Mhc Gene Analysis And Immune Variation In Lepidodactylus Lugubris (The Mourning Gecko), Helen T. Vu

2025 Fall Honors Capstones Projects - Archive

The major histocompatibility complex (MHC) plays a central role in vertebrate immune defense by enabling pathogen recognition and initiating adaptive immune responses. While MHC diversity has been extensively studied in sexually reproducing species, comparatively little is known about its evolution in asexual lineages. The parthenogenetic gecko Lepidodactylus lugubris provides a unique opportunity to investigate how clonality and hybrid origins influence immune gene diversity. By leveraging whole-genome data, this study overcomes the limitations of previous transcriptome-based analyses and provides a more comprehensive view of MHC diversity in a clonal vertebrate. As the first step towards understanding MHC evolution in parthenogenetic lizards, …

The Effect Of 1,25-Dihydroxyvitamin D3 Supplementation On Humoral Immunity Against Bacterial Chondronecrosis With Osteomyelitis Lameness In Broiler Chickens, Bailey Herberger

Animal Science Undergraduate Honors Theses

Bacterial chondronecrosis with osteomyelitis (BCO) lameness, the most predominant locomotive illness in fast-growing broilers, causes substantial animal well-being issues and affects broiler production, leading to huge financial losses. Several strategies, including feed supplementations using organic trace minerals, vitamin D, probiotics, and prebiotics, have been utilized to alleviate BCO lameness incidences in broiler chickens. Previous studies have indicated that feeding an additive of Panbonis®-G-1,25(OH)2D3 from Solanum glaucophyllum containing 1 µg/kg G-1,25(OH)2D3 to broilers for the first 28 days of their production cycle reduced BCO lameness by more than 50%. In this study, we …

Genetic Analysis Of Methionine Restriction-Associated Mitochondrial Unfolded Protein Response Signaling., Josh D. Dodge Jan 2025

Genetic Analysis Of Methionine Restriction-Associated Mitochondrial Unfolded Protein Response Signaling., Josh D. Dodge

Biology Dissertations - Archive

The lifespan of an organism is dictated by various genetic and environmental factors. One such determinant is the health of mitochondria which are organelles that mediate various critical cell functions including a significant role in generating the vast supply of cellular energy. Indeed, the gradual decline of mitochondrial function is believed to be a hallmark and possible cause of aging. Paradoxically, mild mitochondrial dysfunction early in life extends lifespan in various model organisms. Cells use diverse approaches to promote mitochondrial homeostasis, including the transcriptional reprogramming of cells by mitochondrial unfolded protein response (UPRmt) to mitigate mitochondrial stress. Consistently, UPRmt activation …

Immune Outcomes Affected By Selection For Parasite Resistance In Katahdin Sheep, Kelsey Lynne Bentley Jan 2024

Immune Outcomes Affected By Selection For Parasite Resistance In Katahdin Sheep, Kelsey Lynne Bentley

Graduate Theses, Dissertations, and Problem Reports (ETD)

Our understanding of the complex connection between selection for parasite resistance and the overall health outcomes in livestock is unclear. This study investigates the impact of post-weaning fecal egg count (PFEC) estimated breeding values (EBV) on health outcomes and immune responses in Katahdin sheep. Selection for PFEC, favoring Low-PFEC lambs, not only affects survivability to weaning but also suggests broader implications for immunity beyond parasite resistance. The effect of PFEC EBV on antibody production in response to vaccination against Clostridium perfringens type C&D and tetanus toxoid (CD&T) revealed a stepwise decrease in immunoglobin (IgG) from Low-PFEC to High-PFEC groups upon …

Effects Of Cyclic Heat Stress On The Acute Inflammatory Response In Broilers, Alessandro J. Rocchi May 2023

Effects Of Cyclic Heat Stress On The Acute Inflammatory Response In Broilers, Alessandro J. Rocchi

Poultry Science Undergraduate Honors Theses

Heat stress (HS) is a growing concern in broiler production due to increasing environmental temperatures. Little is known of the overall effect of HS on innate immunity. Lipopolysaccharide (LPS), an outer membrane component of Gram-negative bacteria, is commonly used to study the inflammatory response. In avian species, the local tissue and systemic inflammatory activities in response to LPS may be determined concurrently, over time, in an individual, using the growing feather (GF) pulp dermal test along with blood measurements (dual-window approach). To examine the effect of cyclic HS on the local and systemic acute inflammatory responses, Cobb 500 male broiler …

Cluster Homolog Of Immunoglobulin-Like Receptor Genes In Chicken Immune Responses, Brandi A. Sparling Jan 2023

Cluster Homolog Of Immunoglobulin-Like Receptor Genes In Chicken Immune Responses, Brandi A. Sparling

Graduate Theses, Dissertations, and Problem Reports (ETD)

This dissertation explores the identity and role of immunoglobulin-like (Ig-like) receptors in chickens, with focus on their implications in disease and disease progression. These receptors, wisely expressed across immune cells, interact with the major histocompatibility complex (MHC) class I molecules to modulate immune responses in mammals. Due to the insufficient representation of chicken Ig-like receptors in online databases, this study systematically annotates the chicken Cluster Homolog of Immunoglobulin-like Receptors (CHIR) genes using advanced bioinformatic techniques, aligning with the release of the 7th edition of the chicken genome assembly that comprises builds for a broiler and layer chicken. The analysis identifies …

Effects Of A Low Crude Protein Diet With And Without Spirulina Platensis Inclusion On The Concentrations And Proportions Of Circulating Immune Cells In Broilers, Heather Glenn May 2021

Effects Of A Low Crude Protein Diet With And Without Spirulina Platensis Inclusion On The Concentrations And Proportions Of Circulating Immune Cells In Broilers, Heather Glenn

Poultry Science Undergraduate Honors Theses

Immune system health is an important aspect of broiler production, especially with the reduction of the use of antibiotics. Nutritional aspects, like crude protein percentage, and natural alternatives to antibiotics are being investigated to see the effect they have on immunity. Spirulina is an alternative proteinaceous ingredient that producers are looking into, not only due to its high nutritional value, but also its immune boosting properties. The purpose of this study was to determine the effects of a low crude protein diet (LCP) and a Spirulina-supplemented low crude protein (LCP-SP) diet on broiler blood cell measurements, including, concentrations and proportions …

Spirulina microalgae is an alternative protein source under consideration for feed formulation in commercial broiler production. The purpose of this study was to determine the effects of a low crude protein (LCP) diet and a LCP diet formulated with 100 g/kg Spirulina (LCP-SP) on blood cell measurements in broilers. One-day-old Ross 708 male broilers were assigned to three dietary treatments: a standard crude protein (SCP), the LCP, or the LCP-SP diet, with five pens/treatment. When the chickens were 37-days old, blood samples were obtained from 2 birds/pen. Each blood sample was used to determine 1) the concentrations of white blood …

Muscle Mass And Immune Function In The Senior Horse, Alisa Christina Herbst Jan 2021

Muscle Mass And Immune Function In The Senior Horse, Alisa Christina Herbst

Theses and Dissertations--Veterinary Science

Senior horses (≥ 15 years) represent up to one-third of the global equine population, and the proportion of old horses (≥ 20 years) in the U.S. has been steadily increasing. Aging is associated with a loss of skeletal muscle mass in horses, and while age-related muscle loss is comparingly well characterized in humans, little is currently known concerning underlying mechanisms, adverse outcomes, or the prevalence of low muscle mass in senior horses. One factor proposed to play a role in the development of age-related muscle atrophy in humans is inflamm-aging, a low-grade inflammation that affects elderly people and that has …

The Effects Of Genetic Differences In An Innate Immune Receptor On Immune Responses To Haemonchus Contortus In Sheep, Denzel Deshawn Middleton

Graduate Theses, Dissertations, and Problem Reports (ETD)

Little is known about differences observed in early immunological signaling events that result in cellular infiltration and Th2 cytokine production that mediate parasite expulsion. Parasite resistant St. Croix (STC) sheep have been used as model for full host protective immunity as they present the ability to respond to larval stages of H. contortus. RNA sequencing analysis of peripheral blood mononuclear cells (PBMC) stimulated with H. contortus larval antigen (HcLA) revealed significant upregulation in NLRP3 and its pathway component TLR4. Predicted protein structural software identified distinct differences in NLRP3 protein structure in STC and parasite susceptible Suffolk (SUF) sheep. NLRP3 …

Haemonchus Contortus Antigen Identification And Use Of Tropomyosin In Ovine Vaccine Development, Brynnan P. Russ Jan 2020

Haemonchus Contortus Antigen Identification And Use Of Tropomyosin In Ovine Vaccine Development, Brynnan P. Russ

Graduate Theses, Dissertations, and Problem Reports (ETD)

Globally, the small ruminant industry loses hundreds of millions of dollars due to parasitism by the gastrointestinal nematode Haemonchus contortus. This parasite feeds on blood in the host’s abomasum leading to disease including anemia, hypoproteinemia, lethargy, and death. There have been exhaustive efforts to manage this parasite including different management applications, anthelmintic treatment, and now integration of resistance genetics into a flock. Like antibiotics, overuse of anthelmintics has led to resistance of H. contortus to these drugs. Therefore, other methods of control are the main focuses of many research initiatives. St. Croix are a resistant breed of sheep that mount …

Relationships Between Animal Temperament And Systemic Immune Responses In Beef Cattle Exposed To Conditions Associated With Conventional Management, Alexander W. Altman Jan 2019

Relationships Between Animal Temperament And Systemic Immune Responses In Beef Cattle Exposed To Conditions Associated With Conventional Management, Alexander W. Altman

Theses and Dissertations--Animal and Food Sciences

Measures of temperament have been shown to influence physiological responses. Exit velocity (EV) has been identified as an objective, robust measure of temperament that can be used to predict subsequent performance of cattle. Additionally, previous studies from our lab indicate this measure of temperament may be related to production of interferon-γ (IFN-γ), a cytokine associated with cell-mediated immunity (CMI). Whereas research has investigated effects of EV upon immune responses, the overall goal of these studies was to examine this relationship under a variety of scenarios including human handling, transportation, and exposure to endophyte-infected tall fescue (E+) for determination of its …

Ectopic lymphoid structures (ELS) consist of B-cell and T-cell aggregates that are initiated de novo in inflamed tissues outside of secondary lymphoid organs. When organized within follicular dendritic cell (FDC) networks, ELS contain functional germinal centers that can yield autoantibody-secreting plasma cells and promote autoimmune disease. Intranasal instillation of lupus-prone mice with crystalline silica (cSiO2), a respirable particle linked to human lupus, triggers ELS formation in the lung, systemic autoantibodies, and early onset of glomerulonephritis. Here we tested the hypothesis that consumption of docosahexaenoic acid (DHA), an ω-3 polyunsaturated fatty acid with anti-inflammatory properties, influences the temporal profile …

Transcriptomic Response To Immune Challenge In Zebra Finch (Taeniopygia Guttata) Using Rna-Seq, Cassandra Scalf Apr 2018

Transcriptomic Response To Immune Challenge In Zebra Finch (Taeniopygia Guttata) Using Rna-Seq, Cassandra Scalf

Masters Theses & Specialist Projects

Despite the convergence of rapid technological advances in genomics and the maturing field of ecoimmunology, our understanding of the genes that regulate immunity in wild populations is still nascent. Previous work to assess immune function has relied upon relatively crude measures of immunocompetence. However, with next-generation RNA-sequencing, it is now possible to create a profile of gene expression in response to an immune challenge. In this study, captive zebra finch (Taeniopygia guttata; adult males) were challenged with bacterial lipopolysaccharide (2 mg/Kg BW; dissolved in 0.9% saline) or vehicle (0.9% saline) to stimulate the immune system. Two hours after injection, birds …

Finding A Link Between Circadian Rhythms And The Immune System Of Captive Zebra Finches (Taeniopygia Guttata), Amber Bishop

Mahurin Honors College Capstone Experience/Thesis Projects

Circadian rhythms are commonplace in organisms and are normally controlled by a master clock. More recent evidence suggests that autonomous clocks operate in various systems in the body, including the immune system. This study looks for such a connection between the circadian rhythm and the immune system. In this study, captive zebra finches (Taeniopygia guttata) were exposed to different light and dark cycles and blood samples were taken every six hours. Birds were exposed to 12 hours of light and then 12 hours of darkness (LD), 24 hours of darkness (DD), or 48 hours of DD. After collecting …

Aeromonas Hydrophila In Amphibians: Harmless Bystander Or Opportunistic Pathogen, Zachary P. Rivas Jan 2016

Aeromonas Hydrophila In Amphibians: Harmless Bystander Or Opportunistic Pathogen, Zachary P. Rivas

Honors Undergraduate Theses

For several decades amphibian populations have been declining. Historically, the bacterium A. hydrophila (Ah) was hypothesized to be the causal factor in amphibian disease and population declines. However, with the discovery of a chytrid fungus, Batrachochytrium dendrobatidis (Bd) in 1998, which was identified on the skin of amphibians during documented mortality events, Ah research became of minor interest as focus shifted to Bd. Recent studies into the immunocompromising abilities of Bd, however, have opened new questions about its relationship with Ah and their combined effects on a host.

In this study, I explore the …

Parasite Infection Mediates Trait Tradeoffs In Fundulus Heteroclitus, Sarah Dunn May 2015

Parasite Infection Mediates Trait Tradeoffs In Fundulus Heteroclitus, Sarah Dunn

Honors College Theses

To be successful, an animal must eat, grow, and reproduce. With limited resources, there are tradeoffs between these critical life history parameters but the direction of the tradeoffs is largely unknown in a changing environment. To determine whether environmental context affects life-history tradeoffs, I surveyed and quantified investment into reproduction, growth, and a proxy for immunity (parasitism), in the mummichog, Fundulus heteroclitus, a common inhabitant of salt marshes in Georgia. Three salt marsh sites along coastal Georgia (Shellman Bluff, Skidaway Island, and Tybee Island) were selected using a proxy for anthropogenic disturbance (impervious surface), which also fell along a …
