Intraspecific genetic variation is an integral component of diversification and the accumulation of biodiversity. The degree to which isolated populations of the same species are genetically structured in geographical space is impacted by a variety of mechanisms. In this study, I document patterns and discuss possible drivers of genetic structure within Ecpleopus gaudichaudii, a lizard species endemic to the Atlantic Forest of Brazil. For that, I assembled ddRadseq sequences from 48 individuals across much of the range of the E. gaudichaudii and analyzed its population structure. Tissue-specific promoters are a valuable tool in plant biotechnology because they allow for precise spatial control of transgene expression. To identify root-specific promoters in sunflower (Helianthus annuus), sunflower RNA sequence (RNA-seq) data were screened in silico by Pavlidis Template Matching (PTM) with a root-specific template. Eight root-specific genes were identified in sunflower. Gulf and Caribbean Research

Two species of ladyfish occur in the Gulf of Mexico (GOM), Elops saurus and Elops smithi, that are morphologically indistinguishable except for vertebral counts but can also be identified by mitochondrial DNA haplotypes. Here we expand on previous work, most of which has occurred in Florida, and examine the demography, phylogenetics, geographic distribution, and age—structure of ladyfishes in Texas estuaries. Fishery—independent gill net data demonstrated that ladyfishes increase in abundance from north to south along the Texas coast. The abundance of ladyfishes also increased in Texas waters from 1982–2021, which coincides with recent trends of warmer winters. Genetic data …

The American Bullfrog (Rana catesbeiana) is a highly invasive species that has successfully colonized habitats around the world. The genetic variability of immune genes within invasive populations, like that of bullfrogs, may contribute to how resistant a population is to pathogens. The objective of this project was to characterize the genetic diversity of an immune gene in invasive bullfrog populations in California to better understand how persistent these populations might be over time. After myocardial infarction, the massive death of cardiomyocytes leads to cardiac fibroblast proliferation and myofibroblast differentiation, which contributes to the extracellular matrix remodelling of the infarcted myocardium. We recently found that myofibroblasts further differentiate into matrifibrocytes, a newly identified cardiac fibroblast differentiation state. Cardiac fibroblasts of different states have distinct gene expression profiles closely related to their functions. The high level of sparsity in methylome profiles obtained using whole-genome bisulfite sequencing in the case of low biological material amount limits its value in the study of systems in which large samples are difficult to assemble, such as mammalian preimplantation embryonic development. The recently developed computational methods for addressing the sparsity by imputing missing have their limits when the required minimum data coverage or profiles of the same tissue in other modalities are not available. Hybridization offers unique insight into the evolutionary process of speciation and the forces that maintain barriers to reproduction. In northeast Florida, a hybrid zone between two species of killifish, Fundulus heteroclitus and F. grandis, has been identified near Flagler Beach, FL, although the exact boundaries of the hybrid zone remain unknown (Gonzalez et al. 2009). The hard tick (Acari: Ixodidae) Amblyomma quadricavum (Schulze, 1941) is a parasite of snakes and is endemic to the Caribbean region. Morphological characters of this species, namely its rudimentary eye structures, have led to debate about its taxonomic status. The species was originally assigned to the genus Aponomma. However, with revisions of the genus Aponomma, and the recent creation of new genera created for some basal former-Aponomma, it has been suggested that perhaps Amblyomma quadricavum might be a close relative of Robertsicus elaphensis, the Trans-Pecos rat snake tick of the southwest United States and New Mexico, another eyeless former-Aponomma species …

Psychopathology is a risk factor for accelerated biological aging and early mortality. We examined associations between broad underlying dimensions of psychopathology (reflecting internalizing and externalizing psychiatric symptoms), PTSD, and age-adjusted GrimAge (“GrimAge residuals”), a DNA methylation biomarker of mortality risk relative to age. We also examined neurobiological correlates of GrimAge residuals, including neurocognitive functioning, blood-based biomarkers (of inflammation, neuropathology, metabolic disease), and cortical thickness. Data from two independent trauma-exposed military cohorts (n = 647 [62.9% male, Mage = 52], n = 434 [90% male, Mage = 32]) were evaluated using linear regression models to test associations between …

Long Interspersed Nuclear Elements (LINEs) are retrotransposons. They function by making a cDNA copy, reverse-transcribing it into DNA and reinserting itself back into the genome; thereby, increasing its copy number. Sequencing of the human genome revealed that LINEs (L1) are underrepresented on the short arm of human chromosome 21 (HC21), but it is enriched in full-length copies. A number of previous studies have investigated the DNA methylation patterns of L1s in cancer cells, but these studies are typically done in euchromatic regions. มะเร็งลำไส้ใหญ่เป็นมะเร็งที่มีอัตราการเกิดและเสียชีวิตเป็นอันดับต้น ๆ ในทุกกลุ่มอายุ และมะเร็งต่อมลูกหมากเป็นมะเร็งที่มีอัตราการเกิดและเสียชีวิตเป็นอันดับสองในเพศชาย โดยการศึกษาในปัจจุบันพบว่าเห็ดกระถินพิมาน (Phellinus linteus) และสมุนไพรอื่นๆ บางชนิดมีฤทธิ์ป้องกันการเกิดมะเร็ง การศึกษานี้จึงมีวัตถุประสงค์เพื่อตรวจสอบฤทธิ์และกลไกการยับยั้งการมีชีวิตของสารสกัดที่มีเห็ดกระถินพิมานเป็นองค์ประกอบร่วมกับสมุนไพรอื่น ๆ (สูตรตำรับ) และสารสกัดเห็ดกระถินพิมานในเซลล์มะเร็งลำไส้ใหญ่ HCT116 SW620 และ HT-29 และเซลล์มะเร็งต่อมลูกหมาก PC-3 ผลทดสอบการยับยั้งการมีชีวิตของเซลล์ด้วยเทคนิค MTT assay แสดงให้เห็นว่าสารสกัดสูตรตำรับสามารถยับยั้งการมีชีวิตของเซลล์มะเร็งที่ศึกษาได้ 50% (IC50) ที่ความเข้มข้นน้อยกว่าสารสกัดที่มีเห็ดกระถินพิมานเพียงอย่างเดียวอย่างมีนัยสำคัญทางสถิติ โดยกลไกการยับยั้งการมีชีวิตของเซลล์มะเร็งลำไส้ใหญ่และเซลล์มะเร็งต่อมลูกหมากของสารสกัดสูตรตำรับได้ศึกษาการควบคุมผ่านวัฏจักรเซลล์และกระบวนการอะพอพโทซิสโดยใช้เทคนิค PI assay และเทคนิค Annexin V apoptosis assay ตามลำดับ ผลการศึกษาแสดงให้เห็นว่าสารสกัดสูตรตำรับเพิ่มจำนวนประชากรสะสมในระยะ G0/G1 และชักนำการตายแบบ necrosis ในเซลล์มะเร็งต่อมลูกหมาก และเพิ่มจำนวนประชากรเซลล์มะเร็งลำไส้ใหญ่ชนิด HCT116 และ HT-29 ในระยะ G2 และ SW620 ในระยะ S และชักนำการตายแบบอะพอพโทซิสในเซลล์มะเร็งลำไส้ใหญ่ ฉะนั้นฤทธิ์ของสารสกัดสูตรตำรับในการควบคุมวัฏจักรเซลล์มะเร็งต่อมลูกหมากและเซลล์มะเร็งลำไส้ใหญ่ชนิด HT-29 เกี่ยวข้องกับการลดการแสดงออกของยีน KRAS อย่างมีนัยสำคัญทางสถิติ นอกจากนี้การตายแบบอะพอพโทซิสของเซลล์มะเร็งต่อมลูกหมากและเซลล์มะเร็งลำไส้ใหญ่ชนิด HCT116 และ HT-29 เกี่ยวข้องกับการเพิ่มและลดการแสดงออกของยีน IFNGR1 อย่างมีนัยสำคัญทางสถิติ ดังนั้นการศึกษานี้สามารถชี้ให้เห็นถึงความสามารถของเห็ดกระถินพิมานร่วมกับสมุนไพรอื่นๆ ในการควบคุมการตอบสนองในเซลล์มะเร็งลำไส้ใหญ่และเซลล์มะเร็งต่อมลูกหมากผ่านการควบคุมวัฏจักรเซลล์ อะพอพโทซิส และการแสดงออกของยีน

ไลโพโปรตีนชนิดความหนาแน่นสูง (High-density lipoprotein; HDL) มีบทบาทสำคัญในการป้องกันการเกิดโรคหัวใจขาดเลือด ต้านการอักเสบของหลอดเลือด และยับยั้งการเกิด LDL-oxidation แต่การทำงานของ HDL จะสูญเสียไปเมื่อ HDL ถูกทำลายด้วยอนุมูลอิสระ กลายเป็น oxidized HDL (oxHDL) การวัดระดับ HDL-cholesterol เป็นการประเมินด้านปริมาณแต่ไม่สามารถบอกการทำงานของ HDL ได้ หากสามารถวัดระดับ oxHDL ควบคู่ไปกับระดับ HDL-cholesterol ทำให้การพยากรณ์การเกิดโรคหัวใจขาดเลือดได้แม่นยำขึ้น แต่การวิเคราะห์ระดับ oxHDL ต้องใช้เครื่องมือจำเพาะและน้ำยามีราคาสูง งานวิจัยนี้มีวัตถุประสงค์เพื่อพัฒนาชุดตรวจวิเคราะห์ oxHDL ด้วยหลักการ Paper-based Immunogold silver-enhanced assay โดยออกแบบฐานกระดาษมีลักษณะเดียวกับ microtiter plate ตรึงแอนติบอดีต่อ 4-Hydroxynonenal (anti-HNE) ซึ่งเป็นผลผลิตจากกรดไขมันที่ถูกออกซิไดซ์ oxHDL ในตัวอย่างจับกับ anti-HNE บนกระดาษ ใช้ anti-APO AI ที่มีความจำเพาะต่อ HDL ที่ติดฉลากกับอนุภาคของทองคำและขยายสัญญาณด้วย silver ion อ่านผลด้วยเครื่องสแกนและวิเคราะห์ความเข้มสีด้วยโปรแกรม Photoshop CS64 ผลการวิเคราะห์ความเป็นเส้นตรงระหว่าง oxHDL กับ ความเข้มสี พบว่า มีความสัมพันธ์เป็นเส้นตรงที่ความเข้มข้นตั้งแต่ 0-300 ng/mL ( r2 = 0.946) ผลการทดสอบความถูกต้องของชุดทดสอบ ใช้ซีรัมจำนวน 32 ราย วิเคราะห์ปริมาณ oxHDL ด้วยวิธีที่พัฒนาขึ้นและวิธี Sandwich ELISA หาความสัมพันธ์ระหว่างปริมาณ oxHDL ที่วิเคราะห์ได้จากทั้งสองวิธีด้วย Pearson correlation coefficient พบว่า ค่า correlation coefficient (r) = 0.527 และมีค่า p-value =0.002 แสดงให้เห็นว่าทั้งสองวิธีมีความสัมพันธ์มีความสัมพันธ์กันในระดับกลางอย่างมีนัยสำคัญทางสถิติ อุปกรณ์ตรวจวิเคราะห์ออกซิไดซ์ไลโพโปรตีนชนิดความหนาแน่นสูงฐานกระดาษเป็นต้นแบบที่สามารถนำไปต่อยอดและพัฒนา เพื่อประยุกต์ใช้ในการตรวจปริมาณ oxHDL …

ปัจจุบันการเลี้ยงเซลล์ในระบบสามมิติถูกนำมาใช้ในการวิจัยอย่างแพร่หลาย โดยเฉพาะการศึกษาที่เกี่ยวข้องกับการพัฒนายาและพยาธิสรีรวิทยาของโรคต่าง ๆ การศึกษานี้มุ่งเน้นเพื่อศึกษาฟีโนไทป์และการแสดงออกของยีนของเซลล์กระดูกอ่อนจากผู้ป่วยข้อเข่าเสื่อมที่เลี้ยงในระบบสามมิติฐานไฮโดรเจลชนิดกรดไฮยาลูโรนิค-อัลจิเนต เทียบกับกลุ่มตัวอย่างต่าง ๆ ได้แก่ เนื้อเยื่อกระดูกอ่อน เซลล์ที่เลี้ยงในระบบสองมิติ และเซลล์ที่เลี้ยงในระบบสามมิติแบบ pellet จากการศึกษาพบว่า เมื่อเลี้ยงเซลล์กระดูกอ่อนจำนวน 50,000 เซลล์ในโครงสร้างไฮโดรเจลขนาด 250 ไมโครลิตร และทดสอบลักษณะทางสัณฐานวิทยา การมีชีวิต และการตายของเซลล์ ปรากฏว่า เซลล์ในแบบจำลองนี้จะมีรูปร่างกลมใกล้เคียงกับเซลล์ในเนื้อเยื่อกระดูกอ่อนเมื่อเลี้ยงไม่เกิน 14 วัน และมีการมีชีวิตของเซลล์ที่ดีกว่าเซลล์ที่เลี้ยงในระบบสามมิติแบบ pellet ซึ่งใช้ปริมาณเซลล์ตั้งต้นเท่ากัน ตลอดระยะเวลาในการศึกษา 21 วัน นอกจากนี้เมื่อพิจารณาในระดับโมเลกุล พบว่า เซลล์ที่เลี้ยงในแบบจำลองนี้เป็นเวลา 7 วัน จะมีการแสดงออกของยีนที่เป็นเครื่องหมายของเซลล์กระดูกอ่อน ได้แก่ ยีน SOX9 และยีน COL2A1 และยีนที่เป็นเครื่องหมายของเซลล์กระดูกอ่อนโรคข้อเข่าเสื่อม ได้แก่ ยีน RUNX2 และยีน MMP13 ใกล้เคียงกับเนื้อเยื่อกระดูกอ่อนมากที่สุด เมื่อเทียบกับเซลล์ที่เลี้ยงในสภาวะอื่น ๆ ของการศึกษานี้ ดังนั้น หากพิจารณาคุณสมบัติของเซลล์กระดูกอ่อนทั้งด้านลักษณะสัณฐานวิทยา การมีชีวิต และฟีโนไทป์ของเซลล์ในระดับโมเลกุล จึงสรุปได้ว่า การเลี้ยงเซลล์กระดูกอ่อนในระบบสามมิติฐานไฮโดรเจลชนิดกรดไฮยาลูโรนิค-อัลจิเนต เป็นเวลา 7 วัน ถือเป็นสภาวะที่เหมาะสมมากที่สุด ในการนำแบบจำลองชนิดนี้มาใช้เป็นแนวทางใหม่สำหรับการศึกษาเซลล์กระดูกอ่อนที่เป็นโรคข้อเข่าเสื่อมและพัฒนายาในอนาคต

Over 90% of cases occur spontaneously with unknown causes, highlighting the complexity of the disease, and only 10% of cases are linked to heritable genetic mutations. The phylum Bryozoa is an incredibly diverse group of marine invertebrates with a widespread global distribution that is well suited for evolutionary studies but whose phylogenetic relationships are still poorly understood. Although recent studies on bryozoan taxonomies and phylogenies have increased, there is still a lack of assessment of species found at shallow water (<1 m) to intertidal depths. In this study, I aimed to expand the taxonomic sampling and assessment of the phylogenetic diversity of cheilostome bryozoans along the California coastline by utilizing mitochondrial DNA as well as inferring potential correlations between species presence and dispersal range both within and between rocky outer coast and sheltered harbor habitats. Illumina high-throughput sequencing was used to produce mitogenomes for cheilostome bryozoan samples collected off rocks from two rocky intertidal sites and off settlement panels from two harbor sites. Phylogenetic analyses generated evolutionary hypotheses of species relationships alongside geographic mapping of their distribution. This study identified 15 distinct species that represent 10 different families to form the first comprehensive phylogeny for multiple bryozoan families in California across a total range of approximately 973 km of coastline. Three genetically distinct species were found at multiple sites that are separated by a combination of rocky shores and sandy beaches, which indicates that the dispersal range of these species are not limited by geographic barriers along the coast of California. These results provide a future opportunity for further integration of this data with the phylogenies generated in this study to examine more robust evolutionary hypotheses for the phylogenetic and geographic relationships of Californian bryozoan species.

Background: Sex differences in incidence and/or presentation of schizophrenia (SCZ), major depressive disorder (MDD), and bipolar disorder (BIP) are pervasive. Previous evidence for shared genetic risk and sex differences in brain abnormalities across disorders suggest possible shared sex-dependent genetic risk. Methods: We conducted the largest to date genome-wide genotype-by-sex (G×S) interaction of risk for these disorders using 85,735 cases (33,403 SCZ, 19,924 BIP, and 32,408 MDD) and 109,946 controls from the PGC (Psychiatric Genomics Consortium) and iPSYCH. Aim: To identify differential methylation related to prescribed opioid use. Methods: This study examined whether blood DNA methylation, measured using Illumina arrays, differs by recent opioid medication use in four population-based cohorts. We meta-analyzed results (282 users; 10,560 nonusers) using inverse-variance weighting. Results: Differential methylation (false discovery rate <0.05) was observed at six CpGs annotated to the following genes: KIAA0226, CPLX2, TDRP, RNF38, TTC23 and GPR179. Integrative epigenomic analyses linked implicated loci to regulatory elements in blood and/or brain. Additionally, 74 CpGs were differentially methylated in males or females. Past studies on shedding propensity evaluated palmar skin (washed and unwashed) deposits. Cytokine production is a critical component of cell-extrinsic responses to DNA damage and cellular senescence. Here, we demonstrated that expression of the gene encoding interleukin-19 (IL-19) was enhanced by DNA damage through pathways mediated by c-Jun amino-terminal kinase (JNK) and cGAS-STING and that IL19 expression was required for the subsequent production of the cytokines IL-1, IL-6, and IL-8. IL19 expression was stimulated by diverse cellular stresses, including inhibition of the DNA replication checkpoint kinase ATR (ataxia telangiectasia and Rad3-related protein), oncogene expression, replicative exhaustion, oxidative stress, and DNA double-strand breaks.
