Down Syndrome is a human condition caused by an extra copy of a #21 chromosome. At least one to two percent of free (not translocated) trisomy 21 cases are mosaics, i.e., they have two or more distinct cell lines. Usually, one cell line is 47 ,XX or XY ,+21 while the other cell line is normal 46,XX or 46,XY.

On The Distribution And Characteristics Of Isozyme Expression In Mycoplasma, Acholeplasma, And Ureaplasma Species, Stephen J. O'Brien, Janice M. Simonson, S. Razin, Michael F. Barile

A summary of a survey of three genera of mycoplasmatales (Mycoplasma, Acholeplasma, and Ureaplasma) for isozyme expression is presented. Isozyme analysis of mycoplasmas has been employed in at least three distinct areas: (1) as genetic markers for identification, individualization, and taxonomic classification; (2) as markers for cell culture contamination; and (3) as a qualitative measure of the operative metabolic pathways in the diverse species. We have found five ubiquitous enzymes: purine nucleoside phosphorylase, adenylate kinase, inorganic pyrophosphatase, dipeptidase, and esterase. Three enzymes, glucose-6-phosphate dehydrogenase, phosphogluconate dehydrogenase, and superoxide dismutase, were restricted to Acholeplasma species and were not …

Dispermic Origin And Clinical Outcome Of Three Complete Hydatidiform Moles With 46,Xy Karyotype, U. Surti, A. E. Szulman, S. O'Brien

Three new cases of complete, classic hydatidiform mole with a 46,XY karyotype are described. They originated by dispermy as demonstrated by chromosome and enzyme analyses. Levels of human chorionic gonadotropin decreased to normal spontaneously within a short time, indicating a benign course in these three cases. Of a total of 18 cases reported to date, postoperative clinical information was available for 10 patients. Two of these 10 patients had a malignant course with lung metastases. It is of theoretical and clinical importance to establish the magnitude of the risk of malignancy for hydatidiform mole with a 46,XY karyotype. © 1982.

A deletion mutant of the colicin E1-derived plasmid, pDMS6642, exhibited an approximately fourfold increase in copy number. We subsequently isolated hydroxylamine-induced mutants of that plasmid that had a further increase in copy number. Analysis of them suggests that the increased copy number of pDMS6642 is associated with transcriptional readthrough from a Tn3 transposon into the region of ColE1 containing information that influences plasmid replication. The hydroxylamine mutation in one copy number mutant appeared to increase the plasmid copy number by stimulating readthrough transcription from the Tn3 transposon into the ColE1 replication control region, whereas the other hydroxylamine mutation acts by …

Several recent reports have established the biological and molecular genetic similarity between the endogenous AKV virus of strain AKR, and an N-ecotropic endogenous virus found in the genome of feral Japanese mice, Mus musculus molossinus. The similarities are so striking as to suggest a common origin of these viruses, which are present in some, but not all, inbred mouse strains. The virogenes of AKR mice may have been acquired by either: (a) common descent of AKR (and other AKV+ strains) from a common ancestor of AKR and molossinus animals, or (b) horizontal germ line infection of the AKR …

Genetically Controlled Variation Of “Acid” Beta-Galactosidase Detected In Rattus Norvegicus By Isoelectric Focusing, T C Douglas, K A Kimmel, P E Dawson

Two genetically variant forms of rat "acid" beta-galactosidase were found to differ in isoelectric point and pH dependence, but not in thermostability or sensitivity to inhibition by p-mercuribenzoate (PMB). The results of two backcrosses and an intercross indicated that the isoelectric focusing phenotypes are controlled by two codominant alleles at a single autosomal locus, for which we propose the name Glb-1. No significant linkage between Glb-1 and albino (LG I), brown (LG II), or hooded (LG VI) was observed. Strain-specific differences in total levels of kidney beta-galactosidase were detected, but it is not yet known whether the variation is controlled …

Treatment of Schneider's line 3 Drosophila cells with the steroid hormone ecdysterone rapidly stimulated the synthesis and accumulation of the polypeptide previously designated p7 [Berger, E. M., Ireland, R. C. & Wyss, C. (1980) Somatic Cell Genet. 6, 119-129]. In this report, p7 is identified as the 23,000-dalton heat shock polypeptide (hsp23). In addition to hsp23, the synthesis of the low molecular weight heat shock polypeptides hsp22, hsp26, and hsp27 was also stimulated by ecdysterone, although to different extents. Hybridization of a nick-translated genomic clone containing the hsp23 gene to a total RNA blot showed that ecdysterone stimulation of hsp23 …

In this essay I outline the processes of adaptation of animals and of animal populations and discuss their relevance to the problem of animal welfare. Because "animal welfare" has many different aspects including philosophical, ethical, and biological, it is important to examine some of the fundamental issues that underly the concept. Hence, in this essay, I comment on how people come to "know," how information accumulates, and how what we know influences our actions. This study was undertaken to determine specific and intraspecific relationships within the genus Zapus (Mammalia, Rodentia, Zapodidae). Subspecies are based on the biological subspecies concept where the subspecies is an evolutionary step between a population and species. Approximately 20,000 Recent specimens and representatives of fossil taxa were examined. Cranial and postcranial characteristics, colors, and when available bacula, sperms and karyotypes were incorporated into the morphological analyses. In addition, published biological data were summarized and analyzed for evidence of primary and secondary isolating mechanisms. The differential staining of chromosomes with the use of 5-bromo-2'-deoxyuridine (BUdR) was discovered in 1972. Since then, the BUdR technique has been modified and improved. It is used to study cellular kinetics; yet its most extensive use today is focused on studying sister chromatid exchange (SCE). Studies show that the frequency of SCEs increases significantly in the presence of mutagenic and carcinogenic agents. Hence, the BUdR technique (commonly referred to as the SCE technique) has been useful as a diagnostic tool to test for mutagenic anc carcinogenic agents.

Drosophila Alcohol Dehydrogenase: Developmental Studies On Cryptic Variant Lines, Gurbachan Miglani, Franklin Ampy

Thirty-five cryptic variant lines were used to examine the mechanisms involved in genetic modulation of alcohol metabolism in Drosophila. Late third-instar larval, preemergence pupal, and adult stages cultured at 18 and 28 C were examined. Spectrophotometric analyses for native alcohol dehydrogenase (ADH) activity and residual ADH activity after treatment with guanidine hydrochloride and heat were performed. Differential response of cryptic variants to treatment with the denaturants during development suggested that this variation may have an adaptive significance.

Analysis Of Multiple Isoenzyme Expression Among Twenty-Two Species Of Mycoplasma And Acholeplasma, Stephen J. O'Brien, Janice M. Simonson, Marion W. Grabowski, Michael F. Barile

Crude extracts of triple-cloned, purified cultures of 22 species of Mycoplasma and Acholeplasma were examined for expression of 21 isozyme systems routinely used to type mammalian cells. Nine previously described enzymes (purine nucleoside phosphorylase, adenylate kinase, dipeptidase, esterase, glyceraldehyde-3-phosphate dehydrogenase, glucose phosphate isomerase, glucose-6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase, and superoxide dismutase) and three enzymes not previously reported in mycoplasma (triose phosphate isomerase, inorganic pyrophosphatase, and acid phosphatase) were detected in some or all of the species examined. These findings provide new information on the enzymatic expressions of these organisms. A transmission electron microscope analysis of the changes that occur in the nuclear chromatin and the Feulgen-positive areas of the oocyte cytoplasm as the female develops, feeds and mates is described. The description includes analysis of oocytes from three types of females: unfed, unmated; fed, unmated; and fed, mated. In all types, dense material passes through the extremely porous nuclear membrane where it accumulates in the cytoplasm.

Nuclei of previtellogenic oocytes appear the same in all females studied. Fibrillar bodies and up to four large nucleoli are seen. No typical chromatin is seen, but structures resembling nucleosomes appear throughout the …
