In the past 20 years, the number of subterranean taxa discovered in Australia, especially in the Pilbara bioregion, has considerably increased due to incidental environmental surveys often associated with mining development. Bathynellidae are an important component of stygofauna and they occur in most Australian aquifers, but their collection and identification are difficult due to their habitat, and small and fragile bodies with conservative morphology. The study of Pilbaranella ethelensis in the upper Fortescue catchment contributed to a better understanding of the group at local scale, but knowledge at larger catchment scale is still limited. โรคติดเชื้อที่มีสาเหตุมาจาก Mycobacterium avium complex (MAC) มีอุบัติการณ์เพิ่มสูงขึ้นทั่วโลกรวมทั้งในประเทศไทย เนื่องมาจากการเพิ่มขึ้นของจำนวนผู้ป่วยที่มีภาวะภูมิคุ้มกันของร่ายกายต่ำ การวินิจฉัยเชื้อ MAC ในปัจจุบันที่อาศัยการเพาะเลี้ยงเชื้อและทดสอบปฏิกิริยาชีวเคมี นอกจากใช้ระยะเวลานานแล้ว ยังไม่สามารถจำแนกเชื้อได้ถึงระดับสปีชี่ส์ โดยเฉพาะอย่างยิ่งเชื้อ Mycobacterium avium และเชื้อ Mycobacterium intracellulare ซึ่งเป็นเชื้อ MAC ที่สำคัญและเพาะแยกได้บ่อยจากสิ่งส่งตรวจ ดังนั้นวัตถุประสงค์ของการศึกษาในครั้งนี้ คือการพัฒนาเทคนิค Recombinase Polymerase Amplification ร่วมกับการอ่านผลด้วยสี SYBR Green I (RPA/SYBR) ในการตรวจวินิจฉัยเชื้อ MAC ซึ่งอาศัยการเพิ่มปริมาณสารพันธุกรรมภายใต้อุณหภูมิคงที่ ร่วมกับไพรเมอร์ที่จำเพาะต่อเชื้อกลุ่ม MAC เชื้อ M. avium และเชื้อ M. intracellulare และอ่านผลที่เกิดขึ้นด้วยตาเปล่าได้ในทันทีภายหลังการเติมสี SYBR Green I ผลจากการทดสอบกับตัวอย่าง DNA ของเชื้อ MAC จำนวน 120 ตัวอย่าง พบว่าเทคนิค RPA/SYBR สามารถตรวจวินิจฉัยจำแนกสปีชี่ส์ของเชื้อ M. avium และเชื้อ M. intracellulare ได้อย่างมีประสิทธิภาพ โดยมีความไว 100% และความจำเพาะตั้งแต่ 80.7% ขึ้นไป ซึ่งจัดว่ามีความสอดคล้องในเกณฑ์ดีมากเมื่อเปรียบเทียบกับวิธีหาลำดับเบส (สถิติ Kappa มีค่าตั้งแต่ 0.801 ขึ้นไป) ในขณะที่เมื่อเปรียบเทียบกับเทคนิค PCR มีความไวตั้งแต่ 89.2% ขึ้นไปและความจำเพาะตั้งแต่ 97.5% ขึ้นไป และมีความสอดคล้องในเกณฑ์ดีมากเช่นเดียวกัน (สถิติ Kappa มีค่าตั้งแต่ 0.863 ขึ้นไป) นอกจากนี้เทคนิค RPA/SYBR ยังมีค่าความเข้มข้นของ DNA ต้นแบบน้อยที่สุดที่สามารถตรวจได้ เท่ากับ 1 ng/µl และไม่เกิดปฏิกิริยาข้ามกลุ่มกับเชื้อส่วนใหญ่ของ Mycobacterium แม้ว่าเทคนิค RPA/SYBR ที่ได้พัฒนาขึ้น มีขั้นตอนการทดสอบที่ทำง่าย …

เชื้อ Escherichia coli ที่สร้างเอนไซม์ Extended-spectrum β-lactamase (ESBLs) เป็นปัญหาสำคัญที่พบในโรงพยาบาลทั่วโลก การตรวจจีโนไทป์ของเอนไซม์ ESBLs เป็นวิธีที่สำคัญและมีประโยชน์ในการควบคุมการแพร่กระจายของเชื้อดื้อยาและการเลือกใช้ยาต้านจุลชีพอย่างเหมาะสม การศึกษานี้มุ่งหวังจะพัฒนาเทคนิค Multiplex Recombinase polymerase amplification (Multiplex RPA) และ Helicase dependent amplification (HDA) สำหรับตรวจหายีนดื้อยา blaCTX-M, blaOXA, blaSHV และ blaTEM ในเชื้อ E. coli ในสถาบันมะเร็งแห่งชาติ ประเทศไทย เชื้อ E. coli จำนวน 144 ตัวอย่างที่แยกได้จากสิ่งส่งตรวจภายในงานจุลชีววิทยา สถาบันมะเร็งแห่งชาติ ช่วงระหว่างเดือนกุมภาพันธ์ 2017 ถึงเดือนกันยายน 2018 ถูกนำมาทดสอบความไวต่อยาต้านจุลชีพและตรวจคัดกรองการสร้างเอนไซม์ ESBLs ด้วยวิธี Disk diffusion ตามวิธีมาตรฐานของ CLSI ปี ค.ศ. 2017 เชื้อที่สร้างเอนไซม์ ESBLs จำนวน 95 ตัวอย่าง พบดื้อยาสูงสุดในยา Ampicillin (100%), Cefotaxime (100%), Cefdinir (100%) และ Ceftriaxone (99.9%) และเชื้อ E. coli จำนวน 144 ตัวอย่าง มียีน blaCTX-M (100%) รองลงมาคือ blaTEM (42.1%), blaOXA (30.5%) และ blaSHV (2.1%) ตามลำดับ โดยเป็นยีน ESBLs ชนิด CTX-M-15 สูงสุด (51.1%) รองลงมาคือ CTX-M-55 (18.1%), CTX-M-27 (17.0%), CTX-M-14 …

อะไมลอยด์เบต้า 42 (Aβ42) เป็นของเสียที่เกิดจากการย่อยอะไมลอยด์เบต้าพรีเคอเซอร์โปรตีน อาจเสี่ยงต่อการเกิดโรคอัลไซเมอร์เมื่อสะสมที่สมอง โดยมียีนบีเอซีอีวัน (BACE1) ควบคุมการสร้างเอนไซม์เบต้าซีครีเตส เพื่อย่อยอะไมลอยด์เบต้าพรีเคอเซอร์โปรตีน และมียีนเอโปอี (ApoE) เป็นยีนที่ถอดรหัสเป็น Apolipoprotein E ทำหน้าที่ขนส่งอะไมลอยด์เบต้าไปกำจัดที่ตับ คาดว่าการติดเชื้อไวรัสตับอักเสบเรื้อรังอาจส่งผลให้การกำจัดของเสียลดลงด้วย งานวิจัยนี้จึงมีวัตถุประสงค์เพื่อศึกษาระดับอะไมลอยด์เบต้า 42 ในเลือดด้วยหลักการ ELISA และศึกษาความหลากหลายทางพันธุกรรมของยีน BACE1 (rs638405) ด้วยหลักการ PCR-RFLP และยีน ApoE (rs429358 และ rs7412) ด้วยหลักการ real time PCR ในผู้ติดเชื้อไวรัสตับอักเสบบีเรื้อรัง (CHB, N=149) ซีเรื้อรัง (CHC, N=31) และคนปกติ (N=164) โดยเป็นผู้ติดเชื้อไวรัสตับอักเสบบีเรื้อรัง ซีเรื้อรังที่รักษา ณ โรงพยาบาลพระมงกุฎเกล้า และผู้บริจาคโลหิต ณ สถาบันพยาธิวิทยา ในช่วงเดือน สิงหาคม 2561 - มิถุนายน 2562 ผลการศึกษาพบว่าระดับอะไมลอยด์เบต้า 42 ในเลือดของผู้ป่วย CHB มีค่าสูงกว่าคนปกติ (CHB=46.86 pg/ml (N=22), CHC=43.27 pg/ml (N=15) และ คนปกติ =33.72 pg/ml (N=36), p-value=0.002) ข้อมูลจากกลุ่มตัวอย่างที่ตรวจวัดนี้มีจำนวนจำกัด จึงยังไม่สามารถนำไปอ้างอิงได้ นอกจากนี้ผู้วิจัยพบสัดส่วนของจีโนไทป์ของยีน BACE1 ในผู้ป่วย CHB (N=149), CHC (N=31) มีจีโนไทป์ C/G สูงกว่าในคนปกติ (N=164) (p-value = 0.044) และยีน ApoE ในผู้ป่วย CHB (N=149), CHC (N=31) มีจีโนไทป์ ε3ε4 สูงกว่าในคนปกติ (N=148) (p-value=0.001) …

สถานการณ์การดื้อยาปฏิชีวนะของแบคทีเรียเป็นปัญหาใหญ่ที่ส่งผลกระทบไปทั่วโลก จึงจำเป็นต้องศึกษาเพื่อพัฒนาสารทดแทนยาปฏิชีวนะ ตัวเงินตัวทอง (Varanus salvator) เป็นสัตว์เลื้อยคลานมีความสามารถในการอยู่รอดจากสภาวะที่เสี่ยงต่อการติดเชื้อ อาจเนื่องจากสัตว์เหล่านี้มีระบบภูมิคุ้มกันโดยกำเนิดซึ่งมีวิวัฒนาการมาแต่โบราณจึงเป็นจุดเด่นที่น่าศึกษาวิจัย งานวิจัยนี้มีวัตถุประสงค์เพื่อศึกษาฤทธิ์ต้านเชื้อแบคทีเรียขององค์ประกอบเลือดจากตัวเงินตัวทองและโปรตีนหรือเปปไทด์ที่มีคุณสมบัติต้านจุลชีพ ซึ่งข้อมูลของลำดับกรดอะมิโนของตัวเงินตัวทองที่ได้ในการศึกษาวิจัยครั้งนี้จะเป็นพื้นฐานที่สำคัญใน โดยนำพลาสมาและซีรัมจากตัวเงินตัวทองมาทดสอบคุณสมบัติการยับยั้งเชื้อแบคทีเรียที่ทำให้เกิดโรคด้วยเทคนิค agar diffusion พบว่าพลาสมาสามารถยับยั้งการเจริญของเชื้อแบคทีเรียได้ดีที่สุดมีขนาดเส้นผ่านศูนย์กลางของวงใสยับยั้งเชื้อ (inhibition zone) เท่ากับ 17-20 มิลลิเมตร เมื่อทำการทดสอบหาค่าความเข้มข้นต่ำสุดของพลาสมาและซีรัมที่สามารถยับยั้งเชื้อแบคทีเรีย (minimal inhibitory concentration, MIC) ด้วยวิธี broth dilution พบว่าสารสกัดหยาบพลาสมาให้ผลการทดสอบกับเชื้อได้ดีที่สุดโดยมีค่าความเข้มข้นต่ำสุดที่สามารถยับยั้งเชื้อแบคทีเรีย (MIC) เท่ากับ 125 ไมโครกรัมต่อมิลลิลิตร จากผลของการแยกบริสุทธิ์บางส่วนของพลาสมาและซีรัมโดยเทคนิค ion exchange chromatography ด้วย Q - sepharose คอลัมน์ พบว่าทั้งพลาสมาและซีรัมสามารถแยกโปรตีนแฟรคชันได้ 5 แฟรคชันเท่ากัน เมื่อวิเคราะห์ด้วยเทคนิค sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) พบขนาดโมเลกุลอยู่ระหว่าง 16 – 200 กิโลดัลตัน แต่เมื่อทดสอบคุณสมบัติการต้านเชื้อ พบบางแฟรกชันจากพลาสมา (P) และซีรั่ม (S) ซึ่งกำหนดเป็น P1 / P2 / P5 และ S1 / S2 / S4 ตามลำดับ ที่แสดงฤทธิ์ต้านเชื้อแบคทีเรีย เมื่อวิเคราะห์องค์ประกอบของสารออกฤทธิ์ทางชีวภาพด้วยเทคนิค liquid chromatography mass spectrometry (LC-MS) สามารถระบุชนิดโปรตีนได้ทั้งหมด 33 ชนิดและพบโปรตีนที่มีศักยภาพเป็นเปปไทด์ต้านจุลชีพ 15 ชนิด มีเพียง 3 ชนิด จากส่วนของพลาสมา (P1) 2 ชนิด และ (P2) 1 ชนิด ที่มีโครงสร้างเป็น α-helical peptide …

งานวิจัยนี้มีวัตถุประสงค์เพื่อศึกษาองค์ประกอบทางพฤกษเคมีและฤทธิ์ต้านเชื้อรา Candida albicans ของน้ำมันเสม็ดขาวจากใบและกิ่งอ่อนของ Melaleuca cajuputi Powell ที่พบในพื้นที่ภาคตะวันออกของประเทศไทย รวมทั้งประเมินผลกระทบของน้ำมันเสม็ดขาวต่อการแสดงออกของยีนที่เกี่ยวข้องกับกลไกการดื้อยาฟลูโคนาโซลในเชื้อราทดสอบ การวิเคราะห์องค์ประกอบทางพฤกษเคมีด้วยวิธีแก๊สโครมาโทกราฟี-แมสสเปกโตรมิทรี (gas chromatography-mass spectrometry, GC-MS) พบว่าน้ำมันเสม็ดขาวมีองค์ประกอบหลัก 6 ชนิด ได้แก่ 1,8-Naphthyridine derivatives (ร้อยละ 10.46), alpha-Pyrone (ร้อยละ 10.11), Terpinolene (ร้อยละ 9.26), gamma-Terpinene (ร้อยละ 8.00), trans-Caryophyllene (ร้อยละ 6.36) และ beta-Elemene (ร้อยละ 5.09) จากการทดสอบฤทธิ์เบื้องต้นของน้ำมันเสม็ดขาวในการยับยั้ง C. albicans สายพันธุ์คลินิกที่ดื้อต่อยาฟลูโคนาโซล จำนวน 16 ไอโซเลท ด้วยวิธี Disc diffusion พบว่าดิสก์บรรจุน้ำมันเสม็ดขาว ขนาด 1 ไมโครลิตรต่อดิสก์ แสดงฤทธิ์ยับยั้งการเจริญของเชื้อทดสอบทุกไอโซเลท โดยมีขนาดเส้นผ่านศูนย์กลางบริเวณยับยั้งเชื้อเฉลี่ยอยู่ระหว่าง 6.67 ± 0.58 ถึง 10.00 ± 0.00 มิลลิเมตร เมื่อทำการทดสอบหาค่าความเข้มข้นต่ำสุดของน้ำมันเสม็ดขาวที่สามารถยับยั้งเชื้อทดสอบ (minimal inhibitory concentration, MIC) และฆ่าเชื้อทดสอบ (minimum fungicidal concentration, MFC) ด้วยวิธี Broth macrodilution พบว่าน้ำมันเสม็ดขาวแสดงฤทธิ์ฆ่าเชื้อราทดสอบทุกไอโซเลท โดยมีค่า MIC และ MFC อยู่ในช่วง 0.31-1.25 และ 0.63-2.5 ไมโครลิตรต่อมิลลิลิตร ตามลำดับ การทดสอบการเสริมฤทธิ์กันของสารผสมน้ำมันเสม็ดขาวและยาฟลูโคนาโซลในการต้านเชื้อราทดสอบด้วยวิธี Checkerboard microdilution พบว่าสารผสมดังกล่าวมีการเสริมฤทธิ์กันในการยับยั้งเชื้อราทดสอบส่วนใหญ่ (ร้อยละ 60) โดยค่า MIC ของยาฟลูโคนาโซลและน้ำมันเสม็ดขาวจะลดลงประมาณ 8-64 และ 8-16 เท่า ตามลำดับ …

การพัฒนาเทคนิค Helicase Dependent Amplification ควบคู่กับการอ่านผลด้วยตาเปล่าโดยใช้สี Sybr Green I (Hda/Sybr Green I) สำหรับการตรวจการปนเปื้อนเชื้อแบคทีเรียในผลิตภัณฑ์เกล็ดโลหิต, วรางคณา แย้มเกต

ผลิตภัณฑ์เกล็ดโลหิตที่มีการปนเปื้อนของเชื้อแบคทีเรีย อาจก่อให้การติดเชื้อในกระแสโลหิตที่รุนแรงและนำไปสู่การเสียชีวิตได้ ดังนั้นจึงมีการกำหนดมาตรฐานให้งานธนาคารโลหิต ทำการตรวจการปนเปื้อนเชื้อแบคทีเรียในผลิตภัณฑ์เกล็ดโลหิตก่อนนำไปใช้กับผู้ป่วย เพื่อลดอาการไม่พึงประสงค์ที่อาจเกิดขึ้น วิธีการตรวจการปนเปื้อนเชื้อแบคทีเรียในผลิตภัณฑ์เกล็ดโลหิตในปัจจุบันยังมีข้อจำกัดที่พบได้บางประการ อาทิเช่น การเพาะเลี้ยงเชื้อแบคทีเรียที่ไม่สามารถปฏิบัติได้ในห้องปฏิบัติการทางธนาคารโลหิต การทดสอบที่ใช้ระยะเวลานาน ซึ่งไม่เหมาะสมกับอายุการเก็บรักษาผลิตภัณฑ์เกล็ดโลหิตที่สั้นประมาณ 5 วัน และต้นทุนของการทดสอบที่มีราคาแพง วัตถุประสงค์ของการศึกษาครั้งนี้เพื่อพัฒนาเทคนิค Helicase dependent amplification ควบคู่กับการอ่านผลด้วยตาเปล่าโดยใช้สี SYBR Green I (HDA/SYBR Green I) สำหรับตรวจหาการปนเปื้อนเชื้อแบคทีเรียในผลิตภัณฑ์เกล็ดโลหิต โดยใช้ Primer ที่ถูกออกแบบให้จำเพาะต่อยีน 16s rRNA ซึ่งเป็น Universal gene ของเชื้อแบคทีเรีย ผลการทดสอบกับตัวอย่างผลิตภัณฑ์เกล็ดโลหิตที่ปนเปื้อนเชื้อแบคทีเรียแบบจำลอง ณ วันต่าง ๆ จำนวน 96 ตัวอย่าง พบว่าเทคนิค HDA/SYBR Green I มีความไวและความจำเพาะสูง เท่ากับ 96% และ 100% ตามลำดับ และมีความสอดคล้องกันในระดับดีมาก เมื่อเปรียบเทียบกับเทคนิค HDA/AGE เมื่อเปรียบเทียบกับเทคนิค PCR การนับจำนวนโคโลนี และเครื่องอัตโนมัติ BacT/Alert System พบว่าเทคนิค HDA/SYBR Green I มีความจำเพาะสูงเท่ากับ 100% แต่มีความไวอยู่ในช่วงระหว่าง 63-76% จึงทำให้มีความสอดคล้องกันในระดับปานกลางถึงดี อย่างไรก็ดี เมื่อนำเทคนิค HDA/SYBR Green I ตรวจการปนเปื้อนเชื้อแบคทีเรียในผลิตภัณฑ์เกล็ดโลหิตเฉพาะวันที่ 2 ถึง 5 ของการเก็บรักษา พบว่ามีความไวที่สูงขึ้น เท่ากับ 88% และมีความสอดคล้องกันในระดับดีมาก เทคนิค HDA/SYBR Green I มีค่าความเข้มข้นน้อยที่สุดของ DNA ต้นแบบที่สามารถตรวจได้ เท่ากับ 1 ng และไม่เกิดปฏิกิริยาข้ามกลุ่มกับสิ่งมีชีวิตอื่น ๆ ที่มีโอกาสพบการปนเปื้อนได้ในผลิตภัณฑ์เกล็ดโลหิต เทคนิค HDA/SYBR Green I …

การทดสอบความไวต่อยา Pyrazinamide (PZA) ด้วยวิธีมาตรฐาน (culture-based susceptibility tests) นั้นทำได้ยาก เนื่องจากสภาวะเป็นกรดในอาหารเลี้ยงเชื้อสามารถทำให้เกิดผลลบและผลบวกลวง การศึกษานี้มุ่งเน้นศึกษาและพัฒนาชุดทดสอบสำหรับใช้วินิจฉัยเชื้อดื้อต่อยา PZA ในเชื้อ Mycobacterium tuberculosis ด้วยชุดทดสอบ colorimetric biphasic medium assay ที่ผสม Nicotinamide (NIC-CBMA) ความเข้มข้น 250, 500, 1000 และ 2000 µg/mL และวิเคราะห์การกลายพันธุ์ที่สัมพันธ์กับการดื้อยา PZA ในยีน pncA, rpsA และ panD ผลการทดสอบในเชื้อวัณโรคดื้อยาทั้งหมด 150 สายพันธุ์ แบ่งเป็นเชื้อดื้อต่อยา PZA 40 สายพันธุ์ และเชื้อไวต่อยา PZA 110 สายพันธุ์ เมื่อทดสอบด้วยวิธี BACTEC MGIT 960 พบว่าความเข้มข้นของ NIC ≥ 1000 µg/mL เหมาะสำหรับใช้วินิจฉัยเชื้อดื้อยา PZA ด้วยวิธี NIC-CBMA ซึ่งมีความไวของชุดทดสอบเท่ากับ 100% ความจำเพาะเท่ากับ 96.36% เมื่อเทียบกับวิธีมาตรฐาน BACTEC MGIT 960 นอกจากนี้พบการกลายพันธุ์ในยีน pncA ที่สัมพันธ์กับการดื้อยา PZA จำนวน 28 แบบ ในเชื้อจำนวน 37 จาก 40 สายพันธุ์ที่ดื้อยา PZA เป็นการกลายพันธุ์แบบ Nonsynonymous 57.89% การกลายพันธุ์แบบ Indels 21.05% การกลายพันธุ์บริเวณ Promoter 13.16% การกลายพันธุ์แบบ Nonsense 5.26% และการกลายพันธุ์แบบ Double mutation 2.63% นอกจากนี้พบการกลายพันธุ์ใหม่จำนวน 4 แบบ …

ปริมาณโลหิตและส่วนประกอบโลหิตที่เพียงพอในธนาคารเลือดนั้นมีความสำคัญในการรักษาผู้ป่วย ดังนั้นธนาคารเลือดทุกแห่งต้องส่งเสริมให้ผู้บริจาคโลหิตมีสุขภาพดีและมีคุณสมบัติเหมาะสมในการบริจาคโลหิตอย่างสม่ำเสมอจากข้อมูลของศูนย์บริการโลหิตแห่งชาติ สภากาชาดไทยพบผู้ถูกปฏิเสธการบริจาคโลหิตประมาณร้อยละ 15-20 จากจำนวนผู้ประสงค์จะบริจาคโลหิตทั้งหมด ส่วนใหญ่มีสาเหตุจากค่าฮีโมโกลบิน (hemoglobin; Hb) ไม่ผ่านเกณฑ์ นำไปสู่ความผิดหวังความไม่พอใจเพราะผู้บริจาคโลหิตต้องเสียเวลา ค่าใช้จ่ายในการเดินทางแต่ไม่ได้บริจาคโลหิตและส่งผลกระทบต่อการจัดหาโลหิตโดยตรง หากสามารถพยากรณ์ผลตรวจ Hb ได้ล่วงหน้าจะช่วยลดผลกระทบปัญหา เป็นประโยชน์ทั้งต่อผู้บริจาคโลหิต ธนาคารเลือดและผู้ป่วย การศึกษานี้มีวัตถุประสงค์เพื่อพัฒนาและเปรียบเทียบประสิทธิภาพเทคนิคการเรียนรู้ของคอมพิวเตอร์ในการพยากรณ์จำแนกกลุ่มผลตรวจ Hb ของผู้บริจาคโลหิต โดยเก็บข้อมูล 44 ตัวแปรของผู้บริจาคโลหิตจำนวน 2,180 รายจากภาคบริการโลหิตแห่งชาติ 12 แห่งและสถานีกาชาดหัวหินเฉลิมพระเกียรติตั้งแต่ 1 ต.ค. 2561 ถึง 31 พ.ค. 2562 นำมากลั่นกรองข้อมูล คัดเลือกตัวแปร พัฒนาตัวแบบทางเหมืองข้อมูลได้แก่ ต้นไม้ตัดสินใจ ซัพพอร์ตเวกเตอร์แมชชีน การจำแนกแบบเบส์อย่างง่ายและโครงข่ายประสาทเทียม จากนั้นเปรียบเทียบประสิทธิภาพการจำแนกกลุ่มของตัวแบบพยากรณ์ พบว่าต้นไม้ตัดสินใจเป็นตัวแบบการจำแนกกลุ่มที่เหมาะสมที่สุดโดยให้ค่าความถูกต้อง ค่าความไว ค่าความจำเพาะ ค่าการพยากรณ์ผลบวก ค่าการพยากรณ์ผลลบสูงที่สุดและค่า AUC เท่ากับร้อยละ 92.20, 82.98, 94.74, 81.25, 95.29 และ 0.943 ตามลำดับ ซึ่งต้นไม้ตัดสินใจที่ได้จากศึกษานี้อาจนำไปพัฒนาต่อเป็นระบบประเมินออนไลน์ก่อนเดินทางมาบริจาคโลหิตได้

โรคติดเชื้อไวรัสมะเร็งเม็ดเลือดขาวในแมว (Feline Leukemia Virus : FeLV) เป็นโรคติดเชื้อไวรัสในแมวที่สำคัญที่เป็นสาเหตุของโรคมะเร็งเม็ดเลือดขาว และ มะเร็งต่อมน้ำเหลือง การตรวจหา FeLV RNA และโปรไวรัส ด้วยวิธีทางอณูชีววิทยามีความสำคัญในการแยกระยะต่างๆของโรค ซึ่งวิธี Rapid immunochromatographic assay ที่นิยมใช้ตรวจแอนติเจนนั้นสามารถตรวจได้เพียงการติดเชื้อระยะ Progressive ที่มี Viremia เท่านั้น งานวิจัยนี้จึงได้พัฒนาเทคนิค Recombinase polymerase amplification (RPA) และ Nested RPA เพื่อตรวจ exogenous FeLV Provirus DNA และ RT-RPA เพื่อตรวจ FeLV RNA และทดสอบตัวอย่างเลือดแมวจำนวน 122 ตัวอย่าง ผลการศึกษาพบว่าเทคนิค RPA สามารถตรวจหา FeLV Provirus ได้ที่ขีดจำกัด 1 ng/µl และไม่เกิดปฏิกิริยาข้ามกลุ่มกับเชื้อก่อโรคในแมว มีความไว ความจำเพาะ เทียบกับเทคนิค PCR ที่ร้อยละ 95.89 และ 100 ตามลำดับ ขณะที่ Nested RPA เทียบกับเทคนิค Nested PCR มีความไว ความจำเพาะ ที่ร้อยละ 98.89 และ 96.88 ตามลำดับ การตรวจหา FeLV RNA ด้วยเทคนิค RT-RPA มีขีดจำกัดที่ 0.1 ng/µl เมื่อเทียบกับเทคนิค RT-PCR และ Rapid Immunochromatographic assay มีความไว ร้อยละ 93.75 และ 95.24 ตามลำดับ และ ความจำเพาะร้อยละ 100 ค่าความสอดคล้องของเทคนิค RPA …

เชื้อ Klebsiella pneumoniae ที่สร้างเอนไซม์ carbapenemases (KPCs) เป็นปัญหาสำคัญของการติดเชื้อในโรงพยาบาลโดยเฉพาะผู้ป่วยเด็ก เนื่องจากมักดื้อยาหลายขนาน ทำให้มียาที่ใช้รักษาได้จำกัด เป็นสาเหตุทำให้มีอัตราการเสียชีวิตสูงขึ้น งานวิจัยนี้มีวัตถุประสงค์เพื่อพัฒนาเทคนิค multiplex Recombinase Polymerase Amplification (multiplex RPA) สำหรับตรวจหายีนที่สร้างเอนไซม์ carbapenemase ของเชื้อ K. pneumoniae และเปรียบเทียบความไวและความจำเพาะกับ วิธี Modified Carbapenem Inactivation Method Test (mCIM) และ วิธี nucleotide sequencing กับตัวอย่างเชื้อ K. pneumoniae ในสถาบันสุขภาพเด็กแห่งชาติมหาราชินี ประเทศไทย ตั้งแต่เดือนมิถุนายน ถึง ธันวาคม 2018 โดยเก็บเชื้อที่ผลการคัดกรองพบดื้อยา carbapenem แบ่งเป็นจาก perianal ในงานเฝ้าระวังเชื้อดื้อยา จำนวน 129 ตัวอย่าง และจากสิ่งส่งตรวจของผู้ป่วยเด็ก จำนวน 21 ตัวอย่าง พบให้ผลบวกกับวิธี mCIM จำนวน 149 ตัวอย่าง (99.3%) และพบเป็นยีน blaNDM-1 สูงสุดถึงร้อยละ 89.3 รองลงมาคือยีน blaOXA-232 ร้อยละ 8 และยีน blaNDM-1 ร่วมกับ blaOXA-232 ร้อยละ 2 ตามลำดับ เชื้อที่แยกได้จากสิ่งตรวจพบมีการดื้อยาหลายขนาน โดยดื้อยา carbapenem ร่วมกับ gentamicin, trimethoprim/sulfamethoxazole, ciprofloxacin ขณะที่เชื้อที่แยกจากแผนกผู้ป่วยต่าง ๆ ในงานเฝ้าระวังเชื้อดื้อยา พบการแพร่กระจายของยีน blaNDM-1 สูงสุดใน 4 แผนก จาก 9 แผนก การพัฒนาเทคนิค multiplex RPA พบว่าสามารถเพิ่มปริมาณยีน blaKPC, blaNDM, …

The intestinal microbiome of mammals plays a significant role in host health and response to environmental stimuli and can include both beneficial native bacteria as well as parasitic worms. In this study, I examined the intestinal cestode and bacterial communities of two closely related species of shrew, Sorex monticola and Sorex cinereus, over a six month period in 2016. Specimens were collected approximately every three weeks from May to October from the Sangre de Cristo Mountains in Cowles, New Mexico. Carbon-based nanomaterials including carbon nanotubes (CNTs) have been shown to trigger

inflammation. However, how these materials are ‘sensed’ by immune cells is not known. Here we compared the effects of two carbon-based nanomaterials, single-walled CNTs (SWCNTs) and graphene oxide (GO), on primary human monocyte-derived macrophages. Genome-wide transcriptomics assessment was performed at sub-cytotoxic doses. Pathway analysis of the microarray data revealed pronounced effects on chemokine-encoding genes in macrophages exposed to SWCNTs, but not in response to GO, and these results were validated by multiplex array-based cytokine and chemokine profiling. Conditioned medium from SWCNT-exposed cells acted as a chemoattractant for dendritic cells. …

ปัจจุบันพบเชื้อ Escherichia coli ที่สร้างเอนไซม์ extended-spectrum beta-lactamase (ESBL) แพร่กระจายในชุมชนอย่างกว้างขวาง เชื้อแบคทีเรียดังกล่าวสามารถพบได้ว่าเป็นเชื้อประจำถิ่นในลำไส้ของคน และสัตว์ รวมไปถึงสามารถพบได้ในสิ่งแวดล้อมทั่วไป ซึ่งอาจกลายเป็นแหล่งเก็บกักเชื้อแบคทีเรียดื้อสารต้านจุลชีพ ที่สามารถแพร่กระจายในชุมชนต่างๆ ทั่วโลก การศึกษาครั้งนี้ทำการจำแนกลักษณะทางโมเลกุลของเชื้อ E. coli ที่สร้างเอนไซม์ ESBL ซึ่งแยกได้จากสุกรในฟาร์มปศุสัตว์และประชากรในชุมชน ในพื้นที่อำเภอเมือง จังหวัดลำพูน เพื่อศึกษารูปแบบของยีนที่ควบคุมการสร้างเอนไซม์ ESBL และศึกษาหาความสัมพันธ์ของเชื้อในกลุ่มตัวอย่างทั้งสอง เชื้อ E. coli ที่สร้างเอนไซม์ ESBL จำนวนทั้งสิ้น 212 สายพันธุ์ แยกได้จากอุจจาระของสุกรและประชากรที่มีสุขภาพดี และทำการจำแนกยีนที่ควบคุมการสร้างเอนไซม์ ESBL ได้แก่ ยีน blaCTX-M ยีน blaTEM และยีน blaSHV ด้วยเทคนิค multiplex-PCR และเทคนิค whole genome sequencing และศึกษาความสัมพันธ์ของเชื้อ E. coli ที่สร้างเอนไซม์ ESBL ที่แยกได้จากทั้งสองกลุ่มตัวอย่างด้วยเทคนิค multilocus sequence typing ผลการศึกษาพบว่าเชื้อ E. coli ที่สร้างเอนไซม์ ESBL ส่วนใหญ่เป็นเชื้อดื้อยาหลายขนาน และมักดื้อยากลุ่ม fluoroquinolones ยากลุ่ม aminoglycosides และยา trimethoprim/sulfamethoxazole ในขณะที่เชื้อทุกสายพันธุ์ดื้อต่อยา ampicillin ยา ceftriaxone และยา cefotaxime ยีนที่ควบคุมการสร้างเอนไซม์ ESBL ที่พบมากที่สุดได้แก่ ยีน blaCTX-M ซึ่งพบได้ 95.75% รองลงมาคือยีน blaTEM และยีน blaSHV พบได้ 62.73% และ 2.40% ตามลำดับ โดย subgroup ของยีนที่ควบคุมการสร้างเอนไซม์ ESBL ที่พบมากที่สุด ได้แก่ ยีน blaCTX-M-55 …

Cellular immunotherapies promise to transform cancer care. However, they must overcome serious challenges, including: (1) the need to identify and characterize novel cancer antigens to expand the range of therapeutic targets; (2) the need to develop strategies to minimize serious adverse events, such as cytokine release syndrome and treatment-related toxicities; and (3) the need to develop efficient production/manufacturing processes to reduce costs. Here, we discuss whether these challenges might better be addressed through forms of public-private research collaborations, including public-private partnerships (PPPs), or whether these challenges are best addressed by way of standard market transactions. Gene content determination and variant calling in the complex KIR genomic region are useful for immune system function analysis, pathogenesis and disease risk factor elucidation, immunotherapy development, evolutionary investigations, and human migration modeling. Sequence-specific oligonucleotide and sequence-specific primer PCR methods are the de facto standards for KIR presence/absence identification, but the current platforms are unsuitable for SNP calling, impractical for KIR typing large cohorts of DNA samples, and inapplicable for typing repositories in which sequence data, but not cells or cell analytes, are available. Amino acids are the building blocks of life. If DNA is the blueprint, amino acids are the lumber that proteins are built with. Proteins are built with left-handed, L- forms of amino acids. Bacteria have an essential cell wall component that happens to be an exception: peptidoglycan. Bacteria have enzymes called racemases that convert L- amino acid forms into right-handed, D- forms. Amino acids participate in many reactions with keto acids. Transaminases allow conversion between amino acids by transfer of an amino group.

Background: Our recent research showed that antibody response to porcine reproductive and respiratory syndrome (PRRS), measured as sample-to-positive (S/P) ratio, is highly heritable and has a high genetic correlation with reproductive performance during a PRRS outbreak. Two major quantitative trait loci (QTL) on Sus scrofa chromosome 7 (SSC7; QTLMHC and QTL130) accounted for ~40 % of the genetic variance for S/P. : Mesopelagic fishes represent significant ecological links between mesozooplankton and the larger pelagic squids, fishes, and marine mammals. As such, these fishes also play a significant role as intermediate or paratenic hosts for parasites that require a crustacean intermediate host and mature in marine mammals or pelagic fishes. We examined a total of 208 individuals representing 5 species of Sternoptychidae and 88 individuals representing 2 species of Phosichthyidae from 20 locations in the northern Gulf of Mexico (nGOM). Somatic mutations may drive tumorigenesis or lead to new, immunogenic epitopes (neoantigens). The immune system is thought to represses neoplastic growths through the recognition of neoantigens presented only by tumor cells. To study mutations as well as the immune response to mutation-generated antigens, we have created a conditional knockin mouse line with a gene encoding, 5’ to 3’, yellow fluorescent protein (YFP), ovalbumin (which is processed to the immunologically recognizable peptide, SIINFEKL), and cyan fluorescent protein (CFP), or, YFP-ovalbumin-CFP. A frame shift mutation has been created at the 5’ end of the ovalbumin gene, hence YFP should always be expressed, …

Vitamin D is primarily known for its role in bone health, however recently vitamin D has been identified as a potent immunomodulator. Most research studying the effects of vitamin D on immune properties have focused on immune cells. Few have evaluated how vitamin D affects the immune functions of barrier epithelial cells, such as human mammary epithelial (HME) cells, that are exposed to pathogens both locally and systemically. A safe and effective vaccine against human immunodeficiency virus type 1 (HIV-1) is urgently needed to combat the worldwide AIDS pandemic, but still remains elusive. The fact that uncontrolled replication of an attenuated vaccine can lead to regaining of its virulence creates safety concerns precluding many vaccines from clinical application. We introduce a novel approach to control HIV-1 replication, which entails the manipulation of essential HIV-1 protein biosynthesis through unnatural amino acid (UAA*)-mediated suppression of genome-encoded blank codon. We successfully demonstrate that HIV-1 replication can be precisely turned on and off in vitro.

The behavior of three antibiotics (bacitracin, chlortetracycline, and tylosin) and two classes of antibiotic resistance genes (ARGs), tet and erm, were monitored in swine manure slurry under anaerobic conditions. First-order decay rates were determined for each antibiotic with half-lives ranging from1 day (chlortetracycline) to 10 days (tylosin). ARGs were monitored in the swine manure slurry, and losses of approximately 1 to 3 orders of magnitude in relative abundance were observed during the 40 day storage period. First-order degradation profiles were observed for chlortetracycline and its corresponding resistance genes, tet(X) and tet(Q). Tylosin was degraded to approximately 10% …
