The organic anion transporting polypeptide (OATP) 1B1 and OATP1B3 are hepatic transporters that mediate the uptake of numerous therapeutics like antibiotics and statins. Various post-translational modifications, eg, phosphorylation and glycosylation, have been shown to impact OATP1B1 and OATP1B3 function and localization. S-palmitoylation (also known as S-acylation or palmitoylation) is a reversible post-translational modification that adds palmitic acid (C16:0) to cysteine residues and can influence a protein's function, localization, and protein-protein interactions. Polythene bags are highly inert plastic materials made from monomers of ethylene. Due to their durability, lightweight, and low-weight characteristics, polythene bags are in high demand among consumers and retailers worldwide. This widespread use of polythene bags has led to the generation of polythene waste in the environment. Microplastics have become widely dispersed in our environment, and even in the air we breathe. The present research aims at identifying and determining the amounts of microplastics present in atmospheric fallout over Lahore city, which is characterized by a large population and several manufacturing industries. In order to find out the existence of microplastics, samples of atmospheric fallout were collected from various sites across hustling municipalities of Lahore that included urbanized settlements. The analysis disclosed that there are various forms of microplastic fibers, fragments and granules indicating different sources of pollution. Human health may be at risk if drinking water contains excessive amounts of heavy metals (HMs). To determine risks related to heavy metals, fifteen water samples were taken every month between July and September, 2024 from the communities of Abobo (Site A), Osara (Site B) and Osaragada (Site C) close to Confluence University of Science and Technology, Osara. Background: Acute gout arthritis, which is caused by hyperuricemia, is a common inflammatory joint disease, leading to monosodium urate (MSU) crystal deposition. These crystals trigger inflammation, causing pain and significantly impairing the patient’s quality of life. Colchicine is commonly used to treat and prevent acute gout arthritis, but it has several side effects. Air pollution exposure has been linked to many surgically managed diseases and adverse health events that occur in the postoperative period. Despite this, the impact of air pollution on postoperative recovery is poorly understood. This knowledge gap may be one of the underlying mechanisms for persistent disparities in surgical outcomes. Motion sickness is a prevalent neurophysiological condition affecting approximately one-third of the worldwide population, resulting from sensory mismatch and the brain’s internal model of motion. It is characterized by nausea, vomiting, dizziness, and autonomic symptoms during exposure to actual or virtual motion, which may significantly impair daily functioning and quality of life. Current guidelines recommend the prevention and treatment of motion sickness through pharmacologic, herbal, and non-pharmacologic strategies. The coagulation cascade comprises intrinsic, extrinsic, and common pathways that converge when thrombin converts fibrinogen (factor I) into fibrin, leading to fibrin mesh formation, stabilization of the platelet plug, and ultimately thrombus formation. Inhibition of the coagulation cascade can be achieved with a variety of anticoagulant medications, including direct oral anticoagulants (DOACs). The mechanism of action of DOACs is to inhibit either factor Xa or thrombin. Paroxysmal supraventricular tachycardia (PSVT) is characterized by sudden, brief, intermittent episodes that result in a heart rate of 150 to 250 beats per minute (bpm). PSVT is difficult to diagnose, making proper treatment challenging. According to the 2015 American Heart Association (AHA)/American College of Cardiology (ACC)/Heart Rhythm Society (HRS) guidelines, current pharmacologic recommendations for PSVT include adenosine, verapamil, diltiazem, or nonpharmacologic vagal maneuvers.1 These treatments each have a variety of adverse effects, contraindications, and dosing considerations, which highlight a need for novel therapies for the acute treatment and maintenance of PSVT.1 The NODE-301 trial assessed the safety and …

Background and Purpose: Mercury is a pervasive environmental contaminant, and human epidemiologic studies have linked chronic low-level exposure to increased autoimmune disease-risk. Dysregulated lymphocyte activation and impaired signaling tolerance are central mechanisms in human autoimmunity. Protein phosphorylation governs lymphocyte differentiation and function, alterations in phosphoproteomic networks may represent a signature of immune disruption. This study evaluates how low dose mercury exposure modifies B and T cell abundance and intracellular signaling patterns in BALB/c and Diversity Outbred (DO) mice.

Rett syndrome (RTT) is a severe neurodevelopmental disorder that is associated with loss-of-function mutations in the methyl-CPG binding protein 2 (MECP2) gene. There are limited therapeutic options to address the wide range of social, cognitive, motor, and respiratory symptoms that patients experience throughout their lifetime. To identify novel therapeutic targets, we leveraged a rare form of RTT known as MeCP2-mutation-negative or atypical RTT. Traditionally, RTT is first diagnosed clinically and then confirmed through the identification of mutations in the MECP2 gene. Francisella tularensis is a highly infectious intracellular bacterium and the causative agent of tularemia, with pulmonary infection resulting in severe disease and high mortality if untreated. While protective immunity to F. tularensis has been largely attributed to CD4⁺ T cell–mediated IFN-γ responses, the contribution of γδ T cells to mucosal immunity and vaccine-induced protection remains poorly defined. The objective of this study was to characterize the role of γδ T cells during primary pulmonary infection with F. tularensis live vaccine strain (LVS) and following intranasal immunization with inactivated F. tularensis (iFt) prior to LVS challenge.

Acute myeloid leukemia (AML) is a fatal blood cancer with cytotoxic chemotherapy offering at best 25% 5-year survival. While targeted BCL2 and FLT3 inhibitors venetoclax and gilteritinib are used upfront in the treatment of a subset of adult patients with AML and help to extend the survival of some patients, a curative treatment combination with minimal side effects has yet to be discovered. We find that use of the dual histone acetyltransferase p300/CBP bromodomain inhibitor CCS1477 (inobrodib), together with venetoclax and gilteritinib, virtually eliminates leukemia stem cells in an aggressive preclinical model of DNMT3A/FLT3-mutant AML by impairing pro-oncogenic survival and …

Autoimmune disorders disproportionately affect females, a bias largely attributed to estrogen's modulation of B lymphocyte development and tolerance. Bisphenol A (BPA), a common plastic monomer, shows estrogenic activity and acts as an endocrine disruptor and epigenetic modifier. This thesis examines how BPA may increase autoimmune risk by mimicking estrogen such that it can excite estrogen receptors which rescue autoreactive B lymphocytes from apoptosis. Endometriosis is a disease characterized by pelvic pain and the formation of endometrial-like tissue on the outside of the uterine lining. This condition affects nearly 10% of women across the globe and confirming whether someone has endometriosis requires visualization in surgery. This thesis examines the disproportionate burden of lead exposure carried by low income, POC communities. The systemic nature of this problem is a symptom of a longstanding legacy of environmental injustice in the United States. Tetrahydrocannabivarin (THCV), a minor cannabinoid found in Cannabis sativa, is gaining attention for its potential therapeutic applications, yet its effects on early development remain poorly understood. This study investigated how the timing of THCV exposure influences morphological, behavioral, and bioaccumulation outcomes in zebrafish (Danio rerio) embryos. Embryos were exposed to 2 μM THCV during discrete developmental windows (6–30, 30–54, 54–78, 78–102) and continuous 6–102 hours post-fertilization (hpf), designed to model early stages of human gestation. Herbularyo, a card game teaching about herbal plants (HPs), was created and evaluated as a viable educational card game based on Tuomisto and Aksela’s criteria. A quasi-experiment was performed on 81 Grade 8 students from a private high school and 50 Grade 11 from a public high school. Independent t-test revealed that the control group (CG) and experimental group (EG) (p=0.386), and EG1 (played ≤5) and EG2 (played >5) (p=0.681) are not comparable with each other. Paired t-test showed EG performed better (p=0.001) than CG (p=0.901) whereas ANCOVA showed that playing Herbularyo improved students’ information retention (p=0.006). EG1 and EG2 …

Background: Unintentional therapeutic duplication represents an underrecognized yet preventable contributor to medication-related harm, particularly in settings characterized by widespread over-the-counter (OTC) medicine use and self-medication practices. Objective: To estimate the prevalence of unintentional therapeutic duplication among adults in Saudi Arabia and identify sociodemographic and behavioral predictors associated with increased therapeutic duplication risk. Methods: A national crosssectional study was conducted between February 10 and April 28, 2025, using a structured, self-administered online questionnaire distributed via social media platforms. This paper examines the hidden environmental, geopolitical, economic, and social costs of the cobalt supply chain. Fueled by the demand for battery technology and electric vehicles, cobalt has become an essential element to the green energy transition. With the largest share of global cobalt deposits, the Democratic Republic of the Congo bears a disproportionate share of the industry’s harms. This research was conducted to explore the possibility of constructing floating treatment wetlands (FTWs) without using plastic materials. FTWs are floating platforms that hold plants on the water’s surface, letting roots dangle within the water column, to help remove contaminants from contaminated water, especially in stormwater ponds. Many conventional FTWs are made with plastic components, such as mats and plastic cup containers, which can release microplastics into the environment over time. The US population faces simultaneous public health threats from both aging and environmental pollutants. Chief among these is hexavalent chromium [Cr(VI)], whose toxic effects in peripheral tissues are well-established but its effects on the brain are still understudied. Following previous results indicating hippocampal metals dyshomeostasis after Cr(VI) exposure, we aim to assess Cr(VI)’s impact on the blood-brain barrier (BBB), a key structure for brain protection. Pharmaceutical contaminants are increasingly prevalent in aquatic environments, yet their sublethal effects on ecologically relevant behaviors remain poorly understood, particularly in estuarine systems. Selective serotonin reuptake inhibitors (SSRIs), such as fluoxetine, are of particular concern due to the conserved role of serotonin in regulating behavior across vertebrates. Here, we investigated how acute exposure to environmentally relevant concentrations of fluoxetine influences anti-predator behavior in Gulf killifish (Fundulus grandis), a key estuarine fish commonly found in saltmarsh habitats. Brownfields, or properties that are or possibly are contaminated with dangerous materials, represent a common obstacle for communities in Mississippi. The real or potential presence of contaminants at these sites often deters investors due to costly cleanup requirements (EPA, n.d. -b). The Environmental Protection Agency (EPA), through its Brownfield Revitalization Program, implemented guidelines for covering the costs and labor associated with the remediation of these properties. The EPA is not, however, responsible for redevelopment that supports long-term environmental and economic sustainability.
