Background Complex mosaic structures of HIV-1 were found in the Democratic Republic of Congo (DRC). Currently, there is limited information on the circulating HIV-1 strains, the distribution of these strains and antiretroviral (ART) resistant viruses in different regions of the country, and the HIV-1 strains harbored by the high-risk groups like female sex workers (FSW) reported to be the source of recombinant and ART resistant viruses.

Despite ongoing efforts, sub-Saharan Africa faces a higher cervical cancer burden than anywhere else in the world. Besides HPV infection, definitive factors of cervical cancer are still unclear. Particular states of the cervicovaginal microbiota and viral infections are associated with increased cervical cancer risk. Notably, HIV infection, which is prevalent in sub-Saharan Africa, greatly increases risk of cervicovaginal dysbiosis and cervical cancer. Kaposi’s sarcoma-associated herpesvirus (KSHV) is the etiological agent of Kaposi’s sarcoma (KS), one of the most prevalent cancers of people living with HIV/AIDS in sub-Saharan Africa. The seroprevalence for KSHV is high in the region, and no prophylactic vaccine against the virus is available. In this study, we characterized the antigenic targets of KSHV-specific neutralizing antibodies (nAbs) in asymptomatic KSHV-infected individuals and KS patients with high nAbs titers. We quantified the extent to which various KSHV envelope glycoproteins (gB, ORF28, ORF68, gH, gL, gM, gN and gpK8.1) adsorbed/removed KSHV-specific nAbs from the plasma of infected individuals. Tanzania faces one of the highest cervical cancer burdens in the world. Recent work has suggested that the bacterial family Mycoplasmataceae is associated with higher levels of human papillomavirus (HPV), human immunodeficiency virus (HIV), and pre-cancerous cervical lesions. Mycoplasmataceae infection in Tanzania is not well understood, especially when considering the differences between sexually transmitted species of Mycoplasmataceae. Multiple sclerosis (MS) is the most common autoimmune disorder affecting the central nervous system. Epstein-Barr virus (EBV) is a causative agent for infectious mononucleosis (IM) that is associated with MS pathogenesis. However, the exact mechanism by which EBV, specifically in IM, increases the risk for MS remains unknown. EBV immortalizes primary B lymphocytes in vitro and causes excessive B lymphocyte proliferation in IM in vivo. In asymptomatic carriers, EBV-infected B lymphocytes still proliferate to certain degrees, the process of which is tightly controlled by the host immune systems. On average, there are 3–5 million severe cases of influenza virus infections globally each year. Seasonal influenza vaccines provide limited protection against divergent influenza strains. Therefore, the development of a universal influenza vaccine is a top priority for the NIH. Here, we report a comprehensive summary of all universal influenza vaccines that were tested in clinical trials during the 2010–2019 decade. Of the 1597 studies found, 69 eligible clinical trials, which investigated 27 vaccines, were included in this review. Oncolytic adenoviruses (Ad) have shown promising results in the therapeutic treatment of cancer. Ad type 5 (Ad5) is the most extensively utilized Ad type. However, several limitations exist to using Ad5 as an oncolytic virus, including high levels of anti-Ad5 neutralizing antibodies in the population, binding of the Ad5 hexon to blood coagulation factor X leading to liver sequestration and toxicity, and reduced expression of the primary receptor CAR on many tumors. Therapeutic strategies are being clinically tested either to eradicate the latent HIV reservoir or to achieve virologic control in the absence of antiretroviral therapy (ART). Attaining this goal will require a consensus on how best to measure the levels of persistently-infected cells with the potential to cause viral rebound upon ART cessation to assess the results of cure-directed strategies in vivo. Studies on whip spider navigation have focused on their ability to locate goal locations in the horizontal plane (e.g., when moving along the ground). However, many species of tropical whip spiders reside and move along surfaces in the vertical plane (e.g., trees). Two cosegregating single-nucleotide polymorphisms (SNPs) in human TLR4, an A896G transition at SNP rs4986790 (D299G) and a C1196T transition at SNP rs4986791 (T399I), have been associated with LPS hyporesponsiveness and differential susceptibility to many infectious or inflammatory diseases. However, many studies failed to confirm these associations, and transfection experiments resulted in conflicting conclusions about the impact of these SNPs on TLR4 signaling. Skeletal muscle atrophy can result from a range of physiological conditions, including denervation, immobilization, hindlimb unweighting, and aging. To better characterize the molecular genetic events of atrophy, a microarray was performed using skeletal muscle isolated from mice after 3 and 14 days of denervation and compared to control muscle to identify novel atrophy-induced genes. The microarray revealed that FGGY carbohydrate kinase domain containing (Fggy) is expressed in skeletal muscle and is induced in response to denervation. Fam83d and Dupd1 have been identified as novel genes in skeletal muscle that are upregulated in response to neurogenic atrophy in a mouse model. qPCR analysis reveaed both genes are expressed in skeletal muscle with Fam83d expression being highest during myoblast proliferation, while Dupd1 expression is highest during myotube differentiation. Overexpression of either protein results in inhibition of proper muscle cell differentiation as evidenced by repression of both myosin heavy chain and myogenin expression. Characterization of transcriptional activity revealed both genes are modulated by myogenic regulatory factors and additionally, Dupd1 expression is enhanced by dexamethasone treatment. Negative human–wildlife interactions can be better managed by early detection of the wildlife species involved. However, many animals that pose a threat to humans are highly cryptic, and detecting their presence before the interaction occurs can be challenging. We describe a method whereby the presence of the estuarine crocodile (Crocodylus porosus), a cryptic and potentially dangerous predator of humans, was detected using traces of DNA shed into the water, known as environmental DNA (eDNA). Climate change has been considered a key driver for adaptations in water usage efficiency (WUE) in plants. Perceptions persist that C4 photosynthesis is indicative of some level of WUE. Here, we examined the interaction and evolution of WUE within the grass family, Poaceae, as it pertains to photosynthetic pathway. Unique to this study is the magnitude of taxa sampled, presence of intrageneric sampling, and use of complete plastid genomes (plastomes) for phylogenomic analyses.

Glutamate Carboxypeptidase II (GCPII) is a transmembrane, zinc metallopeptidase that is expressed in a wide range of organisms, including roundworms, mice, and humans. In humans, GCPII is primarily expressed in the prostate, kidneys, small intestine, and central nervous system. Within the small intestine, the expected function of GCPII is to aid in the absorption of dietary folate from the intestinal lumen. GCPII cleaves excess glutamates from folate to yield monoglutamated folate which is then readily transported into the enterocyte. INTRODUCTION: Women with pathogenic germline gene variants in

METHODS: We searched Medline, EMBASE, CENTRAL, PubMed, and clinicaltrials.gov up to October 3, 2019. We identified 2775 records and included 21.

RESULTS: There is limited evidence related to these factors and ovarian cancer risk. For breast cancer risk, evidence suggests higher diet quality, adulthood weight-loss of ≥10 pounds, and activity during adolescence and young-adulthood may be linked with decreased risk. Higher meat intake and higher daily energy intake may be linked with increased risk.

CONCLUSIONS: There is not enough evidence to suggest tailored recommendations for dietary habits or weight management among women …

Motor protein-based active transport is essential for mRNA localization and local translation in animal cells, yet how mRNA granules interact with motor proteins remains poorly understood. Using an unbiased yeast two–hybrid screen for interactions between murine RNA-binding proteins (RBPs) and motor proteins, here we identified protein interaction with APP tail-1 (PAT1) as a potential direct adapter between zipcode-binding protein 1 (ZBP1, a β-actin RBP) and the kinesin-I motor complex. Human induced pluripotent stem cells offer a model for human brain development and disease by differentiation into brain organoids; however, current neural culture systems lack the microenvironment, neuronal circuits and connectivity, vascular circulation, and immune system that exist in vivo. Detailed comprehensive knowledge of the structures of individual long-range telomere-terminal haplotypes are needed to understand their impact on telomere function, and to delineate the population structure and evolution of subtelomere regions. However, the abundance of large evolutionarily recent segmental duplications and high levels of large structural variations have complicated both the mapping and sequence characterization of human subtelomere regions. Background: Phenotypic heterogeneity and complicated gene-environment interplay in etiology are among the primary factors that hinder the identification of genetic variants associated with cocaine use disorder. Methods: To detect novel genetic variants associated with cocaine use disorder, we derived disease traits with reduced phenotypic heterogeneity using cluster analysis of a study sample (n = 9965). We then used these traits in genome-wide association tests, performed separately for 2070 African Americans and 1570 European Americans, using a new mixed model that accounted for the moderating effects of 5 childhood environmental factors. Klebsiella pneumoniae is a Gram-negative bacterium responsible for nosocomial infections such as blood stream infections, meningitis, and septicemia. Clinical isolates confirm K. pneumoniae is increasingly antibiotic resistant (AR) making treatment more difficult. While bacterial exposure to antimicrobial substances is a known cause of the AR phenotype, there still exists gaps in knowledge about the genetic changes responsible for this fitness change. The laboratory Klebsiella pneumoniae 43816 (ATCC 43816) was exposed to sublethal concentrations of Cephalothin for a 14-day period. การตรวจความเข้ากันได้ของ Human Leukocyte Antigen (HLA) ก่อนการปลูกถ่ายไตทำขึ้นเพื่อตรวจจับ Donor Specific Antibody (DSA) ต่อผู้บริจาค ที่อาจก่อให้เกิดปฏิกิริยาปฏิเสธอวัยวะ โดยเทคนิคที่ใช้ในปัจจุบันคือเทคนิค Complement-dependent microcytotoxicity crossmatch (CDC-XM) และ Anti-Human Globulin Complement-dependent microcytotoxicity crossmatch (AHG-CDC) มีความไวจำกัด ไม่สามารถตรวจจับ DSA ปริมาณน้อยที่ส่งผลต่ออัตราการรอดของไตได้ ขณะที่เทคนิคโฟลไซโตเมทรี (Flow cytometry crossmactch, FCXM) มีความไวสูงกว่า แต่พบปัญหาผลบวกปลอมจากแอนติบอดีที่ไม่เกี่ยวข้องทำให้ผู้ป่วยเสียโอกาสที่จะได้รับการปลูกถ่ายไต งานวิจัยนี้จึงมีวัตถุประสงค์เพื่อปรับปรุงความไวและความจำเพาะของเทคนิค FCXM โดยศึกษาในลิมโฟซัยต์จากผู้บริจาคไตสมองตายกับน้ำเหลืองตัวอย่างที่ทราบผล DSA จำนวน 20 ตัวอย่างเพื่อประเมินเทคนิคแล้วทดสอบเปรียบเทียบกับเทคนิค CDC,AHG-CDC ในกลุ่มผู้ป่วยจำนวน 199 ตัวอย่าง พบว่าในขั้นตอนการประเมินผลเทคนิค FCXM มีความไวและความจำเพาะในกลุ่ม T cell 71% และ 100% ตามลำดับ ส่วน B cell พบว่าความไวเท่ากับ 75% และความจำเพาะ 100% และสอดคล้องกับผล DSA อย่างมีนัยสำคัญทางสถิติ (p< 0.05) เมื่อทดสอบกับผู้ป่วยให้ผลสอดคล้องกับเทคนิค CDC, AHG-CDC จำนวน 190 ตัวอย่าง คิดเป็น 95.47% พบผลบวกปลอม จำนวน 8 ตัวอย่าง คิดเป็น 4.0% ผลการทดสอบความเข้ากันได้และประเมินประสิทธิภาพเทคนิค FCXM นี้สรุปได้ว่าเหมาะสมที่จะนำไปทดลองใช้ทดแทนเทคนิค CDC และ AHG-CDC แต่ควรทำการศึกษาเพิ่มเติมในกลุ่มที่มี DSA ก่อนนำไปใช้จริงในอนาคต

วัคซีนที่มีความปลอดภัยก่อนนำไปใช้ในมนุษย์ต้องปราศจากเชื้อปนเปื้อนแฝง (extraneous agents) โดยเชื้อ Avian adenoviruses เป็นเชื้อปนเปื้อนแฝงชนิดหนึ่งที่อาจปนเปื้อนมากับไข่ไก่ฟักซึ่งเป็นวัตถุดิบในการผลิตวัคซีนไข้หวัดใหญ่ชนิดเชื้อตาย (Inactivated Influenza Vaccine, IIV) การตรวจหาเชื้อ Avian adenoviruses ในปัจจุบันทำการทดสอบโดยวิธีเซลล์เพาะเลี้ยง ซึ่งวิธีดังกล่าวสามารถตรวจได้เฉพาะเชื้อ Avian adenovirus ในกลุ่มที่ 1 (Fowl adenovirus) ใช้แรงงานจำนวนมาก และใช้เวลาในการทดสอบนาน ด้วยเหตุนี้จึงได้พัฒนาวิธี nested PCR และ real-time PCR เปรียบเทียบกับวิธีเซลล์เพาะเลี้ยงในการตรวจหาการปนเปื้อนของเชื้อ Avian adenoviruses ในวัคซีนไข้หวัดใหญ่ชนิดเชื้อตาย โดยพบว่าวิธี nested PCR และ real-time PCR สามารถตรวจหาเชื้อ Avian adenoviruses ในกลุ่มที่ 1 ได้แก่ Fowl adenovirus ซีโรไทป์ 1 2 และ 4 เชื้อ Hemorrhagic enteritis virus ในกลุ่มที่ 2 และเชื้อ Egg drop syndrome ในกลุ่มที่ 3 ได้อย่างจำเพาะ โดยไม่เกิดปฏิกิริยาข้ามกัน (cross-reaction) กับเชื้อก่อโรคชนิดอื่นในไก่ ได้แก่เชื้อ Mycoplasma gallisepticum Mycoplasma synoviae Infectious laryngotracheitis virus Chicken anemia virus Chicken bronchitis virus โดยวิธีเซลล์เพาะเลี้ยง nested PCR และ real-time PCR มีความไวในการตรวจหาเชื้อ Fowl adenovirus 1 ซึ่งมีการติดเชื้อในไก่แบบแฝงและตรวจพบได้ยาก ได้เท่ากับ 10-2 10-8 และ 10-18 TCID50/0.1ml …

เชื้อแบคทีเรีย Mycobacterium tuberculosis เป็นสาเหตุของวัณโรค ซึ่งเป็นโรคติดเชื้อที่เป็นปัญหาทางสาธารณสุขทั้งในประเทศไทยและในระดับโลก โดยเฉพาะเมื่อโรคพัฒนาสู่ภาวะวัณโรคดื้อยาหลายขนาน (Multidrug-resistant Tuberculosis: MDR-TB) ยา Isoniazid เป็นหนึ่งในยาขนานแรกที่มีประสิทธิภาพสูงสุดในการรักษาผู้ป่วยวัณโรค ดังนั้นการดื้อยา Isoniazid จึงเป็นอุปสรรคที่สำคัญในการควบคุมการแพร่ระบาดของวัณโรค และยังพบว่าผู้ป่วยวัณโรคที่ดื้อต่อยา Isoniazid มีโอกาสสูงที่จะพัฒนาไปเป็นผู้ป่วยวัณโรคดื้อยาหลายขนาน และมีโอกาสในการรักษาสำเร็จที่ลดลง สาเหตุหลักของการดื้อยา Isoniazid จากการกลายพันธุ์ของยีน katG และยีน inhA ตามลำดับ การศึกษานี้ได้พัฒนาเทคนิค Multiplex Allele Specific Polymerase Chain Reaction (MAS-PCR) ร่วมกับแผ่นทดสอบ Dipstick Chromatography เพื่อใช้วินิจฉัยการกลายพันธุ์ของยีน katG ที่ตำแหน่งโคดอน 315 และยีน inhA ที่ตำแหน่งเหนือยีน (-15) ของเชื้อ M. tuberculosis ซึ่งสัมพันธ์กับการดื้อยา Isoniazid ผลการทดสอบกับดีเอ็นเอที่สกัดได้จากโคโลนีของเชื้อ M. tuberculosis จำนวน 250 ตัวอย่าง พบว่าเทคนิค MAS-PCR ร่วมกับแผ่นทดสอบ Dipstick Chromatography มีความไวและความจำเพาะในการวินิจฉัยการกลายพันธุ์ที่สัมพันธ์กับการดื้อยา Isoniazid เท่ากับ 91.67% และ 100% ตามลำดับ เมื่อเปรียบเทียบกับวิธีทดสอบทางฟีโนไทป์ ในขณะที่เมื่อเปรียบเทียบกับเทคนิค Sanger DNA sequencing ในการตรวจหากลายพันธุ์ของยีน katG ณ ตำแหน่งโคดอน 315 มีค่าความไวและความจำเพาะเท่ากับ 100% และ 99.28% ตามลำดับ และในการตรวจหายีน inhA ที่ตำแหน่งเหนือยีน (-15) มีค่าความไวและความจำเพาะเท่ากับ 100% และ 99.51% ตามลำดับ โดยเทคนิค MAS-PCR ร่วมกับแผ่นทดสอบ Dipstick Chromatography มีค่าความสอดคล้องในระดับดีมากกับทุกเทคนิคที่นำมาเปรียบเทียบ เทคนิค MAS-PCR …

วัณโรคเป็นโรคติดต่อที่มีการระบาดทั่วโลก และยังคงเป็นปัญหาสาธารณสุขที่สำคัญของประเทศไทย วัณโรคยังเป็นสาเหตุของการเสียชีวิตจากโรคติดต่อสูงเป็นอันดับสองรองจากโรคเอดส์ โดยมีสาเหตุมาจากเชื้อแบคทีเรีย Mycobacterium tuberculosis complex (MTBC) การตรวจวินิจฉัยวัณโรคทางห้องปฏิบัติการ เป็นหนึ่งปัจจัยที่สำคัญในการควบคุมการแพร่ระบาดของโรค การนำเทคนิคทางอณูชีววิทยาเข้ามาใช้วินิจฉัยทางห้องปฏิบัติการ ซึ่งมีความรวดเร็วมากกว่าการเพาะเลี้ยงเชื้อด้วยวิธีดั้งเดิมและสามารถวินิจฉัยโรคได้ภายใน 1 วัน มีบทบาทที่สำคัญในการวินิจวัณโรคในปัจจุบัน การศึกษานี้มีวัตถุประสงค์ในการพัฒนาแถบตรวจ MTB Strip แบบอ่านผลด้วยตาเปล่าอย่างรวดเร็ว สำหรับวินิจฉัยวัณโรคจากปฏิกิริยา Multiplex-recombinase polymerase amplification (M-RPA) โดยใช้ไพรเมอร์ที่จำเพาะต่อ IS1081 และ IS6110 ซึ่งมีความจำเพาะต่อเชื้อ MTBC และตรวจสอบผลผลิตของทั้งสอง IS ร่วมกัน เพื่อเพิ่มความไวของเทคนิคในการวินิจฉัยวัณโรค การทดสอบเบื้องต้นกับดีเอ็นเอที่สกัดจากโคโลนีของเชื้อ แสดงให้เห็นว่าเทคนิค M-RPA ร่วมกับแถบตรวจ MTB Strip สามารถจำแนกเชื้อ MTBC ออกจาก NTM ได้อย่างถูกต้อง เมื่อนำเทคนิค M-RPA ร่วมกับแถบตรวจ MTB Strip มาวินิจฉัยกับดีเอ็นเอที่สกัดจากสิ่งส่งตรวจเสมหะจำนวน 131 ตัวอย่าง พบว่ามีความไวและความจำเพาะ เท่ากับ 91.03% และ 84.91% ตามลำดับ โดยมีค่าความสอดคล้องในเกณฑ์ดี (κ=0.762) เมื่อเปรียบเทียบกับการย้อมสีทนกรด ในขณะที่มีความไวและความจำเพาะเท่ากับ 100.0% และ 94.55% ตามลำดับ และมีค่าความสอดคล้องในเกณฑ์ดีมาก (κ=0.953) เมื่อเปรียบเทียบกับเทคนิค Real-time PCR เทคนิค M-RPA ร่วมกับแถบตรวจ MTB Strip ใช้ระยะเวลาทำปฏิกิริยาเพียง 25 นาที ภายใต้อุณหภูมิ 37 องศาเซลเซียส และตรวจสอบผลผลิตด้วยแถบตรวจ MTB Strip ภายในเวลา 15 นาที โดยตรวจสอบความเข้มข้นดีเอ็นเอตั้งต้นน้อยที่สุดได้เท่ากับ 0.1 ng/µl และไม่เกิดปฏิกิริยาข้ามกลุ่มเชื้อแบคทีเรียสายพันธุ์อื่นที่มักก่อโรคในระบบทางเดินหายใจ ดังนั้นเทคนิค M-RPA ร่วมกับแถบตรวจ MTB Strip จึงเป็นเทคนิคที่มีความรวดเร็ว ไม่ซับซ้อน …
