The purpose of this study is to analyze the concentration of microcystins from cyanobacteria along with factors that increase the likelihood of microcystins found in water systems at the Turnbull National Wildlife Refuge (TNWR) near Cheney, Washington. Cyanobacteria are oxygen-producing bacteria that use sunlight as an energy source to convert CO2 into biomass. This bacterium has been around for around 3 billion years. Bacillary dysentery, commonly known as shigellosis, is a potentially perilous and extremely contagious bacterial infection of the colon caused by—but not limited to—bacteria shigella, bacillus, E. coli, Yersinia, and the parasite amoeba. This paper formulates and analyses a mathematical model for the transmission dynamics of dysentery epidemic that incorporate the effects of weather variations. It examines the stability of equilibria and compute the basic reproduction number that is coupled with the time-periodic model, and establishes results on the threshold dynamics. Bottom ashes originating from medical waste incineration contain heavy metals capable of causing adverse effects to human beings and the environment. The main goal of this study is to establish levels of heavy metals in bottom ashes from medical waste incinerators to build a basis for the development of sustainable, safe, and efficient methods for healthcare waste management. The study assessed the level of heavy metals in the bottom ashes of thermally treated medical waste from six health care facilities in Dar es Salaam city, Tanzania. Soils contaminated with lead (Pb) from human activities including industrial byproducts create an environmental and human health risk. Phytoextraction through chemical acquisition of Pb in soils offer a potentially cost effective and ecologically driven approach to remediation. The aim of this work was to evaluate the transformation of manganese oxide (hausmannite) by microscopic filamentous fungus Aspergillus niger and the effects of the transformation on mobility and bioavailability of arsenic. Our results showed that the A. niger strain CBS 140837 greatly affected the stability of hausmannite and induced its transformation into biogenic crystals of manganese oxalates—falottaite and lindbergite. The transformation was enabled by fungal acidolysis of hausmannite and subsequent release of manganese ions into the culture medium. While almost 45% of manganese was bioextracted, the arsenic content in manganese precipitates increased throughout the 25-day static cultivation of fungus. …

Drug use, of prescription or illicit varieties, alter bodily functions in different ways, especially in the nervous system. These drugs are found in sources of drinking water because they are not removed by wastewater treatment facilities. To investigate the effects of these drugs as they appear in mixtures, we exposed groups of zebrafish embryos to cocktails of drugs and screened for genes that were differentially expressed between the experimental and control groups and could thus give more insight into their biological functions. …

Cytochrome P450 2D6 (CYP2D6) is a highly polymorphic gene whose protein product metabolizes more than 20% of clinically used drugs. Genetic variations in CYP2D6 are responsible for interindividual heterogeneity in drug response that can lead to drug toxicity and ineffective treatment, making CYP2D6 one of the most important pharmacogenes. Prediction of CYP2D6 phenotype relies on curation of literature-derived functional studies to assign a functional status to CYP2D6 haplotypes. As the number of large-scale sequencing efforts grows, new haplotypes continue to be discovered, and assignment of function is challenging to maintain. Purpose: Donepezil, a drug frequently used to treat dementia, is mainly metabolized by cytochrome P450 2D6 (CYP2D6). This study investigated the relationships between CYP2D6 genotype and activity scores as well as predicted phenotype of plasma donepezil concentrations in 86 Thai dementia participants.
Materials and methods: CYP2D6 was genotyped using bead-chip technology (Luminex xTAG® v.3). Steady-state trough plasma donepezil concentrations were measured using high-performance liquid chromatography.
Results: Sixteen genotypes were found but the most frequent genotypes detected among our participants were CYP2D6*10/*10 (27.9%) and *1/*10 (26.7%). One-third of the participants had an activity score of 1.25 which predicted that they were …

Detection of drugs of abuse in urine samples depends on their composition, absorption, distribution, physical and chemical properties, concentrations, elimination properties, and the techniques employed. This study compared the performance of centrifugation and solid phase extraction (SPE) methods in the determination of levels of drugs of abuse and their metabolites in urine samples collected from drug abusers. Analysis was performed using liquid chromatography-tandem mass spectrometry (LC-MS-MS). Calibration was done using both internal and external standards. The recovery values were generally greater in centrifugation method than in SPE method, but the recoveries in both methods were within an acceptable range. Linearity …

Fisheries populations and their ecosystems are negatively impacted by both chronic and acute inputs of pollutants, including oil spills such as the Deepwater Horizon platform blowout in 2010 in the Gulf of Mexico (GoM). After Deepwater Horizon, toxicological studies of demersal fishes of the northern GoM were undertaken to characterize impacts and to establish baseline contaminant levels in the aftermath of the spill. Greenhouse gas emissions have increased rapidly since the industrial revolution. This has led to an unnatural increase in the global surface temperature, and to other changes in our environment. Acknowledging this observation, the United Nations Framework Convention on Climate Change started an international environmental treaty. This treaty was extended by Kyoto protocol, which was adopted on 11 December 1997. Using the stochastic frontier analysis, we analyze the efficiencies of countries in terms of achieving the lowest greenhouse gas emission levels per GDP output in the years between 1990–2015. Chemotherapy-induced peripheral neuropathy (CIPN) is a prevalent, painful side effect which arises due to a number of chemotherapy agents. CIPN can have a prolonged effect on quality of life. Chemotherapy treatment is often reduced or stopped altogether because of the severe pain. Currently, there are no FDA-approved treatments for CIPN partially due to its complex pathogenesis in multiple pathways involving a variety of channels, specifically, voltage-gated ion channels. Semicarbazide was brought to the forefront of scientific discussions by the scientific community in the early 2000’s as the substance was discovered in several food products from both synthetic and natural sources. Faecal sludge reuse could promote responsible waste management and alleviate resource shortages. However, for this reuse to be carried out at scale, it needs to be financially viable. This paper reviews the financial values of resource recovery from 112 data points from 43 publications from academic and grey literature. The results found 65% of the existing literature is projected rather than being based on observed data from products in practice, with limited studies providing actual experiences of revenue in practice. Halophytes are succulent native plants to the northern region of the Kingdom of Saudi Arabia. They have therapeutic properties and can be potentially useful as a new source of bioactive compounds. Our study aims to investigate the chemical composition of the volatile fraction of Herb and Flowers of Mesembryanthemum forskahlei by the application of Gas Chromatography method. Our study showed that palmetic acid (420mg/ kg) and squaline (352,32mg/kg) are the main components in the herb. Furthermore, we identified that palmitic acid is the main component of the flower. Like so many autoimmune diseases, the exact cause of systemic lupus erythematosus (SLE) remains unknown. Evidence points to both genetics and environment playing roles in the onset of the disease, but neither acts independent of the other. Genetics are the easier of the two to study with recent advances in the field making it easier to isolate genes shared by individuals with the disease. However, genetic studies reveal that there is almost certainly an environmental component to the development of SLE. © 2020 Purpose: Koetjapic acid is an active compound of a traditional medicinal plant, Sandoricum koetjape. Although koetjapic acid has a promising anticancer potential, yet it is highly insoluble in aqueous solutions. To increase aqueous solubility of koetjapic acid, we have previously reported a chemical modification of koetjapic acid to potassium koetjapate (KKA). Antibiotic resistance is a growing problem with the current global death count topping 700,000. In the United States alone there are 2.8 million antibiotic resistant bacterial (ARB) infections each year and approximately 35,000 deaths. In 2017, 55% of the global population were without safely managed sanitation services. On-site sanitation solutions, such as pit latrines, provide the majority of sanitation coverage across developing countries. Appropriate technologies are required in order to safely empty these latrines without damage to people or the environment. The design of appropriate emptying technologies can be hampered by a lack of knowledge of the mechanical properties of the waste, such as its strength. Tuberculosis is an infectious disease with a common occurrence in population of Azad Jammu and Kashmir. The current study was designed to find out the prevalence of TB among the general population of a less developed District Haveli. Samples were collected from different hospitals and TB centers located in district Haveli and tested through sputum smear microscopy for conformation of positive for TB. Background: A major challenge in implementing personalized medicine in pediatrics is identifying appropriate drug dosages for children. The majority of drug dosing studies have been based on adult populations, often with modification of the dosing for children based on size and weight. However, the growth and development experienced by children between birth and adulthood represents a dynamically changing biological system, with implications for effective drug dosing, efficacy as well as potential drug toxicity. Exercise in humans and animals increases plasma bilirubin levels, but the mechanism by which this occurs is unknown. In the present study, we utilized rats genetically selected for high capacity running (HCR) and low capacity running (LCR) to determine pathways in the liver that aerobic exercise modifies to control plasma bilirubin. The HCR rats, compared to the LCR, exhibited significantly higher levels of plasma bilirubin and the hepatic enzyme that produces it, biliverdin reductase-A (BVRA). The HCR also had reduced expression of the glucuronyl hepatic enzyme UGT1A1, which lowers plasma bilirubin. Recently, bilirubin has been shown to activate the peroxisome …

Collagen prolyl 4-hydroxylase 1 (C-P4H1) is an α-ketoglutarate (α-KG)-dependent dioxygenase that catalyzes 4-hydroxylation of proline on collagen. C-P4H1-induced prolyl hydroxylation is required for proper collagen deposition and cancer metastasis. Therefore, targeting C-P4H1 is considered a potential therapeutic strategy for collagen-related cancer progression and metastasis. However, no C-P4H1 inhibitors are available for clinical testing, and the high content assay is currently not available for C-P4H1 inhibitor screening. In the present study, we developed a high-throughput screening assay by quantifying succinate, a byproduct of C-P4H-catalyzed hydroxylation.
