Studies Of The Depolarization-Induced Release Of Beta Adrenergic Receptor Blocking Drugs From In Vitro Neural Preparations, Patricia Sherwood Bright

The objective of this study was to evaluate the hypothesis that nerve terminals may serve as a site for storage and release of beta adrenergic receptor blocking drugs. Experiments were conducted with two different in vitro neural preparations, cultured superior cervical ganglia from adult rats and synaptosomes prepared from rat cerebral cortex. The release of propranolol by depolarizing stimuli was examined and compared with the release of the more hydrophilic, cardioselective beta receptor antagonist atenolol. In both neuronal models, norepinephrine release was monitored as an index of exocytosis. Cultured superior cervical ganglia of the rat represent an in vitro model …

The Effects Of Triethyltin Bromide On Red Cell And Brain Cyclic Amp-Dependent Protein Kinases, Kevin R. Siebenlist, Fumito Taketa

Triethyltin bromide activates the cyclic AMP-dependent protein kinases of human red cell membranes and of bovine brain. Additions of 25-500 microM triethyltin to red cell ghosts resulted in enhanced phosphorylation of ghost proteins. When added to partially purified cyclic AMP-dependent protein kinases from red cell ghosts or bovine brain, stimulation of the phosphorylation of calf thymus histone was observed. The enhancement of kinase activity was due to release of catalytic subunits from the intact protein kinase. Brief exposure of the partially purified enzymes to triethyltin, followed by DE52 chromatography, resulted in elution profiles for regulatory and catalytic subunits that were …

Identification Of Four Complementary Rna Species In Akabane Virus-Infected Cells, Asit K. Pattnaik, G. Abraham

The analysis of RNA extracted from purified Akabane virus demonstrated the presence of three size classes of single-stranded RNAs with sedimentation coefficients of 31S (large, L), 26S (medium, M), and 13S (small, S). Molecular weights of these RNA species were estimated to be 2.15 x 10⁶,1.5 X 10⁶, and 0.48 x 10⁶ for the L, M, and S RNAs, respectively. Hybridization analysis involving viral genomic RNA and RNA from virus-infected cells resulted in the identification of four virus-specific cRNA species in infected cells. The zinc status of twelve lacto-ovo-vegetariah pregnant women and seventeen non-vegetarian pregnant women was evaluated. Zinc intakes were calculated from three-day dietary records and twenty-four hour dietary histories. Plasma zinc levels were drawn during the last trimester of pregnancy and approximately ten weeks postpartum. Gestational age at birth, birthweight, and APGAR scores of the infants born were obtained from the medical charts. Dietary zinc intakes for the vegetarians (10.5 ± 4.0 mg) did not differ significantly from the intakes of the non-vegetarians (10.9 ± 3.0 mg). The vegetarian last trimester plasma zinc levels (47.9 ± 12.4 mcg/dl) and postpartum plasma …

Development Of A Mathematical Model Of Renal Function For Clinical Application, Barbara A. Holshouser

This model describes the orthoiodohippurate (OIH) distribution and clearance in the renovascular system. The model is described by five compartments: the blood, left and right kidneys, bladder and the red blood cell compartment. Data for these compartments, except the RBC compartment, are collected with scintillation detectors monitoring OIH tagged with 1-131 as the radiopharmaceutical passes through each compartment. Time/ activity curves are plotted for the data and used as inputs to the model parameter estimation routine. Food habits of larvae of the northern hog sucker, common shiner, rosyface shiner, and Micropterus sp. from the Middle Fork of Drake's Creek, Kentucky were identified from 18 March to 12 August 1982. Eighteen taxa of animals were observed in the stream drift and a total of seventeen taxa, including larval fish, were identified in the stomach analysis. Eggs, rotifers, hydracarina, and diptera represented the greatest component of the drift comprising 34%, 32%, 8%, and 6%, respectively. The major organisms observed in the gut analysis included eggs (fish, rotifer, and copepod species), rotifers (Euchlanis sp.), diptera (Chironomidae), annelida (Naididae), copepoda …

The sedative hypnotics may exert their effects through a number of different mechanisms. Diazepam interacts with a specific receptor linked to a GABA receptor and a CL ionophore (Skolnick and Paul, 1981) and enhances the binding affinity of the GABA receptor for its ligand. Barbiturates may act at an additional receptor linked to this complex (Olsen, 1981). The sites of action of methaqualone have yet to be defined. Recently Hunkeler et al. (1981) synthesized a new class of compounds, the imidazodiazepines, the prototype being Ro 15-1788. They showed that Ro 15-1788 inhibits 3H-Diazepam binding to brain synaptosomes, reverses diazepam-induced protection …

The present studies were undertaken in order to a) verify the presence of a human Thy 1 antigen, b) demonstrate its similarity to murine Thy 1, and c) biochemically characterize the molecule. Previously, a 25,000 molecular weight molecule had been isolated from a human T lymphoblastoid cell line, Molt 3, using isolation procedures most frequently used for the isolation of rodent Thy 1. The human Thy 1 (p25) was demonstrated to be biochemically similar to murine Thy 1.2 by molecular weight and ability to bind Lens Culinaris lentil lectin. The Utilization Of Response Surface Techniques In Survival Experiments, Leonard Edward Buchner

Linear regression procedures are described which give an unbiased estimate of the percent functional viability of rat polymorphonuclear leukocytes (PMN's). These statistical techniques allow discrimination between variables which result in increased PMN survival after freezing. A review of current response surface techniques was made as they relate to optimizing the functional viability of PMN's after freezing. Experimental designs based on the steepest ascent and simplex methodologies were considered. Since the hill climbing efficiency of the steepest ascent and simplex methods are comparable, the steepest ascent procedure was selected as it is more readily adaptable to a batch mode analysis.

Antibiotic Susceptibility Of Black-Pigmented Bacteroides Isolates From The Human Oral Cavity, Linda Jean Laatsch, Paul R. Hohenfeldt, William L. Kos

The minimal inhibitory concentrations of penicillin and six other antibiotics were determined for 66 oral black-pigmented Bacteroides isolates by using the National Committee for Clinical Laboratory Standards proposed standard agar dilution technique. These results plus iodometric determination of β-lactamase activity showed that oral isolates of black-pigmented Bacteroides are remaining relatively susceptible to commonly used antibiotics.

Some aspects of anuran metabolism are examined, with special emphasis on possible limitations to aerobic metabolism and the effect of chronic hypoxia acclimation on maximal rates of aerobic metabolism and the metabolic fate of lactic acid accumulated after anaerobic metabolism.
