Enhancement Of Zika Virus Infection By West Nile And Dengue Virus Antibody Positive Cord-Blood Samples From Mothers In El Paso-Texas, Jeanette Orbegozo
Open Access Theses & Dissertations
The recent Zika virus (ZIKV) outbreak and its association with microcephaly cases and other neurological syndromes are important public health concerns. A phenomenon known as antibody-dependent enhancement (ADE) involves preexisting flavivirus antibodies to a specific flavivirus that enhances the severity of a second infection with a different flavivirus. Since ZIKV is related to flaviviruses like West Nile virus (WNV) and dengue virus (DENV), prior exposure to these viruses could elicit cross-reactive antibodies that may enhance a subsequent ZIKV infection. Therefore, this study aimed to determine if WNV and DENV antibodies from cord blood enhanced ZIKV infection in vitro. Initially, a …
Vertical Transmission Of Zika Virus In Monodelphis Domestica, Mohini Moulick
Vertical Transmission Of Zika Virus In Monodelphis Domestica, Mohini Moulick
Theses and Dissertations
Zika Viral infection in pregnant women may lead to infants born with microcephaly and other neurological complications, making Zika viral research imperative. Prior research demonstrated Monodelphis domestica to be a unique animal model for Zika virus studies, and viral proteins were detected in brain tissue of juveniles after intracerebral inoculation of infants. We wanted to determine if we can detect a viral protein, non-structural protein 1 (NS1), in brain tissue after intraperitoneal inoculation and we found it to be possible through immunohistochemistry (IHC) analysis. We also investigated if inoculated dams (mothers) would give rise to Zika positive pups (natural inoculation), …
Pseudo-Dumbbell And Dumbbell Rna Structures : Modulators Of Zika Virus Translation And Replication, Marissa Eileen Louis
Pseudo-Dumbbell And Dumbbell Rna Structures : Modulators Of Zika Virus Translation And Replication, Marissa Eileen Louis
Legacy Theses & Dissertations (2009 - 2024)
Upon its discovery in 1947, Zika virus has remained in the shadows until its re-emergence in 2015, where Zika virus wreaked havoc on the lives of mothers and their infants around the world. The pathologies related to this virus range from neurological defects, such as Guillain-Barré syndrome, and congenital birth defects, such as Congenital Zika Syndrome, spontaneous miscarriages and fetal demise.Since 2015, this virus has been largely studied, but secondary structure and function of RNA structures at the end of the 3’UTR, namely the pseudo-dumbbell and the dumbbell (DB) structures, remains elusive. Thus, this dissertation addresses several questions pertaining to …
The Role Of Conserved Poly(A) Sequences In The 3'Utr Of Zika Virus Gene Expression, Kristen Kaytes
The Role Of Conserved Poly(A) Sequences In The 3'Utr Of Zika Virus Gene Expression, Kristen Kaytes
Legacy Theses & Dissertations (2009 - 2024)
Following its discovery in 1947, Zika virus garnered little attention in the global stage until 2015, when a rapidly spreading epidemic brought the virus, and its devastating clinical manifestations to light. Zika virus pathologies include Congenital Zika Syndrome, a known causative of microcephaly, as well as spontaneous miscarriages and the development of Guillain Barré syndrome. Despite recent advances in research, the molecular biology of Zika virus is largely unknown.
Rna Modifications As A Novel Mediator Of Viral Infection, Rachel Netzband
Rna Modifications As A Novel Mediator Of Viral Infection, Rachel Netzband
Legacy Theses & Dissertations (2009 - 2024)
Viral epitranscriptomics is a novel field of RNA biology. Chemical groups are appended to RNA nucleotides under different conditions in cells, and I examine these changes using a mass spectrometry-based approach. Various changes are found to occur in post-transcriptional modifications (PTMs) during viral infection. The field primarily studies a small number of PTMS, such as m6A, while I explore a variety of novel modifications and their impact on viral infection. Hepatitis C virus (HCV) infection induces a variety of modification changes, including the novel presence of dimethylcytosine (m5Cm/m44C). One Writer enzyme for this modification is determined to likely be METTL15, …

Globalization and climate change have contributed to the rampant spread of mosquitos from tropical and subtropical climates into more moderate climates, and thus also the spread of a number of vector-borne viruses. As of 2016, the World Health Organization (WHO) estimates that of all human infections world-wide, 16% result from vectors such as flies, ticks, and mosquitos. The spread of such viruses has encouraged expedient evolutionary pressures resulting in more pathogenic viruses that continually pose a serious threat to public health. First discovered in 1947, Zika virus (ZIKV) has only recently presented as a detriment to public health. The importance …
Zika Modulates Arthropod Histone Methylation In Mosquito Cells, Telvin Lee Harrell
Zika Modulates Arthropod Histone Methylation In Mosquito Cells, Telvin Lee Harrell
Biological Sciences Theses & Dissertations
Epigenetics is the heritable series of covalent modifications that affect chromatin structure, gene expression, and protein function. Methylation is one such epigenetic modification that involves the addition of chemical modifying entities, such as methyl groups, on nucleic acids or proteins. Recent studies have reported that Zika virus (ZIKV) modulates methylation of human and viral RNA important for its replication in vertebrate cells. However, little is known whether ZIKV exerts methylation in arthropod vectors. In this study, I show that ZIKV modulates S-adenosyl methionine (SAMe) synthase, an enzyme involved in the production of SAMe, and histone methylation for its survival in …

Trace elements such as copper and cobalt have been associated with virus-host interactions. However, studies to show the effect of conjugation of copper(II) or cobalt(III) metal centers to thiosemicarbazone ligand(s) derived from either food additives or mosquito repellent such as 2-acetylethiazole or citral, respectively, on Zika virus (ZIKV) or dengue virus (serotype 2; DENV2) infections have not been explored. In this study, we show that four compounds comprising of thiosemicarbazone ligand derived from 2-acetylethiazole viz., (E)-N-ethyl-2-[1-(thiazol-2-yl)ethylidene]hydrazinecarbothioamide (acetylethTSC) (compound 1), a copper(II) complex with acetylethTSC as a ligand (compound 2), a thiosemicarbazone ligand-derived from citral (compound 3) and a cobalt(III) complex …

Zika virus (ZIKV) remained largely quiescent for nearly six decades after its first appearance in 1947. ZIKV reappeared after 2007, resulting in a declaration of an international “public health emergency” in 2016 by the World Health Organization (WHO). Until this time, ZIKV was considered to induce only mild illness, but it has now been established as the cause of severe clinical manifestations, including fetal anomalies, neurological problems, and autoimmune disorders. Infection during pregnancy can cause congenital brain abnormalities, including microcephaly and neurological degeneration, and in other cases, Guillain-Barré syndrome, making infections with ZIKV a substantial public health concern. Genomic and …

The unexpected emergence of Zika virus (ZIKV) in the Pacific Islands and Latin America and its association with congenital Zika virus syndrome (CZVS) (which includes microcephaly) and Guillain-Barré syndrome (GBS) have stimulated wide-ranging research. High densities of susceptible Aedes spp., immunologically naive human populations, global population growth with increased urbanization, and escalation of global transportation of humans and commercial goods carrying vectors and ZIKV undoubtedly enhanced the emergence of ZIKV. However, flavivirus mutations accumulate with time, increasing the likelihood that genetic viral differences are determinants of change in viral phenotype. Based on comparative ZIKV complete genome phylogenetic analyses and temporal …
