Gut dysbiosis characterized by reduced abundance of beneficial butyrate-producing bacteria has been independently linked to HIV-1 infection and heavy alcohol drinking. Further, gut dysbiosis results in loss of gut barrier integrity, microbial translocation and host-specific systemic inflammation. Therefore, to evaluate the functional consequences of structural changes in the gut microbiome, integrated data analysis is imperative. In this dissertation, we perform integrated analyses using data from multi-omics platforms to examine the structural and functional features of the gut microbiome of PWH. Perfluorooctanesulfonic acid (PFOS) has been phased out of production in the United States, but because of its resistance to degradation and its persistence in the environment, PFOS can be found in drinking water across America. Oral exposure to PFOS has been linked to adverse developmental, immunological, hepatic, and endocrine health. Oral PFOS exposure may alter the gut microbiota and cause dysbiosis, which is implicated in the pathology of inflammatory diseases like Crohn’s disease, diabetes, and ulcerative colitis. Type 1 Diabetes (T1D) is a polygenic and multifactorial disease, traditionally attributed to genetic susceptibility and diet. Over the past decade, novel studies have placed a higher significance on the role of gut microbiome in T1D pathogenesis. Furthermore, diabetic mouse models have shown higher incidence of T1D in females compared to males, attributed to the differences in gut microbial community structure. Interestingly, female mouse models elicit male-like protection from T1D when transplanted with the male gut microbiome.
