This report is concerned with the management of waste-waters and subsurface drainage from the City of Las Vegas and its environs and the BMI complex, all in Clark County, Nevada. Gross primary productivity, heterotrophic bacterial numbers, and net phytoplankton densities of seven ponds in Coles County, Illinois, were studied in relation to physical, chemical, and biological habitat variables (light intensity and duration, turbidity, water temperature, pH, dissolved oxygen, sulfur, nitrogen, phosphorus, production, bacteria, and phytoplankton). Ten observations were made for each pond (except where otherwise noted) from 17 June through 25 August 1974. Stepwise multiple linear regression analyses of the data were used in order to determine those environmental factors which were important in predicting (i.e., significantly correlated with) bacterial and phytoplankton densities, and production. This report describes investigations into the physiology of microorganisms that are commonly involved in the oxidation of inorganic sulfur compounds. The metabolic activities of these bacteria play a potent role in several fields of economic importance such as strip mining operations, water pollution, corrosion, metallurgy, petroleum technology and soil fertility processes.
The oxidation of inorganic sulfur compounds was studied in the chemolithotrophs Thiobacillus denitrificans, Thiobacillus A2, Thiobacillus neapolitanus, and a photolithotroph Rhodopseudomonas palustris. Cell suspensions from all of the thiobacilli catalyzed completely the oxidation of sulfide, thiosulfate, and sulfite to sulfate. The oxidation of thiosulfate in R. palustris …

The diurnal vertical movement of zooplankton was first recorded in freshwater lakes by Weismann (1877) in Lake Constance (Bodensee), although Cuvier was credited with observing the migration of Daphniae in 1817 (Gushing, 1955). The migration is best observed in deep oligotrophic lakes and migrations of 50 m per day are described (Worthington, 1931). In addition, two species of chaetognaths are reported to migrate 400 m a day in marine waters off of Lisbon (Waterman and Berry, 1967) while Birge (1895) found little evidence of the phenomenon at all in Lake Mendota, Wisconsin. Phytoplankton samples were collected in Lake Mend 6 times from September 1910 to June 1971 for 8 stations at depths of 0. 3, 5, 10, 20, and 30 m. These samples were processed through a Millipore filter apparatus and 79 planktonic algae were identified. Algal divisions represented were Bacillariophyta, 42 species; Chlorophyta, 18 ; Cyanophyta, 9; Chrysophyta, 3; Cryptophyta, 3; Pyrrophyta, 2; and Euglenophyta, 2. Blue-green algae were dominant in late summer and fall; green algae, diatoms, and, cryptomonads in winter; and green algae in spring. The early summer flora was best represented by the Chlorophyta, Cryptophyta, and Chrysophyta. Palmer's …

The screening of pesticides to determine which were most inhibitory to bacteria was accomplished by testing the effects of 10 pesticides on 9 different organisms using the disk assay method. Results showed that Gram positive bacteria were more sensitive to all pesticides tested than were the Gram negative bacteria. In addition, the hormone herbicides were found to be the most inhibitory to these bacteria and 2,4-D (2,4-dichlorophenoxyacetic acid), a hormone herbicide, was chosen for subsequent in vivo studies.

Phytoplankton successions, applications of the general growth equation, and physical measurements have been employed to investigate events occurring at the interface between industrial and sewage effluent contained in Las Vegas Wash and the waters of Lake Mead, Nevada. The data indicate that the entering waters tend to form a density current interrupted at intervals by dynamic effects generated in the lake. The dynamic relationships described here for the spring months suggest that a much more thorough understanding of physical, chemical and biological interactions is necessary to permit solution of the numerous problems of Las Vegas Bay discussed or alluded to …

The temporal and spatial changes in chemical and biological properties of Lake Mead have been investigated, thereby indicating the sources of water pollution and the time of highest pollution potential. Planktonic organisms have been shown to indicate the presence of water problems. Macro- and micro-nutrient analyses have shown that primary productivity is not inhibited by limiting concentrations. A mathematical model has been developed, tested with one set of independent data, and shown worthy of management utility. Although the model works very well for the Lake Mead area, the physical reality of the Multiple Linear Regression equation should be tested on …

Progressive increases in concentration of dissolved solids in the Colorado River water from Lake Powell to Imperial Dam seem to alter plankton dynamics and biological productivity of the river. Also, changes in biological productivity and micronutrients concentrations occur within the same reservoir. Development of a digital simulation model to predict micronutrients concentrations and biological productivity is necessary for diagnosing changes in plankton population and effluent-carrying capacity of the system. The microflora of Cane Branch of Beaver Creek in McCreary County, Kentucky, which drains an area that was strip-mined between 1955 and 1959, was studied and compared with that of Helton Branch which drains a comparable area where there has been no mining. Differences include: the establishment of Ferrcbacillus ferrooxidans, for which procedures were developed for direct colony isolation from the stream; fewer saprophytic bacteria; more numerous and more diversified filamentous and unicellular fungi; and characteristic differences in algal flora. Representatives of 42 genera of filamentous fungi were identified. Of these, 21 were isolated only from Cane Branch. Representatives of …

In induction experiments with naturally occurring and artificially lysogenic strains of S. potsdam, cells were irradiated in buffer, transferred to complete medium, and then assayed for free phage. Irradiation was followed by half hourly samples for free phage from the growing populations of irradiated cells. For an estimation of burst sizes, the irradiated cells were diluted out to a final concentration of 1 cell per ml and distributed to tubes in 0.5 ml quantities. A series of experiments involving Listeria monocytogenes was undertaken to study first the fecal shedding of Listeria by conventional mice challenged intraperitoneally with Listeria and second the fecal shedding and ability to survive of germ free mice with similar challenge. In addition, mites infesting Listeria infected mice were cultured for their possible contamination by Listeria.
Conventional mice were challenged intraperitoneally with a single near LD50 dose of Listeria. Fecal Listeria were detected by collecting specimens from each mouse daily for the first 4 days and every second day thereafter. The feces was held at 7°C for 14 …

During the fall of 1959, monolayer behavior studies were made on two lakes in the southwest; Boulder Basin of Lake Mead, Nevada, and Lake Sahuaro near Phoenix, Arizona. These studies were made to evaluate the effect of geographical and climatological conditions on movement and behavior of monolayers. The Lake Sahuaro tests also were used to provide data for design and development of improved methods of application and maintenance of a film on this lake in anticipation of full scale, evaporation-reduction tests to be performed there during the summer of I960. Laboratory tests were made to determine the effectiveness of different compounds and microbial groups in increasing the stability of Peorian loess lumps against the action of falling water drops. The influence of these on percolation tests in the laboratory was also determined.
Many organic substances-dextrose, sucrose, starch, peptone, cullulose, and gum arabic-did not themselves contribute directly to soil-structure stability, though these substances do furnish energy material for soil microorganisms, which can convert them readily into either microbial tissue or decomposition products that increase soil-structure stability. Lignin, proteins, oils, fats, waxes, resin, and paraffin increased the stability of lumps of …

In this investigation a survey of Nebraska soils has been made in order to determine the distribution of the aerobic nonsymbiotic and symbiotic nitrogen fixers of the genera Azotobacter and Rhizobium respectively. In connection with this survey, some of the characteristics of the soils which may bring about this distribution were studied. Up to this time practically no research has been reported on the microflora of the soils of Nebraska.
