The Purification, Properties And Subunit Structure Of Glycerol Dehydrogenase, Michael James Barrett

The inducible, NAD-linked glycerol dehydrogenase (E.C. 1.1.1.6) of a guanine requiring mutant of A. aerogenes has been purified to homogeneity. The molecular weight of the pure enzyme was found to be 3.4 x 10⁵ daltons. The sedimentation coefficient of the enzyme was 10.7 x 10^-13 sec.^-1. The diffusion constant was 3.07 x 10^-7 cm²/sec.

The partial specific volume calculated from the amino acid composition was 0.72 ml/g. The following kinetic parameters were determined; K_m glycerol, 2.4 x 10^-3 m, K_m NAD, 2.8 x 10^-4 M, K_m dihydroxyacetone, 5.1 …

Previous studies in the Lome Linda University laboratory showed that obese subjects on an exercise program lost approximately one pound more per week as compared to a non-exercise group. Energy expenditure calculations of the exercise did not account for the total caloric deficit required to lose the additional pound. The present study was designed to investigate changes in BMR and metabolic rate with time for normal weight and overweight subjects involved in a practical exercise program. The relationship of changes in BMR and metabolic rate in coping with the problems of obesity were also studied.

Induction of hepatic microsomal hydroxylative enzymes is an important aspect of detoxication of fat-soluble toxicants. The magnitude of induction depends on numerous factors, such as the nature and dose of toxicant as well as dietary factors. Research was conducted on (1) endrin tolerance in rats, (2) preliminary comparisons of inductive effects of various organochlorine pesticides in rats to select compounds for further study in guinea pigs, (3) general effects of various dietary factors on induction, and (4) effects of ascorbic acid deficiency on induction of hepatic microsomal hydroxylative enzymes by organochlorine pesticides in guinea pigs. Cercariac of Schistosoma mansoni were studied for glycogen utilization, uptake of glucose from solution, lactic acid production, and respiration.

Using the anthrone method of determination of carbohydrate, the amount of glycogen used by a single cercaria in 4 hours is determined as 43.2 x 10^-4 μg ± 6.6 x 10^-4 μg.

Glucose was not observed to have been used from solution under the conditions of the experiment, even in the presence of trehalose. The determinations were made by the Glucostat method.

The response of the normal human esophagus to an obstructing intraluminal bolus was investigated and compared to the response evoked by transient intraluminal distention. A balloon, immobilized within the esophagus by external attachment to a force transducer, was inflated with from 3 to 25 ml of air for from 3 to 210 sec. Pressure phenomena occurring in the esophagus were simultaneously recorded from the body of the esophagus above and below the balloon.

The purpose of this study was to identify the effects of particular sounds upon certain indices of cardio-pulmonary function and the degree and pattern of changes which occurred in the indices. The specific physiological parameters chosen to be monitored were heart rate, respiratory rate, mean tidal volume, instantaneous minute volume, and oxygen consumption. The hypothesis was that particular sounds will cause changes in certain indices of cardio-pulmonary functions. We carried out clearance studies in nine healthy adults and four patients with hypoparathyroidism before and after inducing stable metabolic acidosis with either NH₄Cl or acetazolamide. Clearances were repeated in seven normal subjects and three of the patients 3 days after stopping these agents.

During acidosis in the normal subjects, serum ultrafilterable calcium concentration rose significantly, but inulin clearance fell to a greater extent, so that the calculated filtered load of calcium fell significantly. Despite this, urinary calcium excretion rose. Urinary calcium excretion remained elevated in the recovery studies when the serum ultrafilterable calcium concentration and filtered load …

Small but statistically significant negative calcium balances were found in each of eight studies in seven patients with chronic azotemic renal disease when stable metabolic acidosis was present. Only small quantities of calcium were excreted in the urine, but fecal calcium excretion equaled or exceeded dietary intake. Complete and continuous correction of acidosis by NaHCO₃ therapy reduced both urinary and fecal calcium excretion and produced a daily calcium balance indistinguishable from zero.

Apparent acid retention was found throughout the studies during acidosis, despite no further reduction of the serum bicarbonate concentration. The negative calcium balances that accompanied acid retention …

Scientific Approach To 4-H Food Projects, Grace Brill

The scientific approach to 4-H food projects resulted from the stress on science from federal extension personnel of the U.S. Department of Agriculture, and from surveys and consultations with representatives of the National Science Foundation. The result has been six bulletins prepared by the extension nutritionists of the University of Minnesota for the approximately 24,000 4-H members, aged 9 to 19, enrolled in 4-H food projects in Minnesota. The scientific approach has, consequently, added the question of "why" to the traditional 4-H approach of "how-to-do-it."

Localization of Mycoplasma pulmonis in the lungs of infected gnotobiotic mice by electron microscopy. J. Bacteriol. 92:1164–1176. 1966.—Lesions in lungs of gnotobiotic mice inoculated intranasally with Mycoplasma pulmonis were examined by electron microscopy after osmic acid fixation. At 1 week after infection, mycoplasma cells were found in large numbers in the bronchi at the surface of bronchial epithelial cells and, in smaller numbers, in the alveoli where active phagocytosis by polymorphonuclear leukocytes (PMN) occurred. …

The study of plasma electrolytes notably, the potassium and sodium ions during hemorrhagic stress and other forms of shock, were once the subject of intense investigation (42). However, the advent of new investigative techniques enabled more complex, experimental designs and methods to be used in which it was possible to study the many other variables which exist as a result of shock regardless of cause. The proponents of the probable electrolyte mechanism in shock subsequently diminished in numbers until recent times.

The purpose of this research was to find out what effects the TURP has on ejaculatory ability, frequency of coitus and the emission of seminal fluid. The sample consisted of 50 men, age 46-65, who were married, and had no cancer or other possible physical causes for impotence. Each man was personally interviewed.

Forty-six patients (92 percent) were able to ejaculate. A decrease in the frequency of coitus was reported by 40 per cent of the men. A normal ejaculation of seminal fluid was experienced by thirteen (26 percent) of the men. Considering all the changes in the sexual function …

Three hundred Swiss Webster albino mice three weeks of age were placed on low (8%), medium (24%), high (48%) protein levels and a commercial laboratory animal feed, Purina Chow for various lengths of time: (1) simultaneously with infection, and four weeks thereafter, (2) three weeks before infection and throughout the four-week infection period, and (3) six weeks prior to infection and during the infection period. The protein source was casein. This study was conducted to discover the effects of (1) an interpersonal relationship and (2) an interpersonal relationship and music therapy on preoperative patients in a selected hospital.

The descriptive survey was chosen as the method of study. A checklist and a tape recording of selected music were used as a tool of research. Two groups of preoperative patients were given nursing care by the researcher. Patients in Group One received care with an aim toward developing an interpersonal relationship, while patients in Group Two received the same as Group One with the addition of music therapy.

Theses

The discovery of gamma-aminobutyric acid (GABA) in mammalian brain and its rigorous identification was reported in 1950 (1,2,3). Enzymatic experiments with brain acetone powder revealed that GABA was formed by the decarboxylation of glutamic acid and that the enzyme, glutamic acid decarboxylase, required pyridoxal phosphate as the co-factor (4,5). Radioactive tracer studies in vivo and in vitro with glutamic acid and glucose verfied the fact that the former was the precursor of GABA (6,7).
