Historical and current extraction operations and infrastructure, including natural gas pipelines, conventional and unconventional oil and natural gas drilling, longwall coal mining, and strip coal mining, occur throughout the Mingo Creek watershed of Washington County, Pennsylvania, including Mingo Creek County Park. Mingo Creek and several associated tributaries flow through Mingo Creek County Park, eventually discharging into the Monongahela River. An independent water quality study was conducted from June 2020 through July 2023 in Mingo Creek County Park to determine if historical and current extraction activities impact Mingo Creek and its associated tributaries. Background: At the time of this study, the Commonwealth of Kentucky had over 3,800 active public bathing or swimming pool facilities. In compliance with 902 KRS 10:010 Kentucky Swimming and Bathing Facility regulation, all bathing and swimming pools for public access are subject to inspection and monitoring by local and state health agencies. The regulation requires the facilities to be inspected for code violations on the protection and safety of swimming patrons. It mandates inspections on: water supply and wastewater disposal; bathhouses and attendant structures; safety; and general facility operations. Despite the opioid crisis taking headlines, the abuse of cocaine and amphetamines remains rampant and laboratory instruments must be able to effectively and reliably handle the overwhelming case load. In this study, an Agilent 6475 Triple Quadrupole (QQQ) LCMS, recently acquired by the Tarrant County Medical Examiner’s Office (TCMEO) in Fort Worth, Texas, was validated for identifying and quantifying CocAmps. Results show that the instrument was fit-for-purpose and that the instrument passed all validation tests. Reproducibility was ≤ 20% RSD for 12 QC replicates, 6 of blood and 6 of urine. Accuracy was ± 20%. A 98.9% recovery was yielded …

Δ9-tetrahydrocannabinol (Δ9-THC) is used during pregnancy to mitigate symptoms such as nausea, pain, and insomnia. It is established that Δ9-THC exposure early in life causes adverse effects in children’s development including behavioral disorders. Our goal was to determine the latent and generational impacts on growth, behavior, and mitochondrial bioenergetics following exposure to Δ9-THC. Our group worked alongside community partners Shaunna Barnhart and Jim Dunn during the Spring 2024 semester to assist with their annual event Fireflies at RPA Natural Area. RPA Natural Area is a local park in South Williamsport, Pennsylvania that was recently acquired by Southside Recreational Authority. Since taking over the park, the organization has begun hosting an event each July to educate the general public about firefly biology and conservation by showing a film and guiding attendees through a firefly tour through the park. This paper addresses the increasing vulnerability that coastal communities face regarding climate crises and rising sea levels. Specifically, this paper investigates the environmental crises facing Charleston, South Carolina, and New York City. The geographical location of these cities places a more severe threat upon their environment, as opposed to urban collectives removed from the immediate effect of rising sea levels. The following paper provides insight into the Flint, Michigan water crisis of 2014 through the lens of environmental racism. The Flint, Michigan water crisis saw the contamination of the municipal water supply of the town of Flint as a result of the decision to switch the main water source from Lake Huron to the Flint River as a temporary measure as the town of Flint switched to the Karegnondi Water Authority contract to supply their municipal water. This study aims to assess seasonal and spatial variations, contamination status, ecological risks, and metal sources (Ni, Pb, Cr, Cu, Mn, and Zn) in human-afforested mangrove sediments in a deltaic region. Five sampling locations were sampled during dry and wet seasons. Heavy metal concentrations followed the order: Mn > Zn > Ni > Cr > Cu > Pb. Metal loads, except Cu and Pb, were higher during the dry season, aligning with national and international recommendations. Sediment quality guidelines, contamination factor, geoaccumulation index, enrichment factors, and pollution load index indicated uncontaminated sediment in both seasons. Quantitation of EPZ was performed on a Shimadzu LC-MS/MS system utilizing electrospray ionization operated in positive mode. The validated assay was linear from 0.2 to 500 ng/mL. Metabolic stability studies demonstrated slow intrinsic clearance of EPZ in mouse and human liver microsomes, suggesting no CYP-mediated metabolism. Additionally, EPZ exhibited low binding affinity to plasma and brain proteins and high apparent permeability across artificial membranes, indicating efficient membrane penetration. While antiretroviral therapy (ART) has significantly reduced human immunodeficiency virus type one (HIV-1) co-morbidities and mortality, adherence to daily oral therapy remains a key barrier to all therapeutic and preventative outcomes. Over the past years, the emergence of long-acting (LA) ART has gained considerable enthusiasm amongst people living with HIV (PLWH) and could potentially simplify dosing regimens and improve treatment outcomes. Chronic inflammation, when left untreated, may result in significant morbidity and in severe cases, mortality. While glucocorticoids (GCs) are still being extensively used clinically as a class of very potent anti-inflammatory agents, they are also notorious for diverse adverse side effects. To mitigate these side effects, we have developed a polyethylene glycol (PEG)-based prodrug (ZSJ-0228) of dexamethasone (Dex). When tested on rodent models of Lupus Nephritis and Focal Segmental Glomerulosclerosis (FSGS), ZSJ-0228 was found to provide potent and sustained therapeutic efficacy without any detectable typical GC-associated side effects. This tradition has fundamental roots in the culture and customs of Guatemala. Unfortunately, these methods contribute to air pollution. Consequently, it also results in smoke inhalation, which causes health problems. Multiple myeloma (MM) is a type of cancer that affects plasma B cells. Patients with MM often experience frequent relapses and can develop resistance to drugs. As a medical researcher, it is important to understand the role of Mcl-1 in preventing intrinsic apoptosis and drug resistance. Mcl-1 belongs to the anti-apoptotic subgroup of Bcl-2 family proteins and plays a crucial role in these processes. The opioid epidemic remains a pressing public health crisis, prompting the search for alternative pharmacotherapies for Opioid Use Disorder (OUD). This study explores the potential of the dopamine D3 receptor (D3R) partial agonist, (±)VK4-40, as a novel treatment option. We investigated the impact of intra-nucleus accumbens (NAc) microinfusion of VK4-40 on basal locomotion in mice. Results indicate that VK4-40 did not significantly alter basal locomotion, suggesting that its therapeutic effects may not be mediated through disruptions in generalized motor function. Future research will focus on elucidating the neuropharmacological mechanisms underlying VK4-40's therapeutic actions and exploring its effects on psychostimulant-induced hyperlocomotion. …

Industrial pollution is a growing environmental issue in the United States, having health, economic, and social consequences on the surrounding communities. This study explores how communities respond to industrial pollution by examining two case studies in New York: the Navy/Grumman Plume of Bethpage and the IBM Plume of Endicott. Document analysis provided information to create complete timelines of the pollution and remediation in each community. Interviews were also conducted to provide personal insight from residents, employees of polluting industries, or those involved with remediation efforts. Ultimately, the research suggests that industrial actors should take a proactive stance in addressing contamination. …

The use of wetlands as a treatment approach for nitrogen in runoff is a common practice in agroecosystems. However, nitrate is not the sole constituent present in agricultural runoff and other biologically active contaminants have the potential to affect nitrate removal efficiency. In this study, the impacts of the combined effects of four common veterinary antibiotics (chlortetracycline, sulfamethazine, lincomycin, monensin) on nitrate-N treatment efficiency in saturated sediments and wetlands were evaluated in a coupled microcosm/mesocosm scale experiment. Pesticides are used widely to control weeds and pests in agricultural settings but harm non-target aquatic organisms. We examined the effect of short-term exposure (one week) to pesticide mixtures (low dose: 0.5 μg/L Roundup®, 0.5 μg/L 2,4-D, and 0.4 μg/L atrazine; high dose: 1 μg/L Roundup®, 1 μg/L 2,4-D, and 0.8 μg/L atrazine) on American oyster (Crassostrea virginica, an important shellfish and seafood species) under controlled laboratory conditions. Histological analyses demonstrated the atrophy in oysters' gills, digestive glands, and connective tissue. Defense of mucosal tissues from microbial infection and allergy is reliant on continual production of antibodies. The aryl hydrocarbon receptor (AhR) is known to regulate B cell development and is associated with suppression of systemic humoral immunity. Recent attention has been paid to the role of the AhR in altering expression of cell adhesion molecules (CAMs). B cells express CAMs and chemokine receptors to migrate around the body for localized secretion of antibodies. Oil spilled in aquatic environments undergoes abiotic and biotic oxidation processes that produce hydrocarbon oxidation products (HOPs). HOPs rapidly diffuse in the water column, persisting and readily bioavailable to aquatic organisms. Characterizing HOPs is challenging because most of the mixture resides in the unresolved complex mixture (UCM). Understanding of the formation, fate, and toxicity of HOPs in high latitude environments remains limited. Simparica TRIO®, a canine endectocide manufactured by Zoetis, contains sarolaner, moxidectin, and pyrantel pamoate salt in an orally administered chewable tablet. These compounds are all toxic to both cats and dogs in sufficient amounts. As Simparica TRIO® rises in popularity among pet owners, Felis catus (felines, cats) in multi-pet households (households with both cats and dogs) are at increased risk of overdosage and toxicosis from accidental ingestion of Simparica TRIO® due to their smaller weight.

Cerebral malaria (CM), a severe manifestation of Plasmodium infection, prompts our investigation into the nuanced role of pipecolic acid in its pathophysiology. To unravel the molecular intricacies, we conducted in vitro lysine labeling techniques of mice infected with P. berghei ANKA parasites, and human P. falciparum grown in vitro, aiming to discern the impact of Plasmodium on pipecolic acid production. Oral fluid has been gaining importance in forensic and clinical toxicology for many different drug testing scenarios. Oral fluid is composed of saliva, along with gingival fluid, bacteria, and food residues.
