This study reviews and compares percussion literature pertaining to polyrhythms and scientific literature pertaining to bimanual coordination. There exists a gap in the pedagogical approach to polyrhythms, and there is much disagreement between common instructional methods, especially when considered against the findings of several bimanual coordination studies. The purpose of this study is to reveal insight to the percussion community that the learning of polyrhythms is facilitated by the brain in novel ways, and the uniqueness of this learning process requires a rethinking of the current pedagogical approach. Irritable bowel syndrome (IBS) is a functional gastrointestinal affliction that affects millions around the globe, impacting health-related quality of life (HRQoL). The biopsychosocial model, Folkman and Lazarus’ transactional model of stress, Leventhal’s common sense model of self-regulation health and illness, and Bronfenbrenner’s human ecology theory are relevant to conceptualize interactions among environmental factors, biopsychosocial responses, and health-related quality of life (HRQoL). This quantitative, cross-sectional, complex correlational study sought to provide more understanding of how and whether IBS symptoms of pain, cognitive functioning, and interpersonal functioning serve as mediators between work stress and HRQoL. For women, two of the greatest risk factors for affective disorders are adversity experienced during puberty and later becoming pregnant. We have created a translationally relevant mouse model where we address these complex risk factors. Previously, we discovered that pregnant mice (dams) that experienced chronic variable stress (CVS) during puberty display a blunted hypothalamic-pituitary-adrenal (HPA) response when exposed to an acute stressor. Interestingly, this alteration only first becomes apparent during pregnancy, which is a sensitive period for these effects due to normative neuroendocrine changes. Evidence suggests that rats given long access (LgA) sessions to self-administer (SA) opioids escalate their intake, while also showing greater withdrawal severity and drug-induced reinstatement compared to rats maintained on short access (ShA) daily SA sessions. Little is known about the neural changes that occur during opioid escalation that may impact withdrawal and relapse. Past work examining opioid SA using ShA sessions in rodents has identified the central amygdala (CeA) as an area of interest that becomes hyperactive in acute withdrawal and may be involved in the incubation of craving that occurs after protracted withdrawal. Although people have smoked cannabis for millennia, administration of cannabis by heating the dried plant material or distilled extracts to a temperature just below the combustion point (i.e., vapourization or "vaping") has rapidly increased over the past few years (Javadi-Payder, Cole, and Taffe, 2018; Manwell, Charchoglyan, Brewer, Matthews, Heipel, and Mallet, 2014). Cue reactivity is an important predictor of addiction course and relapse. However, cue reactivity is only observed after an addiction develops. As such, it is unclear to what degree cue reactivity represents a state stemming from the addiction process versus a trait-like propensity towards developing cue-reward associations. Work in animal models has pointed to important individual differences in trait-like inclination to attribute incentive salience to reward-predictive cues that is associated with addiction-relevant behavioral and neurobiological features. In this dissertation, I aim to advance our understanding of human agency and how it is computed. First, I probe the limits of what behavior we can and cannot produce agentically. Secondly, I explore microdosing with psychedelic drugs as a method of perturbing and studying changes to the sense of agency. Thirdly, I use transcranial magnetic stimulation to induce body movements and investigate how the sense of agency is affected. Finally, I summarise my findings and synthesize insights to further our understanding human agency.

Adolescent stress exposure increases the likelihood of alcohol misuse and alcohol use disorder (AUD) in adulthood; however, it is not clear how genetic and environmental factors interact to increase risk. This study examined how adolescent social isolation affects adult binge-like ethanol drinking and levels of the stress hormone corticosterone in male and female mice with a genetic predisposition toward high alcohol preference (HAP). Twenty-eight HAP mice were separated into group-housed (GH) and socially isolated (SI) conditions (n = 13, 13). Binge drinking was assessed using a drinking in the dark (DID) procedure. Spatial information is known to be encoded in the hippocampus, and small changes in the environment can alter the way that it is represented by our hippocampal place cells in a process called remapping. Hearing is an important sense that can be used to orient ourselves and react to the environment accordingly. Major depressive disorder (MDD) is a leading cause of disability worldwide, with a lifetime prevalence rate of approximately 20%. Inadequate pharmacological treatment methods for MDD are a significant debilitating factor. Patient estimates suggest that the treatment resistance rate for pharmacological interventions is over 30%. Postmortem analyses of human tissue of individuals diagnosed with MDD have shown an increase in Poly (ADP-ribose) polymerase 1 (PARP-1) mRNA gene expression in prefrontal cortical white matter when compared to psychiatrically normal brain tissue. The present study experimentally investigated sex differences in lateral hypothalamic glucose concentrations following cumulative dosing of methamphetamine. Male (n=17) and female (n=11) rats were surgically implanted with enzyme-based glucose biosensors in the lateral hypothalamus. Rats then received a saline injection followed by four methamphetamine doses (0.025 mg/kg, 0.05 mg/kg, 0.1 mg/kg, and 0.2 mg/kg) spaced 55 minutes apart. We found that while there was a general decrease in glucose baseline throughout the study for both sexes, the magnitude of the decrease was significantly greater in females compared to males. In the target article Crump et al. present 8 criteria to assess whether decapods experience pain. Four of these -- sensory integration, motivational trade-offs, flexible self-protection, and associative learning -- could be used to assess sentience in general. In this commentary I discuss difficulties with using these criteria to provide evidence of sentience in decapods, particularly if this evidence is to change public opinion and policies. Crump et al. define eight criteria indicating sentience in animals, with a focus on pain. Here, we point out the risk of false negative or false positive diagnoses of pain. Criteria of different levels of inclusivity are useful for using the precautionary principle in animal welfare considerations, and for more formal scientific evidence of pain. We suggest tightening the criteria -- from more general evidence of sentience to pain alone -- because crucial evidence for animal welfare decisions might otherwise be missed for animals subjected to invasive and injurious procedures.

Background: Post-traumatic stress disorder (PTSD) is a debilitating disorder defined by the onset of intrusive, avoidant, negative cognitive or affective, and/or hyperarousal symptoms after witnessing or experiencing a traumatic event. Previous voxel-based morphometry studies have provided insight into structural brain alterations associated with PTSD with notable heterogeneity across these studies. Furthermore, how structural alterations may be associated with brain function, as measured by task-free and task-based functional connectivity, remains to be elucidated.

Anxiety disorders can impair cognition and emotional control, particularly in the face of novel situations, which is concerning due to the high prevalence of such diagnoses. Exposure to unpredictable chronic stress can provide a way to induce anxiety-like behaviors in rodents by disrupting the stress response systems, including the HPA axis and autonomic nervous system. Some models of associative learning attempt to explain effects such as blocking and unblocking as reflecting a prediction error (PE): Associative strength, and thereby learning, are said to be greater when the unconditioned stimulus (US) received following a cue deviates from one’s expectations. Some models of PE-motivated behavior and PE-relevant brain activity represent the US as a single quantitative variable for outcome value, and that learning therefore only occurs when outcome value differs from expectations. Chromosome 22q11.2 deletion syndrome (22q11.2DS) results from a hemizygous deletion located on the long arm of chromosome 22. The most common deletion sizes affect between 30 and 90 genes. Individuals with 22q11.2DS may develop serious developmental and psychiatric disorders. The phenotype is highly variable, however, and may be influenced by allelic variation of the retained copies of genes covered by the deletion. Post-traumatic stress disorder (PTSD) is a trauma-related disorder characterized by intense fearful memory formation. Women are twice as likely as men to develop PTSD, indicating there may be sex differences in the underlying neural circuits. Given that serotonin (5-HT) dysfunction is implicated in PTSD, and 5-HT modulates fear learning, we investigated whether there are sex differences in the modulation of fear learning by 5-HT. We administered the selective serotonin reuptake inhibitor (SSRI) citalopram (20mg/kg or 10m/kg, i.p.) once to male and female mice prior to auditory fear conditioning and tested the effects on fear memory the next day. Depression is a debilitating disorder that can cause motivational deficits such as psychomotor retardation, anergia, apathy, and fatigue. Recent research indicates that these motivational deficits, and potential pathways of therapeutic intervention, can be studied in animal models involving rats and mice. Treatments with the VMAT-2 inhibitor tetrabenazine (TBZ) and cytokine interleukin-1ß (IL-1ß) can create a low-effort bias and impair effort-related motivation (Nunes et al. 2013, 2014). A number of high-affinity DA transport inhibitors such as d-amphetamine, methylphenidate, and cocaine can restore extracellular DA, albeit with the cost of undesirable effects such as high abuse liability.
