Advanced Technologies Of Single-Cell Metabolomics Unveiling Cellular Metabolic Heterogeneity For Biological And Biomedical Research, Shang-Che Kuo, Ting-Wei Tsao, I-Fan Wang, Ying-Lan Chen

Journal of Food and Drug Analysis

Metabolomics provides direct insights into cellular physiology, yet it faces greater analytical challenges compared to genomics and transcriptomics due to the chemical diversity, instability, and environmental sensitivity of metabolites. Conventional bulk metabolomics averages signals across cell populations, thereby masking cellular heterogeneity critical for understanding disease mechanisms. Recent breakthroughs in single-cell metabolomics (SCM), driven by advances in mass spectrometry, microfluidics, isotope tracing, and spatial omics, have enabled the detection of metabolic diversity at unprecedented resolution. SCM has uncovered cell-type-specific biomarkers, revealed metabolic reprogramming in cancer and immunity and revealed disease progression. These studies highlight SCM’s transformative potential in biomarker discovery, clinical …

Background

RNA therapeutics are a rising drug category with potential use for a range of conditions encompassing infectious diseases to therapies for cancer, diseases, and genetic disorders. RNA-lipid nanoparticles (RNA-LNPs) are the prominent delivery method for these therapeutics approved products include mRNA vaccines and polyneuropathy treatments. The emergency use authorizations and orphan drug status of current RNA-LNP drugs has allowed approval without finalization of the regulatory analytical procedures for quality monitoring. The objective of the study was to develop an LC-MS assay to simultaneously measure identity and concentration of two therapeutically relevant intact RNA constructs extracted from RNA-LNPs to enhance …

Bongkrekic acid (BKA) poisoning is a severe foodborne illness with a high mortality rate. This study aimed to identifyBKA in postmortem tissues from poisoning victims in Taiwan and to develop a reliable analytical method for detectingBKA in biofluids to aid clinical diagnosis and treatment. BKA was identified in postmortem samples using high-reso-lution mass spectrometry (HRMS) and confirmed with an independent ultrahigh-performance liquid chromatographycoupled with tandem mass spectrometry (UHPLC-MS/MS) method. To meet clinical needs, a UHPLC-MS/MS methodwas developed and validated for BKA detection in plasma and urine. Method optimization included adjustments to ionsource conditions for multiple reaction monitoring (MRM) transitions and …

Antityrosinase Activity And Lc-Ms/Ms Analysis Of Optimized Ultrasound-Assisted Condition Extracts And Fractions From Strawberry Tree (Arbutus Unedo L.), Serdar Demir, Halil Koyu, Mustafa Abdullah Yilmaz, Abbas Tarhan, Sura Baykan Ozturk Jun 2024

Antityrosinase Activity And Lc-Ms/Ms Analysis Of Optimized Ultrasound-Assisted Condition Extracts And Fractions From Strawberry Tree (Arbutus Unedo L.), Serdar Demir, Halil Koyu, Mustafa Abdullah Yilmaz, Abbas Tarhan, Sura Baykan Ozturk

Journal of Food and Drug Analysis

Investigation of utilization possibilities of natural sources has been an important area for research. Tyrosinase inhibitory activity plays a key role in food and medicine industry. Strawberry tree (Arbutus unedo), a widely distributed plant among Mediterranean countries, possess fruits and leaves with rich bioactive phytochemicals, especially polyphenolic compounds. In this study, we aimed to investigate the antityrosinase activity of the fruit and leaf extracts of the plant, and to determine the phenolic compounds that contribute to the antityrosinase activity. In this regard, we evaluated the effect of solvent composition on the extraction of phenolic compounds from A. unedo …

Identification And Quantitation Of Unspecified Impurities Discovered In Novel Oral Irinotecan By Lc-Ms/Ms And Uhplc, Laura B. Miller

Forensic Science Master's Projects

Irinotecan is a cancer medication approved for medical use in the United States in 1996. It is currently administered via intravenous injection. However, intravenous dosing has disadvantages such as requirement of administration by a trained professional in a medical facility, possible severe adverse effects, pain at the injection site, and possible hemolysis if injected too rapidly. Previously oral administration was not feasible because the p-glycoprotein (p-GP) on the cell membrane of stomach cells acts as a defense mechanism against harmful substances by pumping the irinotecan back into the intestinal lumen for excretion. With the discovery of the p-GP inhibitor encequidar …

Analysis Of Heterocyclic Aromatic Amines Using Selective Extraction By Magnetic Molecularly Imprinted Polymers Coupled With Liquid Chromatography – Mass Spectrometry, Shih-Ci Jhu, Jing-Ying Wang, Hsueh-Ting Wang, Sung-Fang Chen

Journal of Food and Drug Analysis

Heterocyclic aromatic amines (HCAs) are highly carcinogenic and mutagenic chemicals. This study reports on the development of magnetic molecularly imprinted polymers (MMIPs) for the purification and quantification of HCAs. A novel magnetic molecularly imprinted polymer was successfully prepared using a surface molecular imprinting method using functionalized Fe particles as the magnetic cores. 2-Amino-3-methylimidazo[4,5-f]quinoline (IQ) was used as a molecular template; methacrylic acid (MAA), ethylene glycol dimethyl acrylate (EGDMA), 2, 2′-Azobis (2-methylpropionitrile) were used as the functional monomer, crosslinker, and initiator, respectively. The use of the template/functional monomer/crosslinking agent at a ratio of 1:4:20 resulted in a product with better adsorption …

Polydopamine And Silica Nanoparticles Magnetic Solid Phase Extraction Coupled With Liquid Chromatography-Tandem Mass Spectrometry To Determine Phenolic Acids And Flavonoids In Fruit Wine, Hui-Ling Lee, Chih-Yuan Kang, Yen-Jung Kuo, Shan-Ni Tseng

Journal of Food and Drug Analysis

Magnetic solid phase extraction (MSPE) have been widely applied in a variety of sample preparation techniques. Herein, Fe3O4@pDA as the sorbents for MSPE, were developed for the determination of phenolic acids and flavonoids in fruit wine samples in combination with LC-MS/MS. The Fe3O4@pDA were characterized by Fourier transform infrared spectroscopy (FT-IR), powder X-ray diffraction (PXRD), transmission electron microscopy (TEM), Superconducting Quantum Interference Device Magnetometer (SQUID) and thermogravimetric analysis (TGA) in detail. In the present study, a new, rapid, and efficient MSPE by LC-MS/MS was established for the extraction and sensitive detection of phenolic acids and flavonoids. Under the optimized condition …
