2,3,7,8-Tetrachlordibenzo-p-dioxin (TCDD) is a potent toxin which inhibits the antibody response of B cells. The 3'IgHRR which is involved in transcriptional regulation of the heavy-chain polypeptide of antibodies is inhibited by TCDD. The hs1,2 enhancer region, isolated from the 3'IgHRR, is also inhibited while the isolated hs4 is activated by TCDD. This project sought to determine if that dichotomy in effects results from interactions at enhancer-specific binding sites for AhR, thought to mediate transcriptional effects of TCDD, and NFκB, a transcription factor involved in B cell activation. Arsenic is a metalloid toxicant that is widely distributed throughout the earth's crust and causes a variety of health and environment problems. As an adaptation to arsenic-contaminated environments, organisms have developed resistance systems. In bacteria and archaea various ars operons encode ArsAB ATPases that pump the trivalent metalloids As(III) or Sb(III) out of cells. In these operons, an arsD gene is almost always adjacent to the arsA gene, suggesting a related function. Addiction is a chronic, relapsing disorder for which strikingly few effective therapies exist, and there are no FDA-approved pharmacotherapies for methamphetamine (Meth) addition. There is an immense need to identify the neurobiological underpinnings of psychostimulant addiction and develop efficacious drug therapies to compliment the current mainstay treatment of behavioral/cognitive therapy.

Re-exposure to cues associated with psychostimulants (e.g., drug paraphernalia) increase neuronal activity and can elicit drug-craving and -seeking; an effect which is profound and long-lasting. A mechanism to disrupt those brain processes which are necessary to maintain the association may reduce the incidence of cue-elicited relapse.

Organophosphate (OP) nerve agents such as sarin, soman, tabun, and O-ethyl S-[2-(diisopropylamino) ethyl] methylphosphonothioate (VX) do not react solely with acetylcholinesterase (AChE). Evidence suggests that a wide range of cholinergic-independent pathways are also targeted, including serine proteases. These proteases comprise nearly one-third of all known proteases and play major roles in synaptic plasticity, learning, memory, neuroprotection, wound healing, cell signaling, inflammation, blood coagulation and protein processing. Inhibition of these proteases by OPs was found to exert a wide range of noncholinergic effects depending on the type of OP, the dose, and the duration of exposure. In Carbon Monoxide (CO) induced Delayed Neurological Sequelae (DNS) clinical signs develop 1 to 6 weeks after CO has cleared the body. The aim of this experiment was to develop a model of CO induced DNS which closely mimics "real world" conditions both in exposure and treatment. The model was challenged with hyperbaric or normobaric oxygen, or room air. Basic behaviors were measured by Open Field test on days 1, 7, 14 post exposure and treatment. No significant difference in behavior was observed between exposed and control animals or between treatment groups. Histological analyses showed no DNA or necrotic damage …

This study determined the tissue-specific effect of adipose stem cells (ASC) within a melanoma environment for development of a cell-based melanoma therapy. Analysis included a subcutaneous B16-F1 melanoma model using thirty-one C57BL/6 wild-type mice. Melanoma xenografts were treated with cell-based therapies of CFDA-SE-labeled human fibroblasts HF20x (control), non-differentiated ndASC or hematopoietic-differentiated HdASC. No tumor regression was observed in presence of cell-based therapies, thus, the HdASC group demonstrated an increase in tumor growth accompanied with an up-regulated macrophage response, and increased angiogenesis. In addition, this group demonstrated a decrease in Melan-A tumor marker and interferon-γ expression suggesting that ASC-supported tumor angiogenesis …

The relationships between population growth rate and the life-cycle traits contributing to it are nonlinear and variable. This has made it difficult for ecologists to consistently predict changes in population dynamics from observations on changes in life-cycle traits. We show that traits having a high sensitivity to chemical toxicants tend to have a low elasticity, meaning that changes in them have a relatively low impact on population growth rate, compared to other life-cycle traits. This makes evolutionary sense in that there should be selection against variability in population growth rate. In particular, we found that fecundity was generally more sensitive …

Current approaches to ecological risk assessment (ERA) are not sufficient to address environmental protection goals stated in current regulations in the European Union, North America and elsewhere. For example, the data used to estimate the likelihood of adverse ecological effects typically include responses of survival, growth, or reproduction of individuals measured under constant and typically favorable laboratory conditions. But these organism-level endpoints are far removed from the ecological features that the process aims to protect (i.e., the long-term persistence of populations of species in space and time under naturally varying field conditions and in the presence of other stressors). Background: The effects of landscape modifications on the long-term persistence of wild animal populations is of crucial importance to wildlife managers and conservation biologists, but obtaining experimental evidence using real landscapes is usually impossible. To circumvent this problem we used individual-based models (IBMs) of interacting animals in experimental modifications of a real Danish landscape. Worldwide, environmental risk assessment strategies are based on the assumption that measuring direct effects of single substances, using a few single species tests, in combination with safety factors correcting for extrapolation inconsistencies, can be used to protect higher levels of biological organization, such as populations and even ecosystems. The fate of the fragrance material, acetyl cedrene (AC), in sediment was examined in a 16 day laboratory experiment using the sediment microbial community subjected to the following treatments: AC (nominal concentration; 0 and 50 μg g−1 dw sediment) and macrofaunal worms (with/without Capitella teleta (formerly Capitella sp. I)). Furthermore effects of AC on microbial respiration in the system were determined by examining CO2 flux. T-RFLP (terminal restriction fragment length polymorphism) was used to analyze PCR (polymerase chain reaction) amplified 16S DNA gene fragments from the sediments to detect changes in the structure and diversity of the bacterial community. …

This article examines the history and current use of the Special 301 program to restrict access to generic medicines in developing countries, specifically the 2009 and 2010 reports released under the Obama Administration. The news for access to medicines advocates is not good overall. Background: Recent studies demonstrate that diverse antidepressant agents increase the cellular production of the nucleolipid CDP-diacylglycerol and its synthetic derivative, phosphatidylinositol, in depression-relevant brain regions. Pharmacological blockade of downstream phosphatidylinositide signaling disrupted the behavioral antidepressant effects in rats. However, the nucleolipid responses were resistant to inhibition by serotonin receptor antagonists, even though antidepressant-facilitated inositol phosphate accumulation was blocked. Could the neurochemical effects be additional to the known effects of the drugs on monoamine transmitter transporters?
