Population Coding And Spectrotemporal Tuning In The Owl’S Optic Tectum, Olivia K. Murray, Roland Ferger, Michael V. Beckert, Keaunu Shadron, José L. Peña

Undergraduate Research Symposium Posters

Barn owls (Tyto furcata) are a model organism to study sound location and provide access to a neural population representing auditory space. They use interaural time differences (ITD) and interaural level differences (ILD) to localize sound. Neurons in the barn owl’s optic tectum (OT), a part of the midbrain, which respond to combinations of these binaural cues, form a neural map of auditory space which supports sound-orienting behavior. Understanding population responses in the auditory system involves identifying the frequency components of acoustic signals which evoke spikes. Together, these frequency components and the time between their occurrence in the stimulus and …

The Effects Of Long-Term Treadmill Exercise Training On Amyloid-Beta Plaque Levels And Behavioral Profile In 15-Month Tg2576 Mice, Riya R. Thomas

Graduate Theses/Dissertations

Alzheimer’s disease (AD) is a progressive, neurodegenerative disease that is characterized by progressive loss of memory and cognitive function. The causative factors of this disease include modifications in neuronal cell metabolism, such as the progression of neurotic injury by amyloid beta (Aβ), and tau tangle deposition in the brain. Recent studies show that brain atrophy and other pathologic conditions are responsible for dementia in older individuals (80 years of age or older). Non-pharmacological interventions may have a role in both the prevention and slowing disease progression in AD. The role of exercise training in disease prevention has been evaluated in …

Spinal Cord Trauma: An Overview Of Normal Structure And Function, Primary And Secondary Mechanisms Of Injury, And Emerging Treatment Modalities, Daniel Morin May 2018

Spinal Cord Trauma: An Overview Of Normal Structure And Function, Primary And Secondary Mechanisms Of Injury, And Emerging Treatment Modalities, Daniel Morin

Senior Honors Theses

The structures of the spinal cord and vertebral column are designed to provide flexibility, while still providing ample protection for the spinal cord deep within. While it does offer remarkable protection against most routine trauma, the spinal cord is still vulnerable to high-force etiologies of trauma and may become damaged as a result. These events are referred to as primary injury. Following the initial injury, the body’s own physiological responses cause a cascade of deleterious effects, known as secondary injury. Secondary injury is a major therapeutic target in mitigating the effects of spinal cord injury (SCI), and much research is …

Genetic Testing Practices Of Genetic Counselors, Geneticists, And Pediatric Neurologists With Regard To Childhood-Onset Neurogenetic Conditions, Sara Wofford

Dissertations and Theses (Open Access)

Identifying genetic diagnoses for neurological conditions with a considerable hereditary component, such as autism spectrum disorder (ASD), intellectual disability, and epilepsy, is critical to providing proper medical management for these patients and their families. However, many patients with these conditions are not tested appropriately or receive no genetic testing at all. The current study was designed to characterize the genetic testing practices of the providers most likely to evaluate or order genetic testing for these patients: pediatric neurologists, geneticists, and genetic counselors. The study noted significant variance between the testing strategies selected by pediatric neurologists compared to those of geneticists …

A Role Of Vitamin B2 In Reducing Amyloid-Beta Toxicity In A Caenorhabditis Elegans Alzheimer’S Disease Model, Muhammad Tukur Ameen May 2018

A Role Of Vitamin B2 In Reducing Amyloid-Beta Toxicity In A Caenorhabditis Elegans Alzheimer’S Disease Model, Muhammad Tukur Ameen

Electronic Theses and Dissertations

Alzheimer’s disease (AD) is associated with amyloid-beta peptide deposition and loss of mitochondrial function. Using a transgenic C. elegans AD worm model expressing amyloid-beta in body wall muscle, we determined that supplementation with either of the forms of vitamin B2, flavin mononucleotide (FMN) or flavin adenine dinucleotide (FAD) protected against amyloid-beta mediated paralysis. FMN and FAD were then assayed to determine effects on ATP, oxygen consumption, and reactive oxygen species (ROS) with these compounds not significantly improving any of these mitochondrial bioenergetic functions. Knockdown of the daf-16/FOXO transcriptional regulator or the FAD synthase enzyme completely abrogated the …

The Combined Neuropharmacology And Toxicology Of Major 'Bath Salts' Constituents Mdpv, Mephedrone, And Methylone, Serena Allen May 2018

The Combined Neuropharmacology And Toxicology Of Major 'Bath Salts' Constituents Mdpv, Mephedrone, And Methylone, Serena Allen

Electronic Theses and Dissertations

The synthetic cathinones, 3,4- methylenedioxypyrovalerone (MDPV), 4-methylmethcathinone (mephedrone), and 3,4- methylenedioxymethcathinone (methylone), gained worldwide notoriety as the psychoactive components of ‘bath salts;’ a marketing term used to circumvent federal drug laws and permit their legal sale. Previous studies have shown that these drugs share pharmacological characteristics with cocaine and the amphetamines, however, descriptions of their neurotoxic properties are limited. Moreover, while forensic analysis has revealed that the most frequently abused bath salts ‘brands’ contain binary and ternary mixtures of MDPV, mephedrone, and methylone, the majority of preclinical research has focused on explicating the individual effects of these drugs. Therefore, the …

Diel Rhythmicity Found In Behavior But Not Biogenic Amine Levels In The Funnel-Web Spider Agelenopsis Pennsylvanica (Araneae, Agelenidae), Alexander E. Demarco May 2018

Diel Rhythmicity Found In Behavior But Not Biogenic Amine Levels In The Funnel-Web Spider Agelenopsis Pennsylvanica (Araneae, Agelenidae), Alexander E. Demarco

Electronic Theses and Dissertations

Quantifying individual differences in behavior and the extent that behavior is influenced by circadian control is of paramount importance in behavioral ecology. In addition, the proximate mechanisms underlying behavior are also critical in order to obtain a more complete picture of how behavior evolves. Biogenic amines (BAs) are simple nitrogenous compounds derived from amino acids and have been consistently and extensively linked to behavior. For this study, we analyzed temporal patterns of BAs in relation to the antipredator (boldness) and aggressive behavior in female Agelenopsis pennsylvanica, a funnel-web spider. Using HPLC-ED, we compared behavioral responses to temporal patterns of …

Examining The Appropriate Recovery Interval Following Maximal Exertion For Baseline Computerized Neurocognitive Testing (Cnt), Samantha Mohler May 2018

Examining The Appropriate Recovery Interval Following Maximal Exertion For Baseline Computerized Neurocognitive Testing (Cnt), Samantha Mohler

Graduate Theses and Dissertations

Background: Computerized neurocognitive testing is part of the recommended multi-faceted approach to SRC assessment. Prior research has suggested that maximal exertion negatively effects CNT test scores. Purpose: To identify the appropriate timing of the administration of CNT following maximal exertion in healthy college-aged students. Study Design: Random cross-over, repeated measures design. Methods: Participants will be administered CNT on four different visits, with at least one week between administrations. A VO2 max treadmill test will be performed before CNT administration during three of the four trials. Following the VO2 max test, participants will rest for <2 minutes (immediate), 10-minutes, or 20-minutes before taking CNT. The fourth trial, without maximal exertion preceding CNT administration, will serve as the control. All trials will be randomly-counterbalanced to negate practice effects. RESULTS: There was a significant within-subjects effect for prescribed post-exertion recovery intervals on total symptom scores (Wilks λ = .62, F [3, 23] = 4.64, p = .01, η2= .38). Total symptom scores were significantly higher at the immediate (p < .002), 10-minutes (p = .018), and 20-minutes (p = .011) post-exertion recovery intervals compared to baseline. Additionally, a significantly positive within-subjects effect for prescribed post exertion recovery was observed for processing speed (p=.009, Wilks λ = .60, F [3, 27] = 5.9, η2 = .396). No significant effect was observed for visual memory (p = .07), verbal memory (p = .06), or reaction time (p = .40). CONCLUSION: Baseline symptom scores were negatively influenced processing speed was enhanced by maximal exertion. These changes continue to be elevated 20 minutes post-exertion. Moreover, cognitive performance was not significantly impaired following maximal exercise. To obtain more accurate baseline symptom scores, and allow processing speed composites to return to normal, sports medicine professionals should wait at least 20 minutes following maximal exertion before administering CNT.

Arsenic Toxicity And Mitochondrial Metabolism In Astrocytes, Reilly Grealish May 2018

Arsenic Toxicity And Mitochondrial Metabolism In Astrocytes, Reilly Grealish

Honors Program: Senior Projects (Public)

The various cells that support neurons in both the central and peripheral nervous systems are known collectively as neuroglia, or glia. Astrocytes, a type of glial cell found exclusively in the central nervous system (i.e. the brain and spinal cord), are particularly abundant and serve many functions. One of the most important is the role these cells play in the brain as a defense against harmful substances, many of which can be found in the environment and make their way into the human body as xenobiotic substances. Elemental arsenic in its inorganic form occurs naturally throughout the Earth’s crust, but …

Combining Microdialysis And Electrophysiology In Cerebral Cortex To Delineate Functional Implications Of Acetylcholine Gradients, Tazima Nur May 2018

Combining Microdialysis And Electrophysiology In Cerebral Cortex To Delineate Functional Implications Of Acetylcholine Gradients, Tazima Nur

Graduate Theses and Dissertations

The neuronal network in cerebral cortex is a dynamic system that can undergo changes in collective neural activity as the organism changes its behavior. For example, during sleep and quiet restful awake state, many neurons tend to fire together in synchrony. In contrast, during alert awake states, firing patterns of neurons tend to be more asynchronous, firing more independently. These changes in population-level synchrony are defined as changes in cortical state. Response to sensory input is state-dependent, i.e., change in cortical state can impact the sensory information processing in cortex and introduce trial-to-trial variability in response to the same repeated …

Sanders-Brown Center on Aging Faculty Publications

Neurovascular integrity, including cerebral blood flow (CBF) and blood-brain barrier (BBB) function, plays a major role in determining cognitive capability. Recent studies suggest that neurovascular integrity could be regulated by the gut microbiome. The purpose of the study was to identify if ketogenic diet (KD) intervention would alter gut microbiome and enhance neurovascular functions, and thus reduce risk for neurodegeneration in young healthy mice (12–14 weeks old). Here we show that with 16 weeks of KD, mice had significant increases in CBF and P-glycoprotein transports on BBB to facilitate clearance of amyloid-beta, a hallmark of Alzheimer’s disease (AD). These neurovascular …

Reconstitution Of Gabaergic Postsynapses In Host Cells, Karthik Kanamalla Apr 2018

Reconstitution Of Gabaergic Postsynapses In Host Cells, Karthik Kanamalla

Honors Scholar Theses

Type A GABA receptors (GABAARs) can be found embedded in postsynaptic membranes or in a variety of extrasynaptic locations. Receptors with synaptic function are recruited to the postsynapse by submembranous scaffolds composed of gephyrin and collybistin (CB). This study was aimed at assessing whether the ability to interact with the scaffold differentiates synaptic from non-synaptic receptors. Using HEK293 cells as an expression system, and indirect immunofluorescence (IF), co-localization of extrasynaptic receptors α1β3δ and α4β3δ with the CB-gephyrin scaffold was assessed and compared with that of the synaptic receptor α1β3γ2. Results indicated that both extrasynaptic receptors were able to colocalize with …

Psychopathy To Altruism: Neurobiology Of The Selfish–Selfless Spectrum, James W. H. Sonne, Don M. Gash Apr 2018

Psychopathy To Altruism: Neurobiology Of The Selfish–Selfless Spectrum, James W. H. Sonne, Don M. Gash

Neuroscience Faculty Publications

The age-old philosophical, biological, and social debate over the basic nature of humans as being “universally selfish” or “universally good” continues today highlighting sharply divergent views of natural social order. Here we analyze advances in biology, genetics and neuroscience increasing our understanding of the evolution, features and neurocircuitry of the human brain underlying behavior in the selfish–selfless spectrum. First, we examine evolutionary pressures for selection of altruistic traits in species with protracted periods of dependence on parents and communities for subsistence and acquisition of learned behaviors. Evidence supporting the concept that altruistic potential is a common feature in human populations …

Efficient Reduced Bias Genetic Algorithm For Generic Community Detection Objectives, Aditya Karnam Gururaj Rao Apr 2018

Efficient Reduced Bias Genetic Algorithm For Generic Community Detection Objectives, Aditya Karnam Gururaj Rao

Theses

The problem of community structure identification has been an extensively investigated area for biology, physics, social sciences, and computer science in recent years for studying the properties of networks representing complex relationships. Most traditional methods, such as K-means and hierarchical clustering, are based on the assumption that communities have spherical configurations. Lately, Genetic Algorithms (GA) are being utilized for efficient community detection without imposing sphericity. GAs are machine learning methods which mimic natural selection and scale with the complexity of the network. However, traditional GA approaches employ a representation method that dramatically increases the solution space to be searched by …

Factors That Determine Spreading Depolarization Propagation In Brain Slices, Linday Selters Apr 2018

Factors That Determine Spreading Depolarization Propagation In Brain Slices, Linday Selters

Biomedical Engineering ETDs

In the US alone, more than 750,000 people had a stroke in 2017, more than 1.7 million reported a traumatic brain injury, and more than 9 million suffered from migraines with aura. While all three of these neurological conditions have vastly different causes and possible outcomes they all have a common phenomenon occurring within the brain. A wave that slowly propagates through grey matter, hitting neurons with a large burst of energy, opening them up to a flood of ions and silencing them for an extended period of time. These waves are known as spreading depolarizations (SD). Within well-nourished tissue, …

Bilateral Carotid Artery Stenosis Causes Unexpected Early Changes In Brain Extracellular Matrix And Blood-Brain Barrier Integrity In Mice, Jill M. Roberts, Michael E. Maniskas, Gregory J. Bix Apr 2018

Bilateral Carotid Artery Stenosis Causes Unexpected Early Changes In Brain Extracellular Matrix And Blood-Brain Barrier Integrity In Mice, Jill M. Roberts, Michael E. Maniskas, Gregory J. Bix

Neuroscience Faculty Publications

Bilateral carotid artery stenosis (BCAS) is one experimental model of vascular dementia thought to preferentially impact brain white matter. Indeed, few studies report hippocampal and cortical pathology prior to 30 days post-stenosis; though it is unclear whether those studies examined regions outside the white matter. Since changes in the blood-brain barrier (BBB) permeability precede more overt brain pathology in various diseases, we hypothesized that changes within the BBB and/or BBB-associated extracellular matrix (ECM) could occur earlier after BCAS in the hippocampus, cortex and striatum and be a precursor of longer term pathology. Here, C57Bl/6 mice underwent BCAS or sham surgeries …

Background: Neuroendocrine tumors (NETs) have a low incidence but relatively high prevalence. Over the last three decades, the incidence of NETs has risen 6-fold in the United States. We conducted an observational study to compare the incidence of NETs reported to the Kentucky Cancer Registry (KCR) versus that reported to Surveillance, Epidemiology, and End Results Program (SEER). We also provide a systematic review of the state of neuroendocrine tumors worldwide, and compare the available global and local published data.

Methods: KCR and SEER databases were queried for NET cases between 1995 and 2015. A detailed literature review of epidemiological data …

Moderate Prenatal Alcohol Exposure Impairs Performance In An Object-Place-Paired-Associate Task, Lilliana May Sanchez Apr 2018

Moderate Prenatal Alcohol Exposure Impairs Performance In An Object-Place-Paired-Associate Task, Lilliana May Sanchez

Psychology ETDs

Memory impairments, including spatial and object processing, are often observed in individuals with Fetal Alcohol Spectrum Disorder. Much attention has been directed towards the hippocampus, which displays significant alterations after moderate prenatal alcohol exposure (PAE). In the present study, we tested a moderate PAE rat model in an object-place-paired-associate (OPPA) task, previously shown to require hippocampal processing. The OPPA task was composed of training rats to discriminate between an identical pair of objects presented in 180° opposite arms of a radial arm maze. Animals were given a total of 10 trials per day over 14 consecutive days of training and …

Inhibition Of Tnf-Alpha Decreases Microglia Activation In Rats Neonatally Treated With Poly I:C, Heath W. Shelton, Russell W. Brown Apr 2018

Inhibition Of Tnf-Alpha Decreases Microglia Activation In Rats Neonatally Treated With Poly I:C, Heath W. Shelton, Russell W. Brown

Appalachian Student Research Forum

Introduction: Current medical treatment for individuals diagnosed with schizophrenia (SCHZ) primarily relies on the inhibition of the dopamine D2 receptor that has been shown to be supersensitive in these patients. Treatment occurs through the use of antipsychotic medication which leads to a number of debilitating dose-dependent side effects, such as weight gain, agranulocytosis, and seizures. Patients diagnosed with SCHZ have also been shown to have increased inflammation in their central nervous system (CNS), particularly within specific brain regions such as the prefrontal cortex and hippocampus. This is in large part due to the interaction between a pro-inflammatory cytokine called …

Unimanual And Bimanual Haptic Shape Discrimination, Catherine Jane Dowell Apr 2018

Unimanual And Bimanual Haptic Shape Discrimination, Catherine Jane Dowell

Masters Theses & Specialist Projects

In the current study 24 younger adults and 24 older adults haptically discriminated natural 3-D shapes (bell peppers, Capsicum annuum) using unimanual (one hand used to explore two objects) and bimanual (both hands used, but each hand explored separate objects) successive exploration. Haptic exploration using just one hand requires somatosensory processing in only one cerebral hemisphere (the hemisphere contralateral to the hand being used), while bimanual haptic exploration requires somatosensory processing in both hemispheres. Previous studies related to curvature/shape perception have found either an advantage for unimanual exploration over bimanual exploration or no difference between the two conditions. In contrast …

Ambiguities In The Efficacy Of Selective Serotonin Reuptake Inhibitors In The Treatment Of Depression, Stephanie Giblin Apr 2018

Ambiguities In The Efficacy Of Selective Serotonin Reuptake Inhibitors In The Treatment Of Depression, Stephanie Giblin

Writing Across the Curriculum

Major depressive disorder (MDD) is the most debilitating mental illness worldwide. Primary treatment of MDD is the administration of selective serotonin reuptake inhibitors (SSRIs). Deemed as the safest psychotropic drug thus far, SSRIs have been shown to reduce depressive symptoms and enhance the quality of life in youth, adult and geriatric populations. However, with rising numbers of individuals taking maintenance doses of SSRIs for five or more years, researchers are beginning to discover severe adverse side effects associated with prolonged treatment. The overall risks of long-term SSRI therapy include tachyphylaxis, tardive dysphoria, structural neurological abnormalities and suicide. SSRIs also have …
