Edamame are an emerging crop in Arkansas, with 2013 planting estimates at 680 hectares. Although edamame are the same species as conventional soybeans grown in Arkansas, differences in use, maturity, and harvest may require creation and implementation of insect pest management approaches that differ from those used in conventional soybean production systems in order for a quality crop to be produced. Rice false smut (FS), a disease caused by Ustilaginoidea virens (Cke.) Takahashi (1896), was first reported in northeastern Arkansas counties in 1997. The first objective of this research was to establish a collection of U. virens isolates from geographically diverse regions of Arkansas. Three U. virens isolates and chlamydospores from `Templeton' and `Clearfield-151' rice cultivars were used to determine the effects of temperature and pH on mycelial growth and germination. The use of soil fumigation for nematode management in cotton (Gossypium hirsutum) has become increasingly popular in recent years in the absence of effective resistant cultivars. While soil fumigation is relatively expensive, lint yields have consistently been improved to make this practice profitable in fields with severe nematode pressure. Growers in southern Arkansas have observed changes in cotton growth patterns when severely infested fields are fumigated. The most noticeable change has been excessive (rank) growth resulting in an increased need for growth regulators, especially where the nitrogen fertilization exceeds standard recommendations. Two-spotted spider mites (Tetranychus urticae Koch, 1836) are pests of vegetables, ornamentals, and row crops around the world. Two-spotted spider mites have become an important long-season pests of cotton, causing injury to cotton from an early vegetative stage. In the past eight years, Arkansas cotton acreage treated for spider mites has more than doubled and most of the increase has been attributed to early season infestations. Seed-applied insecticides are the standard control method used to prevent rice water weevil (Lissorhoptrus oryzophilus Kuschel) injury to rice (Oryza sativa L.) roots, and often results in greater yields than rice that receives no seed-applied insecticide. Yield increases from seed-applied insecticides often occur even when insect pressure is low and should not cause yield loss. Bovine cytochrome P450 3A28 is responsible for metabolizing ergot alkaloids that cattle ingest when feeding on endophyte-infested tall fescue grass. The objective of this research was to determine associations among genotype, transportation, and stress responses. Angus crossbred steers (n = 47) were genotyped (CC, CG, or GG) for a single-nucleotide polymorphism (C994G) in cytochrome P450 3A28. Genotypes were determined by polymerase chain reaction (PCR) amplification followed by restriction enzyme (Alu1) digestion. Steers were backgrounded on a mixed-cultivar tall fescue pasture. Most forages commonly used to feed horses have potential detriments including blister beetles or excessive fiber concentrations. Teff grass (T), a warm-season annual forage, has the potential to be a good alternative for horses because of its lack of observed disorders. Our objective was to compare preference by horses for T harvested under different conditions with that of bermudagrass (B) harvested at two maturities. Aphids are a group of sap-feeding insects that attack most of the world’s crops. The loss of function of fatty acid desaturase7 (FAD7) in Solanum lycopersicum (tomato plant) induces aphid resistance that is dependent upon the accumulation of plant defense hormones such as salicylic acid (SA). Tomato lacks most of the genetic resources found in the model plant Arabidopsis (Arabidopsis thaliana). Increasing salinity is an important factor limiting agricultural productivity worldwide. In addition to direct effects on growth and yield, diseases also may be affected. This study characterized the effects of soil salinity on seedling disease of soybean and rice caused by Pythium spp. Controlled environment experiments on soybean used two cultivars which differed in chloride tolerance and soil treated with a CaCl2 solution to create a range of electrical conductivity (EC) levels. GLS is a serious foliar disease of maize, a major staple crop grown commercially in the USA for both human and animal feed production, and increasingly, for ethanol production. The disease is caused by two Cercospora species, C. zeae-maydis and C. zeina, both of which infect maize in the USA and in other parts of the world, with yield losses potentially greater than 50%, depending on local conditions. Aerial blight is caused by Rhizoctonia solani AG1-IA. This pathogen also causes sheath blight of rice. In Arkansas, many soybean and rice fields undergo an annual rotation of these two crops which facilitates a continuous source of inoculum from one year to the next. Aerial blight is a two stage disease in that R. solani AG1-IA colonizes the plant during the early vegetative growth stages and then aerial blight develops after the soybean canopy closes in the later reproductive stages of development. Consequently, field trials were conducted to determine the response of soybean (cv. AG 4703) to imazosulfuron drift and carryover (at Fayetteville, Marianna, Keiser and Pine Tree, AR) from rice. Johnsongrass, once the most persistent and troublesome grass weed of row crops throughout the southern U.S., has previously been confirmed resistant to the acetolactate synthase (ALS) and acetyl-coenzyme A carboxylase-inhibiting herbicides in the Midsouth and has recently evolved resistance to glyphosate in Arkansas. The goal of this research was to establish the geographical distribution of herbicide-resistant johnsongrass in Arkansas and to develop herbicide programs for controlling glyphosate-resistant johnsongrass in Arkansas soybean. Japanese beetles are a relatively new pest to Arkansas. During my Ph.D. research I investigated the pathogens and environmental factors influencing Japanese beetle populations in the state. The prevalence of various pathogens and parasitoids attacking Popillia japonica were recorded annually from wild populations. Of specific interest was the microsporidian pathogen Ovavesicula popilliae, which I introduced as a biological control agent in the state. Details of the relationship between this pathogen and the Japanese beetle were investigated, including dose response, host range, and spore production. The superfamily Criconematoidea has been studied since 1886. It is composed of two families: Criconematidae (subfam. Criconematinae, Hemicycliophorinae) and Tylenchulidae (subfam. Tylenchulinae, Paratylenchinae and Tylenchocriconematinae). Multiple species in genera have been identified and differences and similarities have been found. Species belonging to genera Mesocriconema and Criconemoides show very few differences making their identification difficult. Seventy two populations were studied. They were collected in Arkansas and/or received from the following states: California, Florida, Kansas, Missouri, North Carolina and Tennessee. In the US, Arkansas is the largest rice producing state; however, an estimated 62% of the rice fields in the state are infested with red rice, and can cause up to 80% yield reduction in rice. In the mid-2000's, glyphosate-resistant Palmer amaranth biotypes began to emerge in many southern states. In 2006, glyphosate-resistant Palmer amaranth was identified in a field in Mississippi County, Arkansas. A greenhouse experiment was conducted in 2008 to screen Palmer amaranth accessions, collected in this survey, for glyphosate resistance. Inflorescence were collected from a total of 276 plants from fields were glyphosate failure occurred, representing 74 accessions in 14 counties, including Clay, Craighead, Crittenden, Greene, Jackson, Jefferson, Lawrence, Lee, Mississippi, Phillips, Poinsett, Randolph, St. Francis, and White Counties. Females drill through pine bark to deposit eggs, a symbiotic fungus, Amylostereum, and phytotoxic mucus into the tree. In their native range, these insects are not viewed as primary pests because they attack dead or dying trees. Soybean vein necrosis disease (SVND) is widespread in major soybean-producing areas in the U.S. The typical disease symptoms exhibit as vein clearing along the main vein, which turn into chlorosis or necrosis as season progresses. Double-stranded RNA isolation and shot gun cloning of symptomatic tissues revealed the presence of a new tospovirus, provisionally named as Soybean vein necrosis associated virus (SVNaV). Sirex females drill through the bark, phloem and into xylem tissues where they deposit eggs along with a symbiotic Amylostereum fungus. The presence of Amylostereum is necessary for successful development of Sirex immatures as the larvae are unable to derive adequate nutrition from xylem in the absence of the fungus. The root-knot nematode, Meloidogyne incognita, and the seedling pathogen, Thielaviopsis basicola, commonly co-exist in Arkansas cotton fields and may interact resulting in increased losses. The primary objective of this research was to evaluate the effects of soil physical parameters on these soilborne pathogens and cotton growth in controlled environmental, field, and microplot studies. Controlled environmental experiments used two soil bulk densities and four pathogen treatments: non-infested soil, soil infested with M. incognita or T. basicola and soil infested with both pathogens. Charcoal rot of soybean, caused by the soilborne fungus Macrophomina phaseolina (Tassi) Goid, is a disease associated with high soil temperature and low soil moisture. Above-ground symptoms, which can be difficult to distinguish from drought symptoms, include low vigor, dead leaves that remain attached to the plant, early senescence and yield loss. The European woodwasp, Sirex noctilio F. (Hymenoptera: Siricidae), which is a known destructive pest of pine in the southern hemisphere was recently discovered in the eastern United States. Before we can understand how S. noctilio may affect pine forests throughout the United States, we need a better understanding of native Sirex and the role they play in the ecosystem. Rice blast, caused by Pyricularia oryzae (teleomorph: Magnaporthe grisea), is one of the most economically important diseases of rice worldwide, including Arkansas. Bird cherry-oat aphid is a common pest of Arkansas winter wheat. This aphid vectors barley yellow dwarf virus which may cause extensive crop damage and yield loss when wheat is infested by virulent aphids in the fall. Some suggest this damage may be avoided using insecticide seed treatments if growers are unable to delay planting, as is recommended. Field population dynamics of bird cherry-oat aphid during fall 2009 and 2010 was assessed through random sampling of whole plants and pan trapping methods to evaluate aphid immigration. Plants are frequently under attack from pests and deploy various defense mechanisms to fight off predators. Many plant defenses are induced following herbivory or pathogen infection. An investment in defense could potentially lead to a diversion of metabolic resources away from primary metabolism, including reduction in plant commitment to photosynthesis. In this thesis, changes in expression levels of representative genes for primary and secondary metabolism were studied using real-time polymerase chain reaction (PCR) in Medicago truncatula Gaertner subjected to beet armyworm (Spodoptera exigua H¨¹bner) damage or Phoma medicaginis Malbr. & Roum. infection.
