Available water for urban landscape irrigation is likely to become more limited because of inadequate precipitation and the ever-increasing water demand of a growing population. Recent droughts in the western United States have also increased the demand for low-water-use landscapes in urban areas. Penstemon species (beardtongues) are ornamental perennials commonly grown in low-water-use landscapes, but their drought tolerance has not been widely investigated. The objectives of this study were to determine the effects of water availability on the morphology, physiology, and canopy temperature of Penstemon barbatus (Cav.) Roth ‘Novapenblu’ (Rock Candy Blue® penstemon), P. digitalis Nutt. ex Sims ‘TNPENDB’ …

Department of Agronomy and Horticulture: Faculty Publications

Glyphosate-resistant (GR) Palmer amaranth (Amaranthus palmeri S. Watson) is widespread in the Central Great Plains. Introduction of newly developed dicamba/glufosinate/glyphosate (DGG)-resistant soybean varieties allows postemergence (POST) applications of dicamba and glufosinate for in-season control of GR Palmer amaranth. Limited information exists on the effectiveness of glufosinate applied late-POST for tall (70–90 cm) GR Palmer amaranth control in DGG-resistant soybean. The objectives of this study were to (1) determine the effectiveness of late-POST glufosinate-based programs for GR Palmer amaranth control, and (2) determine the impact of those programs on soybeans grain yields. Ten glufosinatebased programs were tested in a field …

Plants Detect And Adapt, But Do Not Feel, Paul C. Struik Apr 2023

Plants Detect And Adapt, But Do Not Feel, Paul C. Struik

Animal Sentience

Plant sentience is a hot topic in scientific and popular media. There are moral reasons to respect both the service of plants to humanity and their natural integrity as creatures playing their own significant role in a complex ecosystem. However, to infer that plants have certain cognitive capacities that are present also in certain human and nonhuman animals calls for scientific rigor beyond mere analogy. The unique capacities of plants identified by Segundo-Ortin & Calvo are not necessarily linked to sentience. Nor is it likely that sentience is an evolutionary trait that is present to some extent in all living …

IGC Proceedings (1977-2023)

Lignification of plant cell walls has been identified as a major factor limiting forage digestibility. It limits the amount of digestible energy available to livestock, resulting in an incomplete utilisation of cellulose and hemicellulose by ruminant animals. Modification of the lignin profile of ryegrasses (Lolium spp.) and fescues (Festuca spp.) is undertaken through modulating the expression of genes encoding enzymes involved in the biosynthesis of monolignols.

Identification Of Putative AtTt2 R2r3-Myb Transcription Factor Orthologues In Tanniferous Tissues Of L. Corniculatus Var. Japonicus Cv Gifu, D. N. Bryant, P. Bailey, P. Morris, Mark P. Robbins, C. Martin, T. Wang Apr 2023

Identification Of Putative AtTt2 R2r3-Myb Transcription Factor Orthologues In Tanniferous Tissues Of L. Corniculatus Var. Japonicus Cv Gifu, D. N. Bryant, P. Bailey, P. Morris, Mark P. Robbins, C. Martin, T. Wang

IGC Proceedings (1977-2023)

R2R3-MYB plant transcription factors are sequence–specific DNA-binding proteins, which regulate the expression of specific gene(s) following the R2R3 DNA-binding domain interacting with the corresponding promoter sequence(s). The biosynthetic pathway leading to the production of anthocyanins has been demonstrated to be under MYB transcriptional regulatory control (Cone et al., 1986), while the accumulation of proanthocyanidins (PAs) in Arabidopsis seed coats is determined by the R2R3-MYB AtTT2 (Nesi et al., 2001). Using an informatics approach, partial sequences of putative AtTT2 orthologues have been identified and cloned from the forage legume Lotus corniculatus var. japonicus cv Gifu.

A Leaf-Level Spectral Library To Support High-Throughput Plant Phenotyping: Predictive Accuracy And Model Transfer, Nuwan K. Wijewardane, Huichun Zhang, Jinliang Yang, James C. Schnable, Daniel P. Schachtman, Yufeng Ge

Department of Agronomy and Horticulture: Faculty Publications

Leaf-level hyperspectral reflectance has become an effective tool for high-throughput phenotyping of plant leaf traits due to its rapid, low-cost, multi-sensing, and non-destructive nature. However, collecting samples for model calibration can still be expensive, and models show poor transferability among different datasets. This study had three specific objectives: first, to assemble a large library of leaf hyperspectral data (n=2460) from maize and sorghum; second, to evaluate two machine-learning approaches to estimate nine leaf properties (chlorophyll, thickness, water content, nitrogen, phosphorus, potassium, calcium, magnesium, and sulfur); and third, to investigate the usefulness of this spectral library for predicting external datasets …

Protease Inhibitor Asp Enhances Freezing Tolerance By Inhibiting Protein Degradation In Kumquat, Hua Yang, Ke-Wei Qiao, Jin-Jing Teng, Jia-Bei Chen, Ying-Li Zhong, Li-Qun Rao, Xing-Yao Xiong, Huang Li

Center for Plant Science Innovation: Faculty Publications

Cold acclimation is a complex biological process leading to the development of freezing tolerance in plants. In this study, we demonstrated that cold-induced expression of protease inhibitor FmASP in a Citrus-relative species kumquat [Fortunella margarita (Lour.) Swingle] contributes to its freezing tolerance by minimizing protein degradation. Firstly, we found that only cold-acclimated kumquat plants, despite extensive leaf cellular damage during freezing, were able to resume their normal growth upon stress relief. To dissect the impact of cold acclimation on this anti-freezing performance, we conducted protein abundance assays and quantitative proteomic analysis of kumquat leaves subjected to cold acclimation …

Senesced Leaves Of An Invasive Species (Lonicera Maackii) Alter Community Composition And Ecosystem Function In Experimental Wetlands, Cy Mott Apr 2023

Senesced Leaves Of An Invasive Species (Lonicera Maackii) Alter Community Composition And Ecosystem Function In Experimental Wetlands, Cy Mott

2023 Midwest Ecology & Evolution Conference

Secondary compounds produced by terrestrial invasive plants can cross terrestrial-aquatic boundaries via senesced leaves, leading to altered patterns of aquatic biodiversity and ecosystem functioning. Prior attempts to examine the impacts of invasive leaves on wetland communities have primarily employed the use of leaf leachates and focal taxa inhabiting relatively low trophic positions, and additional studies are needed to examine impacts on higher-order consumers and potential additive/synergistic effects of leachates and leaf decomposition. Using cattle-tank mesocosms across multiple experiments, we observed that the addition of senescent leaves of Amur Honeysuckle (Lonicera maackii) to native leaves resulted in mass mortality …

Investigating Pollinator Mismatch In Cardamine Concatenata And Claytonia Virginica, Nada Elgousi Apr 2023

Investigating Pollinator Mismatch In Cardamine Concatenata And Claytonia Virginica, Nada Elgousi

2023 Midwest Ecology & Evolution Conference

Global warming is gradually affecting the local climate causing pollinators and plants to change their emergence and flowering times. This can potentially cause a phenology mismatch between pollinator activity and open flowers leading to limitation in seed production in plants. However, other factors such as intense predation or competition can also shift phenology. As part of a preliminary study exploring the mechanisms of phenology mismatch, Cardamine concatenata and Claytonia virginica, were observed to determine if there is a phenology shift due to competition with an invasive plant that flowers early and overlaps these two native species. In early spring, …

A Tale Of Two Scents: Scent Preferences Of Naïve And Experienced Pollinators, Scout N. Steinhiser Apr 2023

A Tale Of Two Scents: Scent Preferences Of Naïve And Experienced Pollinators, Scout N. Steinhiser

2023 Midwest Ecology & Evolution Conference

Mounting evidence of pollinator decline and its repercussions on agriculture and plant diversity means that scientists must work to understand the details of plant-pollinator interactions. Though there are many facets to the relationship between plants and pollinators, one major mediator of this relationship is scent. As much is already known about the scent make-up of Narcissus and its pollinator partners, it is a great model system for additional research. This is also true for the insect species Vanessa cardui, a generalist pollinator found worldwide that has most likely come into contact with Narcissus and its scent. The objective of this …

How Accurately Structure Can Identify Hybrids When Comparing Different Numbers Of Species And Marker Types And Numbers, Chloe Hendrikse Apr 2023

How Accurately Structure Can Identify Hybrids When Comparing Different Numbers Of Species And Marker Types And Numbers, Chloe Hendrikse

2023 Midwest Ecology & Evolution Conference

Hybridization is a common problem in conservation. It can be difficult to identify hybrid individuals but doing so is often necessary in order to make conservation recommendations. For example, botanical gardens and seed banks want to identify if individuals are hybrids of two or more species to determine if offspring could be used for conservation or restoration, and to analyze the risk of possible extinction via introgression. Our project looks at how well the program STRUCTURE can detect and cluster hybrids, their parent species, and other pure species, which we created using simulations. We build on past studies, which typically …

Variation In Synthetic Floral Microbiome Composition Is Largely Ignored By Foraging Bumblebees, Nevin P. Cullen Apr 2023

Variation In Synthetic Floral Microbiome Composition Is Largely Ignored By Foraging Bumblebees, Nevin P. Cullen

2023 Midwest Ecology & Evolution Conference

Most flowers are occupied by diverse communities of microbes, which can alter floral attractiveness to key pollinators and floral fitness by interacting with floral traits. Different species of flower often acquire unique communities of microbes. Consequently, pollinators are frequently forced to choose between two flowers with distinct floral microbiomes while foraging. However, it is currently unknown whether pollinators have an inherent preference for flowers containing one floral microbiome over another, and whether microbial preference is modified by variation in floral traits. We investigated this knowledge gap using distinct bacteria and yeasts cultured from a nickel-hyperaccumulating plant and its non-accumulating relative. …

Quantifying The Relationship Between Arbuscular Mycorrhizal Fungi And A Native And Invasive Thistle, Nicole Baker, Lucia Vera, Ian Rosales Apr 2023

Quantifying The Relationship Between Arbuscular Mycorrhizal Fungi And A Native And Invasive Thistle, Nicole Baker, Lucia Vera, Ian Rosales

2023 Midwest Ecology & Evolution Conference

Arbuscular mycorrhizal fungi (AMF) associate with 80% of vascular plant roots and are an important part of nutrient exchange. Our research quantified the relationship between (AMF) and native Pasture Thistle (Cirsium discolor) and invasive Canada Thistle (Cirsium arvense). We hypothesized that Pasture Thistle would have a stronger relationship with AMF than Canada Thistle because of the degraded mutualism hypothesis, which states that invasive species have reduced relationships with AMF. For our experiment, we grew inoculated and non-inoculated plants at 10 different levels of soil phosphorus concentrations. Pasture Thistle and Canada Thistle were both facultatively dependent upon …

Geographic Analysis Of Sex Chromosome Polymorphism In The Virginian Wild Strawberry, Trezalka A. Budinsky Apr 2023

Geographic Analysis Of Sex Chromosome Polymorphism In The Virginian Wild Strawberry, Trezalka A. Budinsky

2023 Midwest Ecology & Evolution Conference

The evolution of sex chromosomes is a ubiquitous and essential diversifying mechanism in eukaryotes. Yet early stages of sex-chromosome differentiation are poorly understood. The subdioecious octoploid, Fragaria virginiana, provides a rare opportunity to study the beginnings of sex chromosome evolution due to its young, polymorphic ZW sex system. Females are ZW and males/hermaphrodites are ZZ. F. virginiana has a small sex-determining region (SDR) located on the female W chromosome that has undergone three transposition events between the strawberry's four subgenomes, resulting in three SDR haplotypes: alpha (the result of a first transposition and closest to a pre-sex chromosome ancestral …

Spatial Variation In Flowering Age In Cirsium Pitcheri: Does This Imperiled Dune Thistle Hedge Its Bets?, Lucas A. Priemer Apr 2023

Spatial Variation In Flowering Age In Cirsium Pitcheri: Does This Imperiled Dune Thistle Hedge Its Bets?, Lucas A. Priemer

2023 Midwest Ecology & Evolution Conference

For perennial plants, the optimal timing of flowering is a tradeoff between survivorship and fecundity, a classic example of bet-hedging. For many monocarpic perennials, there is an advantage in delaying flowering until the plant reaches a larger size and is thus capable of a higher fecundity. However, in delaying flowering, there is a risk that the plant will die before reproducing at all. Here, we investigate this fitness trade-off in the imperiled dune thistle, Cirsium pitcheri. Using a long-term, spatially-explicit demographic data set with yearly censuses, we found that C. pitcheri individuals on the foredune flowered at a younger …

Effects Of Heavy Metal Uptake On Growth, Chlorophyll Content, And Calcium Oxalate Crystals In Lemna Minor (Duckweed), Olivia F. Miller

Undergraduate Honors Thesis Projects

Lemna minor L., duckweed, is a common aquatic plant used for the phytoremediation of wastewater. Heavy metal contamination poses a significant issue, and numerous studies have been conducted on the efficiency of L. minor’s hyperaccumulation ability of these metals. Calcium oxalate crystals are an important aspect of the growth and development of L. minor, but how they are influenced by the uptake of heavy metal has not been extensively studied. This study aims to determine the effects that lead, cadmium, and copper have on the formation of calcium oxalate crystals, vegetative growth, and chlorophyll a and b content on L. …

In a multivariate analysis of ten on-farm research sites, we …

Foliar Expression Of Candidate Genes Involved In Condensed Tannin Biosynthesis In White Clover (Trifolium Repens), Stephen N. Panter, J. Simmonds, A. Winkworth, A. Mouradov, G. C. Spangenberg Mar 2023

Foliar Expression Of Candidate Genes Involved In Condensed Tannin Biosynthesis In White Clover (Trifolium Repens), Stephen N. Panter, J. Simmonds, A. Winkworth, A. Mouradov, G. C. Spangenberg

IGC Proceedings (1977-2023)

Bloat disease in cattle and sheep is caused by the rapid microbial degradation of protein-rich fodder in the rumen. This leads to the production of protein foams that trap gases, causing bloat, a condition that is often fatal to livestock and costly to farmers. Condensed tannins (CTs) are phenolic polymers produced by the phenylpropanoid pathway of plants (Figure 1). CTs bind to proteins under acidic to neutral conditions, such as those present in the rumen, slowing their breakdown. A diet with a CT content of between 2% and 4% by dry weight, which is provided by some pasture legumes (e.g. …

Novel Genotypes Of The Subtropical Grass Eragrostis Curvula For The Analysis Of Apomixis (Diplospory), S. Cardone, P. Polci, J. P. Selva, M. Mecchia, S. C. Pessino, P. Voigt, G. C. Spangenberg, V. Echenique Mar 2023

Novel Genotypes Of The Subtropical Grass Eragrostis Curvula For The Analysis Of Apomixis (Diplospory), S. Cardone, P. Polci, J. P. Selva, M. Mecchia, S. C. Pessino, P. Voigt, G. C. Spangenberg, V. Echenique

IGC Proceedings (1977-2023)

Eragrostis curvula (Schrad.) Nees is a variable grass native to Southern Africa. Its several forms, known as lovegrasses, were introduced to Australia, USA and Argentina as forage perennial grasses. Apomixis is a common trait in the genus Eragrostis, with diplospory being the most frequent type. Sexual reproduction also occurs in Eragrostis, although not frequently. Since most tetraploid Eragrostis lines are apomictic, the generation of a sexual tetraploid strain is a requirement for linkage analysis of the gene(s) governing the apomictic character. Furthermore, isogenic lines of the same ploidy, reproducing alternatively by sexuality or apomixes, represent an ideal system …

Isolation And Characterisation Of Genes Encoding Malate Synthesis And Transport Determinants In The Aluminum-Tolerant Australian Weeping-Grass (Microlaena Stipoides), R. M. Polotnianka, E. Ribarev, L. Mackin, K. A. Sivakumaran, G. D. Nugent, U. P. John, G. C. Spangenberg

IGC Proceedings (1977-2023)

Acid soils cover some 40% of the Earth’s arable land where they represent a major limitation to plant production. Plant growth on acid soils is primarily limited due to aluminium (Al) solubilized by acidity into toxic Al3+ cations which will inhibit root growth resulting in poor uptake of water and nutrients. Many important pasture species lack sufficient Al tolerance within their germplasm to allow effective breeding for this character.

Development And Use Of A Tool For Automated Alignments Of Genes In The Rice Bac's Genbank Card Against Other Species, P. Barre, G. Darrieutort, J. Auzanneau, B. Julier Mar 2023

Development And Use Of A Tool For Automated Alignments Of Genes In The Rice Bac's Genbank Card Against Other Species, P. Barre, G. Darrieutort, J. Auzanneau, B. Julier

IGC Proceedings (1977-2023)

In many cases, the analysis of the genetic bases of any trait requires molecular markers and if possible co-dominant PCR-based ones. In perennial fodder species, the number of publicly available markers (microsatellites and Sequence Tagged Site (STS)) is limited. Our goal is to use sequences from model grass species, i.e. rice, wheat, maize, barley, in L. perenne in order to develop STS markers in interesting regions such as under a QTL (Quantitative Trait Loci) or around a candidate gene,. As the genome sequence of rice is now available, the objective was to use the sequences of genes included in the …

Application Of Molecular Markers Derived From Medicago Truncatula In White Clover (Trifolium Repens L.), C. Jones, M. T. Abberton Mar 2023

Application Of Molecular Markers Derived From Medicago Truncatula In White Clover (Trifolium Repens L.), C. Jones, M. T. Abberton

IGC Proceedings (1977-2023)

White clover is the major forage legume of temperate areas. Genome maps have been produced recently (Jones et al., 2003; Barrett et al., 2004) and the location of QTL for important agricultural traits reported (Abberton et al., 2004). White clover is closely related to the model legume Medicago truncatula and there is likely to be considerable benefit in applying genomic resources from model to crop. However, the extent of synteny between the species must be established. Here we present preliminary results detailing progress towards this goal.

Screening Of Perennial Grasses And A Mutant Maize Collection By Fourier-Transformed Infrared (Ftir) Spectroscopy For Improved Biofuel Traits, S. C. Thain, P. Morris, S. Hawkins, C. Morris, I. S. Donnison Mar 2023

Screening Of Perennial Grasses And A Mutant Maize Collection By Fourier-Transformed Infrared (Ftir) Spectroscopy For Improved Biofuel Traits, S. C. Thain, P. Morris, S. Hawkins, C. Morris, I. S. Donnison

IGC Proceedings (1977-2023)

Currently the potential of biomass crops, including grasses, is limited because most species have not been bred for this purpose. However traits such as lignification, phenolic cross-linking and carbohydrate accessibility, which are also important for nutritive quality in forage grasses, can affect potential biofuel quality in applications such as combustion, fast-pyrolysis or fermentation. A collection of Lolium and Festuca species known to exhibit a range of lignin, cell wall phenolic and carbohydrate concentrations have been used to test optimum characteristics for biofuel processing. This collection formed a “calibration” set for subsequent high through-put FTIR chemical screening of additional plant lines: …
