A Genetic And Physiological Study Of An Arsenite Resistant, Uncoupled Mutant Of Escherichia Coli, Daniel Gordon Smiley

Dissertations and Theses

Chromosomally determined arsenate resistance in Escherichia coli is well characterized. Little, however, is known about chromosomally determined arsenite resistance in E. coli. Accordingly, spontaneous arsenite resistant mutants were selected in a plasmid free strain of E. coli. One mutant strain was analyzed in detail, genetically and physiologically. The mutation confering arsenite resistance was shown to be a single gene mutation. Genetic mapping studies using conjugation and transduction showed that the mutation was closely linked to the ilv region of the E. coli map.

Physiological studies of this mutant showed that it shared characteristics common to those of mutants …

Streptococcus and Staphylococcus were collected from patients hospitalized at the Oregon Burn Center, Emanuel Hospital, Portland, Oregon, between October 27, 1980 and January 1, 1981. Forty-five isolates of Staphylococcus and twenty-one isolates of Streptococcus, Lancefield Group D, were included in the investigation.

The objectives of the investigation were: (i) to identify to the species level all isolates, (ii) to determine their susceptibility to antimicrobial agents, (iii) to test the organisms. over time for increasing resistance to silver, (iv) to determine, if present, a bacterium harbored within the burn unit (nosocomial bacterium), and (v) to determine any recognizable pattern of successive …

A fluorescent metabolite present in seven members of the genus Rhizopus was isolated. This compound appeared green before spray treatment and purple after spray treatment with p-anisaldehyde in visible light. Subsequent purification and structural elucidation of the isolated compound yielded 1-[2,6,10,14-tetramethyl-17-carbomethyl heptadecyl]-1-[2,6,10,14-tetramethyl-17-methanoyl heptadecyl]-benzene.

Interpopulation Variation In Calcareous And Proteinaceous Shell Components In The Stream Limpet, Ferrissia Rivularis, Carl M. Way, Daniel J. Hornbach, Albert J. Burky

Natural populations of the North American stream limpet, F. rivularis, were studied in upstate New York [USA], in a set of localities whose waters have a 15-fold range of dissolved Ca (4.6-67.6 mg/liter) and also range from oligotrophy to eutrophy. Shell component analyses (CaCO3, total organic C and total N) are reported as component mass-fractions (mg/g or .mu.g/g dry weight) and as values for a standard limpet shell of 35 mm aperture length (AL). Seasonal metabolic rates were examined for the sphaeriid clam, Musculium partumeium (Say) from a permanent and a temporary pond. For the fall-born generation from the permanent pond and for the single generation from the temporary pond, metabolic rates peak during periods of greatest growth and reproduction. Metabolic rates were consistently higher for the permanent pond population. These rates for Musculium partumeium extend the available information on the family Sphaeriidae to the genus Musculium.

Clarke (1973) states that Pisidium casertanum (Poli) is perhaps the most common species of Pisidium and is truly cosmopolitan, occurring throughout Eurasia, Africa, Australia, and South, Central and North America. Populations of these clams are reported in habitats ranging from ephemeral ponds to benthic zones of deep lakes. Bacterial strains resistant to arsenite, arsenate (or both) are readily isolated from nature. The resistance, in many cases, can be shown to be determined by conjugal plasmids. The mechanisms by which these plasmids confer resistance to arsenic compounds are entirely unknown. This study was undertaken to shed light on some of the physiological and genetic aspects of this resistance using plasmid-bearing strains of Escherichia coli which are highly resistant to both arsenite and arsenate. Most studies were carried out using E. coli J53 (R773) or the plasmid-free strain J53.

A study was conducted to contribute to the taxonomy of the nonfruiting myxobacteria and to determine the effects of fire on the occurrence and characteristics of nonfruiting myxobacteria isolated from pine forest soil Three years after a prescribed burn, the population of nonfruiting myxobacteria was decreased in the burn site. The organisms isolated were distinct from any of the known species of nonfruiting myxobacteria, and apparently are new species.

Methods were devised for the isolation and pure culture in defined mineral medium of Closteriurn ehrenbergii Menegh (Chlorophyceae). The effect of pH, type of nitrogen source, nitrate concentration, total hardness, and various buffers on growth were investigated. Best growth rates were obtained at pH 8 to 9, but good growth was observed even at pH 9.4. Nitrate, ammonia, or urea nitrogen could be utilized, but at concentrations greater than 10-2M urea was toxic to growth. At higher concentrations of ammonia and nitrate longer lag phases of growth were obtained but eventual higher cell yields were obtained. Good growth …

The pleomorphism of Coprinus radians was studied. The three distinct growth forms--sporophore, oidial stage, and ozonium stage--were examined in relation to ultrastructure, morphology and development. Scanning and transmission electron microscopy are correlated with photomicrographs of developmental stages. Development of the oidia and of the sporophore are of particular interest. The multinucleate oidia develop quite differently from oidia as generally described for Basidiomycete imperfect stages. Development of the oidia of this Coprinus is holoblastic and sympodial. The mushroom primordia are differentiated internally into gill, cap, and stipe hyphae while only l-2mm in diameter. At this stage they look superficially like sclerotial …

The ampullariid Pomacea urceus uses its ctenidium and lung in respiration. The snails reach the surface with their siphon and ventilate their lung by withdrawing their head-foot in a pumping action. The lung gas also serves to change the buoyancy of the snails. After a ventilation a snail does not necessarily return to the same buoyancy level. The diurnal vertical movement of zooplankton was first recorded in freshwater lakes by Weismann (1877) in Lake Constance (Bodensee), although Cuvier was credited with observing the migration of Daphniae in 1817 (Gushing, 1955). The migration is best observed in deep oligotrophic lakes and migrations of 50 m per day are described (Worthington, 1931). In addition, two species of chaetognaths are reported to migrate 400 m a day in marine waters off of Lisbon (Waterman and Berry, 1967) while Birge (1895) found little evidence of the phenomenon at all in Lake Mendota, Wisconsin. This paper presents a general limnological survey of a subalpine lake ecosystem. Physical, chemical and biological parameters were considered. Chemical and physical factors were dominated by the influence of snow-ice cover and high amounts of solar radiation. Abiotic factors result in a small phytoplankton standing crop and limitation of activity by photosynthetic organisms to the short summer season. Zooplankton and benthic communities are simplified in species composition and population size in response to the small primary producer population.

A study of the spatial distribution and temporal occurrence and diversity of rotifers in relation to certain chemicophysical parameters was conducted in Barren Lake, Kentucky, a flood control lake, from January, 1970, through January, 1970. Paired samples were collected from six depths in the main pool area of the reservoir and from an automatic plankton sampler installed in the tailwater area where, from April through September, the effluent was derived from the upper 3 m of the pool . Amino acid absorption across rat intestine in vitro was inhibited or stimulated by insulin depending on the hormone serosal concentration.

The absence of sodium ions from the incubating solution resulted in a significant decrease of L-alanine absorption. However, L-alanine was enhanced in a sodium-free medium by the addition of insulin on the serosal side of the intestine.

A sizable decrease in L-alanine and L-lysine absorption was produced by introduction of D-glucose (0.2 % and 2 %) in the Krebs-Ringer's incubating buffer. Nevertheless, the presence of D-glucose did not prevent the stimulatory effect of insulin on amino acid absorption.

Progressive increases in concentration of dissolved solids in the Colorado River water from Lake Powell to Imperial Dam seem to alter plankton dynamics and biological productivity of the river. Also, changes in biological productivity and micronutrients concentrations occur within the same reservoir. Development of a digital simulation model to predict micronutrients concentrations and biological productivity is necessary for diagnosing changes in plankton population and effluent-carrying capacity of the system. A new histozoic myxosporidian parasite (Class Mysoxporidea), Myxobolus meglitschi sp. nov., infecting young-of-the-year and yearling bluegill sunfish, Lepomis macrochirus Raf., from Shanty Hollow Lake, Warren County, Kentucky was described. Phenology was investigated from January 20 to December, 1969.

The magnitude of infection varied seasonally in bluegill. Incidence was highest in July and August (43.92) in host populations, declining, but not disappearing from September to December 20, 1969. The infection exhibited a yearly mean incidence of 22.318.

Cercariac of Schistosoma mansoni were studied for glycogen utilization, uptake of glucose from solution, lactic acid production, and respiration.

Using the anthrone method of determination of carbohydrate, the amount of glycogen used by a single cercaria in 4 hours is determined as 43.2 x 10-4 μg ± 6.6 x 10-4 μg.

Glucose was not observed to have been used from solution under the conditions of the experiment, even in the presence of trehalose. The determinations were made by the Glucostat method.

Lily Lake, a subalpine lake in the Wasatch National Forest Tl5, R9E, S31, of the Uinta Mountain Region, Utah, is located about one-half mile west of Trial Lake at an elevation of 10,000 feet. This Lake, which is one of the acidic, cold, lentic bogs characteristic of this area, has been selected for a taxonomic and seasonal distribution study of acquatic phycomycetes.
