Woody breast (WB) myopathy results in poor muscle quality. The increasing incidence of WB over the last several years indicates a need for improved prediction or early diagnosis. We hypothesized that the use of body fluids, including blood, may be more suitable than breast muscle tissue in developing a minimally invasive diagnostic tool for WB detection. To identify potential early-age-biomarkers that may represent the potential onset of WB, blood samples were collected from 100, 4 wks old commercial male broilers. At 8 wks of age, WB conditions were scored by manual palpation. A total of 32 blood plasma samples (eight …
Regression Analysis Of Pacing When Running A Marathon, Hawkin Starke
Regression Analysis Of Pacing When Running A Marathon, Hawkin Starke
Industrial Engineering Undergraduate Honors Theses
Regression analysis can be an effective way of examining performance in the marathon event. By splitting up the race into segments or in runner terminology “splits” the significance of each segment as it relates to the total finish time can be explored. Because the idea of splits is already ingrained into the minds of runners, it makes intuitive sense to use these as the metrics to define a race. Additionally, marathons generally make participant age and gender date publicly available which can then be used to find trends within specific demographics. This tailors trends to smaller groups of people, making …

Using a mouse model of conditional and inducible in vivo fluorescent myonuclear labeling (HSA-GFP), sorting purification of nuclei, low-input reduced representation bisulfite sequencing (RRBS), and a translatable and reversible model of exercise (progressive weighted wheel running, PoWeR), we provide the first nucleus type-specific epigenetic information on skeletal muscle adaptation and detraining. Adult (>4 mo) HSA-GFP mice performed PoWeR for 8 wk then detrained for 12 wk; age-matched untrained mice were used to control for the long duration of the study. Myonuclei and interstitial nuclei from plantaris muscles were isolated for RRBS. Relative to untrained, PoWeR caused similar myonuclear CpG …

Supersize It! Developing A "Supercytoplast" Through Platelet Fusion, Taulbee B. Gunter
Animal Science Undergraduate Honors Theses
Stem cell research opens a wide range of possibilities from lab-grown tissues for medical purposes to cloned animals for livestock production. The issue with one of the most popular forms of cell recombination into a stem cell, known as somatic cell nuclear transfer (SCNT), is that it requires an oocyte. The requirement of an oocyte is often the host of public scrutiny for its economic and ethical issues. Platelets may offer a less expensive and more accessible alternative to oocytes as sources of naturally enucleated cells. Thus, the objective of this explorative research project is to isolate and fuse platelets …
The Effect Of Epa And Dha On Skeletal Muscle Physiology And Energy Balance, Saeed Jebor Ghnaimawi
The Effect Of Epa And Dha On Skeletal Muscle Physiology And Energy Balance, Saeed Jebor Ghnaimawi
Graduate Theses and Dissertations
Eicosapentaenoic acid (EPA) and Docosahexaenoic acid (DHA) supplementation commonly suggested during pregnancy confers many health benefits on offspring and more recently improves fetal BAT development and induces myoblast trans-differentiation into adiopocytes through activating PPARs expression. Embryonic myoblast is a highly flexible cell showing great capacity to change its phenotype into adipocyte in response to EPA and DHA supplementation in many in vitro models. However, a little is known about the identity of derived adipocytes, whether, they change into brown or white adipocyte, and in vivo evidence is still missing. Given that the myogeneis process is initiated at the mid of …

Respiratory Chain Activity And Protein Expression In Skeletal Muscles From High And Low Feed Efficient Swine Within The Same Genetic Line, Earsie L. Alcorn De Feliz
Graduate Theses and Dissertations
Current as well as past studies have provided evidence for a connection between macroeconomic/agricultural and cellular/biochemical basis of feed efficiency (FE; gain:feed) in farm animal production. Our investigation of respiratory chain activity, mitochondrial protein oxidation and protein expression was an expansion to the subcellular/molecular level. In this study we investigated the entire complex (Cx) of the electron transport chain (ETC) for differences in Cx activities in mitochondria isolated from skeletal muscle in high (HFE) and low (LFE) feed efficient swine within the same genetic line. Review of literature provided information for the connection between feed efficiency, mitochondrial function, and muscle …
Phenotypic Plasticity Of Japanese Medaka Gill In Response To Changing Salinities, Laura V. Ellis
Phenotypic Plasticity Of Japanese Medaka Gill In Response To Changing Salinities, Laura V. Ellis
Graduate Theses and Dissertations
Japanese medaka (Oryzais latipes) are euryhaline fish, meaning they are capable of surviving in a variety of salinities from fresh water to seawater. The ability to maintain an internal osmotic concentration stems from the phenotypic plasticity of the osmoregulatory organs, the gill, kidney, intestine, and integument. The gill is the main site of osmotic and ionic regulation in fish due to the three-dimensional structure, the direct contact with the outside environment, and the composition of the gill cells. Fish gills are multifunctional as they regulate water movement, acid/base exchange, nitrogenous waste excretion, and ion fluctuations. In freshwater environments, fish are …
Genetics Of Physiological Traits Associated With Drought Tolerance In Soybean (Glycine Max), Sumandeep Kaur Bazzer
Genetics Of Physiological Traits Associated With Drought Tolerance In Soybean (Glycine Max), Sumandeep Kaur Bazzer
Graduate Theses and Dissertations
Soybean (Glycine max L.) is one of the major row crops in the United States, and its production is often limited by drought stress. Physiological traits from exotic germplasm that confer drought tolerance may be useful in improving commercial soybean production. For example, carbon isotope ratio (δ13C) is positively correlated with water use efficiency (WUE), and nitrogen isotope ratio (δ15N) is negatively correlated with N2 fixation; canopy temperature (CT) is an indicator for genetic variation in transpiration and stomatal conductance. Therefore, the objectives of this research were to identify the genomic regions associated with: (1) δ13C and δ15N using a …

Effects Of A Noni-Supplemented Diet On Intestinal Tight Junction Proteins And Stress Biomarkers In Heat-Stressed Broiler Chickens, Guillermo Tellez
Poultry Science Undergraduate Honors Theses
Heat stress hampers gut health by impeding its ability to absorb nutrients which leads to a lower feed efficiency and sicker birds. Therefore, there is a critical need to identify mechanism-based strategies to alleviate these effects. Polynesian poultry farmers have been supplementing broiler feed with Morinda Citrifolia (Noni), a medicinal plant high in anti-oxidants, to address this problem. The purpose of the present study was to evaluate the effect of Morinda Citrifolia (Noni)-supplementation on the expression of tight junction proteins. Male Broilers (Cobb 500, 3 wks, n=480) were subjected to 2 environmental conditions (TN, 24°C vs. HS, 35°C, 1 …

Cancer cachexia is the rapid, drastic loss of muscle mass associated with cancer and is not reversible by conventional nutritional means (Brown et al., 2018; Brown et al., 2017). It occurs in ~80% of cancer patients and is responsible for 20-40% of cancer-related deaths (Brown et al., 2018; Brown et al., 2017). This condition leaves patients with fatigue, functional impairment, reduced quality of life, and a decrease in survival rates. Cachexia occurs through an imbalance between protein degradation and protein synthesis (Brown et al., 2018), which is associated with inflammation and altered metabolic processes.
Several studies have investigated the effects …

Eastern Collared Lizard (Crotaphytus Collaris) Population Declines In Ozark Landscapes: An Assessment Of Environmental Constraints., Casey L. Brewster
Graduate Theses and Dissertations
The Eastern Collared Lizard (Crotaphytus collaris) has experienced extensive population declines over the past half century in the Ozark Mountains. Previous research suggests that glade habitat degradation through woody vegetation encroachment is an important factor resulting in population declines. In this dissertation I used information on time-energy budgets to investigate the link between habitat degradation and shifts in life history traits likely resulting in population declines of Eastern Collared Lizards (Crotaphytus collaris) in the Ozarks. In chapter one, I addressed the influence of dense woody vegetation encroachment on age-specific growth, body size, body condition and reproduction of C. collaris in …

Comparative Plasma Proteomics In Muscle Atrophy Induced By Cancer Cachexia And Hindlimb Unloading, Kirsten Rene Dunlap
Graduate Theses and Dissertations
Introduction: Muscle atrophy results from a dysfunction in protein turnover that leads to loss of mass and function and occurs concurrently with multiple pathologies such as cancer and extended bed rest. Atrophy reduces overall quality of life while increasing morbidity and mortality. Currently, efficacious therapeutic interventions to treat and prevent muscle wasting in all its forms are lacking, however if conserved mechanisms can be identified between wasting conditions, this would aid in the development of multipurpose therapeutics to ameliorate this pathology. Purpose: To examine circulating factors present across atrophic pathologies. Methods: 35 male C57BL/6J mice were assigned to hindlimb unloading …

Methanogens, Plausible Extraterrestrial Life Forms On Mars, And Their Tolerance To Increasing Concentrations Of Illite Clay, Chandler Kern
Biological Sciences Undergraduate Honors Theses
Methanogens, some of Earth’s most primitive prokaryotic organisms, are candidates for possible life forms capable of inhabiting Mars. Specifically, four different species (Methanobacterium formicicum, Methanococcus maripaludis, Methanosarcina barkeri, Methanothermobacter wolfeii) were analyzed for their tolerance to the presence of illite clay. Illite is a crystalline mineral that has been identified from regions of Mars’s surface. Results indicated that all four species grew with some success in the illite at different concentrations. This experimentation with methanogens’ abilities to survive and reproduce in the presence of illite allows for a more accurate understanding of the potential capability of microbial …

Cellular Mechanisms Of Ionoregulation In The Gill Of Japanese Medaka And Rainbow Trout, Rebecca Jo Bollinger
Cellular Mechanisms Of Ionoregulation In The Gill Of Japanese Medaka And Rainbow Trout, Rebecca Jo Bollinger
Graduate Theses and Dissertations
Euryhaline fishes are capable of adapting to a wide range of salinities such as freshwater, brackish water or seawater. Through the combined effort of the gill, kidney and intestine, they are able to osmoregulate to maintain a constant internal hydromineral balance. As the gill is in direct contact with the external environment, it is continuously working to maintain ion and acid/base balance, gas exchange and eliminate nitrogenous waste. Fish in freshwater are subjected to osmotic water gain and diffusional ion loss across the gill and experience the opposite in seawater. Therefore, the gill exhibits extreme plasticity when experiencing a change …

Noninvasive Measures Of Stress And Lameness In Broilers, Shawna L. Weimer
Noninvasive Measures Of Stress And Lameness In Broilers, Shawna L. Weimer
Graduate Theses and Dissertations
The concept of broiler chicken welfare has evolved from a component of animal husbandry to a label on a chicken product package. Recent attention from the public has led to the need for higher welfare standards for animal production. A primary concern of broiler production is low activity/locomotion attributed to fast growth rates leading to poor leg health. To collect the scientific data necessary to determine conditions providing the best welfare for the bird, multiple methods of assessment are required. Stress is influenced by external (temperature, humidity, lighting, stocking density) and internal (metabolism, thermoregulation, hormonal balance) factors. This series of …
Role Of Trpv4 In Astrocyte Extracellular Matrix Production, Abby Terlouw
Role Of Trpv4 In Astrocyte Extracellular Matrix Production, Abby Terlouw
Biomedical Engineering Undergraduate Honors Theses
Traumatic Brain Injury (TBI) is an alteration of brain pathology following damage of the central nervous system (CNS) by an external force. In the CNS, glial scar formation often occurs following TBI, and astrocytes are widely believed to contribute to this scar formation. While the role of astrocytes in extracellular matrix (ECM) production is known, the exact mechanism(s) for this event remain unclear. One possible method is the activation of transient receptor potential vanilloid 4 (TRPV4). TRPV4 is a channel protein found in the astrocyte membrane which has been shown to generate intracellular calcium ions following mechanical stimulation. Previous research …
Histology Of Rathke’S Glands In The Razor-Backed Musk Turtle, Sternotherus Carinatus (Chelonia: Kinosternidae), With Comments On Lamellar Bodies, S. E. Trauth
Journal of the Arkansas Academy of Science
I examined the histology and ultrastructure of Rathke’s glands in two adult male razor-backed musk turtles (Sternotherus carinatus) collected in northeastern Arkansas. This species possesses two pairs of Rathke’s glands that are embedded beneath marginal bones and are named according to their anatomical location (i.e., axillary and inguinal). These integumentary glands are similar anatomically to one another. Each gland is comprised of a single, highly vascularized secretory lobule, which is surrounded by a thin tunic of asymmetrically arranged, striated muscle. Two types of large secretory vacuoles characterize most of the holocrine cells produced by a relatively thin secretory …
Extracellular Matrix Remodeling And The Inflammatory Response During Skeletal Muscle Regeneration In Sarcopenic Obese Mice, Lemuel Arthur Brown
Extracellular Matrix Remodeling And The Inflammatory Response During Skeletal Muscle Regeneration In Sarcopenic Obese Mice, Lemuel Arthur Brown
Graduate Theses and Dissertations
AIM: Sarocpenic obesity is a national concern within the United States because this metabolic syndrome is tied with reduced mobility and quality of life. Both obesity and aging are associated with insulin-resistance, chronic low-grade inflammation and muscle weakness. Skeletal muscle regeneration is a process that involves the coordinated effort of myogenic regulatory factors (MRFs), inflammatory signaling, and extracellular matrix (ECM) remodeling for optimal regeneration. It has been demonstrated that obesity and aging have a reduction in muscle regeneration. It has not been examined if sarcopenic obesity will further reduce muscle mass and the regenerative process. The purpose of this study …
Nuclear Factor [Erythroid-Derived 2]-Like 2’S (Nfe2l2) Gene Expression In Feed Efficiency And Oxidative Stress In Avian Muscle, Devin Wayne Cook
Nuclear Factor [Erythroid-Derived 2]-Like 2’S (Nfe2l2) Gene Expression In Feed Efficiency And Oxidative Stress In Avian Muscle, Devin Wayne Cook
Graduate Theses and Dissertations
Oxidative stress may play a role in the phenotypic expression of feed efficiency (FE). The transcription factor NFE2L2 (nuclear factor erythroid-derived 2-like 2) coordinates antioxidant response to oxidative stress and its activity is tightly regulated in part by KEAP1 (Kelch like-ECH protein 1) and the E3 ligase CUL3 (Cullin3). Thus, one objective was to determine mRNA expression of NFE2L2, KEAP1, and CUL3 as well as three antioxidant targets [glutathione peroxidase (GPx-1), superoxide dismutase 1 (SOD1), and superoxide dismutase 2 (SOD2)] in breast muscle of immature pedigree broiler males (8 wk), immature Japanese quail males (4 wk) divergently selected for high …

Response Of Leukocyte Profiles To Corticosterone Manipulation In The Prairie Rattlesnake (Crotalus Viridus), Jill E. Guillette
Biological Sciences Undergraduate Honors Theses
Prior work has shown that when animals are stressed they have altered leukocyte profiles. The use of leukocyte profiles could provide an inexpensive and efficient method for determining stress. Here we present a validation of the use of leukocyte profiles for showing induced stress in Prairie Rattlesnakes (Crotalus viridus). Snakes (N=19) with masses ranging from 682g to 137g were used in a repeated measures design to examine the effect of hormone manipulation on leukocyte profiles. During each trial snakes were dosed with either corticosterone and sesame oil (1.1µg/g) or only sesame oil. Prior to dosing, baseline blood samples …
