Circadian rhythms are internal biological rhythms that drive oscillations in physiology and metabolism. Maintained at the level of individual cells throughout the body, the circadian clock is necessary for driving tissue-specific temporal programs of energy balance. Though loss of the circadian transcription factor, BMAL1, in the paraventricular nucleus (PVN) of the hypothalamus has revealed its importance in driving rhythms in energy intake and metabolism, its transcriptional functions across time and in individual cell types in the PVN are not well understood. The lateral hypothalamic area (LHA) is an area crucial for coordinating key behaviors such as sleep, wakefulness, stress, and feeding. Within this area in the brain, Melanin concentrating hormone (MCH) neurons are exclusive to the LHA and also contribute to these key homeostatic behaviors. These neurons can be divided into two different subpopulations: MCH neurons that express cocaine amphetamine-regulated transcript (CART) tachykinin-3 receptor (NK3R) and MCH neurons that do not express CART and NK3R. These neuronal subpopulations have been implicated in coordinating key behaviors but have not been well studied.

Most biological processes are regulated by an internal circadian rhythm. This body clock synchronizes daily rhythms in physiology with the environment. Circadian outputs can differ between females and males, but the underlying mechanisms for these differences are unknown. Glucocorticoids, a class of circadian stress hormones in vertebrates, provide a model for studying sexual dimorphisms in circadian timing. Female rodents show a higher mean and peak-to-trough amplitude of corticosterone than males. This is most prevalent during proestrus when estrogen levels are high. To test the hypothesis that male-female differences in daily corticosterone secretion arise within circadian cells in the brain, we …

Feeding results from the integration of both nutritional and affective states, and is guided by complex neural circuitry in the brain. The hypothalamus is a critical center controlling feeding and motivated behaviors. We found that targeted photostimulation of projections from the lateral hypothalamus (LH) to the paraventricular hypothalamus (PVH) in mice elicited voracious feeding and repetitive self-grooming behavior. GABA neurotransmission in the LH->PVH circuit mediated the evoked feeding behavior, and elicited behavioral approach, whereas glutamate release promoted repetitive self-grooming, which was stress-related in nature. Optogenetic inhibition of LHGABA ->PVH circuit reduced feeding after fasting, whereas photostimulation abruptly …

This article, which includes novel unpublished data along with commentary and analysis,focuses on approaches to link transcriptomic, proteomic, and peptidomic datasets mined frombrain tissue to the original locations within the brain that they are derived from using digital atlasmapping techniques. We use, as an example, the transcriptomic, proteomic and peptidomicanalyses conducted in the mammalian hypothalamus. Following a brief historical overview, wehighlight studies that have mined biochemical and molecular information from the hypothalamusand then lay out a strategy for how these data can be linked spatially to the mapped locations in acanonical brain atlas where the data come from, thereby allowing …

Fragile X Syndrome (FXS) is an inherited developmental disorder characterized by disturbances in emotional and social behavior. Our studies have revealed suppressed hippocampal PKCε expression in Fmr1 knockout (KO) mice, the leading model of FXS. To compensate for this deficiency, we stimulated PKCε in neonatal KO mice by administering a selective PKCε activator, dicyclopropyl-linoleic acid (DCP-LA), and studied its effect on ventral hippocampal neurons and a proximal target of the ventral hippocampus, the hypothalamus, which regulates social and emotional behavior. We observed that at postnatal day 18 (P18), vehicle-treated KO mice displayed increased surface localization of the 3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) …
