Central African tropical forests are rich in biodiversity and home to some of Earth’s most threatened mammals. However, mammals in the region are under increasing threat from habitat loss and degradation, coupled with an increased demand for wild meat. To conserve mammals, wildlife managers and community-led conservation efforts must understand the distribution and diversity of mammal communities. However, funding and logistical challenges make contemporary survey methods, e.g. ground surveys, difficult and expensive. Noninvasive methods allow efficient mammal diversity surveys with minimal field effort. This dissertation was written on topics related to the genus Quercus with a primary focus on Quercus ellipsoidalis (northern pin oak) and Quercus rubra (northern red oak). Within this dissertation are chapters related to the setup of experimental common gardens within the Ford and Kellogg experimental forest, a literature review describing drought adaptations of Quercus sect. Short Tandem Target Mimic (STTM) is a commonly technique used for functional studies of a number of genes in several plant model systems. However, very little is known about application of STTM technique in tree species. In this study, STTM was applied to knock down microRNA396 (miR396) in transgenic poplar trees for the first time. STTM396 expression resulted in dramatic decrease in miR396 expression levels leading to taller plants with larger leaves and larger leaf cell size. 'Northern Hardwoods,’ are a characteristic composition of multiple tree species and a part of the rich and diverse northeastern forests. Hardwoods e.g. oaks (Quercus: Fagaceae and Sugar maple), serve as foundation species and offer ecological and economical gains to wildlife and humans.
