Heterogeneity In Genetic Diversity Among Non-Coding Loci Fails To Fit Neutral Coalescent Models Of Population History, Jeffrey L. Peters, Trina E. Roberts, Kevin Winker, Kevin G. Mccracken Feb 2012

Heterogeneity In Genetic Diversity Among Non-Coding Loci Fails To Fit Neutral Coalescent Models Of Population History, Jeffrey L. Peters, Trina E. Roberts, Kevin Winker, Kevin G. Mccracken

Biological Sciences Faculty Publications

Inferring aspects of the population histories of species using coalescent analyses of non-coding nuclear DNA has grown in popularity. These inferences, such as divergence, gene flow, and changes in population size, assume that genetic data reflect simple population histories and neutral evolutionary processes. However, violating model assumptions can result in a poor fit between empirical data and the models. We sampled 22 nuclear intron sequences from at least 19 different chromosomes (a genomic transect) to test for deviations from selective neutrality in the gadwall (Anas strepera), a Holarctic duck. Nucleotide diversity among these loci varied by nearly two …

Himalayan grey goral (Naemorhedus goral bedfordi) is a Red List (Near Threatened) member of Caprinae endemic to Asia, occupying the south-facing slopes of the Himalaya Mountains. The species is listed as Vulnerable on the National Pakistan Red List, but the distribution and abundance of the species within the nation is poorly known. We conducted a national census of Himalayan grey goral in Pakistan during 2002 – 2004, with the objective of documenting the geographic locations of populations, estimating population size, and describing group sizes and sex ratios. We conducted a direct count census in 98 sites distributed over …

Time Of Day Regulates Subcellular Trafficking, Tripartite Synaptic Localization, And Polyadenylation Of The Astrocytic Fabp7 Mrna, Jason R. Gerstner, William M. Vanderheyden, Timothy Lavaute, Cara J. Westmark, Labib Rouhana, Allan I. Pack, Marvin Wickens, Charles F. Landry

Biological Sciences Faculty Publications

The astrocyte brain fatty acid binding protein (Fabp7) has previously been shown to have a coordinated diurnal regulation of mRNA and protein throughout mouse brain, and an age-dependent decline in protein expression within synaptoneurosomal fractions. Mechanisms that control time-of-day changes in expression and trafficking Fabp7 to the perisynaptic process are not known. In this study, we confirmed an enrichment of Fabp7 mRNA and protein in the astrocytic perisynaptic compartment, and observed a diurnal change in the intracellular distribution of Fabp7 mRNA in molecular layers of hippocampus. Northern blotting revealed a coordinated time-of-day-dependent oscillation for the Fabp7 mRNA poly(A) tail throughout …

Correlation Of Air Displacement Plethysmography With Alternative Body Fat Measurement Techniques In Men And Women, Brittany L. Reinert, Roberta L. Pohlman, Lynn K. Hartzler Jan 2012

Correlation Of Air Displacement Plethysmography With Alternative Body Fat Measurement Techniques In Men And Women, Brittany L. Reinert, Roberta L. Pohlman, Lynn K. Hartzler

Biological Sciences Faculty Publications

Obesity has reached epidemic proportions with serious health consequences. Techniques used to measure body fat (BF) yield variable BF estimates, and this variability may lead to underestimation or overestimation of BF and subsequent treatment options. The measurements that are most accurate (Dual-Energy X-ray Absorptiometry (DEXA) and Air Displacement Plethysmography (ADP)) are expensive and often unavailable. The purpose of this study is to find the commonly available BF measurement that is the most accurate and practical for individual body types in the general population and compare these measurements to ADP (BOD POD®) as the standard. Field measurements include skinfolds …

Prmt5 And The Role Of Symmetrical Dimethylarginine In Chromatoid Bodies Of Planarian Stem Cells, Labib Rouhana, Ana P. Vieira, Rachel H. Roberts-Galbraith, Phillip A. Newmark Jan 2012

Prmt5 And The Role Of Symmetrical Dimethylarginine In Chromatoid Bodies Of Planarian Stem Cells, Labib Rouhana, Ana P. Vieira, Rachel H. Roberts-Galbraith, Phillip A. Newmark

Biological Sciences Faculty Publications

Planarian flatworms contain a population of adult stem cells (neoblasts) that proliferate and generate cells of all tissues during growth, regeneration and tissue homeostasis. A characteristic feature of neoblasts is the presence of chromatoid bodies, large cytoplasmic ribonucleoprotein (RNP) granules morphologically similar to structures present in the germline of many organisms. This study aims to reveal the function, and identify additional components, of planarian chromatoid bodies. We uncover the presence of symmetrical dimethylarginine (sDMA) on chromatoid body components and identify the ortholog of protein arginine methyltransferase PRMT5 as the enzyme responsible for sDMA modification in these proteins. RNA interference-mediated depletion …

Morphological Variation Of Rusty Crayfish Orconectes Rusticus (Cambaridae) With Gender And Local Scale Spatial Gradients, Luke Etchison, Stephen J. Jacquemin, Michael Allen, Mark Pyron

Biological Sciences Faculty Publications

We tested for the influence of gender, stream, and urbanization on morphological variation in rusty crayfish (Orconectes rusticus) in an east-central Indiana, US watershed. We used geometric morphometrics to characterize shape and tested for differences among and within sites. Males had shallower rostrum, increased head width and length, decreased abdomen and cephalothorax width and length, and increased telson length compared to females. Morphology of males did not vary with stream or along an urban gradient. The morphology of females varied with stream and along an urban gradient. Female shapes from small creek sites were stouter and less fusiform …

The Sign And Strength Of Plant-Soil Feedback For The Invasive Shrub, Lonicera Maackii, Varies In Different Soils, Kelly Schradin, Don Cipollini Jan 2012

The Sign And Strength Of Plant-Soil Feedback For The Invasive Shrub, Lonicera Maackii, Varies In Different Soils, Kelly Schradin, Don Cipollini

Biological Sciences Faculty Publications

Plants alter soil characteristics causing changes in their subsequent growth resulting in positive or negative feedback on both their own fitness and that of other plants. In a greenhouse study, we investigated whether the sign and strength of feedback changed across two distinct soil types, and whether effects were due to shifts in biotic or abiotic soil traits. Using soils from two different locations, we examined growth of the exotic invasive shrub, Lonicera maackii and the related native shrub, Diervilla lonicera, in unconditioned soils and in soils conditioned by previous growth of L. maackii, D. lonicera, and …

Biological Sciences Faculty Publications

The nematode Caenorhabditis briggsae is an emerging model organism that allows evolutionary comparisons with C. elegans and exploration of its own unique biological attributes. To produce a high-resolution C. briggsae recombination map, recombinant inbred lines were generated from reciprocal crosses between two strains and genotyped at over 1,000 loci. A second set of recombinant inbred lines involving a third strain was also genotyped at lower resolution. The resulting recombination maps exhibit discrete domains of high and low recombination, as in C. elegans, indicating these are a general feature of Caenorhabditis species. The proportion of a chromosome's physical size occupied by …

Biological Sciences Faculty Publications

Background: A deficiency of specific glycosylphosphatidyl inositol-anchored proteins in paroxysmal nocturnal hemoglobinuria may be responsible for most of the clinical features of this disease, but some functional consequences may be indirect. For example, the absence of certain glycosylphosphatidyl inositol-anchored proteins in paroxysmal nocturnal hemoglobinuria cells may influence expression of other membrane proteins. Membrane-bound proteinase 3 co-localizes with glycosylphosphatidyl inositol-linked neutrophil antigen 2a, which is absent in patients with paroxysmal nocturnal hemoglobinuria. Design and Methods: We compared expression of proteinase 3 and neutrophil antigen 2a by flow cytometry and western blotting in normal and paroxysmal nocturnal hemoglobinuria cells and measured cytoplasmic …

The Effects Of Arbuscular Mycorrhizal (Am) Fungal And Garlic Mustard Introductions On Native Am Fungal Diversity, Alexander M. Koch, Pedro M. Antunes, Kathryn Barto, Don Cipollini, Daniel L. Mummey, John N. Klironomos Jul 2011

The Effects Of Arbuscular Mycorrhizal (Am) Fungal And Garlic Mustard Introductions On Native Am Fungal Diversity, Alexander M. Koch, Pedro M. Antunes, Kathryn Barto, Don Cipollini, Daniel L. Mummey, John N. Klironomos

Biological Sciences Faculty Publications

Introduced, non-native organisms are of global concern, because biological invasions can negatively affect local communities. Arbuscular mycorrhizal (AM) fungal communities have not been well studied in this context. AM fungi are abundant in most soils, forming symbiotic root-associations with many plant species. Commercial AM fungal inocula are increasingly spread worldwide, because of potentially beneficial effects on plant growth. In contrast, some invasive plant species, such as the non-mycorrhizal Alliaria petiolata, can negatively influence AM fungi. In a greenhouse study we examined changes in the structure of a local Canadian AM fungal community in response to inoculation by foreign AM fungi …

Effects Of Xs® Energy Drink On Aerobic Exercise Capacity Of Athletes, Kevin M. Sheehan, Lynn K. Hartzler

Biological Sciences Faculty Publications

A small increase in performance is often the difference between winning and losing for athletes, and energy supplements are marketed to give them a competitive edge. One such supplement is XS® Energy Drink which contains B vitamins, caffeine, taurine, and an herbal adaptogen blend. XS® Energy Drink is primarily designed and marketed as an alertness stimulus; however, it is also marketed to athletes as a performance enhancing supplement. The hypothesis of this investigation was that the consumption of XS® Energy Drink before exercise would increase aerobic capacity (an increase in Vo2max) and time to fatigue, and would decrease time for …

Background

Human scabies is a debilitating skin disease caused by the “itch mite” Sarcoptes scabiei. Ordinary scabies is commonly treated with topical creams such as permethrin, while crusted scabies is treated with topical creams in combination with oral ivermectin. Recent reports of acaricide tolerance in scabies endemic communities in Northern Australia have prompted efforts to better understand resistance mechanisms and to identify potential new acaricides. In this study, we screened three essential oils and four pure compounds based on eugenol for acaricidal properties.

Methodology/Principal Findings

Contact bioassays were performed using live permethrin-sensitive S. scabiei var suis mites harvested from …

Biological Sciences Faculty Publications

Background

The nematode C. briggsae serves as a useful model organism for comparative analysis of developmental and behavioral processes. The amenability of C. briggsae to genetic manipulations and the availability of its genome sequence have prompted researchers to study evolutionary changes in gene function and signaling pathways. These studies rely on the availability of forward genetic tools such as mutants and mapping markers.

Results

We have computationally identified more than 30,000 polymorphisms (SNPs and indels) in C. briggsae strains AF16 and HK104. These include 1,363 SNPs that change restriction enzyme recognition sites (snip-SNPs) and 638 indels that range between 7 …

Isoform-Specific Regulation And Localization Of The Coxsackie And Adenovirus Receptor In Human Airway Epithelia, Katherine J.D.A. Excoffon, Nicholas D. Gansemer, Matthew E. Mobily, Philip H. Karp, Kalpaj R. Parekh, Joseph Zabner Mar 2010

Isoform-Specific Regulation And Localization Of The Coxsackie And Adenovirus Receptor In Human Airway Epithelia, Katherine J.D.A. Excoffon, Nicholas D. Gansemer, Matthew E. Mobily, Philip H. Karp, Kalpaj R. Parekh, Joseph Zabner

Biological Sciences Faculty Publications

Adenovirus is an important respiratory pathogen. Adenovirus fiber from most serotypes co-opts the Coxsackie-Adenovirus Receptor (CAR) to bind and enter cells. However, CAR is a cell adhesion molecule localized on the basolateral membrane of polarized epithelia. Separation from the lumen of the airways by tight junctions renders airway epithelia resistant to inhaled adenovirus infection. Although a role for CAR in viral spread and egress has been established, the mechanism of initial respiratory infection remains controversial. CAR exists in several protein isoforms including two transmembrane isoforms that differ only at the carboxy-terminus (CAREx7 and CAREx8). We found low-level …

Cellular And Molecular Dissection Of Pluripotent Adult Somatic Stem Cells In Planarians, Norito Shibata, Labib Rouhana, Kiyokazu Agata Jan 2010

Cellular And Molecular Dissection Of Pluripotent Adult Somatic Stem Cells In Planarians, Norito Shibata, Labib Rouhana, Kiyokazu Agata

Biological Sciences Faculty Publications

Freshwater planarians, Plathelminthes, have been an intriguing model animal of regeneration studies for more than 100 years. Their robust regenerative ability is one of asexual reproductive capacity, in which complete animals develop from tiny body fragments within a week. Pluripotent adult somatic stem cells, called neoblasts, assure this regenerative ability. Neoblasts give rise to not only all types of somatic cells, but also germline cells. During the last decade, several experimental techniques for the analysis of planarian neoblasts at the molecular level, such as in situ hybridization, RNAi and fluorescence activated cell sorting, have been established. Moreover, information about genes …
