Favorable climate and soils for rainfed crop production, together with a relatively low population density,results in 70–90% of Argentina grain production being exported. Legumes form symbiotic and pathogenic interactions with microbes. Understanding the molecular mechanisms underlying the legume-microbe interactions would help us to improve crop production in a sustainable manner. This thesis covers two independent research projects. The first project was to study the role of alternative splicing in RCT1-mediated disease resistance. RCT1 is a TIR-NBS-LRR-type plant resistance (R) gene in Medicago truncatula that confers broad-spectrum resistance to Colletotrichum trifolii, a fungal pathogen that causes anthracnose disease in Medicago. RCT1 undergoes alternative splicing at both coding and 3'-untranslated regions, thereby producing multiple transcript variants in its expression profile. …

In an effort to maximize yields, many soybean growers have begun moving to intensive, input-based soybean management systems. However, limited reliable information exists about the effect of these inputs on soybean yield. The purpose of this study was to evaluate the effect of individual inputs and combinations of inputs as part of high-yield management systems on soybean seed yield and to determine the effect of one of these inputs, lactofen, on soybean physiology. Gene structural variation (SV) has recently emerged as a key genetic mechanism underlying several important phenotypic traits in crop species. We screened a panel of 41 soybean (Glycine max) accessions serving as parents in a soybean nested association mapping population for deletions and duplications in more than 53,000 gene models. Array hybridization and whole genome resequencing methods were used as complementary technologies to identify SV in 1528 genes, or approximately 2.8%, of the soybean gene models. Although SV occurs throughout the genome, SV enrichment was noted in families of biotic defense response genes. Among accessions, SV was nearly eightfold less …

With the increase in saline soils worldwide, understanding the mechanisms for salt tolerance in plants is important to reduce yield loss due to salt stress. Soybean, Glycine max (L.) Merr., genotypes differ in chloride uptake with genotypes that take up chloride into foliar tissues tending to be salt-sensitive whereas those that partially exclude chloride from the leaves are more salt-tolerant. Background:

The homeodomain leucine zipper (HD-Zip) transcription factor family is one of the largest plant specific superfamilies, and includes genes with roles in modulation of plant growth and response to environmental stresses. Many HD-Zip genes are characterized in Arabidopsis (Arabidopsis thaliana), and members of the family are being investigated for abiotic stress responses in rice (Oryza sativa), maize (Zea mays), poplar (Populus trichocarpa) and cucumber (Cucmis sativus). Findings in these species suggest HD-Zip genes as high priority candidates for crop improvement.

Seed carbohydrate content is an important aspect in breeding for food-grade soybeans commercialized in the soyfood market. Sucrose and stachyose are the primary carbohydrates in soybean seed. Sucrose affects the quality and taste of various soyfoods such as tofu, soymilk, and natto; however, consumption of soy-based products with high stachyose concentrations can cause diarrhea and flatulence. A mutant line (V99-5089) with high-sucrose and low-stachyose has been identified. Both rainfed and irrigated soybean production are important in Nebraska (western US Corn Belt), accounting for a respective 48 and 52% of the state’s soybean production of 7 Mt on a respective 55 and 45% share of the state soybean area of 1.9 Mha. To date, no assessment of factors that may account for regional and inter-annual variation in yield and irrigation amount has been performed. Resistance to SCN is complex, polygenic, race and cultivar specific, and it is controlled by several quantitative trait loci (QTL). Our objective was to identify and map QTL for SCN resistance to races 3 (HG Type 0) and 5 (HG Type 2.5.7) using a high density SNP-based genetic linkage map based on the PI438489B by ‘Hamilton’ (PIxH, n=50) recombinant inbred line population. In soybean, nitrogen fixation is more sensitive to drought than other physiological processes like photosynthesis. The sensitivity of nitrogen fixation to drought has been associated with high shoot concentrations of ureide and nitrogen under well-watered conditions. Previous research by Hwang et al. (2013) detected quantitative trait loci (QTLs) in a KS4895 by Jackson population associated with shoot ureide and nitrogen concentrations. The present research evaluated the use of these QTLs in selecting genotypes with drought tolerant nitrogen fixation. Growers are constantly seeking ways to improve yield in soybean [Glycine max (L.) Merr.]. There has been much interest in the use of selected alternative practices to maximize soybean yield. In the Midsouth, soybean is often grown in close proximity to rice or in rotation with rice. Herbicides used in rice can injure soybean via drift or carryover. Consequently, field trials were conducted to determine the response of soybean (cv. AG 4703) to imazosulfuron drift and carryover (at Fayetteville, Marianna, Keiser and Pine Tree, AR) from rice. Sucrose is a desirable sugar in soybean seed that affects the quality and taste of soyfoods, while stachyose is a non-digestible sugar that induces flatulence in non-ruminant animals. Therefore, soybean cultivars with high sucrose and/or low stachyose would be valuable for soyfood and meal markets. The objectives of this study were to identify quantitative trait loci (QTL) or genes associated with seed sucrose and stachyose content using simple sequence repeat (SSR) or single nucleotide polymorphism (SNP) markers. A Carlavirus transmitted by Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae) is an important disease of soybean nurseries in Puerto Rico causing substantial germplasm losses. Insecticide bioassay experiments were conducted at Dow AgroSciences (DAS) Research Station, Santa Isabel, Puerto Rico, either by spraying insecticides on B. tabaci infested soybean leaves or introducing B. tabaci adults onto insecticide-sprayed soybean leaves. Moreover, host plants were surveyed to detect the virus in overwintering hosts that serve as a source of inoculums. The soybean genome duplicated ~14 and 45 million years ago and has many paralogous genes, including those in urease activation (emplacement of Ni and CO2 in the active site). Activation requires the UreD and UreF proteins, each encoded by two paralogues. UreG, a third essential activation protein, is encoded by the single-copy Eu3, and eu3 mutants lack activity of both urease isozymes. eu2 has the same urease-negative phenotype, consistent with Eu2 being a single-copy gene, possibly encoding a Ni carrier. Coupling Of Carbon Dioxide And Water Vapor Exchanges Of Irrigated And Rainfed Maize–Soybean Cropping Systems And Water Productivity, Andrew E. Suyker, Shashi B. Verma

Continuous measurements of CO2 and water vapor exchanges made in three cropping systems (irrigated continuous maize, irrigated maize–soybean rotation, and rainfed maize–soybean rotation) in eastern Nebraska, USA during 6 years are discussed. Close coupling between seasonal distributions of gross primary production (GPP) and evapotranspiration (ET) were observed in each growing season. Mean growing season totals of GPP in irrigated maize and soybean were 1738 ± 114 and 996 ± 69 g C m−2, respectively (±standard deviation). Corresponding mean values of growing season ET totals were 545 ± 27 and 454 ± 23 mm, respectively. Irrigation affected GPP …
