The Association Of Semen Factors With The Recovery Of Ureaplasma Urealyticum, Rebecca D. Cintron

Fifty-six semen samples from males under examination for suspected subfertility were examined to determine if alterations in specific semen parameters could be associated with the presence or absence or Ureaplasma urealvticum. Twenty-seven (48.2%) of the specimens cultured positive for U· urealyticum in U9B urease broth and A-7 differential agar. Both positive and negative specimens have a normal average motility rating. Overall sperm counts (millions per ml) were slightly lower and ejaculate volumes higher in U. urealvticum positive samples. Morphological abnormalicies were not found more frequently in U. urealyticum positive samples. No statistically significant difference between seminal parameters of U. …

Insulin Degradation By Adipose Tissue. Studies At Several Levels Of Cellular Organization, Barry J. Goldstein, James N. Livingston

A systematic study of the degradation of physiological concentrations of ¹²⁵I-labelled insulin was performed in intact fat-pads, isolated adipocytes and subcellular fractions of isolated adipocytes. The findings indicate that insulin is rapidly degraded to low-molecular-weight peptides and/or amino acids by the intact tissue and isolated cells. Of the total insulin-degradation products present after incubation with an intact fat-pad, 94% is recovered in the medium, indicating that these products are not retained by the cells or tissue. The plasma membranes do not degrade insulin significantly in the absence of reduced glutathione, and over 99% of the cellular degradative capacity is …

The erythropoietin (Ep) dose response of erythroid colony-forming units (CFU-e) and erythroid burst-forming units (BFU-e) from rabbit bone were similar to murine erythroid precursor Ep requirements. Bone marrow and peripheral blood contained a continuum of erythroid precursors at differing stages of maturation. The earliest BFU-e were assayed on day 10. CFU-e were observed in bone marrow, but not in peripheral blood. The most mature erythroid precursors observed in peripheral blood were intermediate BFU-e, assayed on day 5. Isolated mammalian spinal cord has been shown capable of generating locomotor activity. Propriospinal systems assumed to coordinate fore- and hindlimb activity are poorly understood. This study characterizes the long descending propriospinal (LDP) neurons in terms of the location of the somas and their peripheral inputs by direct neuronal recording. Anatomical studies using axonal retrograde transport of horseradish peroxidase from the lumbar to the cervical spinal cord as a tracer first described these neurons. Two hundred and thirty-one LDP neurons were identified in electrophysiological experiments. Of these, 123 responded to natural stimulation, and about 50% of the others were activated only …

The cat entopeduncular nucleus (EN), which is the main output of the basal ganglia, is known to project to the mesencephalic tegmentum. We have been able to elicit antidromic responses in single EN neurons from the region of the mesencephalic locomotor region (MLR), then transect (precollicular-postmamillary) the brainstem and elicit rhythmic movements of the limbs by stimulation of the same site in the same animal. Injections of the fluorescent dye 2,4 diamidino phenylindole 2 HCL (DAPI) into this area induces retrograde labeling of cell bodies in EN and motor cortex. Speculation: In the fetal lamb during prolonged intrauterine hypoxia, total and regional cerebral blood flows increase to the same extent without evidence of preferential shunting to critical brainstem or subcortical areas. Neuropathologic studies have indicated relative sparing of these areas during similar animal experimental or human neonatal conditions. This suggests that the pattern of hypoxic ischemic insult to the neonatal central nervous system associated with asphyxia may differ from that produced by hypoxia alone. In addition, during asphyxia these pathologic changes may result primarily from hypotension and decreased regional cerebral blood flow, or from regional metabolic derangements …

A multienzyme complex contained in Bakers' yeast (Saccharomyces cerevisiae) which synthesizes CoA has been named the coenzyme A-synthesizing protein complex (CoA-SPC). The CoA-SPC has been shown to be insoluble in the crude Bakers' yeast cell lysate formed by exposing the yeast cell to ether and dry ice. Only after solubilization has this multienzyme complex been shown to catalyze the formation of bound dephospho-CoA utilizing the substrates adenosine triphosphate, D-pantothenic acid and L-cysteine. A low molecular weight component or components of the soluble fraction of the yeast cell and chloride ion appears to be responsible for the solubilization of CoA-SPC. This …

Polymorphisms, Linkage And Mapping Of Four Enzyme Loci In The Fish Genus Xiphophorus (Poeciliidae), D C Morizot, M J Siciliano

Electrophoretic variants at four additional enzyme loci--two esterases (Est-2, Est-3), retinal lactate dehydrogenase (LDH-1) and mannose phosphate isomerase (MPI)--among three species and four subspecies of fish of the genus Xiphophorus were observed. Electrophoretic patterns in F1 hybrid heterozygotes confirmed the monomeric structures of MPI and the esterase and the tetrametric structure of LDH in these fishes. Variant alleles of all four loci displayed normal Mendelian segregation in backcross and F2 hybrids. Recombination data from backcross hybrids mapped with Haldane's mapping function indicate the four loci to be linked as Est-2--0.43--Est3--0.26--LDH-1--0.19--MPI. Cell mediated immune (CMI) cytotoxic reactivity of Balb/c mice against H238 cells, a Herpes simplex virus (HSV) Type 2 - induced sarcoma, was measured by the ¹²⁵IUdR release assay. The Balb/c mouse response to the growing HSV-induced tumor, treated with radiation and , was measured by survival rate, tumor growth and immune cytotoxicity of spleen and peritoneal exudate cells (PEC) as determined by ¹²⁵IUdR release assay. Subcutaneous (s.c.) inoculation of 1 x 10⁶ H238 cells (high dose) produced progressive tumor growth while s.c. inoculation of 1 x 10⁴ H238 cells (low dose) produced no tumors. A …

Physicians were interviewed to assess their willingness to risk adverse drug reactions among patients. These untoward reactions were ranked according to severity and weighted against the primary illness being treated. A specially designed questionnaire in the form of a matrix was used. Severity was divided into seven classes denoted by progressively increasing numerical scores, W₁ to W₇, whose values could be calculated from analysis of the completed questionnaires. Interplanetary Magnetic Field Sector Polarity And Neuropsychiatric Adaption: A Reanalysis, Hans W. Wendt

Journal of the Minnesota Academy of Science

Recent Russian studies reported associations between adaptation syndromes and (solar activity-related) interplanetary magnetic field (IMF) sector polarity. Certain ambiguities in the original work suggested some reanalyses, based on the graphs and data made available. Syndrome status exhibited a biphasic (semi annual) trend. It also correlated with specific geomagnetic changes during magnetic storms and possibly after sudden impulses. Moreover, lag effects and higher order interactions were noted among all parameters investigated. The restudy tends to support some of the original findings but also suggests new conclusions regarding potential relationships between geophysical/solar and psychophysiological processes.

Lyb 5 is a B-cell alloantigen which is expressed on 50-60% of B cells. It was defined originally on the basis of cytotoxicity. We have described a new reactivity within the anti-Lyb 5 serum on the basis of selective inhibition of antibody responses in vitro by this antiserum in the absence of complement. This inhibitory activity of anti-Lyb 5.1 serum appears to be due to recognition of antigenic determinants different from the prototype antigens detected in the cytotoxicity assay. Anti-Lyb 5 serum incorporated into spleen cell cultures selectively inhibits antibody responses to a class of thymus-independent antigens (TI-2) previously characterized …

Staphylococcal protein A and several different immunoglobulins have been radio-labeled to high specific activities (> 106 cpm/pg) by reductive methylation with tritiated (3H) sodium borohydride. The proteins retain excellent functional and antigenic properties. The utility of these reagents in a variety of assays for cell surface antigens is illustrated. The results indicate that this radiolabeling procedure may become the method of choice for many cell surface and solution immunoassays.

This study was undertaken to investigate the contribution of carbon dioxide to respiratory control made by the carotid bodies and vagus nerves. Small eupneically breathing dogs, anesthetized with IV ketamine HCL were subjected to inspired CO2 concentrations of 3% while recording simultaneously tidal volume, inspiratory and expiratory airflow rates, and single neuron activity in or near the ventral respiratory group. The burst activity of the ventral respiratory group neuron was considered to be representative of the final integrated motor output of the brain stem respiratory complex. The objective of this thesis is to provide for the cardiologist polarcardiograms and additional diagnostic criteria which will enable him to diagnose heart disease more easily. Other contributions are those of: (a) providing a system of data acquisition and (b) analyzing Dr. Gordon Dower’s criteria for myocardial infarction using the programs provided in this thesis.

The problem inherent in the reading of a polarcardiogram is the identification of the "loop" patterns with the associated heart disease. This can be accomplished by reading and interpreting both vectorcardiograms and electrocardiograms as well as angiograms, then relating the interpretation to the polarcardiogram patterns. …

Abstract

We studied lymphocyte capping in 61 patients with Duchenne, Becker, limb-girdle, facioscapulohumeral and congenital muscular dystrophies. All showed a markedly diminished percentage of capped cells when compared with 86 normal controls, providing support for previous evidence that an alteration in membrane fluidity may be a common pathogenic feature in several genetically distinct forms of proximal muscular dystrophy.

Heterozygous carriers of Duchenne muscular dystrophy showed diminished capping that was indistinguishable from that of afflicted males and was often present even when serum enzyme levels were normal. Studies in 25 families with 16 suspected sporadic cases indicated that no more than …

Taste-Masking: Function Of Exaggerated Prandial Drinking In Desalivate Mice, Richard C. Lewis

Taste thresholds for the bitter compound sucrose octaacetate (SOA) were elevated by desalivation in mice. Thresholds were determined for control and experimental animals both before and after ligation of all salivary ducts. There was a significant increase in SOA thresholds in the desalivate mice, and the pre- to post-operative differences in threshold between the control and experimental groups were significant. The altered response to SOA by desalivate mice is shown to be due to the assumption of a prandial pattern of drinking as a result of desalivation.
