Arkansas Agricultural Experiment Station Research Series
Wheat cultivar performance tests are conducted each year in Arkansas by the Arkansas Agricultural Experiment Station, Department of Crop, Soil and Environmental Sciences. The tests provide information to companies developing cultivars and/or marketing seed within the state and aid the Arkansas Cooperative Extension Service in formulating cultivar recommendations for small-grain producers.

Reduced population size can result in the loss of genetic diversity, which in turn can make organisms more susceptible to environmental challenges. In Iowa less than one percent of the original tallgrass prairie remains, isolated in a few remnant prairies. We are undertaking a long term project to assess the genetic variability in native prairie plants on remnant prairies in Northwest Iowa. We are testing six microsatellite sequences for butterfly milkweed (Asclepias tuberosa) from the Steele Prairie State Preserve, Broken Kettle Grasslands, a private prairie near Cherokee IA, a prairie restoration project on the Dordt College campus and populations from …

Heat stress causes major losses to cotton seed and lint yield. Introduction of heat stress tolerance to Arkansas cotton varieties is highly desirable. However, very little is known about the molecular basis of heat stress tolerance in cotton. The present study attempted to identify heat stress tolerance genes in two heat-tolerant cotton cultivars, VH260 and MNH456, originating from Pakistan. Towards this, the expression profile of the cotton orthologs of sevenArabidopsisheat stress tolerance genes was studied in these two cultivars, and compared with the two heat-susceptible cotton cultivars, ST213 and ST4288, originating from Mississippi Delta region. The shoot apical meristem of grasses produces the primary branches of the inflorescence, controlling inflorescence architecture and hence seed production. Whereas leaves are produced in a distichous pattern, with the primordia separated from each other by an angle of 180◦, inflorescence branches are produced in a spiral in most species. The morphology and developmental genetics of the shift in phyllotaxis have been studied extensively in maize and rice. However, in wheat, Brachypodium, and oats, all in the grass subfamily Pooideae, the change in phyllotaxis does not occur; primary inflorescence branches are produced distichously. Conserved non-coding sequences (CNS) are islands of non-coding sequence that, like protein coding exons, show less divergence in sequence between related species than functionless DNA. Several CNSs have been demonstrated experimentally to function as cis-regulatory regions. However, the specific functions of most CNSs remain unknown. Previous searches for CNS in plants have either anchored on exons and only identified nearby sequences or required years of painstaking manual annotation. Here we present an open source tool that can accurately identify CNSs between any two related species with sequenced genomes, including both those immediately adjacent to exons and distal sequences separated by …

Cultivated sorghum [Sorghum bicolor (L.) Moench subsp. bicolor] can interbreed with a feral weedy relative shattercane [S. bicolor nothosubsp. drummondii (Steud.) de Wet ex Davidse]. Traits introduced from cultivated sorghum could contribute to the invasiveness of a shattercane population. An experiment was conducted to determine the potential for pollenmediated gene flow from grain sorghum to shattercane. Shattercane with juicy midrib (dd) was planted in soybean [Glycine max (L.) Merr.] fields during 2 yr in concentric arcs at varying distances from a 0.39 ha sorghum pollen source with dry midrib (DD). The arcs …

The parental species and hybrid swarm of native Ulmus rubra Muhl. and the introduced, naturalized and weedy U. pumila L. were investigated in a 65-km transect in Lancaster, Saunders, and Butler counties in Nebraska. Thirty-two trees of U. rubra, 32 of U. pumila, and 50 of the hybrid swarm were sampled for leaves and buds and subsampled for flowers and fruits. Leaves were measured for petiole length, blade length, width, primary and secondary teeth per cm, number of secondary teeth per primary tooth, and texture. Like music, one of the hallmarks of biodiversity is theme and variation. Redbuds—species of the genus Cercis— from around the world illustrate this analogy well. Because all redbud species conform to a certain morphological theme, anyone familiar with one particular species of redbud should be able to recognize without hesitation any other redbud species as a member of the genus Cercis. In brief, the redbud theme consists of broad, basally-lobed, leaves with pulvinar petiole thickenings, and pea-like red-purple (rarely white) flowers that may arise on small twigs or main trunks. Historically, the Republican River Valley in Harlan County, Nebraska, has been extensively explored, but the county flora has continually been under-represented and has been found to lack records for plants that are considered common. During the 2009 growing season, a taxonomic study and comparison of 3 different sites in Harlan County was made. Site #1 was pastureland and Sites #2 and #3 were both riparian, located on the north side of the Republican River. Collections were pressed, dried, identified, and deposited in the University of Nebraska at Kearney Herbarium (NEBK). Deerfield Reservoir in the Black Hills of South Dakota and its tributary system are managed as hatchery supplemented rainbow trout (Oncorhynchus mykiss) fisheries. Three genetically unique strains of rainbow trout (Shasta, Erwin and McConaughy) are stocked into the system. Recently, juvenile rainbow trout of unknown origin were collected in the tributary system above Deerfield Reservoir, indicating potential natural reproduction. Understanding the genetic origins and ability of these rainbow trout to contribute to the fishery is essential in determining the proper management strategy for these waters. Western burrowing owl (Athene cunicularia hypugaea) populations have declined across much of western North America, particularly at the northern and eastern edges of the species’ breeding range (Martell et al. 2001, Murphy et al. 2001, Shyry et al. 2001, Skeel et al. 2001, Klute et al. 2003). In South Dakota, the burrowing owl is a summer resident that historically was relatively common throughout the state, but its range has decreased in recent decades, especially in the eastern half of the state (Whitney et al. 1978, South Dakota Ornithologists’ Union [SDOU] 1991, Peterson 1995). Red-tailed hawks (Buteo jamaicensis) and great-horned owls (Bubo virginianus) can influence the nesting behavior of each other through direct competition for nesting sites. For instance, owls begin nesting before hawks and, thus, can use nests from the previous year and prevent hawks from occupying those sites (Orians and Kuhl- man 1956, Gilmer et al. 1983, Minor et al. 1993). However, both species sometimes kill the nestlings of the other (Craig- head and Craighead 1956, Bosakowski et al. 1989).

Discovery Of Bald Cypress Fossil Leaves At The Gray Fossil Site, Tennessee And Their Ecological Significance., Sara Brandon

This study focuses on fossil Taxodium leaves found at the Gray Fossil Site in northeastern Tennessee where many 7-4.5 million year old plants and animals have been recovered. Identification of the leaves is based on comparison of leaf morphology and confirmed by leaf anatomy. The ecological implications of the fossil are discussed to understand the paleoecology and paleoclimate at the Gray site. It is concluded that the fossil plant along with many other plants lived by a large sinkhole lake under a little warmer-than-today’s climate condition.

American skullcap (Scutellaria lateriflora), a perennial herb native to America, is rich in specialized metabolites that have shown various biological activities. In particular, the flavonoids wogonin, baicalein and baicalin have proven anticancer properties in vitro. However, the regulatory mechanisms and biosynthetic steps leading to these specialized metabolites have not been elucidated. To address this issue, hairy root cultures of S. lateriflora expressing the flavonoid-specific transcription factor AtMYB12 from Arabidopsis thaliana were developed using Agrobacterium rhizogenes ATCC 15834 containing a binary vector harboring the AtMYB12 cDNA under the control of the SuperP:TEV expression system. The release of fatty acids from membrane lipids has been implicated in various metabolic and physiological processes, but in many cases, the enzymes involved and their functions in plants remain unclear. Patatin-related phospholipase As (pPLAs) constitute a major family of acyl-hydrolyzing enzymes in plants. Here, we show that pPLAIIId promotes the production of triacylglycerols with 20- and 22-carbon fatty acids in Arabidopsis (Arabidopsis thaliana). Of the four pPLAIIIs (a, b, g, d), only pPLAIIId gene knockout results in a decrease in seed oil content, and pPLAIIId is most highly expressed in developing embryos. The use of roses as a symbol of emotion (specifically love) and as a result, a model of artificial selection, is rarely questioned. These plants share a rich history with multiple humanities around the world and have been a subject of genetic interest to see exactly what genes are being expressed that signal those beautiful desirable traits.
