Parasite Infection Mediates Trait Tradeoffs In Fundulus Heteroclitus, Sarah Dunn

To be successful, an animal must eat, grow, and reproduce. With limited resources, there are tradeoffs between these critical life history parameters but the direction of the tradeoffs is largely unknown in a changing environment. To determine whether environmental context affects life-history tradeoffs, I surveyed and quantified investment into reproduction, growth, and a proxy for immunity (parasitism), in the mummichog, Fundulus heteroclitus, a common inhabitant of salt marshes in Georgia. Three salt marsh sites along coastal Georgia (Shellman Bluff, Skidaway Island, and Tybee Island) were selected using a proxy for anthropogenic disturbance (impervious surface), which also fell along a …

Parasite control is an important aspect of health management of horses, particularly the control of gastrointestinal (GI) parasites. Recently, treatment recommendations have involved selective use of anthelmintics in horses with fecal egg counts (FEC) greater than a specified threshold. The objectives of this study were; (1) to determine the prevalence of helminths in our area by egg and L3 determinations, (2) to determine if certain horses maintained low FEC, therefore eliminating the need to treat them on a year-round basis and (3) to determine the effectiveness of four common treatments (moxidectin, ivermectin (pioneer and generic), fenbendazole and pyrantel tartrate) via …

Background

Intrahepatic cholangiocarcinoma (ICC) is a highly aggressive tumor of the bile duct, and a significant public health problem in East Asia, where it is associated with infection by the parasiteOpisthorchis viverrini. ICC is often detected at an advanced stage and with a poor prognosis, making a biomarker for early detection a priority.

Methods

We have comprehensively profiled miRNA expression levels in ICC tumor tissue using small RNA-Seq and validated these profiles using quantitative PCR on matched plasma samples.

Results

Baylisascaris procyonis (Bp) is a large roundworm of the common raccoon (Procyon lotor) which serves as the definitive host. Bp is an important cause of clinical larva migrans, including severe neurological disease, across numerous taxa including humans. Other procyonids, as well as occasionally dogs, can act as definitive hosts for this or other Baylisascaris spp. Many of these animals are becoming more common as household pets, posing a risk to people who come in contact with these animals. This paper provides the first report of the invasive Asian fish tapeworm, Bothriocephalus acheilognathi Yamaguti, 1934, in Honduras. The cestode was found in Profundulus portillorum (Cyprinodontiformes: Profundulidae), which represents a new host record, and which is a member of a genus faced with a variety of conservation challenges, now potentially complicated by the presence of this pathogenic cestode. Nearly complete sequence data from the ITS-1 5.8S and ITS-2 regions corroborate the determination based on morphological characteristics. When blood-feeding parasites increase seasonally, their deleterious effects may prevent some host species, especially those living in large groups where parasites are numerous, from reproducing later in the summer. Yet the role of parasites in regulating the length of a host’s breeding season—and thus the host’s opportunity for multiple brooding—has not been systematically investigated. The highly colonial cliff swallow (Petrochelidon pyrrhonota), a temperate-latitude migratory songbird in the western Great Plains, USA, typically has a relatively short (eight to nine week) breeding season, with birds rarely nesting late in the summer. Trypanosoma cruzi is a genetically diverse parasite that causes Chagas disease, one of the most important zoonoses in the Americas. This generalist parasite of mammals belongs to a clade mostly comprised of bat parasites, the T. cruzi clade. The origins (i.e., biogeographic history and evolution of hosts associations) of this parasite are far from being understood, and the main areas that need further study are: species limits within T. cruzi sensu lato, further studies on the diversity of T. cruzi clade members and their hosts, and research on adaptations of the hosts to trypanosome infections. Species in the genus Plasmodium cause malaria in humans and infect a variety of mammals and other vertebrates. Currently, estimated ages for several mammalian Plasmodium parasites differ by as much as one order of magnitude, an inaccuracy that frustrates reliable estimation of evolutionary rates of disease-related traits. We developed a novel statistical approach to dating the relative age of evolutionary lineages, based on Total Least Squares regression. We validated this lineage dating approach by applying it to the genus Drosophila. Background

Hookworms infect nearly 700 million people, causing anemia and developmental stunting in heavy infections. Little is known about the genomic structure or gene regulation in hookworms, although recent publication of draft genome assemblies has allowed the first investigations of these topics to be undertaken. The transcription factor DAF-16 mediates multiple developmental pathways in the free living nematode Caenorhabditis elegans, and is involved in the recovery from the developmentally arrested L3 in hookworms. Identification of downstream targets of DAF-16 will provide a better understanding of the molecular mechanism of hookworm infection.

BACKGROUND: Steinernema carpocapsae are parasitic nematodes that invade and kill insects. The nematodes are mutualistically associated with the bacteria Xenorhabdus nematophila and together form an excellent model to study pathogen infection processes and host anti-nematode/antibacterial immune responses. To determine the contribution of S. carpocapsae and their associated X. nematophila to the successful infection of insects as well as to investigate the interaction of each mutualistic partner with the insect immune system, it is important to develop and establish robust methods for generating nematodes devoid of their bacteria.

Schistosoma haematobium affects more than 100 million people throughout Africa and is the causative agent of urogenital schistosomiasis. The parasite is strongly associated with urothelial cancer in infected individuals and as such is designated a group I carcinogen by the International Agency for Research on Cancer. Using a protein microarray containing schistosome proteins, we sought to identify antigens that were the targets of protective IgG1 immune responses in S. haematobium-exposed individuals that acquire drug-induced resistance (DIR) to schistosomiasis after praziquantel treatment. There has been a strong, positive correlation between opisthorchiasis-associated cholangiocarcinoma and infection with Helicobacter. Here a rodent model of human infection with Opisthorchis viverrini was utilized to further investigate relationships of apparent co-infections with O. viverrini and H. pylori. A total of 150 hamsters were assigned to five groups: i) Control hamsters not infected with O. viverrini; ii) O. viverrini-infected hamsters; iii) non-O. viverrini infected hamsters treated with antibiotics (ABx); iv) O. viverrini-infected hamsters treated with ABx; and v) O. viverrini-infected hamsters treated both with ABx and praziquantel (PZQ). Background. Schistosomiasis is a chronic disease caused by infection with parasitic worms of the genus Schistosoma. In sub-Saharan Africa, infections with S. haematobium are most common. Cystoscopic examination (CE) has been accepted as the gold-standard test for detecting the late manifestations of schistosomiasis, including urothelial cancer of the bladder. However, this procedure is invasive and 10 - 40% of tumours may remain undetected. A non-invasive examination and a new generation of biomarkers are needed for better monitoring of the disease.

Miracidia exhibit observable host-finding behaviors, and their sympatric snails have attributes that either permit or prohibit infection. This study was designed to assess the factors involved in intermediate host finding and host-parasite compatibility in the deer liver fluke (Fascioloides magna). The main aim was to determine the extent to which the parasite and/or the intermediate host are involved in host-parasite compatibility. A secondary goal was to determine what factors may lead to miracidial transformation. The discovery of dipteran remains on mummified individuals can lead to either cause for curatorial concern or to a better understanding of the individual’s post-mortem environment. The present study analyzed insect remains associated with the body of a unique medieval mummy of religious significance, that of the Blessed Antonio Patrizi da Monticiano. A total of 79 puparia were examined and all were identified as Ophyra capensis (Diptera: Muscidae). Additionally, a desiccated moth (Lepidoptera: Tineidae) was encountered. Puparia of O. capensis would be associated with normal decomposition shortly after the death of the mummified individual, and not an infestation beginning during …

Cutaneous leishmaniasis and Chagas disease (American trypanosomiasis) are two vector-borne, protozoal zoonoses whose emergence into the southern United States is a public health problem of increasing significance. Cutaneous leishmaniasis is caused by several species of intracellular protozoa in the genus Leishmania and is most often characterized by the formation of large, ulcerative skin lesions that can result in considerable scarring and permanent disfigurement. Infection with Leishmania is prevalent throughout the world in tropical and sub-tropical regions and in areas where people are regularly exposed to the hematophagous sand fly vectors that transmit the disease. Giardia lamblia is a flagellated protozoan parasite responsible for chronic diarrheal disease in humans and animals. Giardia has minimal lipid synThesis machinery and the majority of its lipids are obtained from the small intestine of humans where the trophozoites colonize. Recent results from our laboratory indicate that sphingolipids (SLs) are also important for Giardia and they regulate encystation and cyst production. Only five SL genes are present in this parasite and they are differentially regulated during encystation. In my studies, I found that giardial serine palmitoyltransferase (gSPT) activity is upregulated in encysting cells, suggesting it might play an important role …

Echinococcus granulosus, formerly regarded as a single species with a high genotypic and phenotypic diversity, is now recognized as an assemblage of cryptic species, which differ considerably in morphology, development, host specificity (including infectivity/pathogenicity for humans) and other aspects. This diversity is reflected in the mitochondrial and nuclear genomes and has led to the construction of phylogenetic trees and hypotheses on the origin and geographic dispersal of various taxa.
