As a graduate student of social work, I have become involved in the delivery of medical care and am interested in the dynamics of psycho-social variables as they affect the structure, delivery, and outcome of medical services.

Three Linked Enzyme Loci In Fishes: Implications In The Evolution Of Vertebrate Chromosomes, D C Morizot, D A Wright, M J Siciliano

A three-point linkage group comprised of loci coding for adenosine deaminase (ADA), glucose-6-phosphate dehydrogenase (G6PDH), and 6-phospho-gluconate dehydrogenase (6PGD) is described in fish of the genus Xiphophorus (Poeciliidae). The alleles at loci in this group were shown to assort independently from the alleles at three other loci--isocitrate dehydrogenase 1 and 2, and glyceraldehyde-3-phosphate dehydrogenase 1. Alleles at the latter three loci also assort independently from each other. Data were obtained by observing the segregation of electrophoretically variant alleles in reciprocal backcross hybrids derived from crosses between either X. helleri guentheri or X. h. strigatus and X. maculatus. Role Of Cytochrome P-450 In Methemoglobin Formation By Primaquine With Hamster Liver Microsomes, Tommy L. Hicks

The role of cytochrome P-450 in in vitro methemoglobin formation by primaquine was investigated through a series of experiments with golden-Syrian hamsters, treated before sacrificing with phenobarbital or 20-methylcholanthrene to increase the cytochrome P-450 content or with cobaltous chloride to cause its decrease. To determine the extent of induction in the liver microsomal cytochrome P-450-dependent mixed-function oxidases, cytochrome P-450 content and formaldehyde from the 0-demethylation of p-nitroanisole were measured. A 90% increase occurred in cytochrome P-450 content with phenobarbital pretreatment and an increase of 102% was observed in p-nitroanisole 0-demethylation. Formaldehyde from primaquine increased 64%. The results of our personal studies of alpha-l-protease inhibitor (APi, formerly termed alpha-l-antitrypsin), the major inhibitor of protease in extracellular fluids, are presented in this dissertation. In the first chapter, we describe techniques used for APi quantitation and determination of Pi phenotypes. After a brief review of the available techniques, we report several improvements on our original technique of Pi phenotyping using thin-layer isoelectric focusing. Particular attention is given to biochemical and immunological methods for verification of the results obtained by isoelectric focusing. Three complementary techniques are reported: protease probe, crossed immunoelectrophoresis, and print immunofixation. Analysis of the results from …

An analog of ATP, 6-mercapto-9+nribofuranosylpurine 5’-triphosphate (SH-TP), functions as a phosphoryl donor for the reaction catalyzed by hexokinase. Incubation of the enzyme with millimolar concentrations of this reagent led to its rapid inactivation. However, the corresponding monophosphate derivative of this reagent was much less effective. A plot of the initial rate of inactivation versus the concentration of SH-TP exhibited saturation, suggesting the formation of a reversible complex between the enzyme and SH-TP prior to the inactivation reaction. The dissociation constant of this enzyme-inhibitor complex was the same as the K, of this reagent with respect to hexokinase in the phosphoryl …

Reaction Of Brain Hexokinase With Tetranitromethane: Oxidation Of Essential Thiol Groups, Subbarao Bondada, Umakant W. Kenkare

Microbiology, Immunology, and Molecular Genetics Faculty Publications

Treatment of bovine brain mitochondrial hexokinase with a fivefold molar excess of tetranitromethane (TNM) at pH 8.0 results in a complete loss of activity of the enzyme. Spectral measurements and amino acid analysis showed that only cysteinyl residues were modified by TNM under these conditions. The products of the reaction of TNM with the enzyme were cysteic acid and disulfides as found by amino acid analysis of the carboxymethylated hexokinase. Failure of 2-mercaptoethanol to reverse the effect of TNM suggested that the thiol group that is oxidized to cysteic acid is the one essential for enzyme activity. Substrates glucose and …

Bacillus subtilis has a constitutive system for the uptake of alpha-keto-beta-methylvalerate, alpha-ketoisovalerate, and (probably) alpha-ketoisocaproate. A mutation, kauA1, which blocks the uptake of alpha-keto-beta-methylvalerate and alpha-ketoisovalerate, is located between metB and citK on the B. subtilis chromosome.

A set of general purpose FORTRAN IV programs have been written during this study to assist in the analysis of complex spectra. An interactive approach is adopted, utilizing the computer and graphics terminals of the Scientific Computation Facility at Loma Linda University. Some procedures carried out are inversion, coordinates scaling, smoothing, differentiation and integration, correction for slanting baseline, addition and subtraction, and spectral simulation.

As an application, the programs are used to assist in the study of free radicals in gamma-irradiated single crystals of 1,2,3,- benzene tricarboxylic acid dihydrate by electron spin resonance spectroscopy. Is is found that the predominant …

A Batch Method For The Isolation Of Immunogenically Pure Igg, Iga, And Igm From Human Sera, Samuel Christopher Powell

A method is described for preparing an enriched fraction of the three major antibodies, immunoglobulins IgG, IgA and IgM, from normal human serum by a batch method. Selective precipitation of these proteins was carried out in successive steps using polyethylene glycol (PEG) of average molecular weight 6,000 at concentrations of 7%, 12% and 14% (16) . Zinc sulfate (10 mM) was used to remove highly immunogenic alpha₂ macroglobulin from the IgM fraction. IgA, IgM and IgG were further purified by adsorption methods using DEAE-cellulose. Quantitation and qualitation of the enriched fractions showed nearly complete separation of each of the …

Food additives are listed by functional category and discussed in terms of the scale of their use and relative risks in terms of long-term human toxicity. The greatest risks appear to lie with the antimicrobial preservative agents, and food colors (which serve only a cosmetic purpose); these and the synthetic sweeteners are discussed in relatively greater detail.

Research results from studies of the physiological effects of aerosal propellants on animals and people are summarized. The papers selected for this summary were published between 1968 and 1975. Effects of the 15 chlorofluorocarbons used as propellants have been recorded on the cardiac, pulmonary and respiratory functions of a number of animal species as well as man. The fifteen propellants have been classified into four groups on the basis of their degree of toxicity. Fluorocarbon 11, the most frequently used propellant, is in group 1, the most toxic. The water permeability was measured for blood cells from normal subjects and for blood cells from patients with leukemia. The method of measurement of the kinetics of water influx employed a stopped flow apparatus. The method of calculation of results was based on irreversible thermodynamics. The measurements and calculations were both performed with the assistance of a computer. The water permeability of the leukemic cells was decreased from normal, and the values were more variable than normal. Leukemia changes the structure of the cell membrane.

Stereospecific Opiate Binding In Human Erythrocyte Membranes And Changes In Heroin Addicts, Leo G. Abood, Holly G. Atkinson, Mary Macneil

Publications and Research

Stereospecific opiate binding has been demonstrated in human erythrocyte membranes, having a Kd of 9-10(-9) M. In most respects the binding characteristics resemble those of synaptic membranes. These included the correlation of binding affinity and pharmacological potency of opiates; competition by naloxone; inhibition by Ca2+ and Na+; and sensitivity to phospholipases and trypsin. A comparison of stereospecific opiate binding in control human subjects and heroin addicts revealed a 43% increase in the addict group.
