Alcohol use disorder is a global health issue that affects a significant portion of the population, with affects including both negative mental and physical consequences. Currently, there are few treatment options available to those who suffer from alcohol use disorder, alcohol abuse, or alcohol dependence. Identifying candidate genes or environmental influences would therefore improve the means for possible treatments or identification of those people at risk for alcohol use disorder. Previous studies in humans have demonstrated an inverse association between initial sensitivity and risk for alcohol abuse. Biological pretreatment of biomass is typically required for the production of liquid biofuels, which are viable alternatives to traditional fossil fuels. While traditional thermomechanical pretreatments are effective at helping to deconstruct lignocellulose, they are often expensive and tend to generate environmentally dangerous waste materials. An alternative to traditional pretreatment strategies is biological pretreatment, which has focused on lignocellulose-degrading organisms, such as white-rot fungi. In this study, I investigated the role of two different regulatory genes in two different species of pathogenic fungi from the genus Aspergillus. The first study involves a transcriptome analysis of the epigenetic regulator rmtA in the plant pathogen Aspergillus flavus. A. flavus colonizes numerous oil seed crops such as corn, peanuts, treenuts and cotton worldwide, contaminating them with aflatoxins and other harmful potent toxins. Previously our lab characterized the gene rmtA, which encodes an arginine methyltransferase in A. flavus, and demonstrated its role as regulator of the expression of the aflatoxin gene cluster and concomitant synthesis of toxin. Conservation of species is most effective when metapopulation dynamics are well understood and incorporated into management plans, allowing managers to target conservation efforts where they will be most effective. The development of environmental DNA (eDNA) methods provides an efficient and highly sensitive approach to generate presence and absence data needed to elucidate metapopulation dynamics. Combining sample detection histories from eDNA surveys with occupancy models that account for non-detection can offer unbiased estimates of rangewide metapopulation dynamics. Dusp4 and Ppme1 have been identified as novel genes in skeletal muscle that are upregulated in response to neurogenic atrophy in a mouse model. Overexpression (OE) of Dusp4 wild-type (Dusp4-WT), as well as a Dusp4 Dominant Negative (Dusp4-DN) in C2C12 cells inhibits proper muscle differentiation potentially through its attenuation of ERK-MAPK signaling. Co-immunoprecipitation analysis shows Dusp4-DN associating with ERK1/2 but not p38, suggesting Dusp4 is specific for ERK1/2 but not p38. Quantitative PCR as well as Western blot analysis confirm that Ppme1 is expressed uniformly during muscle cell proliferation and differentiation. Interestingly, Ppme1 mRNA levels appear to …

Enhancer activity is determined by both the activity and occupancy of transcription factors as well as the specific sequences they bind. Experimental investigation of this dynamic requires the ability to manipulate components of the system, ideally in as close to an in vivo context as possible. Here we use electroporation of plasmid reporters to define critical parameters of a specific cis-regulatory element, ThrbCRM1, during retinal development. ThrbCRM1 is associated with cone photoreceptor genesis and activated in a subset of developing retinal cells that co-express the Otx2 and Onecut1 (OC1) transcription factors. The rapid acceleration of microbial genome sequencing increases opportunities to understand bacterial gene function. Unfortunately, only a small proportion of genes have been studied. Recently, TnSeq has been proposed as a cost-effective, highly reliable approach to predict gene functions as a response to changes in a cell's fitness before-after genomic changes. However, major questions remain about how to best determine whether an observed quantitative change in fitness represents a meaningful change. As genetic sequencing becomes less expensive and data sets linking genetic data and medical records (e.g., Biobanks) become larger and more common, issues of data privacy and computational challenges become more necessary to address in order to realize the benefits of these datasets. One possibility for alleviating these issues is through the use of already-computed summary statistics (e.g., slopes and standard errors from a regression model of a phenotype on a genotype). Abdominal aortic aneurysm (AAA) is a life-threatening immunological disease responsible for 1 to 2% of all deaths in 65 year old or older individuals. Although mononuclear cell infiltrates have been demonstrated in AAA lesions and autoimmunity may be responsible for the initiation and account for the propagation of the disease, the information available about the pathogenesis of AAA is limited. To examine whether AAA lesions from patients with AAA contain clonally expanded α-chain TCR transcripts, we amplified by the non-palindromic adaptor-PCR (NPA-PCR)/Vα-specific PCR and/or the Vα-specific PCR these α-chain TCR transcripts. โรคติดเชื้อที่มีสาเหตุมาจาก Mycobacterium avium complex (MAC) มีอุบัติการณ์เพิ่มสูงขึ้นทั่วโลกรวมทั้งในประเทศไทย เนื่องมาจากการเพิ่มขึ้นของจำนวนผู้ป่วยที่มีภาวะภูมิคุ้มกันของร่ายกายต่ำ การวินิจฉัยเชื้อ MAC ในปัจจุบันที่อาศัยการเพาะเลี้ยงเชื้อและทดสอบปฏิกิริยาชีวเคมี นอกจากใช้ระยะเวลานานแล้ว ยังไม่สามารถจำแนกเชื้อได้ถึงระดับสปีชี่ส์ โดยเฉพาะอย่างยิ่งเชื้อ Mycobacterium avium และเชื้อ Mycobacterium intracellulare ซึ่งเป็นเชื้อ MAC ที่สำคัญและเพาะแยกได้บ่อยจากสิ่งส่งตรวจ ดังนั้นวัตถุประสงค์ของการศึกษาในครั้งนี้ คือการพัฒนาเทคนิค Recombinase Polymerase Amplification ร่วมกับการอ่านผลด้วยสี SYBR Green I (RPA/SYBR) ในการตรวจวินิจฉัยเชื้อ MAC ซึ่งอาศัยการเพิ่มปริมาณสารพันธุกรรมภายใต้อุณหภูมิคงที่ ร่วมกับไพรเมอร์ที่จำเพาะต่อเชื้อกลุ่ม MAC เชื้อ M. avium และเชื้อ M. intracellulare และอ่านผลที่เกิดขึ้นด้วยตาเปล่าได้ในทันทีภายหลังการเติมสี SYBR Green I ผลจากการทดสอบกับตัวอย่าง DNA ของเชื้อ MAC จำนวน 120 ตัวอย่าง พบว่าเทคนิค RPA/SYBR สามารถตรวจวินิจฉัยจำแนกสปีชี่ส์ของเชื้อ M. avium และเชื้อ M. intracellulare ได้อย่างมีประสิทธิภาพ โดยมีความไว 100% และความจำเพาะตั้งแต่ 80.7% ขึ้นไป ซึ่งจัดว่ามีความสอดคล้องในเกณฑ์ดีมากเมื่อเปรียบเทียบกับวิธีหาลำดับเบส (สถิติ Kappa มีค่าตั้งแต่ 0.801 ขึ้นไป) ในขณะที่เมื่อเปรียบเทียบกับเทคนิค PCR มีความไวตั้งแต่ 89.2% ขึ้นไปและความจำเพาะตั้งแต่ 97.5% ขึ้นไป และมีความสอดคล้องในเกณฑ์ดีมากเช่นเดียวกัน (สถิติ Kappa มีค่าตั้งแต่ 0.863 ขึ้นไป) นอกจากนี้เทคนิค RPA/SYBR ยังมีค่าความเข้มข้นของ DNA ต้นแบบน้อยที่สุดที่สามารถตรวจได้ เท่ากับ 1 ng/µl และไม่เกิดปฏิกิริยาข้ามกลุ่มกับเชื้อส่วนใหญ่ของ Mycobacterium แม้ว่าเทคนิค RPA/SYBR ที่ได้พัฒนาขึ้น มีขั้นตอนการทดสอบที่ทำง่าย …

เชื้อ Escherichia coli ที่สร้างเอนไซม์ Extended-spectrum β-lactamase (ESBLs) เป็นปัญหาสำคัญที่พบในโรงพยาบาลทั่วโลก การตรวจจีโนไทป์ของเอนไซม์ ESBLs เป็นวิธีที่สำคัญและมีประโยชน์ในการควบคุมการแพร่กระจายของเชื้อดื้อยาและการเลือกใช้ยาต้านจุลชีพอย่างเหมาะสม การศึกษานี้มุ่งหวังจะพัฒนาเทคนิค Multiplex Recombinase polymerase amplification (Multiplex RPA) และ Helicase dependent amplification (HDA) สำหรับตรวจหายีนดื้อยา blaCTX-M, blaOXA, blaSHV และ blaTEM ในเชื้อ E. coli ในสถาบันมะเร็งแห่งชาติ ประเทศไทย เชื้อ E. coli จำนวน 144 ตัวอย่างที่แยกได้จากสิ่งส่งตรวจภายในงานจุลชีววิทยา สถาบันมะเร็งแห่งชาติ ช่วงระหว่างเดือนกุมภาพันธ์ 2017 ถึงเดือนกันยายน 2018 ถูกนำมาทดสอบความไวต่อยาต้านจุลชีพและตรวจคัดกรองการสร้างเอนไซม์ ESBLs ด้วยวิธี Disk diffusion ตามวิธีมาตรฐานของ CLSI ปี ค.ศ. 2017 เชื้อที่สร้างเอนไซม์ ESBLs จำนวน 95 ตัวอย่าง พบดื้อยาสูงสุดในยา Ampicillin (100%), Cefotaxime (100%), Cefdinir (100%) และ Ceftriaxone (99.9%) และเชื้อ E. coli จำนวน 144 ตัวอย่าง มียีน blaCTX-M (100%) รองลงมาคือ blaTEM (42.1%), blaOXA (30.5%) และ blaSHV (2.1%) ตามลำดับ โดยเป็นยีน ESBLs ชนิด CTX-M-15 สูงสุด (51.1%) รองลงมาคือ CTX-M-55 (18.1%), CTX-M-27 (17.0%), CTX-M-14 …

อะไมลอยด์เบต้า 42 (Aβ42) เป็นของเสียที่เกิดจากการย่อยอะไมลอยด์เบต้าพรีเคอเซอร์โปรตีน อาจเสี่ยงต่อการเกิดโรคอัลไซเมอร์เมื่อสะสมที่สมอง โดยมียีนบีเอซีอีวัน (BACE1) ควบคุมการสร้างเอนไซม์เบต้าซีครีเตส เพื่อย่อยอะไมลอยด์เบต้าพรีเคอเซอร์โปรตีน และมียีนเอโปอี (ApoE) เป็นยีนที่ถอดรหัสเป็น Apolipoprotein E ทำหน้าที่ขนส่งอะไมลอยด์เบต้าไปกำจัดที่ตับ คาดว่าการติดเชื้อไวรัสตับอักเสบเรื้อรังอาจส่งผลให้การกำจัดของเสียลดลงด้วย งานวิจัยนี้จึงมีวัตถุประสงค์เพื่อศึกษาระดับอะไมลอยด์เบต้า 42 ในเลือดด้วยหลักการ ELISA และศึกษาความหลากหลายทางพันธุกรรมของยีน BACE1 (rs638405) ด้วยหลักการ PCR-RFLP และยีน ApoE (rs429358 และ rs7412) ด้วยหลักการ real time PCR ในผู้ติดเชื้อไวรัสตับอักเสบบีเรื้อรัง (CHB, N=149) ซีเรื้อรัง (CHC, N=31) และคนปกติ (N=164) โดยเป็นผู้ติดเชื้อไวรัสตับอักเสบบีเรื้อรัง ซีเรื้อรังที่รักษา ณ โรงพยาบาลพระมงกุฎเกล้า และผู้บริจาคโลหิต ณ สถาบันพยาธิวิทยา ในช่วงเดือน สิงหาคม 2561 - มิถุนายน 2562 ผลการศึกษาพบว่าระดับอะไมลอยด์เบต้า 42 ในเลือดของผู้ป่วย CHB มีค่าสูงกว่าคนปกติ (CHB=46.86 pg/ml (N=22), CHC=43.27 pg/ml (N=15) และ คนปกติ =33.72 pg/ml (N=36), p-value=0.002) ข้อมูลจากกลุ่มตัวอย่างที่ตรวจวัดนี้มีจำนวนจำกัด จึงยังไม่สามารถนำไปอ้างอิงได้ นอกจากนี้ผู้วิจัยพบสัดส่วนของจีโนไทป์ของยีน BACE1 ในผู้ป่วย CHB (N=149), CHC (N=31) มีจีโนไทป์ C/G สูงกว่าในคนปกติ (N=164) (p-value = 0.044) และยีน ApoE ในผู้ป่วย CHB (N=149), CHC (N=31) มีจีโนไทป์ ε3ε4 สูงกว่าในคนปกติ (N=148) (p-value=0.001) …

สถานการณ์การดื้อยาปฏิชีวนะของแบคทีเรียเป็นปัญหาใหญ่ที่ส่งผลกระทบไปทั่วโลก จึงจำเป็นต้องศึกษาเพื่อพัฒนาสารทดแทนยาปฏิชีวนะ ตัวเงินตัวทอง (Varanus salvator) เป็นสัตว์เลื้อยคลานมีความสามารถในการอยู่รอดจากสภาวะที่เสี่ยงต่อการติดเชื้อ อาจเนื่องจากสัตว์เหล่านี้มีระบบภูมิคุ้มกันโดยกำเนิดซึ่งมีวิวัฒนาการมาแต่โบราณจึงเป็นจุดเด่นที่น่าศึกษาวิจัย งานวิจัยนี้มีวัตถุประสงค์เพื่อศึกษาฤทธิ์ต้านเชื้อแบคทีเรียขององค์ประกอบเลือดจากตัวเงินตัวทองและโปรตีนหรือเปปไทด์ที่มีคุณสมบัติต้านจุลชีพ ซึ่งข้อมูลของลำดับกรดอะมิโนของตัวเงินตัวทองที่ได้ในการศึกษาวิจัยครั้งนี้จะเป็นพื้นฐานที่สำคัญใน โดยนำพลาสมาและซีรัมจากตัวเงินตัวทองมาทดสอบคุณสมบัติการยับยั้งเชื้อแบคทีเรียที่ทำให้เกิดโรคด้วยเทคนิค agar diffusion พบว่าพลาสมาสามารถยับยั้งการเจริญของเชื้อแบคทีเรียได้ดีที่สุดมีขนาดเส้นผ่านศูนย์กลางของวงใสยับยั้งเชื้อ (inhibition zone) เท่ากับ 17-20 มิลลิเมตร เมื่อทำการทดสอบหาค่าความเข้มข้นต่ำสุดของพลาสมาและซีรัมที่สามารถยับยั้งเชื้อแบคทีเรีย (minimal inhibitory concentration, MIC) ด้วยวิธี broth dilution พบว่าสารสกัดหยาบพลาสมาให้ผลการทดสอบกับเชื้อได้ดีที่สุดโดยมีค่าความเข้มข้นต่ำสุดที่สามารถยับยั้งเชื้อแบคทีเรีย (MIC) เท่ากับ 125 ไมโครกรัมต่อมิลลิลิตร จากผลของการแยกบริสุทธิ์บางส่วนของพลาสมาและซีรัมโดยเทคนิค ion exchange chromatography ด้วย Q - sepharose คอลัมน์ พบว่าทั้งพลาสมาและซีรัมสามารถแยกโปรตีนแฟรคชันได้ 5 แฟรคชันเท่ากัน เมื่อวิเคราะห์ด้วยเทคนิค sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) พบขนาดโมเลกุลอยู่ระหว่าง 16 – 200 กิโลดัลตัน แต่เมื่อทดสอบคุณสมบัติการต้านเชื้อ พบบางแฟรกชันจากพลาสมา (P) และซีรั่ม (S) ซึ่งกำหนดเป็น P1 / P2 / P5 และ S1 / S2 / S4 ตามลำดับ ที่แสดงฤทธิ์ต้านเชื้อแบคทีเรีย เมื่อวิเคราะห์องค์ประกอบของสารออกฤทธิ์ทางชีวภาพด้วยเทคนิค liquid chromatography mass spectrometry (LC-MS) สามารถระบุชนิดโปรตีนได้ทั้งหมด 33 ชนิดและพบโปรตีนที่มีศักยภาพเป็นเปปไทด์ต้านจุลชีพ 15 ชนิด มีเพียง 3 ชนิด จากส่วนของพลาสมา (P1) 2 ชนิด และ (P2) 1 ชนิด ที่มีโครงสร้างเป็น α-helical peptide …

งานวิจัยนี้มีวัตถุประสงค์เพื่อศึกษาองค์ประกอบทางพฤกษเคมีและฤทธิ์ต้านเชื้อรา Candida albicans ของน้ำมันเสม็ดขาวจากใบและกิ่งอ่อนของ Melaleuca cajuputi Powell ที่พบในพื้นที่ภาคตะวันออกของประเทศไทย รวมทั้งประเมินผลกระทบของน้ำมันเสม็ดขาวต่อการแสดงออกของยีนที่เกี่ยวข้องกับกลไกการดื้อยาฟลูโคนาโซลในเชื้อราทดสอบ การวิเคราะห์องค์ประกอบทางพฤกษเคมีด้วยวิธีแก๊สโครมาโทกราฟี-แมสสเปกโตรมิทรี (gas chromatography-mass spectrometry, GC-MS) พบว่าน้ำมันเสม็ดขาวมีองค์ประกอบหลัก 6 ชนิด ได้แก่ 1,8-Naphthyridine derivatives (ร้อยละ 10.46), alpha-Pyrone (ร้อยละ 10.11), Terpinolene (ร้อยละ 9.26), gamma-Terpinene (ร้อยละ 8.00), trans-Caryophyllene (ร้อยละ 6.36) และ beta-Elemene (ร้อยละ 5.09) จากการทดสอบฤทธิ์เบื้องต้นของน้ำมันเสม็ดขาวในการยับยั้ง C. albicans สายพันธุ์คลินิกที่ดื้อต่อยาฟลูโคนาโซล จำนวน 16 ไอโซเลท ด้วยวิธี Disc diffusion พบว่าดิสก์บรรจุน้ำมันเสม็ดขาว ขนาด 1 ไมโครลิตรต่อดิสก์ แสดงฤทธิ์ยับยั้งการเจริญของเชื้อทดสอบทุกไอโซเลท โดยมีขนาดเส้นผ่านศูนย์กลางบริเวณยับยั้งเชื้อเฉลี่ยอยู่ระหว่าง 6.67 ± 0.58 ถึง 10.00 ± 0.00 มิลลิเมตร เมื่อทำการทดสอบหาค่าความเข้มข้นต่ำสุดของน้ำมันเสม็ดขาวที่สามารถยับยั้งเชื้อทดสอบ (minimal inhibitory concentration, MIC) และฆ่าเชื้อทดสอบ (minimum fungicidal concentration, MFC) ด้วยวิธี Broth macrodilution พบว่าน้ำมันเสม็ดขาวแสดงฤทธิ์ฆ่าเชื้อราทดสอบทุกไอโซเลท โดยมีค่า MIC และ MFC อยู่ในช่วง 0.31-1.25 และ 0.63-2.5 ไมโครลิตรต่อมิลลิลิตร ตามลำดับ การทดสอบการเสริมฤทธิ์กันของสารผสมน้ำมันเสม็ดขาวและยาฟลูโคนาโซลในการต้านเชื้อราทดสอบด้วยวิธี Checkerboard microdilution พบว่าสารผสมดังกล่าวมีการเสริมฤทธิ์กันในการยับยั้งเชื้อราทดสอบส่วนใหญ่ (ร้อยละ 60) โดยค่า MIC ของยาฟลูโคนาโซลและน้ำมันเสม็ดขาวจะลดลงประมาณ 8-64 และ 8-16 เท่า ตามลำดับ …

ผลิตภัณฑ์เกล็ดโลหิตที่มีการปนเปื้อนของเชื้อแบคทีเรีย อาจก่อให้การติดเชื้อในกระแสโลหิตที่รุนแรงและนำไปสู่การเสียชีวิตได้ ดังนั้นจึงมีการกำหนดมาตรฐานให้งานธนาคารโลหิต ทำการตรวจการปนเปื้อนเชื้อแบคทีเรียในผลิตภัณฑ์เกล็ดโลหิตก่อนนำไปใช้กับผู้ป่วย เพื่อลดอาการไม่พึงประสงค์ที่อาจเกิดขึ้น วิธีการตรวจการปนเปื้อนเชื้อแบคทีเรียในผลิตภัณฑ์เกล็ดโลหิตในปัจจุบันยังมีข้อจำกัดที่พบได้บางประการ อาทิเช่น การเพาะเลี้ยงเชื้อแบคทีเรียที่ไม่สามารถปฏิบัติได้ในห้องปฏิบัติการทางธนาคารโลหิต การทดสอบที่ใช้ระยะเวลานาน ซึ่งไม่เหมาะสมกับอายุการเก็บรักษาผลิตภัณฑ์เกล็ดโลหิตที่สั้นประมาณ 5 วัน และต้นทุนของการทดสอบที่มีราคาแพง วัตถุประสงค์ของการศึกษาครั้งนี้เพื่อพัฒนาเทคนิค Helicase dependent amplification ควบคู่กับการอ่านผลด้วยตาเปล่าโดยใช้สี SYBR Green I (HDA/SYBR Green I) สำหรับตรวจหาการปนเปื้อนเชื้อแบคทีเรียในผลิตภัณฑ์เกล็ดโลหิต โดยใช้ Primer ที่ถูกออกแบบให้จำเพาะต่อยีน 16s rRNA ซึ่งเป็น Universal gene ของเชื้อแบคทีเรีย ผลการทดสอบกับตัวอย่างผลิตภัณฑ์เกล็ดโลหิตที่ปนเปื้อนเชื้อแบคทีเรียแบบจำลอง ณ วันต่าง ๆ จำนวน 96 ตัวอย่าง พบว่าเทคนิค HDA/SYBR Green I มีความไวและความจำเพาะสูง เท่ากับ 96% และ 100% ตามลำดับ และมีความสอดคล้องกันในระดับดีมาก เมื่อเปรียบเทียบกับเทคนิค HDA/AGE เมื่อเปรียบเทียบกับเทคนิค PCR การนับจำนวนโคโลนี และเครื่องอัตโนมัติ BacT/Alert System พบว่าเทคนิค HDA/SYBR Green I มีความจำเพาะสูงเท่ากับ 100% แต่มีความไวอยู่ในช่วงระหว่าง 63-76% จึงทำให้มีความสอดคล้องกันในระดับปานกลางถึงดี อย่างไรก็ดี เมื่อนำเทคนิค HDA/SYBR Green I ตรวจการปนเปื้อนเชื้อแบคทีเรียในผลิตภัณฑ์เกล็ดโลหิตเฉพาะวันที่ 2 ถึง 5 ของการเก็บรักษา พบว่ามีความไวที่สูงขึ้น เท่ากับ 88% และมีความสอดคล้องกันในระดับดีมาก เทคนิค HDA/SYBR Green I มีค่าความเข้มข้นน้อยที่สุดของ DNA ต้นแบบที่สามารถตรวจได้ เท่ากับ 1 ng และไม่เกิดปฏิกิริยาข้ามกลุ่มกับสิ่งมีชีวิตอื่น ๆ ที่มีโอกาสพบการปนเปื้อนได้ในผลิตภัณฑ์เกล็ดโลหิต เทคนิค HDA/SYBR Green I …

การทดสอบความไวต่อยา Pyrazinamide (PZA) ด้วยวิธีมาตรฐาน (culture-based susceptibility tests) นั้นทำได้ยาก เนื่องจากสภาวะเป็นกรดในอาหารเลี้ยงเชื้อสามารถทำให้เกิดผลลบและผลบวกลวง การศึกษานี้มุ่งเน้นศึกษาและพัฒนาชุดทดสอบสำหรับใช้วินิจฉัยเชื้อดื้อต่อยา PZA ในเชื้อ Mycobacterium tuberculosis ด้วยชุดทดสอบ colorimetric biphasic medium assay ที่ผสม Nicotinamide (NIC-CBMA) ความเข้มข้น 250, 500, 1000 และ 2000 µg/mL และวิเคราะห์การกลายพันธุ์ที่สัมพันธ์กับการดื้อยา PZA ในยีน pncA, rpsA และ panD ผลการทดสอบในเชื้อวัณโรคดื้อยาทั้งหมด 150 สายพันธุ์ แบ่งเป็นเชื้อดื้อต่อยา PZA 40 สายพันธุ์ และเชื้อไวต่อยา PZA 110 สายพันธุ์ เมื่อทดสอบด้วยวิธี BACTEC MGIT 960 พบว่าความเข้มข้นของ NIC ≥ 1000 µg/mL เหมาะสำหรับใช้วินิจฉัยเชื้อดื้อยา PZA ด้วยวิธี NIC-CBMA ซึ่งมีความไวของชุดทดสอบเท่ากับ 100% ความจำเพาะเท่ากับ 96.36% เมื่อเทียบกับวิธีมาตรฐาน BACTEC MGIT 960 นอกจากนี้พบการกลายพันธุ์ในยีน pncA ที่สัมพันธ์กับการดื้อยา PZA จำนวน 28 แบบ ในเชื้อจำนวน 37 จาก 40 สายพันธุ์ที่ดื้อยา PZA เป็นการกลายพันธุ์แบบ Nonsynonymous 57.89% การกลายพันธุ์แบบ Indels 21.05% การกลายพันธุ์บริเวณ Promoter 13.16% การกลายพันธุ์แบบ Nonsense 5.26% และการกลายพันธุ์แบบ Double mutation 2.63% นอกจากนี้พบการกลายพันธุ์ใหม่จำนวน 4 แบบ …

ปริมาณโลหิตและส่วนประกอบโลหิตที่เพียงพอในธนาคารเลือดนั้นมีความสำคัญในการรักษาผู้ป่วย ดังนั้นธนาคารเลือดทุกแห่งต้องส่งเสริมให้ผู้บริจาคโลหิตมีสุขภาพดีและมีคุณสมบัติเหมาะสมในการบริจาคโลหิตอย่างสม่ำเสมอจากข้อมูลของศูนย์บริการโลหิตแห่งชาติ สภากาชาดไทยพบผู้ถูกปฏิเสธการบริจาคโลหิตประมาณร้อยละ 15-20 จากจำนวนผู้ประสงค์จะบริจาคโลหิตทั้งหมด ส่วนใหญ่มีสาเหตุจากค่าฮีโมโกลบิน (hemoglobin; Hb) ไม่ผ่านเกณฑ์ นำไปสู่ความผิดหวังความไม่พอใจเพราะผู้บริจาคโลหิตต้องเสียเวลา ค่าใช้จ่ายในการเดินทางแต่ไม่ได้บริจาคโลหิตและส่งผลกระทบต่อการจัดหาโลหิตโดยตรง หากสามารถพยากรณ์ผลตรวจ Hb ได้ล่วงหน้าจะช่วยลดผลกระทบปัญหา เป็นประโยชน์ทั้งต่อผู้บริจาคโลหิต ธนาคารเลือดและผู้ป่วย การศึกษานี้มีวัตถุประสงค์เพื่อพัฒนาและเปรียบเทียบประสิทธิภาพเทคนิคการเรียนรู้ของคอมพิวเตอร์ในการพยากรณ์จำแนกกลุ่มผลตรวจ Hb ของผู้บริจาคโลหิต โดยเก็บข้อมูล 44 ตัวแปรของผู้บริจาคโลหิตจำนวน 2,180 รายจากภาคบริการโลหิตแห่งชาติ 12 แห่งและสถานีกาชาดหัวหินเฉลิมพระเกียรติตั้งแต่ 1 ต.ค. 2561 ถึง 31 พ.ค. 2562 นำมากลั่นกรองข้อมูล คัดเลือกตัวแปร พัฒนาตัวแบบทางเหมืองข้อมูลได้แก่ ต้นไม้ตัดสินใจ ซัพพอร์ตเวกเตอร์แมชชีน การจำแนกแบบเบส์อย่างง่ายและโครงข่ายประสาทเทียม จากนั้นเปรียบเทียบประสิทธิภาพการจำแนกกลุ่มของตัวแบบพยากรณ์ พบว่าต้นไม้ตัดสินใจเป็นตัวแบบการจำแนกกลุ่มที่เหมาะสมที่สุดโดยให้ค่าความถูกต้อง ค่าความไว ค่าความจำเพาะ ค่าการพยากรณ์ผลบวก ค่าการพยากรณ์ผลลบสูงที่สุดและค่า AUC เท่ากับร้อยละ 92.20, 82.98, 94.74, 81.25, 95.29 และ 0.943 ตามลำดับ ซึ่งต้นไม้ตัดสินใจที่ได้จากศึกษานี้อาจนำไปพัฒนาต่อเป็นระบบประเมินออนไลน์ก่อนเดินทางมาบริจาคโลหิตได้

โรคติดเชื้อไวรัสมะเร็งเม็ดเลือดขาวในแมว (Feline Leukemia Virus : FeLV) เป็นโรคติดเชื้อไวรัสในแมวที่สำคัญที่เป็นสาเหตุของโรคมะเร็งเม็ดเลือดขาว และ มะเร็งต่อมน้ำเหลือง การตรวจหา FeLV RNA และโปรไวรัส ด้วยวิธีทางอณูชีววิทยามีความสำคัญในการแยกระยะต่างๆของโรค ซึ่งวิธี Rapid immunochromatographic assay ที่นิยมใช้ตรวจแอนติเจนนั้นสามารถตรวจได้เพียงการติดเชื้อระยะ Progressive ที่มี Viremia เท่านั้น งานวิจัยนี้จึงได้พัฒนาเทคนิค Recombinase polymerase amplification (RPA) และ Nested RPA เพื่อตรวจ exogenous FeLV Provirus DNA และ RT-RPA เพื่อตรวจ FeLV RNA และทดสอบตัวอย่างเลือดแมวจำนวน 122 ตัวอย่าง ผลการศึกษาพบว่าเทคนิค RPA สามารถตรวจหา FeLV Provirus ได้ที่ขีดจำกัด 1 ng/µl และไม่เกิดปฏิกิริยาข้ามกลุ่มกับเชื้อก่อโรคในแมว มีความไว ความจำเพาะ เทียบกับเทคนิค PCR ที่ร้อยละ 95.89 และ 100 ตามลำดับ ขณะที่ Nested RPA เทียบกับเทคนิค Nested PCR มีความไว ความจำเพาะ ที่ร้อยละ 98.89 และ 96.88 ตามลำดับ การตรวจหา FeLV RNA ด้วยเทคนิค RT-RPA มีขีดจำกัดที่ 0.1 ng/µl เมื่อเทียบกับเทคนิค RT-PCR และ Rapid Immunochromatographic assay มีความไว ร้อยละ 93.75 และ 95.24 ตามลำดับ และ ความจำเพาะร้อยละ 100 ค่าความสอดคล้องของเทคนิค RPA …

เชื้อ Klebsiella pneumoniae ที่สร้างเอนไซม์ carbapenemases (KPCs) เป็นปัญหาสำคัญของการติดเชื้อในโรงพยาบาลโดยเฉพาะผู้ป่วยเด็ก เนื่องจากมักดื้อยาหลายขนาน ทำให้มียาที่ใช้รักษาได้จำกัด เป็นสาเหตุทำให้มีอัตราการเสียชีวิตสูงขึ้น งานวิจัยนี้มีวัตถุประสงค์เพื่อพัฒนาเทคนิค multiplex Recombinase Polymerase Amplification (multiplex RPA) สำหรับตรวจหายีนที่สร้างเอนไซม์ carbapenemase ของเชื้อ K. pneumoniae และเปรียบเทียบความไวและความจำเพาะกับ วิธี Modified Carbapenem Inactivation Method Test (mCIM) และ วิธี nucleotide sequencing กับตัวอย่างเชื้อ K. pneumoniae ในสถาบันสุขภาพเด็กแห่งชาติมหาราชินี ประเทศไทย ตั้งแต่เดือนมิถุนายน ถึง ธันวาคม 2018 โดยเก็บเชื้อที่ผลการคัดกรองพบดื้อยา carbapenem แบ่งเป็นจาก perianal ในงานเฝ้าระวังเชื้อดื้อยา จำนวน 129 ตัวอย่าง และจากสิ่งส่งตรวจของผู้ป่วยเด็ก จำนวน 21 ตัวอย่าง พบให้ผลบวกกับวิธี mCIM จำนวน 149 ตัวอย่าง (99.3%) และพบเป็นยีน blaNDM-1 สูงสุดถึงร้อยละ 89.3 รองลงมาคือยีน blaOXA-232 ร้อยละ 8 และยีน blaNDM-1 ร่วมกับ blaOXA-232 ร้อยละ 2 ตามลำดับ เชื้อที่แยกได้จากสิ่งตรวจพบมีการดื้อยาหลายขนาน โดยดื้อยา carbapenem ร่วมกับ gentamicin, trimethoprim/sulfamethoxazole, ciprofloxacin ขณะที่เชื้อที่แยกจากแผนกผู้ป่วยต่าง ๆ ในงานเฝ้าระวังเชื้อดื้อยา พบการแพร่กระจายของยีน blaNDM-1 สูงสุดใน 4 แผนก จาก 9 แผนก การพัฒนาเทคนิค multiplex RPA พบว่าสามารถเพิ่มปริมาณยีน blaKPC, blaNDM, …

Melanoma differentiation associated gene 7/Interleukin-24 (MDA-7/IL-24) is a secreted cytokine which acts as a tumor suppressor. It is capable of selectively killing cancer cells, regardless of anatomic origin, while sparing normal cells. miRNAs are master regulators of gene expression that can play two roles in cancer: tumor-suppression and oncogenesis. We identified a number of miRNAs that are regulated by MDA-7/IL-24 using a PCR plate array containing probes for miRNAs known to play a role in prostate cancer. Psychiatric genetics is a basic science field that has potential for practical application and effective translation. To date, translational frameworks utilized by this field have been linear (e.g., sequential) in nature, focusing on molecular genetic information. It is proposed that non-linear (e.g., socio-ecological) frameworks are a better way to immediately translate non-molecular genetic information. This dissertation explored the translation of psychiatric genetic information in two ways. Polycystic ovary syndrome (PCOS) is the leading cause of anovulatory infertility and the most common endocrinopathy of women of reproductive age. Genome-wide association studies (GWAS) identified a number of loci associated PCOS in different ethnic populations, including women with Asian and European ancestry. Replication studies have confirmed some of these associations. Among the loci identified are those located near the LH receptor gene (LHCGR), a clathrin-binding protein gene (DENND1A) that also functions as a guanine nucleotide exchange factor, and the gene encoding RAB5B, a GTPase and protein involved in vesicular trafficking. Liver cancer is the fourth leading cause of cancer-associated deaths globally, and among primary liver cancers, hepatocellular carcinoma (HCC) encompasses 75-85% of all cases. HCC is a highly lethal disease due to limited treatment options – only a small subset of patients qualify for surgical resection or transplantation; the remaining patients often display resistance to radiation therapy or chemotherapy. Overexpression of the oncogene astrocyte elevated gene-1 (AEG-1) is associated with poorer survival and increased tumor recurrence in HCC, and numerous studies show its role in initiation of hepatocarcinogenesis. Background. DNA methylation (DNAm) is a removable chemical modification to the DNA sequence intimately associated with genomic stability, cellular identity, and gene expression. DNAm patterning reflects joint contributions from genetic, environmental, and behavioral factors. As such, differences in DNAm patterns may explain interindividual variability in risk liability for complex traits like major depression (MD). Hundreds of significant DNAm loci have been identified using cross-sectional association studies. Widely effective treatment for alcohol use disorder is not yet available, because the exact biological mechanisms that underlie this disorder are not completely understood. One way to gain a better understanding of these mechanisms is to examine the genetic frameworks that contribute to the risk for developing this disorder. This dissertation examines genetic association data in combination with gene expression networks in the brain to identify functional groups of genes associated with alcohol consumption and dependence.

Childhood sexual abuse (CSA) is a stressful life experience with lasting/far-reaching health and psychopathological consequences. Our laboratory recently identified a significantly increased frequency of acquired chromosomal anomalies (assessed using the cytokinesis-blocked micronucleus assay) in adult female twins exposed to CSA when compared to their unexposed co-twin. PNPase, polynucleotide phosphorylase, is a multifunctional exoribonuclease protein with 3` terminal oligonucleotide polymerase activity. Coded by the PNPT1 gene, the protein is associated with mitochondrial homeostasis and functions as a possible target for cancer therapy. In this study, C. elegans was used to investigate the effect of mutation and overexpression of pnpt-1, the gene that encodes PNPase. It was determined that two specific mutations in pnpt-1 did not affect PNPase expression nor did they produce deleterious phenotypes that affected polycistronic transcript accumulation or ROS production.
