Morphology And Genetic Affinities Of A Novel Chattonella Isolate (Raphidophyceae) Isolated From Iran’S South Coast (Oman Sea), Gilan Attaran Fariman, Christopher John Stanley Bolch

The morphology and genetic affinity of a novel raphidophyte belonging to the genus Chattonella Biecheler is described for the first time from the Oman Sea along the south-east coast of Iran. While morphologically very similar to Chattonella subsalsa Biecheler, the Iranian isolates contain a distinct red eyespot. A comparison of LSU-rDNA and rDNA-ITS show that the Iranian isolate is genetically distinct from other Chattonella subsalsa strains isolated from across a wide global range and indicates that the Iranian isolates represent a distinct species related to Chattonella subsalsa.

A taxonomic account of Acanthaceae subfamily Nelsonioideae based on morphological and phylogenetic data treats five genera with 172 species: Anisosepalum (3), Elytraria (21), Nelsonia (2), Saintpauliopsis (1), and Staurogyne (145). Two other currently recognized genera, Gynocraterium and Ophiorrhiziphyllon, are included within Staurogyne, and the new combinations, Staurogyne guianensis and S. macrobotrya, are proposed. Probable apomorphic and other diagnostic macro- and micromorphological characters are discussed relative to the subfamily and genera. Characters of the inflorescence, androecium (especially pollen), and seed show important phylogenetic and diagnostic signal. A taxonomic reassessment of the Tanacetum aureum (Lam.) Greuter, M.V.Agab. & Wagenitz (Asteraceae, Anthemideae) species group was performed. This group of species is distinguished by herbaceous perennial habit, (1-)2-10(-80) disciform capitula, with compact to lax corymbs and strongly to moderately convex receptacle. A multivariate statistical analysis of 58 morphological characters using Ward cluster, principal component analyses, and molecular phylogenetic analyses of nrDNA ITS and cpDNA rpL32-trnL^{(UAG)} + ndhF-rpL32 spacer sequence datasets were conducted. Recent phylogenetic analyses proved several infrageneric units within the genus Crocus to be para- or polyphyletic. In an attempt to arrive at a system of Crocus that closely reflects species relationships, we provide here phylogenetic, morphometric, geographic, and nomenclatorial data for the species of a narrower-defined, monophyletic Crocus series Reticulati. We sequenced the ETS and ITS regions of the nuclear ribosomal DNA in 9 Reticulati and 19 outgroup species. Three chloroplast loci (trnL-F, rps16-trnQ, matK-trnK) were sequenced in the newly defined series Reticulati species and 1 outgroup. Data were analyzed with Bayesian and parsimony algorithms. Inter-simple sequence repeat (ISSR) and inter-retrotransposon amplified polymorphism (IRAP) techniques were used to investigate the genetic diversity among species of the genus Secale L. Chromosomal distribution of the AAC simple sequence repeat was examined by means of the fluorescent in situ hybridization technique. Comparative analysis revealed differences in the distribution of analyzed sequences in the genomes of Secale species. Using ISSR and IRAP data, a similarity matrix was constructed based on Dice’s coefficient, and the unweighted pair-group method with arithmetic mean clustering was used to create dendrograms. The present survey reports maximum parsimony (MP) and Bayesian phylogenetic analyses of the genus Potentilla L. in Iran using the internal transcribed spacer (ITS) of nuclear ribosomal DNA and the trnL-F intergenic region of chloroplast DNA. A total of 25 taxa from 9 sections, 2 intersectional hybrids, and 4 related genera of Iranian taxa plus 40 previously sequenced data were analyzed for both trnL-F and nrDNA ITS regions. The phylogeny of Astragalus L. section Anthylloidei DC. and its interrelationship with allies were examined. The study was conducted using nrDNA ITS and plastid rpl32-trnL_{(UAG)} sequences. Astragalus sect. Anthylloidei is nonmonophyletic, and its members are scattered across the tree in 4 well-supported clades and intermixed with members of other spiny sections. All the multispecific informal groups of the section, with the exception of A. murinus Boiss. group, are not monophyletic. Morphological character evolution was mapped on the molecular tree. Papaver bracteatum Lindl., P. orientale L., and P. pseudo-orientale (Fedde) Medw., species of section Oxytona Bernh. in the genus Papaver L., are found widely in the native flora of Turkey. They are commercially important for their high alkaloid content. Because genus Papaver section Oxytona has similar morphological characters, it is not easy to distinguish these species. We used the inter-simple sequence repeat (ISSR) molecular marker system to determine the molecular characterisation of this section. The genus Heterocapsa Stein is a relatively small armoured dinoflagellate. Because it has few differences in morphological characteristics among its species, species identification is difficult. In this study, small Heterocapsa sp. cells associated with a dense bloom of Cochlodinium polykrikoides Margalef were collected from the south-east coast of Iran. While Heterocapsa sp. was isolated, C. polykrikoides could not be isolated by single cell or serial dilution methods. Thus, a unialgal strain of Heterocapsa sp. was used for molecular analysis and species identification. In eastern Mongolia, a diploid close relative of the tetraploid (4x) crop species Allium tuberosum and its closest wild relative A. ramosum (4x) was found and characterized by karyotype analysis and with molecular marker techniques. An earlier analyses revealed A. ramosum to be sister of the crop but excluded it as its progenitor. At that time a putative diploid cytotype of A. ramosum was hypothesized as a potential progenitor taxon of the tetraploids. The phylogenetic relationships among some bifurcate hairy sections of Astragalus L. were reconstructed using 38 vegetative and reproductive morphological characters. A data matrix comprising 36 ingroups and 2 outgroups was analysed using the maximum parsimony method. The present results revealed that sections Onobrychoidei DC. and Ornithopodium Bunge are interrelated taxa and form a single monophyletic group. The current status of section Dissitiflori DC. is a nonmonophyletic group. Its members form several subclades and unresolved branches across the tree. The genus Malcolmia W.T.Aiton is taxonomically problematic, and some of its species have recently been transferred to the genera Strigosella Boiss. and Zuvanda Dvorák. In the present study the phylogeny and the biogeography of the genus Lilium L. section Liriotypus Asch. et Graebn. were investigated and divergence times for the section Liriotypus were calculated. The study group covers Lilium species from Europe, the Italian and Balkan peninsulas, Anatolia, and the Caucasus. Plastid DNA sequence data (the trnC-petN intergenic spacer and petN gene) and nuclear DNA ITS sequence data were used to infer the phylogenetic history of the section Liriotypus. In Siberia, the bluegrass genus (Poa) comprises 43 species and 61 subspecies in 12 sections. Diverse modes of speciation, including polyploidy and hybridization, have led to reticulate evolution and adaptive radiation. Cladistic methods that ignore hybridization and reticulate evolution may not be appropriate for morphological data. The number of morphological characters suitable for bluegrass analysis is limited, a majority does not have clear adaptive significance, and the character states cannot be readily polarized, rendering phylogenetic reconstruction very difficult in this group. Other methods of estimating phylogenetic relationships should be used to test hypotheses about relationships and hybridization. Biochemical …

Twenty-three populations belonging to 10 wild Primula L. (Primulaceae) taxa of Turkey, some of which are morphologically quite similar, were investigated based on nrDNA ITS regions. The plant materials were collected from different geographical areas of Anatolia in vegetation periods of 2005 and 2006. Total genomic DNAs were isolated from the healthy leaves of each population. The entire ITS regions of the populations were amplified by universal primers with the aid of a polymerase chain reaction (PCR), and then the PCR products were sequenced. The genus Cyclotrichium, a member of the tribe Mentheae subtribe Menthinae (Lamiaceae, Nepetoideae), was analysed with respect to morphological revision, phylogenetic analysis, and cytogenetic properties. All species of the genus were investigated for morphological characters and ITS (internal transcribed spacers) of nrDNA sequence comparison (except C. hausknechtii for ITS). Six members of the genus were also analysed for chromosome numbers. The combined results strongly suggested that Cyclotrichium is a separate genus in Nepetoideae with distinct morphological, phylogenetic, and cytogenetic characteristics. To help in the clarification of the taxonomic status of Elymus L. and related genera Agropyron Gaertn., Leymus Hochst., Hordelymus (Jess.) Harz, and Brachypodium P.Beauv., the sequence diversity in the internal transcribed spacer (ITS) region of nuclear ribosomal DNA was studied. ITS data of 64 Triticeae accessions including 40 Elymus, 12 Agropyron, 7 Leymus, and 4 Hordelymus, and 1 Brachypodium as an out-group were analysed. To understand evolutionary patterns and processes that account for anatomical diversity in relation to ecology and life form diversity, anatomy of storage roots and stems of the genus Adenia (Passifloraceae) were analyzed using an explicit phylogenetic context. Over 65,000 measurements are reported for 47 quantitative and qualitative traits from 58 species in the genus. Vestiges of lianous ancestry were apparent throughout the group, as treelets and lianous taxa alike share relatively short, often wide, vessel elements with simple, transverse perforation plates, and alternate lateral wall pitting; fibriform vessel elements, tracheids associated with vessels, and libriform fibers as additional tracheary elements; …

In this investigation, our main objective was to estimate evolutionary relationships in tribe Cariceae using DNA sequence data from two spacer regions in nuclear ribosomal genes (ITS and ETS-1f) combined with noncoding chloroplast DNA (trnL intron, trnL–trnF intergenic spacer, and trnE–trnD intergenic spacers). Paspalum (Poaceae: Panicoideae: Paniceae) includes 330 species distributed mainly in tropical and subtropical regions of America. Due to the large number of species and convergence in many characters, an adequate infrageneric classification is still needed. Since the Monocots II meeting in 1998, significant new data have been published that enhance our systematic knowledge of Cyperaceae. Phylogenetic studies in the family have also progressed steadily. For this study, a parsimony analysis was carried out using all rbcL sequences currently available for Cyperaceae, including data for two new genera. One of the four subfamilies (Caricoideae) and seven of the 14 tribes (Bisboeckelereae, Cariceae, Cryptangieae, Dulichieae, Eleocharideae, Sclerieae, Trilepideae) are monophyletic. Subfamily Mapanioideae and tribe Chrysitricheae are monophyletic if, as the evidence suggests, Hellmuthia is considered a member of Cypereae. Some other features of our analysis include: …

Phylogenetic relationships among 85 species representing 35 genera in the grass tribe Andropogoneae were estimated from maximum parsimony and Bayesian analyses of nuclear ITS and chloroplast trnL–F DNA sequences. Ten of the 11 subtribes recognized by Clayton and Renvoize (1986) were sampled. Independent analyses of ITS and trnL–F yielded mostly congruent, though not well resolved, topologies. Arundinella is sister to Andropogoneae in the trnL–F phylogeny and is nested within the tribe in the ITS and combined data trees. Triticeae are renowned for their complicated taxonomy, but their phylogeny is equally intricate and perplexing, and remains largely unresolved. Based on morphology and nucleotide sequences from two plastid genes (rbcL, rpoA), one mitochondrial gene (coxII), and two single-copy nuclear genes (DMC1, EF-G), the most comprehensive hypothesis (both with respect to taxa and data points) of the phylogeny of diploid Triticeae to date is presented. The incongruence length difference tests clearly indicate that the four logical data partitions (morphology and the three genome compartments) are mutually incongruent, except the mitochondrial and nuclear sequences. …

The most robust previously published phylogeny for the overall structure of the grass family (Poaceae) shows three early diverging lineages and two major derived clades, the BEP clade and the PACCAD clade (Grass Phylogeny Working Group 2001). A few key taxa were incompletely sampled, however, and support for the BEP clade was moderate at best and relationships among the major lineages within the PACCAD clade remained unresolved. In addition, recent studies indicated that the sister group to Poaceae may be Joinvilleaceae and/or Ecdeiocoleaceae, the latter of which were not previously sampled. We investigated the phylogenetic relationships of the endemic New Zealand (NZ) species of Festuca (Poaceae, Pooideae) by assessing sequence variation from the nuclear internal transcribed spacers (ITS) and a chloroplast intergenic spacer (trnL–trnF) and by measuring DNA content using flow cytometry. The ITS and trnL–trnF data sets were congruent in showing that the NZ species of Festuca have two origins. One group, containing F. coxii, F. luciarum, F. multinodis, and F. ultramafica, is closely related to Festuca sect. Aulaxyper. The other group includes a clade of five endemic …

A reconstruction of the phylogeny of Bromeliaceae based on sequence data from three noncoding chloroplast DNA markers (trnL intron, trnT–trnL, and trnT–trnF intergenic spacer [IGS]) is presented, including 26 genera and 33 species. Relationships of Bromelioideae and phylogeny within this subfamily were analyzed in more detail on the basis of two of these markers (trnL intron and trnL–trnF IGS) using a set of 37 genera/74 species of Bromeliaceae, including 28 genera/60 species of Bromelioideae. Within the tribe Abildgaardieae, the relationships between Fimbristylis and its relatives have not been certain, and the limits of Fimbristylis have been unclear, with Bulbostylis and Abildgaardia variously combined with it and each other. The relationships and limits of tribes Abildgaardieae and Arthrostylideae and their genera were evaluated across 49 representative species using parsimony and maximum likelihood analyses of ITS (nuclear ribosomal) and trnL–F (plastid) DNA sequence data separately and combined. Bambuseae (woody bamboos), one of two tribes recognized within Bambusoideae (true bamboos), comprise over 90% of the diversity of the subfamily, yet monophyly of the tribe is generally only moderately supported, and phylogenetic relationships within the tribe are poorly understood. In addition, there appears to be some level of conflict between morphological and molecular data within the tribe. The phylogeny of Chloridoideae (Gramineae) was inferred from parsimony analyses of DNA sequences from two genomes—the chloroplast trnL intron, trnL 3' exon, and trnL–F intergenic spacer, and the nuclear ribosomal internal transcribed spacer region (ITS1 + 5.8S + ITS2). Eighty species representing 66 chloridoid genera were sampled, including all but four of the native New World genera. Analyses of the individual and combined data sets were performed. Great advances have been made in our understanding of the phylogeny and classification of Agavaceae in the last 20 years. In older systems Agavaceae were paraphyletic due to overemphasis of ovary position or habit. Discovery of a unique bimodal karyotype in Agave and Yucca eventually led to a reexamination of concepts and relationships in all the lilioid monocots, which continues to the present day. Developments in cytogenetics, microscopy, phylogenetic systematics, and most recently DNA technology have led to remarkable new insights.
