Asymmetric Localizations Of Lin-17/Fz And Mig-5/Dsh Are Involved In The Asymmetric B Cell Division In C. Elegans, Mingfu Wu, Michael A. Herman
School of Biological Sciences: Faculty Publications
LIN-44/Wnt and LIN-17/Frizzled (Fz) function in a planar cell polarity (PCP)-like pathway to regulate the asymmetric B cell division in Caenorhabditis elegans. We observed asymmetric localization of LIN-17/Frizzled (Fz) and MIG-5/Disheveled (Dsh) during the B cell division. LIN-17∷GFP was asymmetrically localized within the B cell prior to and after the B cell division and correlated with B cell polarity. Asymmetric localization of LIN-17∷GFP was dependent upon LIN-44/Wnt and MIG-5/Dsh function. The LIN-17 transmembrane domain and a portion of the cysteine-rich domain (CRD) were required for LIN-17 function and asymmetric distribution to the B cell daughters, while the conserved KTXXXW …

The high prevalence of Duffy negativity (lack of the Duffy blood group antigen) among human populations in sub-Saharan Africa has been used to argue that Plasmodium vivax originated on that continent. Here, we investigate the phylogenetic relationships among 10 species of Plasmodium that infect primates by using three genes, two nuclear (β-tubulin and cell division cycle 2) and a gene from the plastid genome (the elongation factor Tu). We find compelling evidence that P. vivax is derived from a species that inhabited macaques in Southeast Asia. Specifically, those phylogenies that include P. vivax as an ancient lineage from …

The Evolution Of Primate Malaria Parasites Based On The Gene Encoding Cytochrome B From The Linear Mitochondrial Genome, Ananias A. Escalante, Denise E. Freeland, William E. Collins, Altaf A. Lal
Harold W. Manter Laboratory of Parasitology: Library Materials
We report a phylogenetic analysis of primate malaria parasites based on the gene encoding the cytochrome b protein from the mitochondrial genome. We have studied 17 species of Plasmodium, including 14 parasitic in primates. In our analysis, four species were used for rooting the Plasmodium phylogenetic tree: two from closely related genera (Hepatocystis sp. and Haemoproteus columbae) and two other Apicomplexa (Toxoplasma gondii and Theileria parva). We found that primate malaria parasites form a monophyletic group, with the only exception being the Plasmodium falciparum–Plasmodium reichenowi lineage. Phylogenetic analyses that include two species …

Harold W. Manter Laboratory of Parasitology: Library Materials
We report the results of molecular analysis of 39 isolates of Cryptosporidium parvum from human and bovine sources in nine human outbreaks and from bovine sources from a wide geographic distribution. All 39 isolates could be divided into either of two genotypes, on the basis of genetic polymorphism observed at the thrombospondin-related adhesion protein (TRAP-C2) locus. Genotype 1 was observed only in isolates from humans. Genotype 2, however, was seen in calf isolates and in isolates from a subset of human patients who reported direct exposure to infected cattle or consumed items thought to be contaminated with cattle feces. Furthermore, …
Parasite Biodiversity And Emerging Pathogens: A Role For Systematics In Limiting Impacts On Genetic Resources, Eric P. Hoberg
Parasite Biodiversity And Emerging Pathogens: A Role For Systematics In Limiting Impacts On Genetic Resources, Eric P. Hoberg
Harold W. Manter Laboratory of Parasitology: Faculty Publications
Emergence of pathogenic organisms continues as a threat to overall biodiversity and genetic resources. Macroparasites including helminths constitute a potential threat to economically important resources in agriculture and conservation biology. Limitation of this threat can be achieved through survey and inventory for biodiversity and the application of systematics to understand the host range , biogeography and history of faunas. Systematics constitutes the foundation for recognition of endemic and introduced elements of faunas and the basis for predicting the behavior of pathogens introduced to new ecological settings or host groups. The basis for emergence of pathogens has both a deep historical …
Microevolutionary Patterns And Molecular Markers: The Genetics Of Geographic Variation In Ascaris Suum, Steven A. Nadler
Microevolutionary Patterns And Molecular Markers: The Genetics Of Geographic Variation In Ascaris Suum, Steven A. Nadler
Harold W. Manter Laboratory of Parasitology: Faculty Publications
Molecular markers have been used only rarely to characterize the population genetic structure of nematodes. Published studies have suggested that different taxa may show distinct genetic architectures. Isoenzyme and RAPD markers have been used to investigate geographic variation of Ascaris suum at the level of infrapopulations (nematodes within individual hosts), within localities, and among geographic regions. Independent estimates of genetic differentiation among population samples based on isoenzyme and RAPD data showed similar patterns and substantial correlation. Heterozygote deficiencies within infrapopulations and large values for inbreeding coefficients among infrapopulations suggested that the composition of these populations was not consistent with a …
Breeding Sheep For Worm Resistance, John Karlsson, Johan Greeff, Julia Harris
Breeding Sheep For Worm Resistance, John Karlsson, Johan Greeff, Julia Harris
Journal of the Department of Agriculture, Western Australia, Series 4
Sheep production os one of Western Australia's most important agricultural industries. However, it is faced with the serious threat of sheep worm populations becoming increasingly resistant to the available drenches.
Although it's not a 'quick fix' solution, part of the long term answer may be selection for sheep with greater resistance to worms.
Phylogeny Of The Malarial Genus Plasmodium, Derived From Rrna Gene Sequences, Ananias A. Escalante, Francisco J. Ayala
Phylogeny Of The Malarial Genus Plasmodium, Derived From Rrna Gene Sequences, Ananias A. Escalante, Francisco J. Ayala
Harold W. Manter Laboratory of Parasitology: Library Materials
Malaria is among mankind's worst scourges, affecting many millions of people, particularly in the tropics. Human malaria is caused by several species of Plasmodium, a parasitic protozoan. We analyze the small subunit rRNA gene sequences of 11 Plasmodium species, including three parasitic to humans, to infer their evolutionary relationships. Plasmodium falciparum, the most virulent of the human species, is closely related to Plasmodium reiehenowi, which is parasitic to chimpanzee. The estimated time of divergence of these two Plasmodium species is consistent with the time of divergence (6-10 million years ago) between the human and chimpanzee lineages. The …
Method Of Karyotype Determination For Cercaria: Application To Family Azygiidae (Trematoda), James Everett Mick
Method Of Karyotype Determination For Cercaria: Application To Family Azygiidae (Trematoda), James Everett Mick
Masters Theses
The development of a method for karyotype determination of cercaria provides a basis for comparative karyotype analysis between cercaria and adult, possibly eliminating the need for infection experiments for positive identification of cercaria. Snails (Goniobasis livescens) were collected from below the spillway of Lake Charleston, on the Embarras River, near Charleston, Illinois. Of a total of 196 snails collected 42 (21.5%) were found to be infected with undetermined furcocyticercous cercariae. Infected snails were detected through isolation procedure. From the infected snails, emerged and non-emerged forms of the cercaria were recovered. Whole mounts of the cercaria were made and …
