Neuroendocrine Regulation Of The Luteinizing Hormone Surge And Puberty Onset In Sheep, Max J. Griesgraber

Gonadotropin-releasing hormone (GnRH), luteinizing hormone (LH), and follicle stimulating hormone (FSH) are essential for the reproduction of all mammalian species, including both sheep and humans. GnRH is secreted by hypothalamic neurons into the hypophyseal portal vasculature in a pulsatile manner, which then acts within the anterior pituitary to elicit the release of LH and FSH into wider systemic circulation. In females LH and FSH act on the ovaries to promote estradiol (E2) production, follicular development, and under certain conditions in regard to LH, ovulation. Diet-induced obesity and adipose tissue (AT) inflammation that ensues sets the stage for systemic metabolic inflammation, disrupted insulin signaling, and eventual type 2 diabetes (T2D) development. At the center of this pathology lies a disruption in endogenous inflammation resolution pathways characterized by an imbalance in lipid mediator production (greater abundance of pro-inflammatory to pro-resolving lipids). Additionally, obese states are characterized by adrenergic deficiency, especially in response to physical activity. Inflammatory processes have been experimentally mitigated by either the pharmacological inhibition of pro-inflammatory signals (i.e., Leukotriene B4 (LTB4) and its receptor BLT1, which establishes a potent chemotactic stimulus …

Endocrine disrupting compounds (EDCs) are chemicals that interfere with hormone function and are increasingly detected in aquatic environments, where they elicit adverse effects from exposed organisms. The toxicological effects of EDCs can be described as either activational (reversible) or organizational (irreversible), where the latter are associated with adverse outcomes in reproductive performance of adult fish. However, few studies have investigated the organizational impacts of anti-estrogenic or “defeminizing” EDCs, e.g. agrichemicals or pharmacological agents, in an environmentally-relevant or “sentinel” species. Salt inducible kinase 1 (SIK1) has been considered a stress-inducible kinase since it was first cloned in 1999. Continued efforts since this time have been dedicated to characterizing the structure and function of SIK1. Such research has laid the ground work for our understanding of SIK1 action and regulation in tissue and stimuli dependent manners. The fundamental findings of this dissertation continue in this tradition and include investigations of SIK1 regulatory mechanisms in skeletal muscle cells, the cellular and physiological effects of SIK1 loss of function in vitro and in vivo, and intracellular metabolic and mitochondrial regulation by this …

Adrenocortical (interrenal) tissue is composed of steroidogenic cells that produce corticosteroids involved in the stress response and hydromineral balance. Previous research characterizing the elasmobranch interrenal suggests that the number of interrenal bodies that produce the single primary corticosteroid, 1a-hydroxycorticosterone (1a-OHB), varies among species. However, potential species-specific differences in the amount of interrenal tissue and major steroid products produced have been understudied. The hypothalamic-pituitary-adrenal (HPA) axis releases glucocorticoids, including corticosterone (CORT), in response to stress. CORT then negatively feeds back to inhibit its own production by binding to glucocorticoid receptors (GRs) in the hypothalamus and pituitary gland. The HPA axis is subject to “programming” by abnormal stimuli during early development, which may permanently alter how the HPA axis responds to stress. These altered responses have been linked to an increased risk for human psychiatric and metabolic disorders in later life, but the mechanism by which this happens is not fully understood. The objective of this thesis was to evaluate the influence of steroids and gonadotropins on reproduction in beef cattle. In experiment 1, beef heifers were used to determine the influence of growth-promoting implants on growth, reproductive development, estrous behavior, and pregnancy rate. Heifers were assigned to 1 of 4 implant treatment groups: control (CON); trenbolone acetate (TBA); trenbolone acetate plus estradiol (TBA+E2) or zeranol (ZER). Heifers were implanted once, A.I. and exposed to bull during this experiment. Body weight, BCS, HH, RTS, estrous behavior and pregnancy data were collected throughout this experiment.
