Hvj-Liposome-Mediated Gene Transfer To Healing Rat Meniscal Tissue, İlhan Özkan, Norimasa Nakamura, Konsei Shino, Takashi Natsu-Ume, Tetsuya Tomita, Yasufumi Kaneda, Takahiro Ochi
Turkish Journal of Medical Sciences
The success rate of a repaired torn meniscus is not more than 70%. Gene transfer to healing soft tissues appears to be a feasible method for manipulating the healing process. In an attempt to evaluate the feasibility of gene transfer to meniscal tissue we tried to transfer a reporter gene ( Echerichia coli, b-galactosidase gene) by using HVJ-liposome mediated gene transfer. After performing complete radial tear on the medial meniscus of fifty 14-week-old male Wistar rats, HVJ-liposomes with DNA were injected into the femoral artery of 15 rats and into the knee joint of the other 15 rats. HVJ- liposomes …

Contralateral White Noise-Induced Enhancement In The Guinea Pigõs Mlr: A Possible Link To Direc- Tional Hearing, Cüneyt Göksoy, Rezzan Utkuçal
Turkish Journal of Medical Sciences
The evoked potential components in a time window of 10-50 ms following an acoustic stimulus are called middle latency responses (MLRs). It is known that an amplitude enlargement occurs in guinea pig MLRs to monaural clicks when continuous white noise is applied to the other ear. This study was undertaken to see whether this enlargement is due simply to an overall, generalised effect of contralateral white noise (WN), or whether it may have some connection to directional hearing. Recordings were made from chronic guinea pig preparations with epidural electrodes in the temporal regions. Parallel to the results in the literature, …

Turkish Journal of Medical Sciences
The present experimental study was carried out on rats subjected to ischemia and reperfusion. The effects of allopurinol on mucus, an important component of the stomach mucosal barrier, were investigated. Twenty-one Swiss Albino rats, each weighing 200-250 g, were used. On the control group, no treatment was performed, while the sham operation group was subjected to 30 minutes of ischemia and 20 minutes of reperfusion. Then, stomachs were examined for ulcerative lesions, and ulcer scores were recorded. Mucus content was determined by the Corne method. In the sham group, the mucus decreased to a significant extent (p

Investigation Of Various Terminal Carbohydrate Structures In Apolipoproteins Of Human Serum Chylomicrons And Vldl, Tayfun Güldür, Tülay İleri̇ Büyükoğlu, Ramazan Yilmaz
Turkish Journal of Medical Sciences
The roles of terminal carbohydrate moieties of serum glycoproteins in their recognition and uptake by receptors and in their rapid removal from the circulation are well established. However, little is known about the detailed structure of the terminal glycan chains in apolipoproteins and their compatibility with the known ligand specificity of carbohydrate recognition systems. The carbohydrate moieties of apolipoproteins in human serum chylomicrons and VLDL were examined. Chylomicrons and VLDL were isolated from the serum by ultracentrifugation for 1.6x10 ^6 g.min and 5.5x10 ^7 g.min respectively. The top 1.5cm fraction containing chylomicrons or VLDL was recovered. Following delipidization, apolipoproteins were …

Turkish Journal of Medical Sciences
The formation of nitric oxide (NO), a free radical, increases in liver disease, where the L-arginine-nitric oxide (L- arg-NO) pathway is activated by the increased levels of cytokines and endotoxins. In this study, we investigated the relationship between the L-arg-NO pathway and chronic active hepatitis and cirrhosis, which are classified as chronic liver disease. We determined the serum levels of nitrite and nitrate, the end products of the L-arg-NO pathway, in patients with chronic liver disease and compared them with the control group levels. Total nitrite produced by the reduction of nitrate to nitrite in the presence of nitrate reductase …
Effect Of The Combination Of Glibenclamide, An Atp-Dependent Potassium Channel Blocker, And Metoprolol, A Cardioselective B-Adrenoceptor Blocker, During Myocardial Infarction In Conscious Rats, Ömer Bozdoğan, Istvan Lepran, Julğus Gy. Papp
Papp
Turkish Journal of Medical Sciences
We investigated the possible interaction of glibenclamide and metoprolol on the occurrence of life threatening arrhythmias during the acute phase of experimental myocardial infarction. Coronary artery ligation was performed in conscious rats and ECG was recorded for 15 min following ligation. Neither metoprolol (2 mg/kg i.p., 20 min before coronary artery ligation), nor glibenclamide (5 mg/kg i.p., 30 min before ligation) pretreatment increased significantly the survival rate during the acute phase of myocardial infarction (10 % and 22 %, respectively vs. 9 % in controls). Combination of glibenclamide with metoprolol, however, significantly improved the survival rate (62 %, P < 0.05). These results suggest that the combination of a cardioselective b-blocking agent with an ATP- dependent potassium channel inhibitor may result in an enhanced antiarrhythmic effect during the acute phase of myocardial infarction.

Mechanical And Thermal Properties Of Hydroxyapatite-Impregnated Bone Cement, Kemal Şerbetçi̇, Feza Korkusuz, Nesri̇n Hasirci
Turkish Journal of Medical Sciences
Self-curing acrylic cements, consisting mainly of polymethylmethacrylate (PMMA), are widely used in dentistry and orthopedic surgery. One of the major side effects of the standard PMMA application is tissue necrosis at the bone-cement interface due to the rise of temperature during the polymerization reaction. This may also lead to aseptic loosening over time. Therefore, intense research is being carried out in the development of bone cements with new compositions. In this study, the aim was to develop new bone cement compositions that would have low setting temperature and high mechanical strength and be comparable with the commercially available ones. For …

The Effect Of Iron Bound Transferrin Onmegakaryocytopoiesis In Bone Marrow Culture, Selda Deli̇orman, Ruhi̇ Uyar, Yilmaz Altuner
Turkish Journal of Medical Sciences
Megakaryocytopoiesis is a complicated phenomenon which involves the proliferation of progenitor cells, formation of a polyploid nucleus, cytoplasmic changes and formation of thrombocytes. It has been suggested that megakaryocytopoiesis resembles erythropoiesis. It is well known that iron is a required element for the proliferation of erythrocytes, lymphocytes and cancer cells but no consensus has yet been reached about its role in thrombocytopoiesis. Although thrombocytosis is generally found in patients with iron deficiency anemia, thrombocytopenia is also observed. Cell surface receptors for transferrin were initially characterized on cells with high iron requirements including hemoglobin producing cells and placental cells. More recently, …
