Assessment Of Bovine Vascular Serotonin Receptor Populations And Transport Of Ergot Alkaloids In The Small Intestine, Miriam A. Snider

Prior work using a contractility bioassay determined that the serotonin (5-HT) receptor subtype 5-HT2A is present in bovine lateral saphenous veins and plays a role in ergot alkaloid-induced vascular contraction in steers grazing endophyte-infected (Epichloë coenophiala) tall fescue (Lolium arundinaceum). A study was conducted to determine what 5-HT receptors are involved in vasoconstriction of bovine gut vasculature. The findings of this study indicate that 5-HT2A is present and may play a role in ergot alkaloid induced vasoconstriction. Oxidative stress may play a role in the phenotypic expression of feed efficiency (FE). The transcription factor NFE2L2 (nuclear factor erythroid-derived 2-like 2) coordinates antioxidant response to oxidative stress and its activity is tightly regulated in part by KEAP1 (Kelch like-ECH protein 1) and the E3 ligase CUL3 (Cullin3). In fishes, hyperammonemia may occur following feeding or exposure to abnormally high concentrations of environmental ammonia due to sewage effluents, agricultural run-off and in crowded aquaculture pens. Increased internal ammonia can result in hyperactivity, convulsions, coma and death. In mammals, it is also associated with potentially fatal brain edema, in which the accumulation of intracellular water results in swelling, increased intracranical pressure and herniation leading to death. Recently it was shown that rainbow trout (Oncorhynchus mykiss) and goldfish (Carassius auratus) experience brain swelling following exposure to high external ammonia (HEA). Fishes living in freshwater need to actively compensate for the diffusive loss of ions and osmotic gain of water. The gill is the primary organ of ion regulation and contains an array of ion transport proteins to help maintain homeostasis. Two of the more well studied ion pumps are the Na+/K+-ATPase (NKA) and vacuolar type proton ATPase (V-ATPase). This thesis focuses on another ion pump known as the non-gastric H+/K+-ATPase (ngHKA). Muscle growth in young horses is characterized by an increase in muscle cross-sectional area, which can be accomplished through the activation and differentiation of satellite cells. Satellite cells can be stimulated or inhibited in response to different cytokines and growth factors and are key mediators of muscle hypertrophy and regeneration. The aim of this study was to investigate the growth of the longissimus dorsi (LD) muscle in horses under 5 years of age and to obtain preliminary data on satellite cell behavior in foals. The pesticide, 3-trifluoromethyl-4-nitrophenol (TFM), is widely used in the Great Lakes to control invasive sea lampreys (Petromyzon marinus) populations, but much about its sub-lethal effects remains unknown. A better understanding of its toxicity is needed to improve TFM effectiveness and to protect non-target organisms from its potential adverse effects. Contaminated sediments in marine environments have been shown to be good indicators of ecological risk and a means to assess anthropogenic impacts on marine habitats and the animals that inhabit them (Long et al. 1995, Rattner 2009). Estuarine sediments are especially complex media with regard to physical, chemical, and biological characteristics that trap, store, modify and sometimes release contaminants to the biota (Long et al. 1995). Locomotion in mammals relies on a central pattern-generating circuitry of spinal interneurons established during development that coordinates limb movement. These networks produce left–right alternation of limbs as well as coordinated activation of flexor and extensor muscles. Here we show that a premature stop codon in the DMRT3 gene has a major effect on the pattern of locomotion in horses. The mutation is permissive for the ability to perform alternate gaits and has a favorable effect on harness racing performance. Purified brush border membrane vesicles (BBMV) were isolated to characterize primary cellular transport mechanisms for white shrimp. The ultimate goal is to determine the effective components of a shrimp’s diet, thereby enhancing growth, as well as nutrient content. Juvenile shrimp are dependent on plant material as a food source. Potassium is a key component of plants, thus it may play a role in nutrient transport. In addition, divalent metals have been shown to act as co-transporters in several other organisms, thus they may serve as a transport mechanism for shrimp.

Salmonids, such as Chinook salmon (Oncorhynchus tshawytscha) and steelhead (O. mykiss), are a staple economic, recreational, tribal, and environmental resource, yet many populations are unsustainable. This study was part of a broad scale effort to monitor the impact of downstream migration obstacles on juvenile salmonid health and survival, which is an essential step towards increasing Smolt-to-Adult Return ratios (SARs). We characterized 24 polymorphic tetranucleotide microsatellite loci for delta smelt (Hypomesus transpacificus) endemic to the San Francisco Bay Estuary, California, USA. Screening of samples (n = 30) yielded two to 26 alleles per locus with observed levels of heterozygosity ranging from 0.17 to 1.0. Only one locus deviated from Hardy–Weinberg equilibrium, suggesting these individuals originate from a single panmictic population. Linkage disequilibrium was found in two pairs of loci after excluding the locus out of Hardy–Weinberg equilibrium. Four microsatellite-enriched DNA libraries yielded 35 microsatellite loci from 100 primer pairs designed for Pacific lion-paw scallop, Nodipecten subnodosus. The number of alleles ranged from four to 28. Three of the 35 loci were not in Hardy–Weinberg equilibrium and linkage disequilibrium was found for one pair of loci. These microsatellites will be used to analyze the population structure of the species in Mexico’s Baja Peninsula to propose management strategies for scallop aquaculture development. Twenty-six primer pairs cross-amplified in Nodipecten nodosus, whereas none (Argopecten ventricosus) or few cross-amplified in the Argopecten species.

1,25-dihydroxyvitamin D3 [1,25(OH)2D3] has been shown to promote phosphate transport rapidly in the perfused duodenal loop, relative to controls, reaching treated/av basal at T = 40 min = 1.82 ± 0.42 and 1.11 ± 0.21, respectively.

By using isolated chick enterocytes, studies were undertaken to determine whether 1,25(OH)2D3 has a direct effect on isolated intestinal cells that is manifested by either enhanced uptake or extrusion of phosphate.

The decapod crustacean with a relatively well-defined visual system have contributed significantly to our understanding of visual neuron morphology and physiology. The general morphology of the optic lobes of several decapods were initially described by Parker (1897) and Hanstrom (1924). As pointed out by Jackson (1971) and Patton (1972), the key to understanding mechanisms of chromosomal evolution lies in studies of chromosomal variation within populations that can hybridize. Hybrids between cytotypes contain the answer to problems of meiotic pairing and reduced fertility, as well as information concerning the nature of zones of contact. However, before detailed studies can be initiated, chromosomal races and hybrid zones must be located. Few chromosomal races have been described for bats (Baker, 1970a).
