Objective: To measure the physical activity levels of Division I collegiate female soccer players with the use of accelerometers to help determine compensatory changes and help establish appropriate training rhythms through the course of a season.
Design: 25 female Division I soccer players volunteered to participate in a 7-day observational study. Players wore an ActiGraph GT3X+ accelerometer during waking hours. Previous studies evaluating single and multiple GnRH injections at time of CIDR removal/prostaglandin injection using the 7-day CO-Synch + CIDR protocol have reported decreased interval to estrus and increased expression of estrus, or increased conception rates, respectively. Thus, the objective of experiment 1 (Chapter 3) was to compare the effects of supplementing GnRH at prostaglandin injection on expression of estrus and conception rates in beef cows and heifers synchronized with different protocols. Colorectal adenocarcinoma is the fourth most common cancer diagnosed worldwide and is a significant cause of morbidity and mortality. This dissertation performed an exploratory RNA-sequencing analysis comparing gene expression between colon adenocarcinoma tissue and paired normal colon epithelium. After identification of a number of lncRNAs that were increased in expression in colon adenocarcinoma compared to normal colon epithelium, we aimed to validate the expression and investigate their function in vitro. Paralympic athletes (PA) appear to be more prone to chronic overuse injuries from daily wheelchair or crutch use. Over half of these injuries are shoulder related which can deleteriously impact quality of life. Adaptive powerlifters (AP) are a subdivision of Paralympic athletes and are at a higher risk for catastrophic injuries as compared to their counterparts, due to the compound of fatigue and lifting of maximal weights. Juvenile sandbar sharks (Carcharhinus plumbeus) have been caught in salinities ranging from 7 – 40. In Winyah Bay, a partially mixed estuary in Northeast SC, juvenile sandbar sharks tidally alternate between higher tides in middle bay and lower tides in lower bay. To assess salinity preference and duration in eight acoustically-tagged juvenile sandbar sharks in different salinities, acoustic receivers with salinity loggers were placed throughout Winyah Bay. Juvenile sandbar sharks were caught in salinities from 17.2 to 36.1 and acoustic detections were recorded from 11.5 to 24.7 by salinity loggers in middle bay. Load-velocity relationships may vary between people of different strength levels and across different loads. The purpose of this dissertation was to investigate how external loads influence the velocity characteristics of the back squat exercise, and the influence of strength on these variables. Healthy male students with a history of resistance training completed repetitions at specified intensities of their estimated one-repetition maximum (1RM) until they reached 1RM. Post-activation potentiation (PAP) is a stimulus used to enhance performance by performing specific interventions prior to subsequent explosive movements. The purpose of this study was to determine the effect of different PAP methods on power and sprint acceleration in resistance trained, college-aged men and women. A total of 13 subjects participated in the study. After determining back squat 1 repetition max, subjects returned for testing on separate days to complete one of four interventions (dynamic resistance, weighted plyometric, isometric, or control) in a randomized order. Traditionally, post-exercise muscular alterations have been examined using invasive techniques that lack the ability to single out individual muscle groups. Sonomyography, tensiomyography, and electrical impedance myography allow for noninvasive skeletal muscle assessment. This project aimed to examine changes in muscle contractility and composition that occur in the early stages of recovery following unaccustomed exercise. METHODS: Twenty-one untrained adults (21.9 ± 1.9 y) performed exercise – 10 × 10 maximal eccentric knee extensions – with their nondominant leg. The current investigation was an examination of the kinetic and kinematic characteristics of the back squat using accentuated eccentric loading (AEL) and cluster set programming strategies. PURPOSE: A promising therapeutic intervention to counteract endothelial ischemia/reperfusion (I/R) injury is ischemic preconditioning (IPC). Limited data from ovariectomized animal models shows that estrogen deficiency abolishes the cardioprotective effects of IPC, whereas estrogen replacement restores protection. This suggests that alterations in female sex hormones, particularly estrogens, that occur naturally during the menstrual cycle is likely to influence the vascular protective properties of IPC. This dissertation assesses the effects of climate warming and unionized ammonia (NH3), one of the most abundant water pollutants, on the behavior and physiology of two caenogastropod snails: fine-ridged elimia (Elimia semicarinata) and Shawnee rocksnails (Lithasia obovata) (Gastropoda: Neotaenioglossa: Pleuroceridae). This dissertation aimed to explore the usefulness of using force derivatives for onset detection in the isometric mid-thigh pull test. First, we examined applications of three differential calculus principles, first and second derivative, and curvature using visual detection as a reference under different baseline conditions. Second, we compared the best derivative method to a threshold-based method using visual detection as a reference. Results of our first investigation showed trivial differences between many differential calculus methods and visual detection. Ataxia-telangiectasia mutated kinase (ATM), a serine/threonine kinase primarily located in the nucleus, is typically activated in response to DNA damage. Individuals with mutations in ATM gene develop a disease called Ataxia telangiectasia (AT). These individuals are more susceptible to ischemic heart disease and metabolic disorder. Our lab has previously shown that ATM plays a critical role in β-adrenergic receptor (β-AR) - and myocardial infarction (MI)-stimulated myocyte apoptosis and cardiac remodeling. Discovery of an unusual rectal gland in the Atlantic sixgill shark, Hexanchus vitulus, led to examination of rectal glands in 29 species of epipelagic and deep-sea sharks. Eight of 14 deep-sea species of sharks had digitiform glands that were previously assumed to be characteristic of elasmobranchs (N=281; mean width-length ratio ± SD = 0.18 ± 0.07). Hematoxylin-and-eosin stained sections from deep-sea sharks were similar to those from shallow water sharks. Glands from the family Somniosidae were kidney bean-shaped (N = 3; mean width-length ratio ± SD = 0.46 ± 0.05); whereas those from the families Echinorhinidae and Hexanchidae appeared lobulate …

Poikilothermic organisms experience trade-offs by differential physiological demands generated by temperature extremes. Many such organisms exhibit acclimatory effects, adjusting their metabolism and physiology to recently experienced temperatures. One such acclimatory effect is metabolic compensation, the deceleration of biological rates below Arrhenius expectations. Daphnia magna is eurythermal, and if acclimated to mildly stressful temperatures first, survives longer in lethal temperatures. This study examined the effect of ambient temperature (5°C-37°C) and acclimation history (lifetime at 10°C or 25°C) on the oxygen consumption rates of 8 genotypes of Daphnia with high or low acute temperature tolerance. Excessive alcohol consumption is a leading cause of death and disability globally and can lead to diseases such as alcoholic skeletal myopathy. Skeletal muscle is the largest organ in the human body and functions to regulate whole-body energy homeostasis. Additionally, skeletal muscle can function as an endocrine organ via secretion of myokines. Two myokines, myonectin and irisin, present a wide range of effects upon metabolism, inflammation, and tissue survival- signaling. This dissertation is an analysis of skeletal muscle atrophy and a molecular assessment of potential preventative treatments. Chapter I begins with a background of skeletal muscle atrophy along with an analysis of associated molecular pathways. Here, we discuss how skeletal muscle atrophy is the consequence of protein degradation exceeding protein synthesis and can occur when a muscle is abnormally disused. Alcoholic Fatty Liver Disease (AFLD) is caused by excessive alcohol consumption and is a leading cause of liver related mortalities, with currently no treatments available. The goal of this project was to establish the effect of alcohol consumption on adipose tissue-derived secreted factors, adiponectin and C1q TNF Related Proteins 1-3 (CTRP1-3). We propose that excessive alcohol consumption will reduce circulating levels of adiponectin and CTRPs 1-3. The left atrial appendage has been a historically understudied region of the heart until fairly recently with the new understanding of its role in the stroke pathway of patients with atrial fibrillation. The present inquiry uses methods from psychophysiology and machine learning to reduce overall error in classification models. The field of psychophysiology, though rooted in decades of experimentation, has never reached the same level of precision as some aspects of medical inquiry. In fact, while some medical regression models, when determining some way to classify a patient's illness based on certain symptoms, can result in highly significant results with large effect sizes, equal levels are virtually unheard of in psychophysiology. The primary purpose of this study was to compare the metabolic influence of varying work-to-rest ratios during upper body sprint interval training (SIT). Forty-two recreationally trained men were randomized into one of three training groups [10s work bouts with two minutes of rest (10:2, n = 11) or four minutes of rest (10:4, n = 11), or 30s work bouts with four minutes of rest (30:4, n = 10)] or a control group (CON, n = 10). Sympathetic stimulation occurs in the heart after injuries such as ischemia/reperfusion (I/R) and myocardial infarction and affects myocardial remodeling. Prolonged sympathetic stimulation can result in myocardial dysfunction through its effects on cardiac myocyte apoptosis and myocardial fibrosis. Ubiquitin (UB) is well known for its role of tagging old or damaged proteins for degradation via the UB-proteosome pathway. The role of exogenous UB however, is not fully understood. Previously, our lab showed that β-adrenergic receptor (β-AR) stimulation increased levels of extracellular UB in the conditioned media of adult rat ventricular myocytes and that UB inhibits β-AR-stimulated apoptosis. Background: An essential mitochondrial component, Mitochondrial Transcription Factor A (TFAM) is reduced within the failing heart. TFAM maintains regulatory functions of cardiomyocyte physiology and stability. Mitochondrial Transcription Factor A forms a mitochondrial nucleoid anti-oxidant structure and reduces pathophysiological abnormalities to structural and contractile proteins. Overall, this functions to physically stabilize mtDNA. Significant molecular changes such as a loss of TFAM drive pathophysiological concentrations of cytoplasmic Ca2+ and reactive oxygen species. Both of these factors induce proteolytic enzymes to degrade the physiological cardiomyocyte.
