During the past two decades, genetic studies of model organisms have been the most important tool underlying advances in understanding the biological basis of aging and longevity. Drosophila melanogaster, the geneticist's "fruit fly," is a model organism because it has been the focus of genetic studies for more than 90 years. This review argues that studies on D. melanogaster will make an especially important contribution to the field of aging and longevity at the intersection of research on genetics, complex traits, and fly populations.

Herbivore damage is generally detrimental to plant fitness, and the evolutionary response of plant populations to damage can involve either increased resistance or increased tolerance. While characters that contribute to resistance, such as secondary chemicals and trichomes, are relatively well understood, characters that contribute to a plant’s ability to tolerate damage have received much less attention. Using Helianthus annuus (wild sunflower) and simulated damage of Haplorhynchites aeneus (head-clipping weevil) as a model system, we examined morphological characters and developmental processes that contribute to compensatory ability. The purpose of this study was to investigate medfly longevity and reproduction across a broad spectrum of diet restriction using a protocol similar to those applied in most rodent studies. Age-specific reproduction and age of death were monitored for 1,200 adult males and 1,200 females, each individually maintained on one of 12 diets from ad libitum to 30% of ad libitum. Diet was provided in a fixed volume of solution that was fully consumed each day, ensuring control of total nutrient consumption for every fly. The Ute ladies’-tresses, Spiranthes diluvialis, is listed as a threatened orchid in west-central United States by the Federal government. Information on its origin and patterns of genetic variation is needed to develop effective conservation strategies for this species. DNA sequencing and polymerase chain reaction–restriction fragment length polymorphism (PCR-RFLP) was used to evaluate genetic variation and structure of 23 populations of S. diluvialis. In addition, four congeneric species were analyzed to determine possible origins of the putative allotetraploid S. diluvialis. Female Sensitivity To Diet And Irradiation Treatments Underlies Sex-Mortality Differentials In The Mediterranean Fruit Fly, James R. Carey, Pablo Liedo, Hans-Georg Müller, Jane-Ling Wang, Brad Love, Lawrence G. Harshman, Linda Partridge

Large-scale experiments on medflies that were subjected to sterilizing doses of ionizing radiation (plus intact controls) and maintained on either sugar-only or full, protein-enriched diets revealed that, whereas the mortality trajectories of both intact and irradiated male cohorts maintained on both diets are similar, the mortality patterns of females are highly variable. Mean mortality rates at 35 days in male cohorts ranged from 0.2 to 0.3 but in female cohorts ranged from 0.09 to 0.35, depending on treatment. The reputation of the field of psychiatric genetics has recently become tarnished in the view of many human geneticists. Too many linked loci were claimed and withdrawn, too many association studies published and not confirmed, and, more recently, too many new and different chromosomal regions have been implicated for the same disorder. Here, we summarize recent trends, focusing on research that moves away from traditional linkage studies. Some promising strategies include psychopharmacogenetics and consideration of endophenotypes such as neurophysiological and behavioral markers in addition to the clinical diagnosis. Oxidative Stress Resistance: A Robust Correlated Response To Selection In Extended Longevity Lines Of Drosophila Melanogaster?, Lawrence G. Harshman, Beth A. Haberer

Stress resistance is associated with longevity in Drosophila melanogaster and other model organisms used for genetic research. The present study tests for oxidative stress resistance in one set of lines selected for late-life reproduction and extended longevity. Both females and males from the selected lines were appreciably more resistant to oxidative stress than were flies from the control lines. A relative increase in oxidative stress resistance is a correlated response to selection in this laboratory selection experiment. Increased oxidative stress resistance appears to be a relatively robust correlated response to laboratory selection for late-life reproduction and extended longevity.

Unique cytoplasmic filaments are found in the treponeme genus of spirochete bacteria. Their function is unknown, but their location underneath the periplasmic flagellar filaments (PFF) suggests a role in motility and/or cell structure. To better understand these unique structures, the gene coding for the cytoplasmic filaments, cfpA, was identified in various treponemal species. Treponema phagedenis cfpA was 2,037 nucleotides long, and the encoded polypeptide showed 78 to 100% amino acid sequence identity with the partial sequence of CfpA from T. denticola, T. vincentii, and T. pallidum subsp. pertenue. The treponemal fla operon is comprised of numerous motility-related genes; however, the initial gene of this operon, tap1, has no known function. A recently developed system to generate specific mutants in Treponema denticola was utilized to determine if Tap1 was essential for motility. T. denticola tap1 and flanking DNA were identified, cloned, and sequenced, and a suicide plasmid that contained tap1 interrupted with an erythromycin resistance cassette (ermF and ermAM) was constructed. Because of potential polar effects from this cassette, a second plasmid that contained tap1 interrupted with a modified erythromycin resistance cassette that lacked the putative ermF transcription …

Radioimmunoassay was used to determine ecdysteroid titers in mated or unmated Drosophila melanogaster females. Whole-body ecdysteroid titers increase after mating and this response is more pronounced after 12–24 hours than it is immediately after mating. In one experiment, females were mated to transgenic males deficient in accessory gland proteins to test whether these peptides mediate the observed increase in female whole-body ecdysteroid titers. Females mated to such transgenic males do not show a pronounced increase in whole-body ecdysteroid titers. The effect of mating on female hemolymph ecdysteroid titers was also investigated. The hybrid bridge hypothesis suggests that plant hybrids ‘‘bridge’’ the genetic gap between actual and potential host species, and that, for this reason, herbivorous insects are more likely to evolve an expanded host range in the presence of hybrids. While intuitively appealing, the hypothesis has two implicit assumptions: that phenotypic gaps between potential hosts limit host range, and that characters controlling host use are additively inherited in plant hybrids. Evaluation of these assumptions suggests that operation of the hybrid bridge hypothesis is relatively uncommon. Alcoholism is transmitted in families. The complexity and heterogeneity of this disorder has made it difficult to identify specific genetic correlates. One design with the potential to do so is the family-based association study, in which the frequencies of genetic polymorphisms are compared between affected and nonaffected members. Reduced central serotonin neurotransmission is associated with features of an antisocial subtype of alcoholism, although a primary deficit has not been traced to a particular component. Genetic markers related to the serotonergic system have been identified, located, and cloned. If associations can be discovered, the development process for pharmacotherapy could be facilitated. …

The complex mechanisms of heredity are little appreciated by nonspecialists, in some measure, because of misunderstandings that are perpetuated when words used for technical terms have other, more widely understood, folk meanings. When a word has both technical and folk meanings, it is the responsibility of the specialist to avoid promoting confusion by either using extremely cautious and precise language when using the term or, in cases when confusion is inevitable, abandoning the term in favor of one without a widely understood folk meaning. Previously, all of the major fruit fly, Drosophila melanogaster, chromosomes (I, II, and III) have been shown to be associated with geotaxis, but the Y chromosome has not. Using two methods (back-crossing and chromosome substitution), Y chromosomes from lines that have evolved stable, extreme expressions of geotaxis were placed into different genetic and cytoplasmic backgrounds to test the resulting males for geotaxis. The results of the back-crossing do not support the interpretation of Y-chromosome effects on geotaxis. These tests do not have sufficient statistical power, however, to detect small genetic effects. A subregion of the mitochondrial large subunit (16s) rRNA gene was amplified by polymerase chain reaction (PCR) from nine species of blackflies (Diptera: Simuliidae) which serve as natural or experimental vectors of human or bovine Onchocerca parasites. PCR products from each species of blackfly were tested by directed heteroduplex analysis (DHDA), and their genotypes established according to diagnostic banding patterns of the heteroduplex products. As part of a long-term study (38+ years) to identify genetic correlates of geotaxis in Drosophila melanogaster, the investigators report the results of allozyme-level analyses of 2 lines derived from hybridizing high- and low-selected lines that have evolved stable, extreme expressions of geotaxis. Allelic variation at Adh was associated with geotactic performance in 1 hybrid-derived line, but not in another, after 66 free-mating generations beyond the F2 generation. A second-chromosome gene correlate of geotaxis may lie within 1 map unit of Adh. Population genetic analyses suggest that there were different selection pressures on the hybrid-derived lines and …

Mitochondrial DNA was amplified and sequenced from eastern, western, and Egyptian strains of alfalfa weevil, Hypera postica (Gyllenhal). Eastern and Egyptian weevils differed at only 2 nucleotide sites in 1,031 base pairs sequenced; western weevils differed by 5% sequence divergence. Three restriction sites were identified which separated eastern and western haplotypes. No intrastrain polymorphism was detected in 150 weevils from Nebraska. Collections from Lincoln in eastern Nebraska and Scottsbluff in western Nebraska were fixed for the eastern and western haplotypes, respectively. Eastern and western haplotypes were found together in the same fields in a broad overlap region in central Nebraska.

Molecular markers have been used only rarely to characterize the population genetic structure of nematodes. Published studies have suggested that different taxa may show distinct genetic architectures. Isoenzyme and RAPD markers have been used to investigate geographic variation of Ascaris suum at the level of infrapopulations (nematodes within individual hosts), within localities, and among geographic regions. Independent estimates of genetic differentiation among population samples based on isoenzyme and RAPD data showed similar patterns and substantial correlation. Heterozygote deficiencies within infrapopulations and large values for inbreeding coefficients among infrapopulations suggested that the composition of these populations was not consistent with a …

The behavior-genetic analysis of Drosophila melanogaster with geotactic performance as the phenotype is an ideal model system with which to investigate the complex relations between heredity and behavior. As part of a long-term, 38-year study, we report 4 experiments that identify and analyze trait correlations in the selected high- and low-geotaxis lines. We performed F2 correlational analyses and backcrosses to examine 3 types of correlations: (a) genotype-genotype (alcohol dehydrogenase [Adh]-amylase [Amy]), (b) genotype-phenotype (Adh and Amy-geotaxis), and (c) phenotype-phenotype (mate preference–geotaxis). Sequences are given for nine complete genes and one partial mitochondrial tRNA gene of the black fly, Simulium vittatum (Zetterstedt). Sequenced tRNA genes were for alanine, arginine, asparagine, aspartic acid, glutamic acid, glycine, leucine(CUN), lysine, serine(AGN), and valine. Nucleotides were aligned with the same previously sequenced genes in Aedes albopictus Skuse and Drosophila yakuba Burla. A cluster of six tRNA genes, which differ in arrangement in Ae. albopictus and D. yakuba, was amplified by PCR and found to have the same position and orientation in S. vittatum as in D. yakuba. There are significant differences if the distribution of weedy characteristics among weeds, normal plants, and crops. The world’s most serious weeds possess on the average 10 or 11 of these characters, a random collection of British plants have an average seven of the traits, and crop plants only five. For the average crop to become as “weedy” as the average weed, it would need to acquire five weedy traits. Even using the unlikely assumption that those traits are single loci in which a dominant mutation would provide the weedy character, this would require the simultaneous acquisition of five gene substitutions. …

The effects of density and food on remating were investigated using Drosophila melanogaster. The frequency of remating was unaffected by density for some combinations of fly strains but was reduced at low relative densities for other combinations. Until females had used most of their stored sperm, remating was less likely when food was absent or contact with food was prevented. Food availability had little effect on the incidence of remating once stored sperm were depleted and had no effect on initial virgin mating frequency. Bats of the genus Lasiurus present a number of interesting systematic problems that are difficult to resolve by traditional techniques. Members of the genus share a suite of derived morphological (Hall and Jones, 1961; Handley , 1960) and karyotypic (Bickham 1979, 1988) characteristics. However, until 1960 (Handley, 1960), members were placed in two genera- Lasiurus and Dasypterus-based primarily upon the presence or absence of the small, first upper premolar. Populations of Alsophila pometaria (Harris) are largely gynogenetic and composed of asexual females that must mate with a conspecific male to reproduce. Within this asexual contingent, there are a variety of electrophoretically defined multilocus genotypes. In this study we found that populations differ in sex ratio. We also found that there is an indication of differential mating success among asexual genotypes in a population of A. pometaria.

Effects of male Drosophila melanogaster on female fecundity and productivity were examined, considering both females held in containers with males and females exposed to male effects not involving contact. Females were more fecund when male effects were present, and the largest increase was recorded when vials were previously conditioned by males. A survey of spatial and temporal variation in the frequency of electrophoretically defined genotypes in the geometrid moth Alsophila pometaria revealed a high diversity of uncommon or rare asexual genotypes and clinal distributions of two of the more common clones. There was substantial year-to-year variation in genotype frequencies in seven of eleven sites. Progeny tests have revealed that sexual reproduction is uncommon in two populations and that new asexual genotypes arise from the sexual population. The recurrent origin of asexual genotypes is likely to account for the high genetic and ecological diversity of the asexual contingent of this species’ populations, …

A group of Escherichia coli isolates from nature were compared with one another and with laboratory strains of E. coli with respect to size distribution of chromosomal restriction endonuclease fragments and differences in nucleotide sequences in selected small portions of the genomes. The estimated frequency of base substitutions in nucleotide sequences in and near the trp operons of 26 of the 28 E. coli strains examined ranged from 0.008 to 0.066. For the past decade, numerous investigators have attempted to explain the plethora of genetic polymorphism found in nature (see Ayala 1976). However, few researchers have been able to document how polymorphisms have been maintained, whether the polymorphisms were genic or chromosomal. Our investigations into the maintenance of a Robertsonian chromosomal polymorphism in the plains pocket goher, Geomys bursarius major, have revealed evidence that this polymorphism is maintained by differential viabilities of the three chromosomal morphs, with the heterozygote being favored.
