Haemophilus influenzae is an important human pathogen. The lipooligosaccharide (LOS) of H. influenzae has been implicated as a virulence determinant. To better understand the assembly of LOS in nontypeable H. influenzae (NtHi), we have cloned and characterized the rfaD and rfaF genes of NtHi 2019, which encode the ADP-L-glycero-D-manno-heptose-6-epimerase and heptosyltransferase II enzymes, respectively. This cloning was accomplished by the complementation of Salmonella typhimurium lipopolysaccharide (LPS) biosynthesis gene mutants. These deep rough mutants are novobiocin susceptible until complemented with the appropriate gene. To define the role of the surface lipooligosaccharide (LOS) of Haemophilus ducreyi in the pathogenesis of chancroid, Tn916 mutants of H. ducreyi 35000 defective in expression of the murine monoclonal antibody (MAb) 3F11 epitope on H. ducreyi LOS were identified by immunologic screening. One mutant, designated 1381, has an LOS which lacks the MAb 3F11 epitope and migrates with an increased mobility on sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The gene disrupted by the Tn916 element in strain 1381 was identified by cloning the sequences flanking the Tn916 element. The sequences were then used to probe a lambda DASHII genomic library. …

Ventilation Rates For Carassius Auratus During Changes In Dissolved Oxygen, Collin Nolan

This experiment was designed to determine whether dissolved oxygen concentrations would influence the ventilation rate of goldfish, Carassius auratus. Ventilation rate can be determined by counting opercular movements. Opercular movements of the same five goldfish were counted at three different levels of dissolved oxygen to determine whether there were differences in ventilation rates. Average ventilation rates were established for each of the three levels of dissolved oxygen and compared statistically. There were significant differences in the ventilation rates averages at each of the three levels. Carassius auratus will exhibit changes in ventilation rates when dissolved oxygen levels change.

Field sampling was carried out between 0830 and 1500 beginning at the confluence of Las Vegas Wash and the Inner Las Vegas Bay. Ten (10) locations were sampled, each in a similar manner. Locations of sampling stations are in line from the Wash-Bay confluence to a point midway between Saddle and Black Islands. In addition, sampling was done at a location midway between Sentinel Island and the base of Fortification Hill, and at the buoy line in front of Hoover Dam. The major lipooligosaccharides of the sexually transmitted pathogen Haemophilus ducreyi 35000 have been previously found to terminate in N-acetyllactosamine and sialyl-N-acetyllactosamine, Neu5Ac alpha 2-->3Gal beta 1-->4GlcNAc (W. Melaugh, N. J. Phillips, A. A. Campagnari, M. V. Tullius, and B. W. Gibson, Biochemistry 33: 13070-13078, 1994). In this study, mass spectrometry and composition analyses have shown that the lipooligosaccharides from three other H. ducreyi strains also contain N-acetyllactosamine and are highly sialylated (approximately 30 to 50%), although one African strain was found to contain neither of these structural features.

Haemophilus ducreyi, a cause of genital ulcer disease in developing countries, appears to facilitate the heterosexual transmission of the human immunodeficiency virus in Africa. Despite an increase in studies of this gram-negative human pathogen, little is known about the pathogenesis of chancroid. Our studies have shown that the lipooligosaccharides (LOS) of H. ducreyi may play an important role in ulcer formation. Monoclonal antibody and mass spectrometric analyses identified a terminal trisaccharide present on H. ducreyi LOS that is immunochemically similar to human paragloboside. Extraction of the lipids of Legionella pneumophila yields phases unlike those produced from other Gram-negative bacteria. A viscous interface forms between the aqueous (wash) and organic phases. More than half of the hydroxylated fatty acids were found distributed between the aqueous phase and the interfacial material, fractions in which such constituents have not been reported in other Gram-negative species. It was further observed that after the material from the aqueous/interfacial phase was dissolved in methanol or chloroform/methanol (2:1 (V/V)), the addition of acetone would create a white, flocculent precipitate. Soil microorganisms are important sources of the nitrogen trace gases NO and N₂O for the atmosphere. Present evidence suggests that autotrophic nitrifiers such as Nitrosomonas europaea are the primary producers of NO and N₂O in aerobic soils, whereas denitrifiers such as Pseudomonas spp. or Alcaligenes spp. are responsible for most of the NO and N₂O emissions from anaerobic soils. It has been shown that Alcaligenes faecalis, a bacterium common in both soil and water, is capable of concomitant heterotrophic nitrification and denitrification. Heterogeneity in the lipooligosaccharides (LOS) of pathogenic Haemophilus and Neisseria species is evident from the multiplicity of components observed with electrophoretic analyses. Knowledge of the precise structures that make up these diverse LOS molecules is clearly the key to reaching an understanding of pathogenic processes such as phase variation and molecular mimicry. Except for a few cases, little is known about the specific structural features of LOS that underlie phase variation and molecular mimicry, partly because of the inherent difficulties in the structural elucidation of these complex glycolipids. Two-color flow cytometric analysis was performed on paired samples of peripheral blood (PB) and pleural effusions (PE) of patients with metastatic malignancies. The purpose of these analyses was to test the hypothesis that there are significant differences in the distribution of immunocyte populations in the PE compared to that of the PB.

The majority of the pleural fluid immunocytes were CD3+ cells. The ratio of CD4+ to CD8+ cells was higher in the PE compared to PB. Levels of CD8+, CD19+ and CD14+ cells were not significantly different in the PE compared to the levels in the PB. CD16+CD56+ cells …

A pumped storage system consists of an upper reservoir and lower reservoir separated by an elevation difference. During low demand energy periods such as nights and weekends water is pumped from the lower to the upper reservoir using available energy from conventional steam electric power plants. During high energy demand periods, such as mornings and afternoons of weekdays, upper reservoir water is allowed to drop back down through the same system of water conduits and turbines, generating electricity to conveniently meet abrupt electrical energy requirements. In February, 1982 I began a year-long study to determine if growth and reproduction in Daphnia Pulex were limited by the amount of food available in Boulder Basin, Lake Mead. To determine this, I made monthly collections of Daphnia Pulex and natural lake seston from an already established station in Boulder Basin. A 12 month study of Lake Trashmore was conducted on phytoplankton composition, inorganic nutrients and their seasonal associations. The dominant phytoplankton groups observed were diatoms, chlorophyceans and cyanobacteria. Phytoplankton assemblage patterns were coupled with pH, temperature, dissolved oxygen, nitrites and silicates. Diatoms dominated during periods of high nutrient concentrations, cooler temperatures and an unstable water column. Cyanobacteria forms dominated during periods of warmer temperatures, higher pH and decreased nitrogen and dissolved oxygen concentrations. A seasonal community shift occurred from cyanobacterial dominance to chlorophycean dominance when pH levels decreased. Endodontic (root canal) infections are generally considered to be caused by the invasion of microorganisms which destroy pulp tissue and extend to the periapical region. No definitive study has identified a specific bacterium or group of bacteria as etiological agents. We quantitively measured antibodies present in the serum of 30 normal individuals and patients with either chronic (n = 30) or acute (n = 30) periapical lesions with the indirect fluorescent antibody (IFA) and enzyme-linked immunosorbent assay (ELISA) for IgG and IgM against 11 oral microorganisms routinely isolated from such lesions. A high percentage of fat in the diet has been implicated through epidemiological studies in several human cancers. Studies in experimental animals have produced evidence that increasing the fat in the diet decreases the animal’s ability to restrict tumor development and growth. In previous work done in this laboratory with BALB/c mice injected with herpes simplex Type 2-transformed mouse cells (H238), the effects of a diet which greatly enhanced tumor growth was compared with one which restricted tumor growth. Water samples from the Charleston, Illinois water supply and selected agricultural run-off areas were tested for the presence of organic mutagenic chemicals. The Ames Salmonella/microsome mutagenicity assay utilizing strains TA98 and TA100 was used to analyze concentrated water samples. A special set of samples was exposed to the S9 activation system coupled with the TA98 strain to simulate the mammalian metabolic system.

No mutagens were detected in any of the samples tested. Several positive and negative control systems were included in the testing system. Negative control systems were used to determine the average spontaneous reversion rates for each strain for …

The response to periodic desiccation of periphyton communities developed in eddy environments or in areas exposed to variable, direct current was examined. Algal communities were incubated on clay tiles for between 2 and 12 weeks in the Colorado River, directly below Hoover Dam from October, 1983 - January, 1984. Mean daily discharge from the dam decreased over this period. To assess the influence of these discharge changes on community dynamics, samples collected over the first 12 weeks of the investigation were compared to samples of the same age collected over the final 8 weeks.

Monthly water samples collected from four sites in the Lost River Groundwater Basin, a shallow karst aquifer in the Bowling Green-Warren County area of Kentucky, represented samples from sites receiving conduit and diffuse flow. All sites were severely contaminated with bacteria, and on some occasions the surface water criteria for some heavy metals were exceeded.

Of the total 334 bacterial colonies identified 92.1% were verified as Escherichia coli by the API20E system. Zooplankton samples collected from throughout Lake Mead, in 1981-1982, demonstrate the presence of a statistically significant seasonal and spatial heterogeneity in zooplankton densities. Seasonally, the major zooplankton groups were most abundant in the spring and fall, coincident with maxima in chlorophyll-a concentrations. Successions among the various rotifers, cladocerans and copepods present in the reservoir were influenced by food availability, diapause, predation by planktivorous fish and, possibly, water temperatures. Spatial heterogeneity in zooplankton densities was unrelated to water temperature, pH, conductivity and dissolved oxygen concentrations, but was related to the abundance of phytoplankton (chlorophyll-a concentrations) and fish. At the request of the U.S. Bureau of Reclamation, Boulder City, Nevada, the U.S. Environmental Protection Agency's Environmental Monitoring Systems Laboratory at Las Vegas collected multispectral scanner imagery of Las Vegas Wash on October 1, 1982. A combined maximum likelihood classification and editing procedure was used to classify the multispectral scanner imagery into 12 categories of land cover. The classification identified four categories of marsh vegetation, one category of riparian, two categories of mixed scrub, and two desert categories. Charleston, Illinois, a city located in east-central Illinois, recently implemented a new concept in a raw water system. Due to storage capacity-sedimentation problems in the old Lake Charleston-Embarras River raw water supply, the Charleston Pump Storage Reservoir was constructed. The new system involved building a dike separating the Lake Charleston impoundment from the Embarras River. Water to increase the storage capacity of the reservoir can now be selectively pumped into the impoundment from the river.

Previous studies have shown the planktonic organisms of various natural water bodies to be quite sensitive to the levels of solar ultraviolet radiation incident at the water surface in summer. It has been assumed that the majority of the phytoplankton are simply carried about in the mass of the water which they inhabit with little control of their position or sunlight exposure. Photosynthetic microorganisms must strike a delicate balance with regard to solar radiation; they must obtain enough visible light for adequate photosynthesis while avoiding an excessive exposure to the injurious solar UV-B radiation.

A silica gel-petroleum (SGP) medium was developed which is reliable, approximately the same cost as that of the corresponding agar-based petroleum medium, and can be made sufficiently firm to use in streaking or the spread-plate technique. The presence of large numbers of enteric viruses in wastewaters has resulted in concern regarding their significance and consequences for humans. Assessments of relative significance or consequences may most accurately be made following precise measurements of the range of virus types and amounts in wastewaters and sludges and after epidemiological considerations. In this study, results are presented pertaining to enteric virus isolation, concentration, and enumeration from primary wastewater sludges.

Cell mediated immune (CMI) cytotoxic reactivity of Balb/c mice against H238 cells, a Herpes simplex virus (HSV) Type 2 - induced sarcoma, was measured by the ¹²⁵IUdR release assay. The pleomorphism of Coprinus radians was studied. The three distinct growth forms--sporophore, oidial stage, and ozonium stage--were examined in relation to ultrastructure, morphology and development. Scanning and transmission electron microscopy are correlated with photomicrographs of developmental stages. Development of the oidia and of the sporophore are of particular interest. The multinucleate oidia develop quite differently from oidia as generally described for Basidiomycete imperfect stages. Development of the oidia of this Coprinus is holoblastic and sympodial. The mushroom primordia are differentiated internally into gill, cap, and stipe hyphae while only l-2mm in diameter. At this stage they look superficially like sclerotial …

1. The waters of Las Vegas Bay, a heavily utilized recreational resource, receive discharges from a variety of municipal and industrial waste sources. The U.S. Environmental Protection Agency has determined, on the basis of numerous studies, that the present water quality violates state and federal standards and constitutes a public nuisance. Consultants have advised the Sewage and Wastewater Advisory Committee that rapid abatement of the alleged pollution conditions can be achieved by an advanced wastewater treatment (AWT) plant.
