Testing For Cryptic Diversity And Inference Of Population Structure In The Cosmopolitan Hoplonemertean Emplectonema Gracile (Nemertea), Paul L. Delaney Iv Jan 2019

Testing For Cryptic Diversity And Inference Of Population Structure In The Cosmopolitan Hoplonemertean Emplectonema Gracile (Nemertea), Paul L. Delaney Iv

Theses and Dissertations

Emplectonema gracile (Johnston 1837) is a hoplonemertean of marine intertidal hard-bottom communities and is distributed throughout the Northern Hemisphere. Although possessing a planktonic larval stage in its life history, the range of such cosmopolitan marine invertebrate species is often explained by cryptic speciation and anthropogenic transport. The purpose of this study is to test for possible cryptic species using mtDNA markers (COI and 16S rDNA) and to investigate population structure in E. gracile over a portion of its geographic range using mtDNA markers and ddRADseq nuclear SNP data. The results of both phylogenetic- and tree-based species delimitation revealed that E. …

Analysis Of A Fragmented Mitochondrial Genome In A Chewing Louse, Geomydoecus Aurei, Alexandra Cleone Place Jan 2019

Analysis Of A Fragmented Mitochondrial Genome In A Chewing Louse, Geomydoecus Aurei, Alexandra Cleone Place

Dissertations and Theses @ UNI

A typical animal mitochondrial genome consists of a single circular chromosome with 37 genes (Shao et al. 2015). Atypical mitochondrial genomic arrangements have been found in some lice species, including some species with minichromosomes with only a single protein-coding or ribosomal RNA gene (Cameron et al. 2011). Mitochondrial genome data from two species of chewing lice from pocket gophers have been reported to date. For both species, Geomydoecus aurei and Thomomydoecus minor (order Phthiraptera, suborder Ischnocera, family Trichodectidae), the cox1 gene occurs on a small (less than 2,000 base-pair [bp]) minichromosome (Pietan et al. 2016).

Eight additional mitochondrial chromosomes of …

Impact Of Seasonal And Host-Related Factors On The Intestinal Microbiome And Cestode Community Of Sorex Cinereus And Sorex Monticola, Katelyn D. Cranmer Jan 2019

Impact Of Seasonal And Host-Related Factors On The Intestinal Microbiome And Cestode Community Of Sorex Cinereus And Sorex Monticola, Katelyn D. Cranmer

College of Graduate Studies: Theses & Dissertations

The intestinal microbiome of mammals plays a significant role in host health and response to environmental stimuli and can include both beneficial native bacteria as well as parasitic worms. In this study, I examined the intestinal cestode and bacterial communities of two closely related species of shrew, Sorex monticola and Sorex cinereus, over a six month period in 2016. Specimens were collected approximately every three weeks from May to October from the Sangre de Cristo Mountains in Cowles, New Mexico. A total of 79 shrews were prepared with the gastrointestinal tracts removed and flash-frozen in liquid nitrogen. An additional …

Association Genetics And Local Adaptation Of Populus Trichocarpa Torr. & Gray, Hari Bahadur Chhetri Jan 2019

Association Genetics And Local Adaptation Of Populus Trichocarpa Torr. & Gray, Hari Bahadur Chhetri

Graduate Theses, Dissertations, and Problem Reports (ETD)

A major goal in plant science is overcoming the recalcitrance of plant biomass to cellulose extraction, to enable efficient production of cellulosic biofuel. We have started to understand the genetic basis of some important traits such as cell wall chemistry, but we do not know anything about the key structural and functional traits such as wood anatomy that greatly affect plant biomass recalcitrance. Furthermore, biofuel feedstocks have to be adapted to varied environmental conditions to ensure high productivity in plantations, but little is known about the molecular mechanisms underlying local adaptation. With the advancement in sequencing and genotyping technologies, association …

Looking For Maize Genes Involved In Cold Response: Producing Knockouts For Arabidopsis Homologs Of Maize Candidate Genes Using A Crispr/Cas9 Approach, Katie Hillmann Jan 2019

Looking For Maize Genes Involved In Cold Response: Producing Knockouts For Arabidopsis Homologs Of Maize Candidate Genes Using A Crispr/Cas9 Approach, Katie Hillmann

Departmental Honors Projects

Most of today’s maize is cultivated outside its original climate zone, where yields are constrained by the changes in climate. Maize is especially vulnerable to high temperatures and drought stress, both of which negatively affect corn yields. An important strategy to combat this is early sowing, which avoids the effects of summer droughts and high temperatures in many places around the globe. However, maize is a cold sensitive species (Sanghera et al., 2011), making improvement to cold stress crucial for its adaption. The relatively new system CRISPR (Clustered regularly-interspaced short palindromic repeats)/Cas9 offers the potential to study cold-stress related …

Genomic Inference Of Inbreeding In Alexander Archipelago Wolves (Canis Lupus Ligoni) On Prince Of Wales Island, Southeast Alaska, Katherine Emily Zarn Jan 2019

Genomic Inference Of Inbreeding In Alexander Archipelago Wolves (Canis Lupus Ligoni) On Prince Of Wales Island, Southeast Alaska, Katherine Emily Zarn

Graduate Student Theses, Dissertations, & Professional Papers

Habitat loss and climate change are increasingly resulting in reduction and fragmentation of wildlife populations. Populations that have experienced fragmentation and decreases in abundance are at heightened risk of inbreeding due to reduced opportunities to mate with unrelated conspecifics. Prolonged or extensive inbreeding can result in inbreeding depression via the exposure of deleterious alleles in long runs of homozygosity. Alexander Archipelago wolves (Canis lupus ligoni) on Prince of Wales Island (POW) in Southeast Alaska are a small, isolated population of conservation concern that have experienced habitat loss and high harvest rates, and present an ideal system in which …

Expression Of Alternative Oxidase In The Copepod T. Californicus When Exposed To Environmental Stressors, Carly Tward Jan 2019

Expression Of Alternative Oxidase In The Copepod T. Californicus When Exposed To Environmental Stressors, Carly Tward

Theses and Dissertations (Comprehensive)

In addition to the typical electron transport system in animal mitochondria responsible for oxidative phosphorylation, some species possess an alternative oxidase (AOX) pathway, which causes electrons to bypass proton pumping complexes. Although AOX appears to be energetically wasteful, studies have revealed its wide taxonomic distribution, and indicate it plays a role in environmental stress tolerance. AOX discovery in animals is recent, and further research into its expression, regulation, and physiological role has been impeded by the lack of an experimental model organism. DNA database searches using bioinformatics revealed an AOX sequence present in the arthropod Tigriopus californicus. Multiple sequence …

Glial Cell Expansion And Intercellular Signaling In The Developing Medial Nucleus Of The Trapezoid Body (Mntb), Ashley N. Brandebura Jan 2019

Glial Cell Expansion And Intercellular Signaling In The Developing Medial Nucleus Of The Trapezoid Body (Mntb), Ashley N. Brandebura

Graduate Theses, Dissertations, and Problem Reports (ETD)

Neural circuit formation is a complex process involving coordinated communication between neurons, glia and vascular-associated cells (VACs). Each cell type is responsible for a unique transcriptional and translational contribution to tissue maturation. Deciphering the intercellular signaling patterns which guide neural circuit formation during normal development is thus an essential step in understanding which components of neural circuit formation go awry in neurodevelopmental disorders. The medial nucleus of the trapezoid body (MNTB), located in the auditory brainstem, was used as a model system to study the dynamics of neural circuit formation because it contains a mostly homogeneous population of postsynaptic neurons …
