As coordinator, I prepared the following pages for the campaign to abolish the Draize rabbit blinding test. The plan includes background information, short and long range goals and step by step initiatives. This working blueprint was circulated to individuals and organizations interested in supporting the campaign. And we made a point of sharing copies of our plan with the cosmetics industry and any interested observers. It has always been our policy to have an up-front, open agenda.

Summary

The study of trypanosomatid biology would benefit greatly from the acquisition of a tool for culturing such as those used in microbiology. Agar plate culture has been described several times, but until the present its potential for use in this area has been underestimated. The previous work has demonstrated that this culture method may be used to culture a wide variety of species, that these species give rise to identifiable colony phenotypes, and that these are stable regardless of the life cycle stage used to initiate the culture. The periapical tissue reaction to three concentrations of endotoxin solutions and to a sterile saline control solution was examined in six adult cats. The maxillary and mandibular cuspids were isolated and the pulps extirpated with barbed broaches. The four solutions (1, 10, 100 μg/ml endotoxin, and saline) were then injected into the four prepared teeth for each cat and the access cavities sealed with IRM.

At two week intervals, two cats were sacrificed and the specimens removed, while the remaining animals were retreated as previously described. The block tissue sections were radiographed, fixed in 10% buffered formalin, and prepared for …

Four groups of female Swiss Webster mice were injected intraperitoneally with similar volumes of normal saline, sonicated Trypanosoma musculi and saline, sonicated trypanosomes and Freund's Complete Adjuvant (FCA) and FCA. When the mice became immune and thus responded negatively to a T. musculi challenge, sera were collected and administered to four other groups of mice to establish passive immunity in these animals.

An established dose of innoculum of Trichinella spiralis larvae (8.1 larvae/gram of body weight) was given to all the mice by gastric gavage. These mice were killed 26 days later and the musculature digested with artificial gastric juice …

Cell mediated immune (CMI) cytotoxic reactivity of Balb/c mice against H238 cells, a Herpes simplex virus (HSV) Type 2 - induced sarcoma, was measured by the ¹²⁵IUdR release assay. The Balb/c mouse response to the growing HSV-induced tumor, treated with radiation and , was measured by survival rate, tumor growth and immune cytotoxicity of spleen and peritoneal exudate cells (PEC) as determined by ¹²⁵IUdR release assay. Subcutaneous (s.c.) inoculation of 1 x 10⁶ H238 cells (high dose) produced progressive tumor growth while s.c. inoculation of 1 x 10⁴ H238 cells (low dose) produced no tumors. A …

Some of the animal protection societies in England were prepared to accept animal experimentation provided it was performed under anesthesia. The Metcalf-Hatch Act forces tax supported pounds and shelters to turn over unwanted cats and dogs to New York laboratories. This creates difficulties for shelters/pounds. People who are aware of Metcalf-Hatch (MH) will tend to abandon animals rather than risk having them recycled into labs. And this places additional tax burdens on communities for rounding up abandoned strays. The cost of stray animals is already $400 million a year in the USA.

It has been shown previously that a root surface which has been scaled, root planed and demineralized by topical application of citric acid in vitro, is dominated by exposed collagen fibrils. The purpose of this study was to examine the mechanism of healing and possible reattachment of soft periodontal connective tissue to the root planed and subsequently acid treated root surfaces and to determine whether new collagen fibrils produced in the connective tissue could attach to these exposed fibrils.

For the past decade, evidence has accumulated which suggests that the resistance of the teeth to dental decay is in part under systemic control. It was shown that the resistance is accomplished through the optimal operation of a dentinal fluid transport mechanism which is believed to be hormonally controlled by the hypothalamic-parotid gland endocrine axis. Although circumstantial evidence for such an endocrine function was strong, the isolation of a parotid hormone which stimulates the dentinal fluid transport mechanism remained to be accomplished.

Using modern biochemical techniques, a methodology was designed to isolate and purify the hormone from porcine parotid glands. …

It has been concluded from this study that nerve fibers in the superior laryngeal nerves have an influence on the development of C-cells in the mouse thyroid. This conclusion is based on the results of two types of investigation. The first is an analysis of the effect of age on the number of C-cells and the second is an investigation of the influence of denervation and hemithyroid-parathyroidectomy on the age related changes in C-cells numbers.

The possible effects of ablastin, a reproduction-inhibiting antibody which is produced during the infection of the blood parasite, Trypanosoma musculi in the mouse, were investigated by fractionating the parasite into a supernatant fraction (designated as Group A test mice) and a pellet fraction (Group B test mice). These groups A and B mice were injected with the respective fractions, plus Freund’s Complete Adjuvant (FCA). Control mice were injected with FCA only. Administration of Corynebacterium parvum in combination with localized irradiation has been shown to be more efficient than radiation treatment alone in prolonging the survival time of methylethrane induced tumor in C3H mice. Human Placental Alkaline Phosphatase And Steroid Metabolism By Rat Adrenals, Allen G. Meier

Human placental alkaline phosphatase was purified by butanol extraction, methanol precipitation and chromatography on DEAE-cellulose. The pH optimum was found to be 10.9. The enzyme was competitively inhibited by di-ethyl-p-nitrobenzyl-phosphonate and copper sulfate. An increase in enzymatic activity occurred in the presence of the phosphate ion (HPO₄) and ATP.

The enzyme was crystallized from a 40% saturated solution of ammonium sulfate and shown to be homogeneous by sedimentation analysis. Homogeneity of the enzyme was also demonstrated using poly-acrylamide-gel-electrophoresis. At a pH of 7.5% the electrophoretic pattern yielded a single band and showed no significant contamination.

Endodontic therapy was evaluated as a possible control or prevention modality of radiation type dental caries in dogs. Two adult male dogs with permanent dentitions had endodontic therapy performed on alternating teeth in each jaw. The nontreated teeth served as controls.

The dogs were irradiated with 7000 rad Cobalt-60 radiation fractionated over an eight week period. The dogs were killed at 60 and 182 days post-irradiation. The teeth were removed from the jaws and immersed in methylene blue dye, sectioned, and the dye penetration pattern photographed. Diffusion of the dye into the tooth structure was quantified and statistically analyzed.

This study compared the ability of three different bone growth factor combinations with that of Ca(OH)₂ to stimulate apical closure as well as the effectiveness of Mineral Trioxide Aggregate (MTA) as an apical barrier. Fifty-six roots of second and fourth mandibular premolars of seven six- month old beagle dogs were used. Periradicular lesions were created over a four week period. The canals were cleaned and shaped and filled with Ca(OH)₂ for one week. They were then irrigated and received one of seven different treatments in a balanced design. In April and May, 1977, 397 (14 species) amphibians were collected from southwestern and central West Virginia and examined for parasites. Five species of trematodes and two species of cestodes were found from 93 hosts. The total of 397 amphibians was subdivided, for convenience, into six categories of amphibians; aquatic male and female, terrestro-aquatic male and female, and terrestrial male and female. Nine of 24 (37.5%). female aquatic amphibians was the highest incidence found, while terrestro-aquatic females contained the highest mean number (6.0) worms per incidence, and parasite loads of host. Hosts, each platyhelminth specimen are given.

The possible effects of ablastin on Schistosoma mansoni were investigated by (1) establishing a concurrent infection of Trypanosoma musculi and Schistosoma mansoni in mice, and (2) by obtaining Trypanosoma lewisi immune rat serum (ablastin) and injecting it into S. mansoni infected mice. These procedures were followed on days 4, 14 and 28 of S. mansoni infection. For the groups that were given immune rat serum, controls were kept into which normal rat serum was injected.

Ablastin does not have any effect on the development or morphology of the S. mansoni worms as tested in this study. Although ablastin inhibits glycolytic …

Hypothalamic and cortical extracts from various cariogenically grouped rats which had been maintained on either a Purina diet or a high sucrose diet (HSD) for three months were infused intravenously into 28 days old rats to determine if there was a titer of parotid hormone releasing factor (PRF) capable of facilitating fluid movement (FM) in the teeth. The hypothalamic and cortical extracts obtained from Purina-fed rats with 0-caries and the hypothalamic extracts from BSD-fed rats conclusively demonstrated FM stimulatory ability. Other Purina-fed groups were essentially biologically ineffective, however, extracts from HSD-fed caries groups including scores 11-40 demonstrated extremely potent FM …
