to alveolar type II epithelial cells and bronchiolar epithelial cells (Clara cells) of adult lung. We previously (Margana, R. K., and Boggaram, V. (1996) Am. J. Physiol. 270, L601-L612) found that a minimal promoter region (-236 to +39) of rabbit SP-B gene is sufficient for high level expression of chloramphenicol acetyltransferase reporter gene in NCI-H441 cells, a cell line with characteristics of Clara cells. In the present study we used mutational analysis, electrophoretic mobility shift assays, and DNase …

A Heterodimer Of Thioredoxin And Ib2 Cooperates With Sec18p (Nsf) To Promote Yeast Vacuole Inheritance, Zuoyu Xu, Andreas Mayer, Eric Muller, William Wickner Jan 1997

A Heterodimer Of Thioredoxin And Ib2 Cooperates With Sec18p (Nsf) To Promote Yeast Vacuole Inheritance, Zuoyu Xu, Andreas Mayer, Eric Muller, William Wickner

Dartmouth Scholarship

Early in S phase, the vacuole (lysosome) of Saccharomyces cerevisiae projects a stream of vesicles and membranous tubules into the bud where they fuse and establish the daughter vacuole. This inheritance reaction can be studied in vitro with isolated vacuoles. Rapid and efficient homotypic fusion between saltwashed vacuoles requires the addition of only two purified soluble proteins, Sec18p (NSF) and LMA1, a novel heterodimer with a thioredoxin subunit. We now report the identity of the second subunit of LMA1 as IB2, a previously identified cytosolic inhibitor of vacuolar proteinase B. Both subunits are needed for efficient vacuole inheritance in vivo …

Logger Killed When Struck By Tree, Kentucky Injury Prevention And Research Center

Fatality Case Reports--Tractors/Logging

A 59-year-old male was killed when he was struck by a tree while logging on a mountainside. The victim was working with two other men doing contract work for a local lumber company. All three men were full-time loggers and had many years of experience. On the morning of the incident, the men were working on a steep mountainside. One logger was working at the top of the hill operating a skidder that he owned to pull the cut trees up the hill. At the time of the incident, the victim was working about 30-35 feet from the top of …

The Family Situation Of Street Youth In Latin America: A Cross-National Review , Marcela Raffaelli Jan 1997

The Family Situation Of Street Youth In Latin America: A Cross-National Review , Marcela Raffaelli

Department of Psychology: Faculty Publications

One of the greatest social welfare challenges confronting Latin American nations is the growing number of children and adolescents seeking their survival on city streets. The presence of these apparently abandoned youngsters is not new (see Felsman, 1989; Peralta, 1992), but the number of street youths has been increasing steadily in recent years (Connolly, 1994; Durning, 1992). Although the exact numbers are disputed, most experts agree that millions of children and adolescents work and sometimes live on city streets in the developing countries of Latin America (UNICEF, 1989). In the face of what at times appears to be an insurmountable …

Factor Structure Of The Athletic Identity Measurement Scale With Athletes With Disabilities, Jeffrey J. Martin, Robert C. Eklund, Carol Adams Mushett Jan 1997

Factor Structure Of The Athletic Identity Measurement Scale With Athletes With Disabilities, Jeffrey J. Martin, Robert C. Eklund, Carol Adams Mushett

Kinesiology, Health and Sport Studies

Individuals who perceive themselves as "athletes" are thought to have self-schemas composed of, in part, an athletic identity (Brewer, Van Raalte, & Linder, 1993). The recent development of the Athletic Identity Measurement Scale (AIMS) has allowed sport psychology researchers to assess athletic identity. Research with adolescent athletes with disabilities has suggested that the AIMS is composed of 4 factors (Martin, Mushett, & Eklund, 1994). The purpose of the current study was to further examine the psychometric properties of the AIMS. Seventy-eight international swimmers (34 females, 44 males) with disabilities, ranging in age from 12 to 44 (M = 23.4 …

Diffusion Behavior Of Lipid Vesicles In Entangled Polymer Solutions, X. Cao, R. Bansil, D. Gantz, E.W. Moore, N. Niu, Nezam H. Afdhal

Internal Medicine Publications

ABSTRACT

Dynamic light scattering was used to follow the tracer diffusion of phospholipid/cholesterol vesicles in aqueous polyacrylamide solutions and compared with the diffusive behavior of polystyrene (PS) latex spheres of comparable diameters. Over the range of the matrix concentration examined (Cp = 0.1-10 mg/ml), the diffusivities of the PS spheres and the large multilamellar vesicles exhibited the Stokes-Einstein (SE) relation, while the diffusivity of the unilamellar vesicles did not follow the increase of the solution's viscosity caused by the presence of the matrix molecules. The difference between the diffusion behaviors of unilamellar vesicles and hard PS spheres of similar size …

Relative Wages And The Returns To Education In The Labor Market For Registered Nurses, Edward J. Schumacher Jan 1997

Relative Wages And The Returns To Education In The Labor Market For Registered Nurses, Edward J. Schumacher

Health Care Administration Faculty Research

Over the past two decades there have been substantial changes in the health care sector in general and hospitals in particular. These changes in turn have had an impact on the labor market for nurses. Nursing comprises the third largest occupation among women (behind secretaries and teachers) and is the largest occupation in hospitals, accounting for about a quarter of total hospital employment in 1992 (Wootton & Ross, 1995). It is well documented that there were substantial "shortages" of qualified RNs during the 1980s, reaching a peak in the late 1980s (Aiken & Mullinex, 1987; Buerhaus, 1993; Hassanein, 1991; McKibbon, …

Antagonist Muscle Inhibition During Voluntary And Automatic Movements: Implications For Clinical Intervention, Charles Leonard, Charles Leonard, T. Matsumoto, P. M. Diedrich, J. A. Mcmillan

Physical Therapy and Rehabilitation Science Faculty Publications

Various rehabilitation techniques encourage eliciting automatic reactions in the hope that muscle activations elicited in this way will influence voluntary movement. Unlike voluntary movements that consist of willed initiation and involvement of descending corticofugal and and basal ganglia pathways, automatic postural reactions resulting from external perturbations primarily involve peripheral receptors arising from the vestibular apparatus, muscles, joints, and eyes. We were interested to determine whether voluntary and automatic movements, which are initiated very differently, involved similar temporal activation of muscles and the same pattern of reciprocal inhibition between agonist and antagonist motor neuron pools.

Multiple Ionic Conductances Of The Human Dopamine Transporter: The Actions Of Dopamine And Psychostimulants, Mark S. Sonders, Si-Jia Zhu, Nancy R. Zahniser, Michael Kavanaugh, Susan G. Amara

Biomedical and Pharmaceutical Sciences Faculty Publications

Electrophysiological and pharmacological studies of a cloned human dopamine transporter (hDAT) were undertaken to investigate the mechanisms of transporter function and the actions of drugs at this target. Using two-electrode voltage-clamp techniques with hDAT-expressing Xenopus laevis oocytes, we show that hDAT can be considered electrogenic by two criteria. (1) Uptake of hDAT substrates gives rise to a pharmacologically appropriate “transport-associated” current. (2) The velocity of DA uptake measured in oocytes clamped at various membrane potentials was voltage-dependent, increasing with hyperpolarization. Concurrent measurement of transport-associated current and substrate flux in individual oocytes revealed that charge movement during substrate translocation was greater …

Excitatory Amino Acid Transporter 5, A Retinal Glutamate Transporter Coupled To A Chloride Conductance, Jeffrey L. Arriza, Scott Eliasof, Michael Kavanaugh, Susan G. Amara

Biomedical and Pharmaceutical Sciences Faculty Publications

Although a glutamate-gated chloride conductance with the properties of a sodium-dependent glutamate transporter has been described in vertebrate retinal photoreceptors and bipolar cells, the molecular species underlying this conductance has not yet been identified. We now report the cloning and functional characterization of a human excitatory amino acid transporter, EAAT5, expressed primarily in retina. Although EAAT5 shares the structural homologies of the EAAT gene family, one novel feature of the EAAT5 sequence is a carboxy-terminal motif identified previously in N-methyl-d-aspartate receptors and potassium channels and shown to confer interactions with a family of synaptic proteins that promote ion channel …

Aspartate 19 And Glutamate 121 Are Critical For Transport Function Of The Myo-Inositol/H+ Symporter From Leishmania Donovani, Andreas Seyfang, Michael Kavanaugh, Scott M. Landfear Jan 1997

Aspartate 19 And Glutamate 121 Are Critical For Transport Function Of The Myo-Inositol/H+ Symporter From Leishmania Donovani, Andreas Seyfang, Michael Kavanaugh, Scott M. Landfear

Biomedical and Pharmaceutical Sciences Faculty Publications

The protozoan flagellate Leishmania donovani has an active myo-inositol/proton symporter (MIT), which is driven by a proton gradient across the parasite membrane. We have used site-directed mutagenesis in combination with functional expression of transporter mutants in Xenopusoocytes and overexpression in Leishmania transfectants to investigate the significance of acidic transmembrane residues for proton relay and inositol transport. MIT has only three charged amino acids within predicted transmembrane domains. Two of these residues, Asp19 (TM1) and Glu121 (TM4), appeared to be critical for transport function of MIT, with a reduction of inositol transport to about 2% of wild-type …

Tyrosine 140 Of The Γ-Aminobutyric Acid Transporter Gat-1 Plays A Critical Role In Neurotransmitter Recognition, Yona Bismuth, Michael Kavanaugh, Baruch I. Kanner

Biomedical and Pharmaceutical Sciences Faculty Publications

The γ-aminobutyric acid (GABA) transporter GAT-1 is located in nerve terminals and catalyzes the electrogenic reuptake of the neurotransmitter with two sodium ions and one chloride. We now identify a single tyrosine residue that is critical for GABA recognition and transport. It is completely conserved throughout the superfamily, and even substitution to the other aromatic amino acids, phenylalanine (Y140F) and tryptophan (Y140W), results in completely inactive transporters. Electrophysiological characterization reveals that both mutant transporters exhibit the sodium-dependent transient currents associated with sodium binding as well as the chloride-dependent lithium leak currents characteristic of GAT-1. On the other hand, in both …

Mutation Of An Amino Acid Residue Influencing Potassium Coupling In The Glutamate Transporter Glt-1 Induces Obligate Exchange, Michael Kavanaugh, Annie Bendahan, Noa Zerangue, Yumin Zhang, Baruch I. Kanner Jan 1997

Mutation Of An Amino Acid Residue Influencing Potassium Coupling In The Glutamate Transporter Glt-1 Induces Obligate Exchange, Michael Kavanaugh, Annie Bendahan, Noa Zerangue, Yumin Zhang, Baruch I. Kanner

Biomedical and Pharmaceutical Sciences Faculty Publications

Glutamate transporters maintain low synaptic concentrations of neurotransmitter by coupling uptake to flux of other ions. After cotransport of glutamic acid with Na+, the cycle is completed by countertransport of K+. We have identified an amino acid residue (glutamate 404) influencing ion coupling in a domain of the transporter implicated previously in kainate binding. Mutation of this residue to aspartate (E404D) prevents both forward and reverse transport induced by K+. Sodium-dependent transmitter exchange and a transporter-mediated chloride conductance are unaffected by the mutation, indicating that this residue selectively influences potassium flux coupling. The results …
