There is great variation in the extent of surveillance of foodbome disease throughout the world. Some countries contribute important epidemiological and microbiological data to the WHO Surveillance Programme for Foodbome Diseases in Europe. While the UK is a major contributor to this programme, data from Eire has been non-existent. Trends in the incidence of foodbome disease in the UK may have particular relevance to Eire given the similarity of culture. Microbiological and epidemiological data in relation to foodbome pathogens, foods most frequently incriminated in outbreaks, places where outbreaks occur, together witli the main contributory factors are discussed. In 1990-1991, in a national surveillance study, and in 1991-1992, in a followup study, both by Thornsberry et al. (1993), ciprofloxacin data from various geographical and demographical institutions were collected. Several species of bacteria have shown resistance to ciprofloxacin and norfloxacin, but the degree of resistance to these drugs has not been reported for the Stockton area. To determine the extent of this resistance, Dameron Hospital antibiograms generated from 1990 to 1994 were reviewed and compared. Results of the comparison show that susceptibility among the Gram-negative isolates, with the exception of Providencia stuartii, Acinetobacter lwoffi, and to a lesser extent …

Hyla albopunctata is a medium-sized species in the albopunctata group, adults range in size from 42-52 mm SVL (males average 47 mm and females 49 mm). The head is triangular in outline, always longer than wide. The snout is moderately long and acuminate in dorsal view, whereas in lateral view, it is acuminate and protrudes beyond the mandible to form an angle of 45 degrees over the mandible. The canthus rostralis is rounded and distinct, the loreal region is slightly concave, and the internarial region is slightly convex. The eyes are large, approximately twice the diameter of the tympanum. The …

Utilizing the reverse transcriptase-polymerase chain reaction (RT-PCR) procedure, we have synthesized full-length copies of segment 10 from African horsesickness virus (AHSV) serotypes 1,4 and 8. The genes were cloned, sequenced and compared with the sequence of the cognate gene from AHSV serotypes 3 and 9. Sequences were analyzed to assess evolutionary relationships among serotypes using cladistics. Based on this analysis the data support a close relationship between serotypes 4 and 9 and between serotypes 1 and 8 and a closer relationship of serotype 3 to the 4 and 9 group.

This report provides details for the construction of a hot-bead thermistor current meter which is capable of measuring water velocities on a millimeter spatial scale and for the construction of a compact and accurate calibration system. Hot-bead thermistor current meters can be built with response times of 200 ms capable of measuring velocities between 0.1 and 80 cm s-1. The construction of a sturdy probe for application in lotic systems such as high gradient Hawaiian streams was achieved by the use of heavy-duty acrylic tubing, small stainless steel gas-chromatography tubing, and flexible Tygon spaghetti tubing. Soil microorganisms are important sources of the nitrogen trace gases NO and N2O for the atmosphere. Present evidence suggests that autotrophic nitrifiers such as Nitrosomonas europaea are the primary producers of NO and N2O in aerobic soils, whereas denitrifiers such as Pseudomonas spp. or Alcaligenes spp. are responsible for most of the NO and N2O emissions from anaerobic soils. It has been shown that Alcaligenes faecalis, a bacterium common in both soil and water, is capable of concomitant heterotrophic nitrification and denitrification. Using oligonucleotide probes which have previously been shown to be specific for the ospC gene found in the Lyme disease spirochete species Borrelia burgdorferi, B. garinii, and group VS461, we detected an ospC homolog in other Borrelia species including B. coriaceae, B. hermsii, B. anserina, B. turicatae, and B. parkeri. In contrast to the Lyme disease spirochetes, which carry the ospC gene on a 26-kb circular plasmid, we mapped the gene in other Borrelia species to linear plasmids which varied in size among the isolates tested. Coumermycin A1 is an inhibitor of DNA gyrase, an enzyme that catalyzes supercoiling of DNA and is required for bacterial DNA replication. We have investigated the activity of this coumarin antibiotic on Borrelia burgdorferi, a spirochete and the causative agent of Lyme disease. B. burgdorferi was more susceptible than many other eubacteria to coumermycin as well as novobiocin, another coumarin antibiotic; this contrasted with its relative resistance to the DNA gyrase inhibitors nalidixic acid, oxolinic acid, and ciprofloxacin. Coumermycin at 0.2 micrograms/ml inhibited the growth of B. burgdorferi B31 in BSK II medium. A 100-fold-lower concentration induced the relaxation of …

Because of the potential health hazards associated with the use of sludge for agricultural purposes, Dudley et al (1980) published a scheme for the routine analysis of bacteria in municipal sewage sludge. In this study, the Dudley et al scheme (1980) was modified by updating some of the procedures. Aerobically digested sludge generated by the Bowling Green Wastewater Treatment Plant, Bowling Green, Kentucky, was analyzed using the modified scheme. Sludge samples were collected once every two months over a one-year period from October 1989 to August 1990.

The role of microorganisms in the degradation of xenobiotics in the environment is well established. Bacteria from the genus Pseudomonas are particularly well adapted to the degradation of hydrocarbons, aromatics, and numerous other natural and introduced substrates. We have isolated a strain of Pseudomonas putida, designated PC2P15, that uses toluene, phenol, benzene, and a number of other substrates as its sole sources of carbon and energy.

I have investigated the possibility that specific conalbumin (ovotransferrin) iron saturation levels enable less virulent strains of Salmonella to become more virulent. Iron starved cells of two pathogenic Salmonella strains, S. paratyphi B var. java and S. thompson, were cultured in iron limited media at 3 different iron conalbumin saturation levels. Results indicate that strains differ significantly at both low and high iron saturation conalbumin. These differences depict a growth advantage for S. paratyphi B which correlates with reports by the Centers for Disease Control that S. paratyphi B was 3 times more frequent in blood isolates than S. …

A synthetic, 23-bp ecdysterone regulatory element (EcRE), derived from the upstream region of the Drosophila melanogaster hsp27 gene, was inserted adjacent to the herpes simplex virus thymidine kinase promoter fused to a bacterial gene for chloramphenicol acetyltransferase (CAT). Hybrid constructs were transfected into Drosophila S3 cells and assayed for ecdysterone-inducible CAT expression. In the absence of ecdysterone a tandem pair of EcREs repressed the high constitutive level of CAT activity found after transfection with the parent reporter plasmid alone. Recently, using cnida-mediated measurements of adhesive force, Thorington and Hessinger (1984, 1988a, b) identified two different chemoreceptors involved in triggering cnida discharge on the tentacles of the sea anemone, Aiptasia pallida. These two classes of receptors were shown to interact with glycine and N-acetylneuraminic acid (NANA). We now show that the discharge of the microbasic p-mastigophores, one of the three types of cnidae present on the tentacles of A. pallida, is under the controlling influence of these two classes of cnidocyte-associated chemoreceptors. A pumped storage system consists of an upper reservoir and lower reservoir separated by an elevation difference. During low demand energy periods such as nights and weekends water is pumped from the lower to the upper reservoir using available energy from conventional steam electric power plants. During high energy demand periods, such as mornings and afternoons of weekdays, upper reservoir water is allowed to drop back down through the same system of water conduits and turbines, generating electricity to conveniently meet abrupt electrical energy requirements. In February, 1982 I began a year-long study to determine if growth and reproduction in Daphnia Pulex were limited by the amount of food available in Boulder Basin, Lake Mead. To determine this, I made monthly collections of Daphnia Pulex and natural lake seston from an already established station in Boulder Basin. I cultured the Daphnia Pulex under simulated field conditions in a flow-through feeding apparatus using four different food regimes; 1-natural lake seston filtered through 80 um mesh to remove other zooplankton, 2-lake seston (as above) with an enrichment of 103 cells-ml-1 of Chlamydomonas reinhardti, 3-lake …

The phototoxicity of hematoporphyrin derivative (Hpd) to murine bladder tumor (MBT-2) cells was studied in vitro. It was observed that selenium in the form of sodium selenite enhanced Hpd-sensitized photodamage in MBT-2 cells under conditions where the selenite alone was non-toxic. Sodium selenite was found to enhance the fluorescence emission of Hpd and augment the Hpd-sensitized photooxidation of tryptophan. The data indicate that sodium selenite is able to disaggregate Hpd, thereby enhancing Hpd-sensitized phototoxicity.

Protective antigen component of B. anthracis toxin was produced and purified to the >99% level. Toxin was purified from culture supernatant utilizing concentration and liquid chromatography techniques. Purity was determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis.
The purified protective antigen retained biological and antigenic activity as evidenced respectively by lethality in Fischer 344 rats when injected in combination with lethal factor, and by positive results on the Ouchterlony double diffussion assay.
Radioiodinated protective antigen was used both in the in vivo and the in vitro experiments.

We have developed a rapid assay which can detect the function of the origin of 0X174 complementary strand DNA in Ml3 by virtue of its ability to initiate the synthesis of M13 complementary strands in the presence of the antibiotic rifampicin. A segment of DNA carrying the sequence recognized by the replication protein n' has been isolated from bacteriophage ØX174 and cloned into an Ml3 bacteriophage vector. Protein n' is a component of the bacterial primosome, a multi-protein assembly which initiates the synthesis of primers during DNA replication in the bacterium Escherichia coli. Samples of cave water were analyzed for volatile organic compounds during four separate storm events. The major compounds detected were toluene, 1,1,1-trichloroethane, trans-1,2-dichloroethylene, methylene chloride, and 1,1-dicloroethane. Minor compounds detected were trichloroethylene, tetrachloroethylene, 1,1-dichloroethylene, and ethylbenzene. These compounds were detected in varying amounts at each of the three study sites. During a storm event, the levels of each contaminant changed significantly at all three sites studied. Two effects were observed when a storm event occurred. The first effect was the dilution of the volatile organic compounds during the storm event. These effects were related to the amount of rainfall and …

A recently isolated freshwater methanogenium species, Methanogenium wolfei, is characterized. Cells were irregular cocci, measuring 1.5 to 2.0 micrometers in diameter. No motility was observed, but 1 to 2 flagella per cell were observed after staining with Gray's Flagella Stain. Colonies formed by this species were small, shiny, and green-brown in color. Formate or hydrogen plus carbon dioxide served as substrates for growth. The optimal temperature for growth was found to be 45 degrees centigrade with minimal growth below 30 degrees centigrade and above 55 degrees centigrade. The optimal pH for growth was determined to be 6.8. Optimal growth was …

Individuals with Down syndrome (US) suffer from increased incidence of respiratory infections and lymphoblastic leukemia, and a death rate that is particularly high in the first 5 years of life. Relatively few studies have probed immune parameters in young US children. Primary immune defects in DS may be masked by a degree of immune maturity in adults, and hygienic factors may have an effect on immune capability throughout the years. A study of young children can give clearer evidence of the actual primary immune defects in DS.

Seasonal filtration rates of a pond population of the freshwater pisidiid clam, Musculium partumeium (Say), were assessed by measuring the clearance of 2.02 µ.m latex beads from suspension and expressed in terms of FR = a(AFDW}b where FR = filtration rate (ml H20 · hr- 1 ) , AFDW = mg ash-free dry weight of a whole clam and "a" and "b" are constants. The a-values ( = FR of a 1-mg AFDW clam} vary seasonally with the highest rates corresponding to periods of maximum growth and reproduction in the spring and fall. The response to periodic desiccation of periphyton communities developed in eddy environments or in areas exposed to variable, direct current was examined. Algal communities were incubated on clay tiles for between 2 and 12 weeks in the Colorado River, directly below Hoover Dam from October, 1983 - January, 1984. Mean daily discharge from the dam decreased over this period. Monthly water samples collected from four sites in the Lost River Groundwater Basin, a shallow karst aquifer in the Bowling Green-Warren County area of Kentucky, represented samples from sites receiving conduit and diffuse flow. All sites were severely contaminated with bacteria, and on some occasions the surface water criteria for some heavy metals were exceeded.
Of the total 334 bacterial colonies identified 92.1% were verified as Escherichia coli by the API20E system. The influences of dietary deficiencies of folic acid or zinc on rotaviral disease infant mice were studied. Preliminary studies indicated that bovine and simian rotaviruses, but not porcine rotavirus, caused diarrhea in infant mice. Bovine and porcine rotaviruses were not, however, sufficiently infectious to replace murine rotavirus in studies utilizing the murine model. It was also determined that murine rotavirus purified by a cesium chloride gradient was highly infectious and useful for subsequent studies on nutritional influences of rotaviral disease. Zooplankton samples collected from throughout Lake Mead, in 1981-1982, demonstrate the presence of a statistically significant seasonal and spatial heterogeneity in zooplankton densities. Seasonally, the major zooplankton groups were most abundant in the spring and fall, coincident with maxima in chlorophyll-a concentrations. Successions among the various rotifers, cladocerans and copepods present in the reservoir were influenced by food availability, diapause, predation by planktivorous fish and, possibly, water temperatures. Spatial heterogeneity in zooplankton densities was unrelated to water temperature, pH, conductivity and dissolved oxygen concentrations, but was related to the abundance of phytoplankton (chlorophyll-a concentrations) and fish. Solid media were employed to determine the presence and absence of extracellular enzyme production by two genera of fruit-rot fungi, Rhizopus and Mucor. The results of this investigation revealed that phosphatase was released into the cultural medium by all the fungi examined; however, only R. oryzae, R. tritici, M. mucedo, and M. piriformis showed the possibility of being high producers of the enzyme. Protease, urease, ribonuclease, pectate lyase, and polygalacturonase, at varying levels of activity, were detected, in the majority of the fungi, in the cultural medium.
