Turkish Journal of Medical Sciences

Background/aim: Methotrexate (MTX) is a folic acid antagonist that is widely used to treat osteosarcoma, leukemia, breast cancer, and autoimmune and inflammatory diseases. The most important concerns with MTX are its poor solubility and high toxicity, particularly in liver cells. To enhance its solubility and to minimize its toxicity, we encapsulated MTX in niosomes and investigated its hepatotoxicity mechanisms using genetic biomarkers. Materials and methods: Niosomes were successfully prepared using a modified thin film method, and the prepared monodisperse smallsized formulation was subsequently characterized. In vitro cytotoxicity studies were performed both in hepatocarcinoma (HEP3G) and healthy liver (AML12) cell lines. …

Can Amniotic Fluid Protect Developing Fetal Lungs Against The Harmful Effects Of Oxidative Stress?, Levent Korkmaz, Cumali̇ Alan, İsmai̇l Topal, Mahi̇r Tayfur, Ali̇seydi̇ Bozkurt, Cebrai̇l Gürsul, Osman Baştuğ

Turkish Journal of Medical Sciences

Background/aim: Preterm births cause fetuses to be born without completing the development of their organs. Due to this undesirable situation, it is the pulmonary tissue which has to be most exposed to harmful effects of extrauterine environment. Early disappearance of the prophylactic and constructive effects of amniotic fluid (AF) on developing tissues, such as pulmonary tissue, facilitates the formation of pulmonary morbidities resulting from oxygen. Setting out from this knowledge, we wanted, in addition to assessing the beneficent effects of AF on pulmonary tissue, to study the importance of AF in morbidities of this tissue thought to originate from oxygen. …

Hidden Threat In Familial Mediterranean Fever: Subclinical Inflammation, Oxidative Stress And Their Relationship With Vitamin D Status, Mhd Boshr Alesh, Di̇dem Barlak Keti̇, Ayşenur Paç Kisaarslan, Sabahatti̇n Muhtaroğlu, Sema Nur Taşkin

Turkish Journal of Medical Sciences

Background/aim: Vitamin D levels have been investigated in children with familial Mediterranean fever (FMF), but the relationship between vitamin D status and inflammation/oxidative stress indicators could not be clearly demonstrated. This study aimed to investi- gate the relationship between subclinical inflammation/oxidative stress and vitamin D status in children with FMF during an attack-free period. Materials and methods: In the cross-sectional study, ninety children with FMF in the attack-free period and 30 healthy children were included. Patients were grouped according to their vitamin D status (< 20, 20-29, and 30-100 ng/mL). The groups were compared in terms of pentraxin 3 (PTX-3), total oxidant status (TOS), and total antioxidant status (TAS). Multivariable linear regression analysis was performed to identify factors associated with vitamin D status. Results: PTX-3 levels were significantly higher in patients with vitamin D insufficiency (20-29 ng/mL) than in the group with vitamin D sufficient (30-100 ng/mL). Patients with vitamin D deficiency (< 20 ng/mL) had higher TOS. A strong negative correlation was ob- served between vitamin D levels and TOS (p = 0.003). Subclinical inflammation (PTX-3 ≥ 0.640) and high TOS levels were negatively associated with vitamin D levels. Conclusion: Subclinical inflammation and oxidative stress were negatively associated with vitamin D levels in patients with FMF dur- ing an attack-free period. Sufficient vitamin D levels are important in fighting subclinical inflammation and oxidative stress in children with FMF.

Chemical, oxidative, and emotional stressors are important in terms of the clinical course of tinnitus. Apelin is an endogenous peptide which is an oxidative stress mediator. It has been shown that the apelin/APJ (apelin junction receptor) system plays various roles in the physiology and pathophysiology of many organs. However, the role of the apelin/APJ system as an oxidative stress mediator in tinnitus is unknown. We investigated the level of apelin in patients with normal …

Turkish Journal of Medical Sciences

8-Hydroxy-2'-deoxyguanosine (8-OHdG) is a biomarker of oxidative stress and has been implicated in many diseases. The aim of this study was to investigate the clinical value of plasma 8-OHdG level in patients with acute exacerbation of chronic obstructive pulmonary disease (AECOPD).Materials and methods: A total of 154 subjects were enrolled in this study, including 20 healthy volunteers, 24 COPD patients in the stable phase, and 110 AECOPD patients. Peripheral blood samples, demographic information, and clinical characteristics were collected from all subjects at the time of being recruited into the study. Plasma 8-OHdG level was detected by enzyme-linked immunosorbent assay. Results: 8-OHdG …

Ischemic Modified Albumin As A New Biomarker In Predicting Oxidative Stress In Alopecia Areata, Hati̇ce Ataş, Müzeyyen Gönül, Yasi̇n Öztürk, Mustafa Kavutçu

Results show that oxidative stress is a pathophysiologic factor for alopecia areata (AA); however, the markers used can be confounding. Thus, we aimed to investigate the role of oxidative stress in the pathogenesis of AA through an evaluation of ischemia-modified albumin (IMA); other markers of the oxidant/antioxidant system, such as SOD, CAT, GSH-ST, and MDA; and contributing clinical risk factors. Materials and methods: The usefulness of IMA as a new marker for oxidative stress was compared with that of other markers and evaluated in patients with AA.Results: The mean serum level of IMA was of higher statistical significance in AA …

Background/aim: Since the nature of ischemia/reperfusion (IR)-induced tissue damage is multifactorial and complex, in the current study, the effects of multiple treatment strategies via concomitant administration of erythropoietin (EPO) and N-acetylcysteine (NAC) with an ischemic preconditioning (IPC) regimen on renal IR injury were examined.Materials and methods: Thirty male Wistar rats were subjected to bilateral occlusion of the renal pedicles for 50 min followed by reperfusion. EPO (1000 IU/kg) was administered for 3 days, as well as IPC before the IR and NAC (150 mg/kg) administration for 4 days after IR. The animals were randomly allocated into 6 groups (n = …

The Effect Of N-Acetylcysteine On Inflammation And Oxidative Stress In Cisplatin-Induced Nephrotoxicity: A Rat Model, İnayet Güntürk, Cevat Yazici, Sema Kader Köse, Fatma Dağli, Bi̇lal Yücel, Arzu Hanim Yay Jan 2019

The Effect Of N-Acetylcysteine On Inflammation And Oxidative Stress In Cisplatin-Induced Nephrotoxicity: A Rat Model, İnayet Güntürk, Cevat Yazici, Sema Kader Köse, Fatma Dağli, Bi̇lal Yücel, Arzu Hanim Yay

Turkish Journal of Medical Sciences

Background/Aim: Cisplatin is a highly effective chemotherapeutic agent used in the treatment of solid organ cancers. Besides its chemotherapeutic effectiveness, cisplatin administration is associated with numerous side effects. Of those, the most clinically significant and common effect is nephrotoxicity. Recent studies reported that oxidative stress and inflammation are probably the most important mechanisms that contribute to the nephrotoxicity. N-acetylcysteine (NAC) is an antioxidant and antiinflammatory agent. In the present study, the effects of NAC on cisplatin-induced nephrotoxicity were investigated. Materials and methods: Rats were divided into four groups each including eight rats: CONT, NAC-250, CP, and CP+NAC. Rats in experimental …
