The rapid rise of e-cigarette use among young adults has created a significant public health challenge, as the popularity of "vaping" has grown faster than our understanding of its long term cellular consequences. Current research indicates that e-cigarette vapor (ECV) induce high levels of oxidative stress and impair the lungs' ability to clear bacterial pathogens. Furthermore, ECV exposure upregulates specific receptors on the surface of airway cells, essentially making it easier for bacteria to adhere to and infect the lungs. Small RNAs (sRNAs) are noncoding RNA molecules in bacteria that function primarily to regulate gene expression, including those associated with stress. The Borchert Lab has identified 173 sRNAs and > 500 sRNAs that are responsive to desiccation and carbon starvation (c-starvation), respectively. Prokaryotic transcription involves an RNA Polymerase (RNAP) that transcribes DNA into RNA. RNAP involves sigma factors, which recognize promoters for the initial steps of RNA synthesis. The Borchert Lab has determined that there are significant overlaps between sRNAs expressed in cells subjected to both short-term durations and long-term durations of carbon starvation. Salmonella can colonize plant tissue effectively. Understanding the molecular mechanisms underlying Salmonella’s interactions with plants is essential for developing effective strategies to reduce its impact on human health and agriculture. Effector proteins, known for their role in modulating host-pathogen interactions, have been implicated in the virulence and pathogenicity of Salmonella. However, their specific functions and contributions to Salmonella’s interactions with plants remain largely unknown. Here, we aim to investigate the functions of Salmonella effector proteins during the colonization of plants.

Mycoplasma pneumonia is a human bacterial pathogen known to induce chronic upper and lower respiratory infections. This bacterium commonly causes acute, chronic infections, such as atypical pneumonia, and spreads through droplet transmission (> 5 microns). M. pneumoniae damages cells by attacking the epithelium of the respiratory tract, resulting in epithelium shedding. Infection of the airway tract by M. pneumoniae will usually express itself in people with chronic symptoms such as coughing, dyspnea, diarrhea, and epiphora. Staphylococcus aureus (S. aureus) is a biofilm-forming pathogen. S. aureus treatment is marked by the development of antibiotic resistance. The public health impact has increased since the emergence of methicillin-resistant S. aureus (MRSA), which has started to show intermediate resistance to vancomycin in MRSA. Nano-second pulse electric fields (nsPEFs) are low-energy and high-power electric pulses, which have been suggested to sensitize pathogens to antibiotics by creating transient pores in the cell membrane. Our combinatorial treatment includes nsPEF pre-treatment and vancomycin post-treatment of MRSA cells. Our results show that MRSA log phase cells had the highest susceptibility to vancomycin. …

During the construction of VCU’s Hermes A. Kontos Medical Sciences Building in April 1994, nineteenth century ancestral remains were found in an abandoned well on the Medical College of Virginia (MCV) campus. This well, now known as the East Marshall Street Well (EMSW), is thought to have been used as a disposal location for cadaver dissection for surgical and other medical waste in the past. Ustilago maydis, the basidiomycete smut-fungus, can infect and cause tumors in corn plants. For this, mating between compatible haploid cells is important. The mating and subsequent dimorphic transition in U. maydis require starvation for nutrients such as nitrogen, in addition to pheromone-receptor interactions between compatible partners. In this research, the CRISPR-Cas9 gene-editing technique was used to create INDEL mutations (sequence insertion or deletion) in the nitrate transporter gene, um03849, in U. maydis. Cystic Fibrosis (CF) is a genetic disorder characterized by faulty ion channels and result in thick mucus accumulation, especially in lungs. Mucus buildup provides ideal conditions for bacterial infections. Pseudomonas aeruginosa (PA) is the second most prevalent bacterium isolated from people with CF and has a high clinical importance. Most CF pathogens form biofilms which make treatment of infections difficult. Biofilms are clusters of cells attached to a surface enclosed in a structured matrix. These structures are a means to provide shelter for bacteria from the environment, especially antibiotics and the immune system. PA alone can form these biofilms, but …

Respiratory failure caused by chronic and recurrent microbial infections is the most common cause of death for people with cystic fibrosis (CF)1, a disease causing the formation of thick mucus in the lungs2. Most bacteria can form biofilms, collections of sessile cells adhered to a surface by a secreted substance. Biofilm-associated cells develop antibiotic resistance at higher rates3. The gram-negative bacteria Sneathia amnii is a poorly-characterized commensal of the female urogenital tract frequently associated with adverse clinical outcomes such as bacterial vaginosis (BV), amnionitis, and preterm labor. To investigate its potential role in virulence, we sought to identify and characterize virulence determinants produced by S. amnii in an effort to better understand the pathogenesis of infectious preterm birth. Through sequencing of the Sn35 genome (type strain of S. amnii), we identified two genes with amino acid sequence similarity and structural similarity to the filamentous hemagglutinin (FHA) protein of Bordetella pertussis and its Type Vb transporter. CRAYFISH PLAGUE IS A MAJOR DISEASE AFFECTING FRESHWATER CRAYFISH IN SOME AREAS OF THE WORLD. THE DISEASE HAS NEVER BEEN REPORTED IN AUSTRALIA. THE SIMULATION EXERCISE THAT WAS UNDERTAKEN AS PART OF THIS PROJECT WAS A RESPONSE TO A FICTITIOUS OUTBREAK OF CRAYFISH PLAGUE.
Funding for this project was provided to enable an exercise simulating an outbreak of crayfish plague to be conducted in Western Australia.
