Antimicrobial Activity And Characteristics Of Bacteriocins Produced By Vaginal Lactobacilli, Şengül Alpay Karaoğlu, Faruk Aydi̇n, S. Sirri Kiliç, Ali̇ O. Kiliç

Turkish Journal of Medical Sciences

The aim of this study is to test the production of bacteriocin in vaginal Lactobacilli and determining the characterization and antibacterial activity of this bacteriocin. For this reason 100 Lactobacillus strains were isolated and identified from vaginal swab samples of 75 women who attended obstetric and gynecology clinics. It was determined that six of 100 Lactobacillus strains produced bacteriocin. The antibacterial activity of bacteriocins was examined and found to be effective on vaginal L. gasseri, L. acidophilus, Gardnerella vaginalis ATCC 14018 and Pseudomonas aeroginosa ATCC 10145. The aim of this study was to investigate the antibody response following immunization with plasmid vector DNA containing the hepatitis B virus core antigen (HBcAg) gene of a hepatitis B virus (HBV) isolated from a patient in Turkey. A HBcAg gene fragment was cloned into a pcDNA 3 eukaryotic expression vector. After confirmation of the cloning of the HBcAg gene, the expressing vector based HBcAg was used in DNA immunization experiments. Four-to 6-week-old Balb/c mice were immunized with HBcAg expressing plasmids and the anti-HBcAg antibody responses of the mice were evaluated by enzyme immunoassay (EIA). In this study we investigated nitric oxide metabolites nitrate+nitrite and lipid peroxidation product (malonyldialdehyde: MDA) related to the pathogenesis of eclampsia and intrauterine growth retarded (IUGR) pregnancy. Plasma nitrate+nitrite levels were higher in the eclamptic (31.6 ± 16.7 µmol/L p < 0.001) and IUGR (24.3 ± 9.5 µmol/L p < 0.005) groups than in the control group (18.8 ± 6.9 µmol /L). Plasma MDA levels were higher in the eclamptic group (5.5 ± 2.5 nmol/ml (p < 0.05) but not significantly different in the IUGR (3.4 ± 2.4 nmol/ml p > 0.05) group than control group (3.0 ± 1.3 nmol/ml). In addition, the plasma nitrite+nitrate and MDA levels in the eclamptic group were higher than in the IUGR group (p < 0.05). These variations could play different roles in the pathogenesis of eclampsia and IUGR pregnancy.

We have modified a high-performance liquid chromatography (HPLC) procedure for homocysteine assay using 7-fluoro-2, 1, 3-benzoxadiazole-4-sulfonamide (ABD-F) pre-column derivatization. In this study, the thiol compounds were liberated from plasma proteins by reduction with 2-mercaptoethanol (BME) and derivatized with a thiol-specific fluoregenic marker, ABD-F. The derivatives were separated by reversed-phase HPLC on a Hypersil C 18 column (4.6 x 150 mm; 5 µ) using isocratic elution with 0.1 M phosphate buffer pH 6.0 containing 8% methanol and fluorescence detection. Elevated plasma homocysteine concentration either in fasting state or after methionine loading is an independent risk factor for vascular disease. The aim of this study was to evaluate the relationship between fasting total homocysteine (tHcy) and post-methionine load tHcy in healthy control subjects and in patients with angiographically defined coronary artery disease (CAD). In this study the non-peroxide antibacterial factors in Malaysian honey were isolated and identified. The phenolic components were extracted from two different local floral honeys and their effects on the growth of selected pathogens were examined. A solid-phase extraction procedure was applied for the first time to recover honey phenolics. Identification was carried out via high performance liquid chromatography and gas chromatography analysis. Antibacterial activity was determined via the disc-diffusion and broth dilution assays. The phenolic fractions of gelam and coconut honeys showed potent antibacterial activities. Both honeys contain gallic, caffeic, and benzoic acids. Nowadays, modern biopsy techniques such as fine and wide needles are used instead of invasive biopsy techniques for examining malign and benign lesions. This study examines whether wide needle biopsy (Tru-cut) possesses advantages as an alternative method to open biopsy. This study was performed on 40 patients with suspicious intra-jaw lesions. Our purpose was to investigate whether Helicobacter pylori (Hp) has a specific mucosal appearance. Eighty-eight (female / male: 53/35) patients included in the study underwent endoscopy to determine the macroscopic appearance of their gastric mucosa. Biopsies taken from the antrum, incisura angularis and corpus of each patient were stained with toluidine blue and examined for Hp under a light microscope. Thirty were Hp negative and 58 Hp positive. Thirty-six percent of the patients had normal mucosa, 21.59% had spotty erythema in the antrum, 31.81% had a mosaic pattern in the antrum and/or corpus and 10.23% had acute duodenal ulcer. Microalbuminuria is considered to be an early indicator of diabetic nephropathy. We have previously reported the establishment of A-HAlb/98 hybridoma secreting monoclonal antibody that specifically recognizes human serum albumin (HSA). In this report, an in-house enzyme-linked immunosorbent assay (ELISA) was developed using this antibody for the measurement of urinary albumin (UA) and UA was examined in 24 different samples. A-HAlb/98 was used as the capture antibody by immobilizing onto microtitre plates in ELISA format. The objective of this study was to determine the morphological effects of chronic nitric oxide (NO) inhibition on the thoracic aorta of Wistar rats. Fifteen male Wistar Albino rats were divided into three groups. A pilot study was undertaken to investigate the differences in oxidative DNA damage and 2-h DNA repair in peripheral lymphocytes between lung cancer patients and healthy subjects. Twenty-four lung cancer patients and 23 normal controls were recruited from the Queen Mary Hospital, Hong Kong SAR, China. The neural crest origin of the aortopulmonary septum has led us to investigate the septum. We designed the present study to assess the hemodynamic changes following aortopulmonary separation in the rat. We freed the ascending aorta from the pulmonary artery by blunt dissection and placed aluminium foil between them in anesthetized, ventilated, open-chest animals and found that the procedure resulted in a slight but significant increase in systolic and diastolic blood pressure and in an insignificant elevation of the heart rate. The Effects Of Parenting Modes Of Our Middle Class On The Future Behavior Of Early Adolescents: A Behavioral Analytical Point Of View, Sabi̇ha Keski̇n

Using the statements of 352 14-year-old early adolescents, 183 boys and 154 girls, with no history of conduct disorder, we surveyed the implications of how the behavior modification modes of our middle class will affect the future behavior of the subjects. Severe reprimand (53%), privilege removal (31%) and intense painful stimuli (12%) were the behavior managing modes used by the mother (53%), father (25%) or tutor (12%).
