Older adults were tested on a verbal working memory task that used the item-recognition paradigm. On some trials of this task, response-conflict was created by presenting test-items that were familiar but were not members of a current set of items stored in memory. These items required a negative response, but their familiarity biased subjects toward a positive response. Younger subjects show an interference effect on such trials, and this interference is accompanied by activation of a region of left lateral prefrontal cortex. Age-related decline in working memory figures prominently in theories of cognitive aging. However, the effects of aging on the neural substrate of working memory are largely unknown. Positron emission tomography (PET) was used to investigate verbal and spatial short-term storage (3 sec) in older and younger adults. Previous investigations with younger subjects performing these same tasks have revealed asymmetries in the lateral organization of verbal and spatial working memory. Using volume of interest (VOI) analyses that specifically compared activation at sites identified with working memory to their homologous twin in the opposite hemisphere, we show pronounced age differences in this …

Real time, in vivo microvoltammetric studies were performed, using miniature carbon-based sensors, to concurrently detect norepinephrine (NE) release and serotonin (5-HT) release, in 2 separate electrochemical signals, within CA1 region of hippocampus in the freely moving and behaving, male, Sprague Dawley laboratory rat.
2. Concurrently, four parameters of open-field Behavior I.E. Ambulations, Rearing, Fine Movements and Central Ambulatory behavior (a measure of anxiety reduction behavior), were assayed by infrared photobeam detection.

In this study I investigated the effects of 192-lgG saporin injections into the medial septal area.(MSA) and nucleus basalis magnocel/ularis (NBM) on radial arm maze performance in the male rat. The results of the present study reveal that combined injections of 192-lgG saporin into the basal forebrain failed to disrupt RAM performance when compared to vehicle-injected controls. In addition, intraperitoneal injections using a muscarinic receptor blocker, scopolamine, failed to reveal a compensatory response of the cholinergic basal forebrain that may have explained the lack of behavioral effects of 192IgG saporin. Consequently, the results of this study suggest that a selective …

Alzheimer's disease currently afflicts approximately 4 million people in the United States, with 100,000 new cases being reported each year. As post mortem examination of AD patientsI brains has revealed a significant decrease in the number of cholinergic neurons, one approach we have taken is to look at the correlation between the depletion of certain cholinergic markers in animals and the resulting behavioral deficits. Two regions of specific interest are the medial septal area (MSA) and the nucleus basalis magnocellularis (NBM). Neuropathologically, Alzheimer's disease (AD) is characterized by neuritic plaques and neurofibrillary tangles. Evidence has suggested that a protein called B-amyloid (BA) is a major component of the neuritic plaques and may playa role in the neurodegeneration seen in AD. The cellular mechanisms by which BA induces neurotoxicity, however, are still unclear. Recent evidence suggests that the aggregational state of BA may be relevant to its neurotoxicity. Whether portions of the BA protein or the entire sequence produces neurotoxicity in neurons, however, remains a controversy. Auditory imagery for songs was studied in two groups of patients with left or right temporal-lobe excision for control of epilepsy, and a group of matched normal control subjects. Two tasks were used. In the perceptual task, subjects saw the text of a familiar song and simultaneously heard it sung. On each trial they judged if the second of two capitalized lyrics was higher or lower in pitch than the first. The imagery task was identical in all respects except that no song was presented, so that subjects had to generate an auditory image of the song. The results indicated …

The Energetic Costs Of Rough And Tumble Play In The Juvenile Rat, Stephen M. Siviy, Dale M. Atrens

The metabolic costs of rough-and-tumble play behavior were studied in juvenile rats. Using indirect calorimetry, it was determined that energy expenditure during play is increased by 66-104% over the resting metabolic rate, indicating that play accounts for between 2% and 3% of the total daily energy budget of the rat. In a subsequent experiment, food intake and body weight were monitored for three weeks in rats allowed to play for one hour per day and in rats not allowed to play. While the body weights of the two groups did not differ significantly from each other, those rats allowed to …
