Accurate estimation of the hip joint center (HJC) is critical for biomechanical modeling and clinical assessment. Common methods include predictive methods (e.g., regression equations) and functional methods such as sphere-fitting. In infants, femoral head-based methods are often limited due to incomplete ossification of the anatomy and poor visualization in computed tomography (CT) images. This study evaluates an alternative approach using MRI-based sphere fitting of the ossified acetabular rim, comparing its accuracy to traditional femoral head-based methods. Cancer remains a predominant health challenge that is responsible for a significant portion of global morbidity and mortality. Epirubicin (EPI), a chemotherapeutic anti-cancer drug, has shown remarkable efficacy in combating various malignancies. However, it is known to have undesirable side effects on the heart, collectively referred to as cardiotoxicity. This research investigates the adverse effects of chemotherapy on cardiac contractility through an in vitro examination of the mechanics of healthy and infarcted animal heart tissues. Patellofemoral Pain Syndrome (PFPS) is the most prevalent overuse injury in cycling, often linked to altered neuromuscular activation patterns of the quadriceps and hamstrings. While sensory feedback has successfully modified cyclists posture, there is a critical lack of evidence-based interventions targeting the underlying muscle activation imbalances associated with chronic knee pain. This thesis evaluates whether accessible and affordable technologies can provide clinically significant, quantifiable gait analysis comparable to traditional marker-based motion capture systems. Modern high-end systems offer excellent accuracy but remain financially and logistically inaccessible to many clinicians and researchers. To address this gap, a framework was developed using markerless pose estimation using the model, MediaPipe, inertial measurement units (IMUs) processed through an Extended Kalman Filter (EKF), and surface electromyography (sEMG) to measure joint kinematics and neuromuscular activity during gait. Spinal ligaments play a crucial role in maintaining the mechanical stability of the lumbar spine. These dense, collagenous tissues not only resist excessive motion but also distribute loads between spinal components to protect neural structures. Traditionally, the mechanical response of ligaments has been modeled using linear elastic assumptions, which treat the tissue as having a constant stiffness regardless of loading history. While this simplifies analysis, it fails to capture the time-dependent behaviors, such as creep, stress relaxation, and hysteresis, that are characteristic of biological tissues.

This thesis investigates the biomechanical behavior of the human knee under varying conditions of motion, load, and fatigue, with a focus on clinical angle analysis and gait stability. Four studies were conducted using motion capture data and a modified Grood and Suntay Joint Coordinate System (JCS) to evaluate knee joint kinematics—specifically flexion-extension, abduction-adduction, and internal-external rotation. The first study examined the effects of incremental weight loads during isolated knee extension exercises, revealing dose-dependent increases in abduction and internal rotation that may contribute to joint stress. This dissertation addresses four specific aims that collectively attempted to advance the experimental and computational analysis of total knee arthroplasty (TKA) components. The first study focused on the development of a novel whole knee joint simulator capable of simultaneous tibiofemoral and patellofemoral knee loading. This simulator employs custom fixturing to facilitate dynamic, unconstrained, muscle-driven PF articulation alongside controlled TF contact mechanics. Validation against experimental measurements showed strong agreement, demonstrating the simulator's potential as a valuable tool for future TKA design and surgical technique investigations.

Contemporary total knee arthroplasty (TKA) has not fully restored natural patellofemoral (P-F) mechanics across the patient population. Previous experimental simulations have been limited in their ability to create dynamic, unconstrained, muscle-driven P-F articulation while simultaneously controlling tibiofemoral (T-F) contact mechanics. The purpose of this study was to develop a novel experimental simulation and validate a corresponding finite element model to evaluate T-F and P-F mechanics. Osteoarthritis (OA) is a form of arthritis that develops in the joint due to overuse and aging causing pain, discomfort, and disability. Total Knee Arthroplasty (TKA) is a surgical procedure performed when OA symptoms are severe with an estimated 600,000 patients in the United States currently receiving TKA. Studies have reported dissatisfaction of the knee for 14-39% of patients. The glenohumeral joint (GH) is commonly conceptualized as a ball-and-socket joint [1], and its center of rotation (COR) is presumed to coincide with the geometric center of the medial-superior region of the humeral head [2]. Recent research has endorsed improvements in COR estimation through invasive and noninvasive techniques, including cadaver studies, stereophotogrammetry, and motion capture (MOCAP) systems. Brain–body interactions (BBIs) have been the focus of intense scrutiny since the inception of the scientific method, playing a foundational role in the earliest debates over the philosophy of science. Contemporary investigations of BBIs to elucidate the neural principles of motor control have benefited from advances in neuroimaging, device engineering, and signal processing. This dissertation explores how multimodal walking impacts quantitative and qualitative aspects of gait for persons with multiple sclerosis (pwMS) experiencing foot-drop. Foot-drop can dramatically impede mobility and clinicians routinely prescribe ankle-foot-orthosis (AFO) and functional electrical stimulation (FES) devices to alleviate its impacts on daily life. Background: Disease of the lower extremity arteries (Peripheral Arterial Disease, PAD) is associated with high morbidity and mortality. During disease development, the arteries adapt by changing their diameter, wall thickness, and residual deformations, but the effects of demographics and risk factors on this process are not clear.

Falls during walking are a leading cause of injuries across aging. Many of these falls are due to slips and trips. The ability to perceive disturbances to ongoing motion may play an important role in the control of walking balance. However, disturbance perception has been investigated in standing balance, but its role in walking balance due to slip- and trip-like disturbances remains largely unknown. There is no cure for osteogenesis imperfecta (OI), and current treatments can only partially correct the bone phenotype. Stem cell therapy holds potential to improve bone quality and quantity in OI. Here, we conduct a systematic review and meta-analysis of published studies to investigate the efficacy of stem cell therapy to rescue bone brittleness in mouse models of OI. Identified studies included bone marrow, mesenchymal stem cells, and human fetal stem cells. This thesis proposes an approach for Finite Element Analysis (FEA) of mandibular fracture fixation. Using a Computerized Tomography (CT) scan of mandible obtained from a specific person, the material characteristics, density and modulus of elasticity, were determined from a set of discrete points within the mandible that are 1mm spaced based on the Hounsfield Units of these points. The mandible geometry was sectioned to simulate a fracture. Muscle and mastication forces were added to replicate post-surgery loading. Musculoskeletal modeling provides an alternative to in-vivo characteristics that are difficult to directly measure for movements such as running, especially for trunk muscles and joints. The full-body-lumbar-spine (FBLS) model by Raabe and Chaudhari, 2016 is an OpenSim model created for simulations of jogging. The lifting full-body (LFB) model by Beaucage-Gauvreau et al., 2018 is an adaptation of the FBLS created for estimating spinal loads during lifting. BACKGROUND: Knee joint contact (bone on bone) forces are commonly estimated using surrogate measures such as external knee adduction moments (with limited success) or musculoskeletal modeling (more successful). Despite its capabilities, modeling is not optimal for clinicians or persons with limited experience and knowledge. Therefore, the purpose of this study was to design a novel prediction method for knee joint contact forces that is equal or more accurate than modeling, yet simplistic in terms of required inputs. Bone adapts its architecture to the applied load; however, it is still unclear how bone mechano-adaptation is coordinated and why potential for adaptation adjusts during the life course.
