This study investigated changes in the richness and composition of the rumen microbiome in yaks at various developmental stages. Eighteen healthy male yaks at the ages of 5 months (M5), 12 months (M12), and 24 months (M24) had samples of their rumen digesta taken for 16S rRNA gene sequencing. The 11087 amplicon sequence variations (ASVs) in total were found; the M5, M12, and M24 groups had 3585, 1146 and 5545 distinct ASVs, respectively. Alpha diversity indices Chao, Shannon, Sobs, and ace indicated significant differences among the groups. Streptomyces bacteria produce antibiotics that inhibit competing microorganisms. We hypothesized that Streptomyces from the rhizosphere (around plant roots) would be more likely to produce antibiotics than those from bulk soil (further away from roots). Soil samples were collected from rhizosphere and bulk soils in transitional and long-term organic fields of corn and alfalfa. Streptomyces from these soils were cultured on glucose-asparagine agar media. Their antibiotic production was evaluated by testing whether they could inhibit the growth of two target Streptomyces strains. Widespread antibiotic use has led to antibiotic resistance of infectious bacteria. (Young, 2017) As part of the TinyEarth Program, the lab group collected and tested local soil samples for bacteria with hopes of finding novel antibiotic properties. From the soil samples, a bacteria was isolated, subsequently sequenced, and determined to be Pseudomonas parafulva. P. parafulva is a Gram-negative bacterium that has been known to frequent rice paddies of eastern Asian countries and has been collected in select regions of North America, including that of Cache Valley. Intervening proteins (inteins) are translated as subdomains within host proteins and removed through an intein-driven splicing reaction where the flanking sequences (exteins) are joined with a peptide bond. Previously, we developed a self-removing translation reporter for labeling Ebola virus (EBOV). In this reporter, an intein (RadA) containing the fluorescent protein ZsGreen (ZsG) is inserted within the EBOV protein VP30. Upon VP30-RadA-ZsG expression from the viral genome, RadA-ZsG is removed from VP30 through the protein splicing activity of RadA, generating functional, non-tagged VP30 and functional ZsGreen. Introduction

Staphylococcal Scalded Skin Syndrome (SSSS) is a blistering skin condition caused by a toxin-producing staphylococcus species. Epidemiologic studies on pediatric SSSS in the United States are limited, with a reported incidence of 7.67 cases per million children and 45.1 per million infants. Immature renal function, lack of protective antibodies, and increased exotoxin target desmoglein-1 are hypothesized to increase incidence in younger children. We aimed to determine clinical and microbiological characteristics of pediatric SSSS and factors that could help predict disease severity.

The microbiome of the cystic fibrosis (CF) intestine is altered significantly in structure and function beginning in early life. Key taxa associated with health, digestion and immune-training are reduced in abundance, including but not limited to Bacteroides, Faecalibacterium, Akkermansia, Ruminococcus and other short-chain fatty acid producers. Conversely, there is an expansion of microbes with pathogenic potential, such as Escherichia, Veillonella, Pseudomonas and Staphylococcus. The shifts in microbiota composition direct community-wide changes with differences in beta diversity by genotype, whereby children with CF (cwCF) exhibit significantly different gut microbial compositions compared to nonCF children. As antibiotic-resistance becomes an increasing global concern, researchers are looking for alternatives to combat phytopathogenic rhizobacteria – one of which is through the application of phage biocontrol. To elucidate the complex tripartite relationships between phage, bacteria and plant interactions, nine novel phages were isolated from potential phytopathogenic strains of Acidovorax (RBW13) and Brevundimonas (RBW18). These phages were isolated from root samples obtained from the Rocky Branch Creek and the Broad River located in Columbia, SC. Using Pulsed-Field Gel Electrophoresis (PFGE), we identified seven “medium sized” phages (~40 kbp), one “large-sized” phage (~200 kbp), and one “small-sized” phage ( < 25 kbp). Three phages were determined to be lambda-like due to their “ladder-building” patterns on PFGE. Although spot-tests revealed that all phages seemed to have broad host ranges that spanned over multiple genera, plaque-formations must be observed in future experiments to determine if these phages can infect their hosts productively. Seven bacteriophages were studied to determine their host range among Caulobacter and non-Caulobacter bacteria. Recently, Caulobacter-associated phages have been shown to have broader host ranges than previously believed. Thus, it is crucial to thoroughly verify the range of bacteria strains that these phages can infect. To understand the extent of each phage’s host range, phage lysates were spotted on bacterial lawns to determine whether lysis could occur. Host-range analyses in this project conclude that these Dolichocephalovirinae phages were able to infect multiple genera and species of host bacteria. Vibrio vulnificus is a marine bacterial pathogen perpetrating severe wound and gastrointestinal infections in humans. The growing incidence of V. vulnificus infections and the widening ecological range of the species, closely linked to climate change trends, draw attention to an increasing need for methods to combat V. vulnificus infections without further augmenting the threat of antimicrobial resistance. The rising prevalence of antimicrobial resistance (AMR) among Enterobacteriaceae in livestock presents a significant threat to both animal and human health. This study investigates the antibiotic resistance profiles of Enterobacteriaceae isolated from 160 samples collected from the lymph nodes of healthy pigs in Armenia. Fifty isolates were identified, including Escherichia coli (24), Klebsiella spp. (10), Salmonella spp. (6), Raoultella ornithinolytica (4), Pseudomonas aeruginosa (4), and Proteus mirabilis (2). Resistance was evaluated using the disk diffusion method against 22 antibiotics from nine classes. Alarmingly, 94.5% of isolates were resistant to clarithromycin and 90.3% to erythromycin, while resistance to cephalothin and amoxicillin …

Increasing resistance to existing antibiotics has led to a need for new alternative sources. Among the possibilities that need to be explored is the endophytic actinobacteria associated with Ziziphus mauritiana. The present study aimed to evaluate the antibacterial activity of endophytic actinobacteria isolated from Z. mauritiana against five multidrug-resistant bacterial strains. Thirty actinobacteria isolates were obtained from the roots, stem, and leaves of the plant using the serial dilution technique. Therefore, it has been deemed a critical priority pathogen by the World Health Organization (WHO) and the Center for Disease Control (CDC), generating a need for research on its fundamental biology. Geothermal hot springs provide unique microbial habitats characterized by extreme temperatures, variable pH, and diverse geochemical conditions. This study investigates the microbial diversity of three hot spring locations in Diamond Fork Canyon, Utah—Diamond Fork Warm Springs, Fifth Water Hot Springs, and Monk’s Hollow Trailhead—using Oxford Nanopore MinION sequencing of the 16S rRNA gene. Samples were collected in triplicate and analyzed to identify the bacterial composition, with a focus on potential opportunistic pathogens. The aim of the present study was to investigate the effect of adding ethyl lauroyl arginate (ELA), encapsulated in organic nanoparticles, to a semen extender without antibiotics on sperm quality and bacterial load during short-term storage. Three ejaculates from Duroc boars were divided into four equal portions and diluted with Beltsville Thawing Solution to produce semen doses containing 3 billion spermatozoa in 80 ml. Lyme disease, caused by the spirochete Borreliella burgdorferi, is the most common vector-borne infection in North America. A critical determinant of early mammalian infection is outer surface protein C (OspC), a highly variable lipoprotein that is indispensable for spirochete survival but whose precise function remains unresolved. OspC has been proposed to mediate diverse roles, including complement evasion, adhesion, tissue dissemination, and antiphagocytic activity, supporting the view that it is a multifunctional virulence factor. Structural studies have identified two putative ligand-binding domains (LBD1 and LBD2), with LBD2 forming a solvent-exposed crown at the membrane-distal end of the OspC dimer.

The rise in drug-resistant bacteria, together with the decline in antibiotic development, requires new strategies for infectious disease control. Gram-negative pathogens are particularly challenging to combat due to their outer membrane. This study highlights the effectiveness of the silver-containing antimicrobial AGXX® against the Gram-negative bacterium Escherichia coli. AGXX® is a promising silver coating that presumably kills bacteria through the generation of reactive oxygen species (ROS). Prior studies indicated Borrelia burgdorferi SpoVG protein was regulated, SpoVG binds specifically to a number of nucleic acids, and SpoVG binds to the non-variable section of the major virulence factor vlsE. Although a direct transcriptional activator or inhibitor was not elucidated, it is shown that SpoVG is regulated based on cell culture density independent of the growth rate of B. burgdorferi, indicating that SpoVG regulation is outside the scope of the alternative sigma factor RpoS. Using a dCas9 CRISPRi construct to target SpoVG resulted in protein expression trending towards upregulation of the BosR-RpoS regulon, and a marked decrease …

Symbiotic bacteria, known as rhizobia, can colonize the roots of legume plants and then convert atmospheric nitrogen into a usable form by the host through the process called biological nitrogen fixation (BNF). Many of these bacteria produce other plant-growth-promoting benefits, such as nutrient acquisition, phytohormone production, and the modification of plant stress responses. Agricultural inoculants made from these bacteria have helped reduce the need for man-made nitrogenous fertilizers, known causes of pollution and eutrophication. Bacterial two-hybrid (B2H) systems offer a versatile platform for detecting protein-protein interactions (PPIs) in vivo. Originally developed to study bacterial transcriptional regulators, these systems have evolved to support a wide range of applications, including interaction mapping, domain analysis, and high-throughput screening. This review surveys the development and expansion of B2H systems across diverse biological contexts. We highlight technical considerations, platform-specific innovations, and recent adaptations that extend the utility of B2H systems in both prokaryotic and eukaryotic protein interaction research environments. Salmonella enterica is a globally distributed bacterial pathogen that is commonly acquired through consumption of contaminated food and water. The enterica subspecies includes over 1500 unique subvariants, or serovars, that are classified by antigen presentation. Due to its global environmental prevalence, broad host range, and increasing pathogenicity, serovar Typhimurium is of particular concern for human and animal health, as well as food safety. Once in the gastrointestinal tract, Salmonella Typhimurium (STm) exploits host immune mechanisms, implements an arsenal of virulence factors, and outcompetes the microbiota to invade the intestinal epithelium. Pseudomonas aeruginosa is a prominent opportunistic pathogen that is often found in microoxic environments like sites of infection and colony biofilms. P. aeruginosa can grow at oxygen (O2) at concentrations as little as 3 μM and also grows anaerobically by generating energy using nitrate as an alternate electron acceptor. An important way P. aeruginosa adapts to low O2 is through Anr, a transcription factor that upregulates many genes important for microoxic and anoxic fitness.

Klebsiella pneumoniae is the third most common cause of nosocomial infections in the United States. The capsular polysaccharides (CPS) are virulence factors allowing K. pneumoniae to evade host immune systems. The current model of capsule structure indicates that CPS fibrils interact with the external O-antigen of lipopolysaccharide molecules to enhance retention to the bacterial outer membrane. These interactions may influence pathogenicity in clinically relevant strains. This study characterized alterations to LPS and CPS structure, the impact of these changes on capsule retention, membrane permeability, biofilm formation, and macrophage interactions. Clostridium, a genus of anaerobic, spore-forming bacteria, includes clinically and ecologically significant species like Clostridium difficile and Paeniclostridium sordellii. C. difficile causes antibiotic-associated colitis, while P. sordellii is linked to fatal infections and spontaneous abortions. Recent studies found both species in marine sediments, suggesting their role in environmental and wildlife health disturbances, including the 2013 mass mortality of over 300 manatees in the Indian River Lagoon. However, isolating and differentiating P. sordellii is challenging due to the lack of selective media optimized for its growth.
