Black carbon (BC) is a significant component of fine particulate matter (PM2.5) air pollution, which has been linked to a series of adverse health effects, in particular premature mortality. Recent scientific research indicates that BC also plays an important role in climate change. Therefore, controlling black carbon emissions provides an opportunity for a double dividend. This study quantifies the national burden of mortality and morbidity attributable to exposure to ambient BC in the United States (US). We use GEOS–Chem, a global 3-D model of atmospheric composition to estimate the 2010 annual average BC levels at 0.5 x 0.667° …
Impact Of A Diesel Spill On Macroinvertebrate Communities In Ponds In S.E. Alabama, Usa, Katie E. Winkles
Impact Of A Diesel Spill On Macroinvertebrate Communities In Ponds In S.E. Alabama, Usa, Katie E. Winkles
Theses and Dissertations
In this study, macroinvertebrate communities were examined to determine an indication of the water quality in ponds on a wetlands property after a diesel spill. In 2012, a train derailed, and at least 3000 gallons of diesel fuel leached into a small pond of the wetlands. Even after clean-up attempts there remains major concerns that the contamination spread throughout the wetlands. This concern led to the present study. Macroinvertebrates were the chosen focus due to their sensitivity to changes in the environment. Two field trials were conducted in which Hester-Dendy multiplate samplers were placed in ponds to allow macroinvertebrates to …

The number of highly caffeinated products has increased dramatically in the past few years. Among these products, highly caffeinated energy drinks are the most heavily advertised and purchased, which has resulted in increased incidences of co-consumption of energy drinks with alcohol. Despite the growing number of adolescents and young adults reporting caffeine-mixed alcohol use, knowledge of the potential consequences associated with co-consumption has been limited to survey-based results and in-laboratory human behavioral testing. Here, we investigate the effect of repeated adolescent (post-natal days P35-61) exposure to caffeine-mixed alcohol in C57BL/6 mice on common drug-related behaviors such as locomotor sensitivity, drug …

The nearly complete inhibition of Na+/K+ ATPase (NKA) in fetal human lens (FHL) epithelial cells by chelerythrine (CHE), a Bcl-2 homology (BH)3-mimetic quaternary benzophenanthridine alkaloid was proposed to be the consequence of CHE binding at a BH1-like hydrophobic groove at the cytosolic aspect of NKA. This conclusion was based on in silico analysis showing at least two motifs in the N-terminal domain of NKA’s a1 subunit- i) aa 59-71 (ARAAEILARDGPN) and ii) aa 42-48 (DELHRKY) homologous to the BH1 and BH3 motifs of Bcl-2 (B-cell lymphoma) respectively, leading to a novel hypothesis that NKA could interact with these proteins by …

Exploration Of The Srx-Prx Axis As A Small-Molecule Target, Murli Mishra
Theses and Dissertations--Toxicology and Cancer Biology
Lung cancer is a leading cause of cancer-related mortality irrespective of gender. The Sulfiredoxin (Srx) and Peroxiredoxin (Prx) are a group of thiol-based antioxidant proteins that plays an essential role in non-small cell lung cancer. Understanding the molecular characteristics of the Srx-Prx interaction may help design the strategies for future development of therapeutic tools. Based on existing literature and preliminary data from our lab, we hypothesized that the Srx plays a critical role in lung carcinogenesis and targeting the Srx-Prx axis or Srx alone may facilitate future development of targeted therapeutics for prevention and treatment of lung cancer. First, …

Citrate Coated Silver Nanoparticles With Modulatory Effects On Aflatoxin Biosynthesis In Aspergillus Parasiticus, Chandrani Mitra
Theses and Dissertations
The manufacture and usage of silver nanoparticles has drastically increased in recent years (Fabrega et al. 2011a). Hence, the levels of nanoparticles released into the environment through various routes have measurably increased and therefore are concern to the environment and to public health (Panyala, Peña-Méndez and Havel 2008). Previous studies have shown that silver nanoparticles are toxic to various organisms such as bacteria (Kim et al. 2007), fungi (Kim et al. 2008), aquatic plants (He, DorantesAranda and Waite 2012a), arthropods (Khan et al. 2015), and mammalian cells (Asharani, Hande and Valiyaveettil 2009) etc. Most of the toxicity studies are carried …

Background
In Europe, the life cycle of Echinococcus multilocularis is predominantly sylvatic, involving red foxes (Vulpes vulpes) as the main definitive hosts and rodents such as muskrats and arvicolids as intermediate hosts. The parasite is the etiological agent of human alveolar echinococcosis, a malignant zoonotic disease caused by the accidental ingestion of eggs shed by definitive hosts in their feces. The aims of this study were to investigate the prevalence of E. multilocularis in red foxes and raccoon dogs (Nyctereutes procyonoides) and to study the environmental factors favoring the perpetuation of the parasite in Latvia. …
Toxicity Of Engineered Nanomaterials To Plant Growth Promoting Rhizobacteria, Ricky W. Lewis
Toxicity Of Engineered Nanomaterials To Plant Growth Promoting Rhizobacteria, Ricky W. Lewis
Theses and Dissertations--Plant and Soil Sciences
Engineered nanomaterials (ENMs) have become ubiquitous in consumer products and industrial applications, and consequently the environment. Much of the environmentally released ENMs are expected to enter terrestrial ecosystems via land application of nano-enriched biosolids to agricultural fields. Among the organisms most likely to encounter nano-enriched biosolids are the key soil bacteria known as plant growth promoting rhizobacteria (PGPR). I reviewed what is known concerning the toxicological effects of ENMs to PGPR and observed the need for high-throughput methods to evaluate lethal and sublethal toxic responses of aerobic microbes. I addressed this issue by developing high-throughput microplate assays which allowed me …

The Effects Of Manufactured Nanomaterial Transformations On Bioavailability, Toxicity And Transcriptomic Responses Of Caenorhabditis Elegans, Daniel L. Starnes
Theses and Dissertations--Plant and Soil Sciences
In recent decades, there has been a rapid expansion in the use of manufactured nanoparticles (MNPs). Experimental evidence and material flow models predict that MNPs enter wastewater treatment plants and partition to sewage sludge and majority of that sludge is land applied as biosolids. During wastewater treatment and after land application, MNPs undergo biogeochemical transformations (aging). The primary transformation process for silver MNPs (Ag-MNPs) is sulfidation, while zinc oxide MNPs (ZnO-MNPs) most likely undergo phosphatation and sulfidation. Our overall goal was to assess bioavailability and toxicogenomic impacts of both pristine, defined as-synthesized, and aged Ag- and ZnO-MNPs, as well as …

Medical Interns', Residents' And Attending Physicians' Attitudes Towards Integrative Medicine And Recommended Treatments For Patients With Psoriasis, Jennifer L. Pacyon
PCOM Psychology Dissertations
Background: Psoriasis is a multifactorial disease that effect approximately 7.5 million Americans. There are a variety of traditional treatments for psoriasis (e.g. topical medications, biologics) that for many, are effective in combating the disease. However, traditional treatments often have aversive side effects and may not be appropriate for every patient (e.g. pregnant women). Additionally, the efficacy of these medications are variable leaving some looking for adjunctive treatment options. There is increasing evidence to support the use of adjunctive treatments for psoriatic patients (e.g. psychotherapy, mindfulness). However, research is variable in regards to medical professionals’ attitudes towards integrative medicine (IM) as …

Piperlongumine (piplartine, 1) is a small molecule alkaloid that is receiving intense interest due to its antiproliferative and anticancer activities. We investigated the effects of 1 on tubulin and microtubules. Using both an isolated tubulin assay, and a combination of sedimentation and Western blotting, we demonstrated that 1 is a tubulin-destabilising agent. This result was confirmed by immunofluorescence and confocal microscopy, which showed that microtubules in MCF-7 breast cancer cells were depolymerised when treated with 1. We synthesised a number of analogues of 1 to explore structure-activity relationships. Compound 13 had the best cytotoxic profile of this series, …
A Glimpse At Media-Framing Effects On Place Attachment In Rural Northwest Montana, Gregory D. Bruns
A Glimpse At Media-Framing Effects On Place Attachment In Rural Northwest Montana, Gregory D. Bruns
Communication & Leadership Dissertations and Theses
This study examined how outdated media framing can disturb rural place attachment. Evaluating this concept against the backdrop of south Lincoln County, Montana, which is nearing the end of a years-long EPA Superfund cleanup, over 100 regional visitors were surveyed to learn if media framing affected place-attachment sentiments. The investigation revealed that existing news stories, some up to 15 years old, may be playing a lingering and consequential role in attracting first-time destination seekers to the region, in support of framing theory. Furthermore, these findings also lend credence to the significance of sound Internet search-engine optimization practices, and provide an …

Silver nano particles (AgNPs) are the most frequently used nanomaterials (NMs) in industrial and biomedical applications, and extensive knowledge about their properties is fast accumulating. In addition, functionalization of nanoparticles significantly impacts their properties and potential applications. In spite the urge to produce, investigate and apply them, the toxicity of nanoparticles and their interaction with blood components, such as erythrocytes, has been insufficiently investigated. Here, we report on the effect of AgNPs in cellular function, and particularly on ion transport in human red blood cells (HRBCs). Two methods of synthesis were used for the functionalization of AgNPs, Creighton, and Lee-Meisel. …

Identification Of Methylene Diphenyl Diisocyanate Thermal Degradation Products In A Generation Chamber By Liquid Chromatography Coupled With Tandem Mass Spectrometry, Sébastien Gagné, Yves Cloutier
Études primaires
Isocyanate thermal degradation characterization by liquid chromatography coupled with electrospray tandem mass spectrometry has been performed to elucidate the methylene diphenyl diisocyanate (MDI) thermal degradation structure emitted in a generation chamber using a temperature between 50°C and 180°C to produce MDI vapors. [M+H]+ ions containing an isocyanate functional group were studied by tandem mass spectrometry. The [M+H]+ ion analyses based on the combination of full scans and precursor ion scans were useful for identifying all structures. The compounds emitted were identified and validated as a mixture of compounds containing amine and isocyanate functions. Residual MDI, methylene diphenyl amino-isocyanate, …

Implementation And Evaluation Of An Analytical Method For A Novel Derivatizing Agent To Measure 4,4'-Methylene Diphenyl Diisocyanate Atmospheres, Silvia Puscasu, Simon Aubin, Mark Spence, Sébastien Gagné
Études primaires
Accurate measurement of 4,4'-methylene diphenyl diisocyanate (MDI) atmospheres is a challenge since the molecule is both chemically reactive and likely to be present in aerosol form when heated and sprayed because of its low vapor pressure. Meeting this challenge requires optimizing both the sampling device used and the derivatization agent employed to stabilize the isocyanate functional group. This study describes the use of a novel derivatization reagent for isocyanate sampling to address the challenge of MDI aerosol exposure sampling. Like most conventional derivatizing agents for isocyanates, 1,8-diaminonapthalene (DAN) reacts with isocyanate functional groups to form a urea. However, unlike other …

Aberrant intracellular signaling pathway is one of the major driving forces of malignancy through multiple stages of human cancers. Our study demonstrates that in cancer cells, the signaling pathways are profoundly and actively intertwined with each other so they can synergistically affect cell biology, including promoting development of malignancy and compensating the loss of proliferation or survival signals in responses to anti-tumor drug. Moreover, cancer cells can also adopt “non-canonical” mechanisms …
Structural Characterization And Therapeutic Utility Of The Proton-Coupled Folate Transporter, Michael Roy Wilson
Structural Characterization And Therapeutic Utility Of The Proton-Coupled Folate Transporter, Michael Roy Wilson
Wayne State University Dissertations
Folate is a B9 vitamin essential to DNA synthesis. The proton-coupled folate transporter (PCFT) is a newly discovered proton/folate symporter with an acidic pH optimum and broad expression across a variety of solid tumor types, with limited expression in normal tissues. Several antifolate molecules have been developed as cancer therapeutics, although these classical antifolates display numerous off-target effects due to transport by the ubiquitous reduced folate carrier (RFC). In this dissertation, we determine the roles of multiple PCFT structure/function domains, and develop PCFT-specific antifolates to target solid tumors. We utilize substituted cysteine accessibility methods (SCAM) to identify a novel reentrant …

From Seed To Sky: Impacts Of Explosive Compounds On Vegetation Across Spatial And Developmental Scales, Stephen M. Via
Theses and Dissertations
Explosive compounds are broadly distributed across the globe as a result of nearly two centuries of munitions use in warfare and military activities. Two explosive compounds have seen disproportionate use; RDX (hexahydro-1,3,5-trinitro-1,3,5-triazine) and TNT (2-methyl- 1,3,5-trinitrobenzene), being the most commonly found explosives in the environment. The effects of explosives on biota have been studied in great detail; however, there is a general lack of understanding with regard to broader ecological impacts of these contaminants. My dissertation objective was to follow the impacts of explosive compounds on vegetation across scales. Impacts on vegetation at the species scale alter community composition via …
