Heart rate deflection point (HRDP), identified as the second breakpoint in the "intensity / heart rate" relationship, is indicative of the anaerobic threshold (AT). This point can be determined via bi-segmental linear regression (2SEG) or through use of the maximal distance model (D-max); however, the relationship between these methods has yet to be investigated 2. This dissertation attempts to discover the mechanisms between cardiovascular and respiratory motor control post spinal cord injury (SCI): in normal, non-injured (NI) persons, cardiovascular regulation is dependent upon respiration, but there is nothing that suggests the mechanism for this relationship post-SCI. Thus we hoped to evaluate various aspects of cardiovascular regulation to further illustrate how this relationship is changed or unchanged by SCI. Background: Hyperhomocysteinemia (HHcy) is heavily implicated in diabetes and cardiovascular disease. HHcy is known for causing inflammation and vascular remodeling, particularly through production of reactive oxygen species (ROS) and matrix metalloproteinase-9 (MMP-9). Although its impact on skeletal muscle is rather unclear, HHcy is known to cause skeletal muscle weakness and functional impairment. The impact of HHcy on inflammation makes it seem likely that HHcy causes skeletal muscle fibrosis through induction of inflammatory factors and destructive macrophages. According to the CDC, the most common type of heart disease is coronary artery disease, which frequently leads to myocardial infarction (MI). Therapeutic approaches to lessen the resulting cardiovascular injury associated with MI are limited. Recently, the management paradigm for cardiac injury has entered the molecular era and microRNAs (miRNAs) have been shown to act as negative regulators of gene expression by inhibiting mRNA translation and/or stimulating mRNA degradation. A single miRNA can modulate physiological or disease phenotypes by regulating whole functional systems. Controlled frequency breath (CFB) holding is a swim training modality that involves holding one’s breath for ~12 strokes before taking another breath. We looked to examine the effects of CFB training on pulmonary diffusing capacity for nitric oxide (DLNO) and carbon monoxide (DLCO). Elite swimmers (n = 25) were divided into either the CFB or a group that breathed regularly, every ~3rd stroke. The training intervention included 16 sessions of 12 x 50-m repetitions with either breathing pattern. Approximately 60% of the males and ~20% of the females were above the upper limits of normal for diffusing capacity at baseline. …

The mammalian/mechanistic target of rapamycin complex 1 (mTORC1) signaling pathway appears to be the primary regulator of protein synthesis and growth. While resistance exercise paradigms are often divided into hypertrophy (HYP) and strength (STR) protocols, it is unknown whether these protocols differentially stimulate mTORC1 signaling. The purpose of this study was to examine mTORC1 signaling in conjunction with circulating hormone concentrations following a typical lower-body HYP and STR resistance exercise protocol. The successful regeneration of muscle tissue is dependent upon the infiltration of phagocytic CD14++CD16- monocytes that support the proliferation and differentiation of myogenic precursor cells. Physiologically, the magnitude of the cellular response following resistance exercise is dictated by the level of receptor expression on the plasma membrane of the monocyte, as well as the secretion of their cognate ligands from tissue resident cells. Paddlefish often frequent oxbows and backwater habitats between periods of migration. Speculation as to the use and energetic savings associated with this seasonal residence has not been substantiated. This study used radio telemetry to track the seasonal movements and swim tunnel respirometry to estimate metabolic rates and swimming efficiency of paddlefish within an oxbow located just north of Clarksdale, Mississippi (Moon Lake). Paddlefish overwintered in the deepest portions of Moon Lake with little movement into shallower areas. Paddlefish swimming performance was measured at four swimming speeds (76.04, 91.27, 106.50, and 121.72 cm/s) to determine the cost of locomotion over season …

To reconstruct the 3D space and objects in the visual system from the 2D retinal images, surface representation must be a critical intermediate stage in the visual processing stream. The type IIa sodium-phosphate cotransporter (Npt2a) is expressed in the apical membrane of the renal proximal tubule and is responsible for the reabsorption of the majority of the filtered load of phosphate. Parathyroid hormone (PTH) is secreted by the parathyroid gland in response to a decrease in serum calcium or an increase in serum phosphorus, and acutely induces phosphaturia through the rapid stimulation of endocytosis of Npt2a and its subsequent lysosomal degradation. Chronic PTH stimulation leads to sustained phosphaturia, but the mechanisms for the chronic regulation of Npt2a by PTH remain unclear. We hypothesize that PTH decreases Npt2a mRNA levels …

Detrusor-sphincter dyssynergia after chronic spinal cord injury (SCI) leads to over-distention, high bladder pressures and vesico-ureteric reflux into the kidney, resulting in repeated infections and pyelonephritis. Given the importance/limited knowledge of SCI effects on the upper urinary tract, this study assessed expression of two proteins in the kidneys of contused (T9) male rats relative to controls that are indicative of tissue stress/damage (TGFβ, CD11b). Running economy (RE) is the amount of oxygen consumed while running at a submaximal intensity. Along with aerobic capacity (VO2max), RE is an important predictor of running performance. Little research has investigated changes in RE with restricted breathing training [i.e. controlled breath-holding (CFB)] during exercise. RE may improve ~6% amongst a novice swimming cohort through CFB training, but this has not been established in elite swimmers. In our studies we found that stabilized expression of HIF-1α in heart led to better recovery of function and less tissue death after 30 minutes of global ischemia, via mechanisms that preserve the mitochondrial polarization. Our group previously showed that HIF-1α conferred ischemic tolerance by allowing cardiomyocytes to use fumarate as an alternative terminal electron acceptor to sustain anaerobic mitochondrial polarization. The source of fumarate was identified as the purine nucleotide cycle (PNC). Here we discovered that HIF-1α upregulates AMP deaminase 2 (AMPD2), the entry point to the PNC. This project investigated the role of miRNA-21 in the macrophage response to peritonitis; with the goal of understanding whether the associated mechanism has potential benefits in the clinical treatment of peritonitis. A novel therapeutic intervention in the treatment of peritonitis could modulate the host immune response to decrease an exaggerated pro-inflammatory innate response and then prevent or ameliorate the development of sepsis and organ failure. Background: Interruption of cardiac stromal cell-derived factor 1 (SDF1)-CXCR4 axis by chronic AMD3100 administration increased myocardial injury after permanent coronary artery ligation, demonstrating the important role of this chemokine in cardiac regeneration. Hypothesis: Cardiomyocyte-specific conditional overexpression of SDF prevents heart failure after permanent coronary ligation and facilitates cardiac regeneration. Methods and Results: Tetracycline-controlled, a-myosine heavy chain promoter directed overexpression of cardiac SDF resulted in a significant increase of SDF expression (SDF: 8.1 ng / g protein) compared with littermate WT mice (0.02 ng / g protein) four weeks after doxycycline withdrawal. Cross education is the phenomenon by which the untrained limb will experience a gain in strength following a unilateral resistance training program. However, little is known as to the underlying adaptation occurring in the untrained limb. Purpose: To examine the effect of dynamic unilateral resistance training on the strength and neuromuscular adaptations of both the trained and untrained legs. Methods: Eight previously untrained males (22.38±2.92 y, 1.73±0.08 m, 75.26±14.53 kg) completed a four-week unilateral resistance training program, while another eight untrained males (24.00±4.57 y, 1.84±0.05 m, 94.21±16.14 kg) served as controls. Ecological stressors (i.e., high predator density, low food availability) are often associated with increased variation in mate choice by females. The association between environmental stressors and variation in mate choice suggests that circulating stress hormones (i.e., glucocorticoids) may mediate such effects, but this idea remains largely unexplored. I examined how circulating glucocorticoids affect preferences for male advertisement calls in female green treefrogs, Hyla cinerea, and barking treefrogs, Hyla gratiosa. PURPOSE: The purpose of this study was to examine the effects of short-term lower body unilateral resistance training on hormonal, muscle morphological, and performance measures in young men. METHODS: Seventeen healthy, untrained young men (Age: 22.8 ± 3.7 y; BMI: 26.5 ± 4.9 kg/m2) were randomly assigned to one of two groups (UT: 22.9 ± 4.6 y, 25.3 ± 4.2 kg/m2; CON: 24.0 ± 4.6 y, 27.7 ± 5.1 kg/m2). This may pose a potential competitive disadvantage considering that moderate altitude may cause decrements in VO2max, distance covered, and time to exhaustion in endurance athletes. Purpose: To examine the effects of 28 days of β-hydroxy-β-methylbutyrate free acid (HMB) and high-intensity interval training (HIIT) on maximal oxygen consumption (VO2peak), ventilatory threshold (VT), respiratory compensation point (RCP) and time to exhaustion (Tmax) in collegeaged men and women. The purpose of this study was to compare the efficacy of two dose levels of L-Alanyl-LGlutamine in a commercially available sports drink to the sports drink only on time to exhaustion, neuromuscular fatigue and physiological measures during prolonged endurance exercise. Twelve endurance-trained males (23.5±3.7 yrs; 175.5±5.4 cm; 70.7±7.6 kg) performed four trials, each consisting of 1 hr treadmill runs at 75% of VO2peak followed by a run to exhaustion at 90% of VO2peak. Purpose: To examine the effect of three muscle potentiation protocols on changes in muscle architecture and the subsequent effect on jump power performance. Methods: Maximal (1RM) squat strength (Mean SD=178.3 ± 36.6kg), vertical jump power, and muscle architecture were obtained in 12 resistance trained men (25.2±3.6y; 90.67±12.7kg). Participants randomly completed three squatting protocols at 75% (3 x 10 reps), 90% (3 x 3 reps) or 100% (1 x 1) of their 1RM, or no workout (CON), with each protocol being separated by one week.
